A cascade cyclization/nucleophilic aromatic substitution (SNAr) reaction of α,β-unsaturated N-tosylhydrazones with N-heteroaryl chlorides was developed for the synthesis of N-heteroaryl pyrazole derivatives. This one-pot reaction provided bi(heteroaryl) derivatives in good to excellent yields and with excellent regioselectivity. The procedure is operationally simple and applicable to large-scale synthesis.  相似文献

Ketoanilides containing alkyl side chains were readily cyclized to 3-hydroxy-2-oxindoles or spirooxindoles by a single or double intramolecular Friedel-Crafts reaction in the presence of trifluoroacetic acid (TFA) at room temperature or at 45 °C. α-Iminocarboxamides, generated in situ from ketoamides, cyclized similarly to 3-aminooxindoles under identical conditions.  相似文献

The iron-catalyzed hydrocarboxylation of aryl alkenes has been developed using a highly active bench-stable iron(II) precatalyst to give α-aryl carboxylic acids in excellent yields and with near-perfect regioselectivity. Using just 1 mol % FeCl(2), bis(imino)pyridine 6 (1 mol %), CO(2) (atmospheric pressure), and a hydride source (EtMgBr, 1.2 equiv), a range of sterically and electronically differentiated aryl alkenes were transformed to the corresponding α-aryl carboxylic acids (up to 96% isolated yield). The catalyst was found to be equally active with a loading of 0.1 mol %. Preliminary mechanistic investigations show that an iron-catalyzed hydrometalation is followed by transmetalation and reaction with the electrophile (CO(2)).  相似文献

Propylphosphonic anhydride (T3P®) was successfully applied to the synthesis of an isoindolinone library by the utilization of an Ugi four-center, three-component reaction (Ugi-4C-3CR). The use of T3P® significantly shortened the required reaction time and the corresponding products were obtained in good to high yields. Moreover, a side-reaction was observed when phenylethylamine derivatives and tryptamine were used as the amine component. The latter reaction was applied to the microwave-assisted, one-pot synthesis of the isoquinoline alkaloid (±)-nuevamine. Surprisingly, the traditional Ugi four-component reaction (Ugi-4CR) was unsuccessful in the presence of T3P®. In this case an α-amino amide was produced excluding the carboxylic acid from the multicomponent reaction.  相似文献

Novel and convenient methodology for the construction of N-substituted amide derivatives have been developed from nitriles and alcohols using propylphosphonic anhydride (T3P®). This methodology is an alternate approach to the synthesis of amides via Ritter reaction, which is one of the classical methods for the synthesis of N-substituted amides from nitriles and alcohols. In this approach, first T3P® activates the alcohol which is then attacked by nitrile to form N-substituted amides. This methodology can also apply for the synthesis of benzhydryl ether. This developed protocol is one of the novel applications of T3P®.  相似文献

An efficient, rapid and green synthesis of α,α′-bis(substituted-benzylidene)cycloalkanones and their xanthene derivatives is reported under solvent-free conditions using nano titania-supported sulfonic acid (n-TSA) as a reusable catalyst. This method offers many advantages, such as environmental friendliness reaction conditions, simplicity, short reaction times, easy work-up, reusability of catalyst, and high yields of products. CeCl3·7H2O supported on silica(CeCl3·7H2O-SiO2) is used as a heterogeneous,efficient and recyclable catalyst for a three component one-pot reaction of an amine,aldehydes and diethyl phosphite to synthesizeα-diaminophosphonate derivatives under microwave irradiation exploiting neat reaction conditions.Tenα-diaminophosphonates(6a-j) of 4,4’-sulfonyldianiline(Dapsone)(3) were synthesized and structural elucidation was confirmed by spectral data.Antifungal and antioxidant activities were evaluated include minimum inhibitory concentrations and IC50 values,respectively of the titled compounds.Compounds 6h,6i exhibited promising antioxidant activity at lower IC50 values 53.7μg/mL, 53.2μg/mL,respectively as compared with standard IC50 value 51.6μg/mL.  相似文献

Based on the structure-activity relationships and antiangiogenic mechanism of RGD-containing peptides,a series of 5-amino- 1,3-dihydro-1,3-dioxo-2H-isoindole derivatives were synthesized.The structures were characterized by ~1H NMR,MS and elementary analysis.There ability to inhibit angiogenesis were evaluated by chick embryo chorioallantoic membrane assay at 10~(-5) mol/L.Compounds 7a and 7b displayed obvious antiangiogenic activity.  相似文献

An efficient and environment friendly process for the synthesis of α-aminophosphonates has been devised. Through a one-pot three-component condensation of various aldehydes, amines, and triethyl phosphite in the presence of Fe3O4@SiO2-imid-PMAn nanoparticles as magnetic catalysts under solvent-free conditions and ultrasonic irradiation, α-aminophosphonates were obtained with excellent yields. The reactions under solvent-free conditions at room temperature are compared with the ultrasonic-assisted reactions. This new procedure has notable advantages such as short reaction time, excellent yields, easy purification, and the absence of any tedious workup or purification. The aforementioned catalyst could be easily recovered by an external magnetic field and can be reused for six consecutive reaction cycles without significant loss of activity. In addition, SEM and DLS of the catalyst after the reaction cycle were investigated.  相似文献

A versatile, alternative and environmentally benign strategy for the synthesis of a series of pyrazoles has been successfully performed in water using PEG–SO3H as an acidic catalyst. The products are obtained in high yield from the one-pot reaction procedure involving dicarbonyl compounds and hydrazines/hydrazides. This new method totally avoids the use of organic acids and toxic or expensive solvents in this reaction. The catalyst is waste-free, easily prepared, and efficiently re-used.  相似文献

Double alkylation of enantiopure N,N-acetal pyrimidinone (S)-1, a masked chiral derivative of β-alanine prepared from (S)-asparagine, proceeds with high stereoselectivity to give C(5) disubstituted adducts (2S,5R)-6, (2S,5S)-6, (2S,5R)-7, and (2S,5S)-7. Acid hydrolysis of these derivatives affords enantiopure α,α-dialkylated β-amino acids (R)-8, (S)-8, (R)-9, and (S)-9 in very good yields.  相似文献

Mesoporous MCM-41 supported Schiff base and Cu SO_4 5H_2 O shows high catalytic activity in the cyclocondensation of mercaptoacetic acid with imines(or aldehydes and amines) to afford pharmaceutically important thiazolidinone derivatives. This work describes the regioselective synthesis of two new series of 1,1′-oxalylbis[3-(alkyl/aryl/heteroaryl)-4,5-dihydro-5-hydroxy-5-(trihalomethyl)-1H-pyrazoles], where the 3-substituents are H, Me, C6H5, 4-FC6H4, 4-ClC6H4, 4-BrC6H4, 4-NO2C6H4, 4,4′-BiPh, and 2-furyl, in a one-pot methodology with ethanol as solvent, from the reaction of 4-alkoxy-4-(alkyl/aryl/heteroaryl)-1,1,1-trihaloalk-3-en-2-ones with oxalyldihydrazide (51–89%). Complementarily, the dehydration reactions of five examples of the described oxalylbispyrazolines are also reported, which furnished the respective 1,1′-oxalylbis[3-(alkyl/aryl/heteroaryl)-5-(trihalomethyl)-1H-pyrazoles] in 53–78% yields without the two C(O)–N bond cleavages.  相似文献

In this work, titanomagnetite nanoparticles (Fe3-xTixO4) have been used as a novel suppport for the synthesis of a magnetic acidic catalyst. These nanoparticles were functionalized with sulfonic acid groups to prepare the Fe3-xTixO4@SO3H nanoparticles. The synthesized acidic nanoparticles have been explored as new and efficient recyclable heterogeneous catalyst for a one-pot, three-component synthesis of tetrahydrobenzo[b]pyrans known as 4H-chromenes and 1,4-dihydropyrano[2,3-c]pyrazoles. The structure of the catalyst was established by infrared, energy dispersive x-ray (EDX), and scanning electron microscopy analyses. The reactions proceed smoothly to furnish the respective products in excellent yields and short reaction times. The facile reaction conditions, easy isolation of the products, versatility, and easy magnetic separation and reusability of the catalyst with no significant loss of activity are the main merits of the present method.  相似文献

A facile and environmentally benign synthesis of some 2-amino-6-(2-oxo-2H-chromen-3-yl)-4-arylnicotinonitrile derivatives from the reaction of 3-acetylcoumarin, aromatic aldehydes, and malononitrile under solvent-free condition in the presence of silica-supported perchloric acid (HClO4–SiO2) is described. An efficient and direct procedure has been developed for the preparation of amidoalkyl naphthols by a one-pot condensation of aryl aldehydes, 2-naphthol and urea or amides, in the presence of HClO4-SiO2 as a heterogeneous catalyst. The reactions were carried out under reflux and solvent-free conditions. The present methodology offers several advantages such as excellent yields, simple procedure, easy work-up and ecofriendly reaction condition. An efficient synthesis toward highly diversified α-carboline derivatives via a three-component tandem reaction using acid chlorides, terminal alkynes, and 2-aminoindole hydrochlorides has been described. The salient feature of the one-pot strategy involves regioselective [3 + 3]-cyclocondensation and the presence of water in the reaction medium to facilitate cyclization.
