固相萃取-亲水作用色谱法测定废水中四环素类抗生素   总被引:1,自引:0,他引:1  
建立了固相萃取(SPE)-亲水作用色谱法(HILIC)测定废水中金霉素(CTC)、强力霉素(DC)、四环素(TC)和土霉素(OTC) 4种四环素类抗生素(TCs)残留的新方法.水样经Oasis HLB固相萃取柱净化富集后, 采用以氨基色谱柱及高极性有机溶剂-水相缓冲溶液为流动相的亲水作用色谱法(HILIC)进行分析. 对流动相中缓冲溶液的类型和pH值、离子强度、 有机溶剂的浓度以及流速进行了优化, 确定了以V(乙腈)∶V(6.7 mmol/L柠檬酸铵缓冲溶液, pH 4.0)=85∶ 15混合液为流动相的最佳条件.本方法具有良好的线性关系(r> 0.999)和重现性(峰面积RSD<1.0%), 最低检出限(S/N=3)为12~30 μg/L, 4种四环素类药物添加水平在0.5~10 μg/L范围内的标准加入回收率为 90.6%~106.5%; 相对标准偏差为 2.5%~6.2%.本方法简便、准确、流动相离子强度低,适合于与质谱联用,用于屠宰场污水及医院污水等实际样品检测,结果满意.  相似文献

A rapid, specific, and sensitive method has been developed using molecularly imprinted polymers (MIPs) as solid-phase extraction sorbents for extraction of trace tetracycline antibiotics (TCs) in foodstuffs. MIPs were prepared by precipitation polymerization using tetracycline as the template. Under the optimal condition, the imprinting factors for MIPs were 4.1 (oxytetracycline), 7.0 (tetracycline), 7.4 (chlortetracycline), 7.7 (doxycycline), respectively. Furthermore, the performance of MIPs as solid-phase extraction sorbents was evaluated and high extraction efficiency of molecularly imprinted solid-phase extraction (MISPE) procedure was demonstrated. Compared with commercial sorbents, MISPE gave a better cleanup efficiency than C18 cartridge and a higher recovery than Oasis HLB cartridge. Finally, the method of liquid chromatography–tandem mass spectrometry coupled with molecular-imprinted solid-phase extraction was validated in real samples including lobster, duck, honey, and egg. The spiked recoveries of TCs ranged from 94.51% to 103.0%. The limits of detection were in the range of 0.1–0.3 μg kg−1. Monodisperse molecularly imprinted polymers (MIPs) for diphenyl phosphate (DPP) and 1-naphthyl phosphate (1-NapP) have been prepared by a multi-step swelling and polymerization method using 4-vinylpyridine as a functional monomer, glycerol dimethacrylate as a crosslinker and cyclohexanol or 1-hexanol as a porogen. The retention and molecular-recognition properties of these MIPs for organophosphorus compounds were evaluated by HPLC using a mixture of phosphate buffer and acetonitrile as an eluent. In addition to shape recognition, hydrogen bonding and hydrophobic interactions could play an important role in the retention and molecular recognition of DPP and 1-NapP. Furthermore, the MIPs were applied to the separation of adenosine and adenosine phosphates (AMP, ADP and ATP). These phosphates were retained on the MIPs according to the number of phosphate groups in the molecule and were well separated from one another. Hydrogen bonding and hydrophobic interactions seemed to affect the retention and recognition of adenosine phosphates in low acetonitrile content, while hydrophilic interactions affected these properties in high acetonitrile content. Finally, the MIPs were applied to the trapping of phosphopeptides. The MIPs non-selectively trapped phosphopeptides, which have phosphorylated tyrosine, serine or threonine in the sequences, and successfully trapped four phosphopeptides in tryptic digests of bovine α-casein.  相似文献

Tetracycline (TC)‐imprinted microspheres have been synthesized by reversible addition–fragmentation chain‐transfer precipitation polymerization using PEG as a coporogen. In the synthesis, methacrylic acid and ethylene dimethacrylate were used as the functional monomer and cross‐linker, respectively. 2,2′‐Azobisisobutyronitrile was the initiator, and cumyl dithiobenzoate was the chain‐transfer reagent. Although monodispersed microspheres were obtained using acetonitrile as porogen, the particles cannot be used in the column extraction because of the high backpressure. To increase the porosity of the material, PEG was introduced as a coporogen. The influence of the molecular weight and concentration of PEG on the morphology, binding affinity, and porosity of the molecularly imprinted polymers (MIPs) have been studied. The results demonstrated that PEG as a macroporogen increased the porosity of the polymers. Meanwhile, the column backpressure was reduced using the MIPs with higher porosity. The binding affinity of the MIPs was increased when a low concentration of PEG was employed, while it was decreased when the ratio of PEG 12 000/monomers was >0.8%. Under the optimized conditions, TC‐imprinted microspheres with good selectivity and size uniformity have been obtained, which facilitates its application in the column extraction for TC determinations.  相似文献

Two novel molecularly imprinted polymers (MIPs) selected from a combinatorial library of bupivacaine imprinted polymers were used for selective on-line solid-phase extraction of bupivacaine and ropivacaine from human plasma. The MIPs were prepared using methacrylic acid as the functional monomer, ethylene glycol dimethacrylate as the cross-linking monomer and in addition hydroxyethylmethacrylate to render the polymer surface hydrophilic. The novel MIPs showed high selectivity for the analytes and required fewer and lower concentrations of additives to suppress non-specific adsorption compared with a conventional MIP. This enabled the development of an on-line system for direct extraction of buffered plasma. Selective extraction was achieved without the use of time-consuming solvent switch steps, and transfer of the analytes from the MIP column to the analytical column was carried out under aqueous conditions fully compatible with reversed-phase LC gradient separation of analyte and internal standard. The MIPs showed excellent aqueous compatibility and yielded extractions with acceptable recovery and high selectivity.  相似文献

抗生素的滥用及残留对生物体和环境造成极大危害,其含量低、种类多、基质复杂,通常需要进行样品前处理结合色谱分析以实现灵敏测定。分子印迹聚合物(MIPs)能选择性识别、有效富集目标分析物并消除干扰,已广泛用于抗生素的样品前处理中。该文对MIPs制备中面临的挑战进行了总结;对2016年以来抗生素MIPs的固相萃取应用进行了综述和展望,主要包括固相萃取、分散固相萃取、磁固相萃取、基质固相分散萃取、固相微萃取、搅拌棒吸附萃取。此外,该文重点介绍了抗生素MIPs的印迹新策略,如多模板、多功能单体、虚拟模板、刺激响应、亲水性印迹等。最后,该文对抗生素MIPs的制备和前处理应用进行了展望。  相似文献

A method using liquid chromatography-tandem mass spectrometry has been developed for determination of trace levels of tetracycline antibiotics in ground water and confined animal feeding operation waste water. Oxytetracycline (OTC), tetracycline (TC), and chlortetracycline (CTC) were extracted from water samples using both polymeric and C18 extraction cartridges. The addition of a buffer containing potassium phosphate and citric acid improved tetracycline recoveries in lagoon water. Method detection limits determined in reagent water fortified with 1 microg l(-1) OTC, TC, and CTC were 0.21, 0.20, and 0.28 microg l(-1). Method detection limits in lagoon water samples fortified at 20 microg l(-1) for OTC, TC, and CTC were 3.6, 3.1, and 3.8 microg l(-1). Variability in recovery from laboratory fortified blanks ranged from 86 to 110% during routine analysis.  相似文献

Uniformly-sized, molecularly imprinted polymers (MIPs) for atrazine, ametryn and irgarol were prepared by a multi-step swelling and polymerization method using ethylene glycol dimethacrylate as a cross-linker and methacrylic acid (MAA), 2-(trifluoromethyl) acrylic acid (TFMAA) or 4-vinylpyridine either as a functional monomer or not. The MIP for atrazine prepared using MAA showed good molecular recognition abilities for chlorotriazine herbicides, while the MIPs for ametryn and irgarol prepared using TFMAA showed excellent molecular recognition abilities for methylthiotriazine herbicides. A restricted access media-molecularly imprinted polymer (RAM-MIP) for irgarol was prepared followed by in situ hydrophilic surface modification using glycerol dimethacrylate and glycerol monomethacrylate as hydrophilic monomers. The RAM-MIP was applied to selective pretreatment and enrichment of methylthiotriazine herbicides, simetryn, ametryn and prometryn, in river water, followed by their separation and UV detection via column-switching HPLC. The calibration graphs of these compounds showed good linearity in the range of 50-500 pg/mL (r > 0.999) with a 100 mL loading of a river water sample. The quantitation limits of simetryn, ametryn and prometryn were 50 pg/mL, and the detection limits were 25 pg/mL. The recoveries of simetryn, ametryn and prometryn at 50 pg/mL were 101%, 95.6% and 95.1%, respectively. This method was successfully applied for the simultaneous determination of simetryn, ametryn and prometryn in river water.  相似文献

A novel solid phase extraction (SPE) method for determination of tetracyclines (TCs) in milk and honey samples by molecularly imprinted monolithic column was developed. Using tetracycline (TC) as the template, methacrylic acid (MAA) as the functional monomer, ethylene glycol dimethacrylate (EGDMA) as the cross-linker, methanol as the solvent, cyclohexanol and dodecanol as the mixed porogenic solvents, a TC imprinted monolithic column was prepared by in situ molecular imprinting technique for the first time, and the optimal synthesis conditions and the selectivity of TC imprinted monolithic column were investigated. The interfering substances in food samples and TCs can be separated successfully on imprinted column. Molecularly imprinted solid phase extraction (MISPE) coupling with C18 column was used to determinate the TCs in milk and honey. The recoveries of this method for six tetracyclines antibiotics such as tetracycline (TC), oxytetracycline (OTC), minocycline (MINO), chlortetracycline (CTC), metacycline (MTC) and doxycycline (DTC) were investigated, and high recoveries of 73.3-90.6% from milk samples and 62.6-82.3% from honey samples were obtained. A method for determination of TCs at low concentration level in milk and honey samples was successfully developed by using the monolithic column as the precolumn for solid phase extraction of six TCs compounds.  相似文献

A mixture of five tetracycline (TC) derivatives: minocycline (MC), demeclocycline (DMCTC), doxycycline (DC), and sancycline (SC), as well as each TC derivative from its main degradation product were separated by capillary zone electrophoresis (CZE). The influence of the pH and the concentration and nature of the background electrolyte (BGE) on the separations was investigated. Ethylenediaminetetraacetic acid (EDTA; 1 mM) was used as additive in a 25 mM phosphate buffer (pH 2.3) because this BGE enabled the rapid separation of the TC derivatives and of each TC derivative from its respective degradation product in less than 6 min. After optimization of the separation conditions, the analytical characteristics of the method were investigated. The parameters involved were linearity, precision (repeatability and reproducibility), and limits of detection (LODs). LODs obtained for the five TC derivatives studied were about 3 microg/mL. Finally, the CZE method developed was applied to study the stability of TC derivatives and to analyze the TC derivative content in three different pharmaceutical preparations.  相似文献

A molecularly imprinted polymer solid phase extraction (MISPE) method combined with flow-injection chemiluminescence (FI-CL) for the determination of residual tetracycline (TC) in fish samples is presented. The molecularly imprinted polymer (MIP) of TC was synthesized and particles of this MIP were packed into a polytetrafluoroethylene (PTFE) tube, which was connected into the sampling loop of an eight-way injection valve and served as the MISPE column for on-line selective adsorption of TC. The eluent (CH3CN : HNO3 (0.01 mol L(-1)) = 4 ratio 1, v ratio v) was used for extracting the adsorbed TC, which could be detected by its good enhancing effect on the CL reaction between Ce(iv) and rhodamine B. The CL intensity is linear to TC concentration in the range from 4 x 10(-9) to 4 x 10(-7) g mL(-1). The detection limit is 1 x 10(-9) g mL(-1) (3 sigma) and the relative standard deviation is 2.4% (n = 9). The conditions of preconcentration, extraction and CL reaction were carefully studied. The selectivity experiment shows that the selectivity and sensitivity of the CL method could be improved greatly when MIP was used as a recognition material in SPE. However, the MISPE column interacted indiscriminately with oxytetracycline (OTC) with a 49 +/- 2% binding. An intermediate differential pulsed elution (DPE) step using 3% acetic acid as eluent was employed to remove OTC and other interfering substances. The proposed MISPE-CL method has been applied successfully to the determination of TC in fish samples. At the same time, the binding characteristics of the polymer to tetracycline were evaluated by batch and dynamic methods.  相似文献

A simple, rapid and precise analytical method for tetracycline (TC) residues in the liver of slaughtered animals has been established. The recoveries of oxytetracycline (OTC), TC, chlortetracycline (CTC) and doxycycline (DC) from beef liver spiked at the level of 1.0 ppm were 87.7, 87.5, 79.6 and 67.5% with coefficients of variations of 1.01-2.87%. Detection limits in beef liver were 0.05 and 0.1 ppm for OTC and TC and for CTC and DC, respectively. It is also possible to apply this method to the analysis of residual TCs in various foods with the same recovery, accuracy and detection limits as in the case of beef liver.  相似文献

Molecular imprinted polymers (MIPs) were prepared through thermal polymerization by using quercetin as the template molecule, acrylamide (AA) as the functional monomer and ethylene glycol dimethacrylate (EDMA) as the cross-linker in the porogen of tetrahydrofuran (THF). The synthesized MIPs were identified by both Fourier transform infrared (FTIR) and scanning electron microscope (SEM). Systematic investigations of the influences of key synthetic conditions, including functional monomers, porogens and cross-linkers, on the recognition properties of the MIPs were conducted. Scatchard analysis revealed that the homogeneous binding sites were formed in the polymers. Besides quercetin, two structurally similar compounds of rutin and catechol were employed for molecular recognition specificity tests of MIPs. It was observed that the MIPs exhibited the highest selective rebinding to quercetin. Accordingly, the MIPs were used as a solid-phase extraction (SPE) sorbent for the extraction and enrichment of quercetin in cacumen platycladi samples, followed by HPLC-UV analysis. The application of MIPs with high affinity and excellent stereo-selectivity toward quercetin in SPE might offer a novel method for the enrichment and determination of flavonoid compounds in the natural products.  相似文献

Abstract

In this research, hydrophilic molecularly imprinted microspheres (HMIPs) for azoxystrobin were successfully synthesized through precipitation polymerization. The adsorption capacities of HMIPs for azoxystrobin in water medium were higher than ordinary molecularly imprinted microspheres (MIPs), and HMIPs exhibited good hydrophilic properties. HMIPs and non-imprinted microspheres (HNIPs) were characterized by FT-IR, SEM, laser particle size analyzer and TGA. Comparing with HNIPs, azoxystrobin had a significant influence on morphologys and sizes of HMIPs. The Langmuir adsorption isotherm illustrated each binding site of HMIPs had the same adsorption capacity. The Lagergran pseudo-second-order kinetic model indicated the adsorption process between azoxystrobin and HMIPs was chemical adsorption. BET test illustrated HMIPs had bigger specific surface areas than HNIPs. Selective adsorption indicated that HMIPs had highly specific recognition of azoxystrobin. HMIPs successfully exhibited high selectivity and high hydrophilicity in water medium.  相似文献

We report on a protocol for a simultaneous competitive immunoassay for tetracycline (TC) and chloramphenicol (CAP) on the same sensing interface. Conjugates of TC and of CAP with bovine serum albumin were first co-immobilized on a glassy carbon electrode modified with gold nanoparticles. In parallel, monoclonal anti-TC and anti-CAP antibodies were conjugated onto CdS and PbS nanoclusters, respectively. In a typical assay, the immobilized haptens and the added target analytes competed for binding to the corresponding antibodies on the nanoclusters. Subsequently, Cd(II) and Pb(II) ions are released from the surface of the corresponding nanoclusters by treatment with acid and then were detected by square wave anodic stripping voltammetry. The currents at the peak potentials for Cd(II) and Pb(II) were used as the sensor signal for TC and CAP, respectively. This multiplex immunoassay enables the simultaneous determination of TC and CAP in a single run with dynamic ranges from 0.01 to 50 ng mL?1 for both analytes. The detection limits for TC and for CAP are 7.5 pg mL?1 and 5.4 pg mL?1, respectively. No obvious nonspecific adsorption and cross-reactivity was observed in a series of analyses. Intra-assay and inter-assay coefficients of variation were less than 10 %. The method was evaluated by analyzing TC and CAP in spiked samples of milk and honey. The recoveries range from 88 % to 107 % for TC, and from 91 % to 119 % for CAP.

以3种结构类似的抗胆碱能药物盐酸新托品(1116)、盐酸苯环壬酯(8021)和盐酸戊乙奎醚(8018)为模板分子合成分子烙印聚合物,采用固相萃取-高效液相色谱法(SPE-HPLC)考察各MIP对乙腈溶液中结构类似的药物1116、8021、8018及盐酸卡马特灵(1113)、氯苯那敏(CPA)和樟柳碱(AT3)的固相吸附行为,探讨MIP特异性识别的影响因素及机理。结果表明,在MIP合成中,功能单体、交联剂及引发剂的种类和用量、引发方式、模板分子与功能单体的比例等因素对MIP的特异性识别能力均有重要影响。以1116、8021和8018为模板分子、甲基丙烯酸(MAA)为功能单体、三羟甲基丙烷三甲基丙烯酸酯(TRIM)为交联剂,在乙腈中合成的各MIP均表现出较强的特异性识别能力。其中,MIP对待测物的非特异性吸附主要由其网状结构表面游离的极性基团引起;MIP的特异性识别过程中,识别位点与待测物的空间结构匹配起着更为重要的作用。  相似文献

用紫外光谱方法研究了四环素(TC) Cu(II)配合物与DNA的相互作用.吸收光谱研究表明,DNA能与四环素(TC)及Cu(II)形成的配合物发生反应,配合物与DNA的作用方式随着配合物类型及DNA浓度的不同而不尽相同:当四环素与铜形成1∶1型配合物时,较低浓度的DNA能与配合物以嵌插方式相互作用,而较高浓度的DNA与该配合物除了发生嵌插作用外,还存在另外的作用方式;当四环素与铜形成1∶2型配合物时,DNA与该配合物则主要以嵌插方式相互作用,并且这两种配合物与DNA的嵌插作用均是通过四环素配体插入的.  相似文献

研究了四环素(TC)-Al3+配合物与DNA的相互作用.吸收光谱和荧光光谱研究表明,TC与DNA无明显的相互作用,但在Al3+离子存在下,TC能结合到小牛胸腺DNA上.TC先与Al3+作用,生成TC-Al二元配合物,该配合物再与DNA发生作用,形成(TC-Al)-DNA三元配合物.TC-Al二元配合物与DNA的结合常数为6.5×104L/mol,其配位比n(TC):n(Al):n(DNA)=1:1:1.TC-Al配合物与DNA的键合模式主要是静电相互作用.溶液的pH值对TC-Al二元配合物和(TC-Al)-DNA三元配合物的形成有一定的影响.利用TC-Al配合物与DNA作用使体系荧光强度增强的性质测定DNA,其线性范围为1.0×10-6~6.0×10-5mol/L,检测限为5.0×10-7mol/L.分别测定了天然与热变性酵母样品中DNA的含量,结果令人满意.  相似文献
