Parental exposure to polybrominated diphenyl ethers (PBDEs) in animals has been found to be transferred to the offspring. The environmental health risk and toxicity to the offspring are still unclear. The objective of the present study was to identify environmentally relevant concentrations of PBDEs for parental exposure that would cause developmental neurotoxicity in the offspring. Adult zebrafish were exposed to environmentally relevant concentrations of DE-71 (0.16, 0.8, 4.0 μg/L) via water. The results showed that PBDE exposure did not affect larvae hatching, malformation, or survival. The residue of PBDEs was detected in F1 eggs upon parental exposure. Acetylcholinesterase (AChE) activity was significantly inhibited in F1 larvae. Genes of central nervous system development (e.g., myelin basic protein, synapsin IIa, α1-tubulin) were significantly downregulated in larvae. Protein levels of α1-tubulin and synapsin IIa were also reduced. Decreased locomotion activity was observed in the larvae. This study provides the first evidence that parental exposure to environmentally relevant concentrations of PBDEs could cause adverse effects on neurodevelopment in zebrafish offspring.  相似文献

In recent years, polybrominated diphenyl ethers (PBDEs) have been detected at increasing levels in the environment due to their widespread use as flame retardants. PBDEs can affect thyroid hormone homeostasis and the cholinergic neurotransmitter system. In this study, several PBDE congeners were detected in whole brain samples and neuronal cells of herring gulls (Larus argentatus). A herring gull neuronal cell culture method was used to determine the effects of PBDEs on cytotoxicity and mRNA expression. Real-time RT-PCR assays were developed for genes associated with the thyroid hormone pathway (thyroid hormone receptors [TR alpha and beta], transthyretin [TTR]), and the cholinergic system (neuronal nicotinic acetylcholine receptor alpha-7 [nAChR alpha-7]). Administration of T3 resulted in a significant up-regulation of the two TRs and a significant down-regulation of TTR. TTR was also down-regulated by the commercial penta-BDE mixture, DE-71. In contrast, neither DE-71, nor BDE-47, -99, or -100 altered the mRNA levels of the TRs or nAChR alpha-7. The in vitro approach was a relevant model system for assessing the effects of PBDEs on cytotoxicity and mRNA expression. Herring gull neuronal cells were responsive to both T3 and PBDEs although, receptors associated with two predicted mechanisms of PBDE action were not effective molecular biomarkers of exposure.  相似文献

Polybrominated diphenyl ethers (PBDEs) have been used worldwide as additive flame retardants in polymeric materials. Commercial products consist predominantly of deca-, octa-, and pentabromodiphenyl ether mixtures. PBDEs are resistant to degradation in the natural environment and Penta-BDE in particular accumulates in the fatty tissues of fish, birds and mammals (including humans). Several toxic effects on the thyroid system or on neurodevelopment have been reported in experimental animals exposed to PBDEs. It is likely that human exposure is predominantly through the ingestion of contaminated food and/or mother's milk. The potential health effects of dietary exposure to PBDEs have now become a great concern because of the increasing PBDE levels in the biosphere. In this review, published information on the toxicology of PBDEs, levels in foodstuffs and human milk, and analytical methods has been compiled.  相似文献

Prenatal exposure to polybrominated diphenyl ethers (PBDEs) may disrupt thyroid function and contribute to adverse neurodevelopmental outcomes. We conducted a pilot study to explore the relationship between serum concentrations of lower-brominated PBDEs (BDE-17 to -154), higher-brominated PBDEs (BDE-183 to -209), and hydroxylated PBDE metabolites (OH-PBDEs) with measures of thyroid function in pregnant women. Concentrations of PBDEs, OH-PBDEs, thyroid-stimulating hormone (TSH), total thyroxine (T(4)), and free T(4) were measured in serum samples collected between 2008 and 2009 from 25 second trimester pregnant women in California. Median concentrations of lower-brominated PBDEs and OH-PBDEs were the highest reported to date in pregnant women. Median concentrations of BDE-47 and the sum of lower-brominated PBDEs (ΣPBDE(5)) were 43.1 ng/g lipid and 85.8 ng/g lipid, respectively, and the sum of OH-PBDEs (ΣOH-PBDE(4)) was 0.084 ng/mL. We observed a positive association between the weighted sum of chemicals known to bind to transthyretin (ΣTTR binders) and TSH levels. We also found positive associations between TSH and ΣPBDE(5), ΣOH-PBDE(4), BDE-47, BDE-85, 5-OH-BDE47, and 4'-OH-BDE49, and an inverse association with BDE-207. Relationships with free and total T(4) were weak and inconsistent. Our results indicate that PBDE exposures are elevated in pregnant women in California and suggest a relationship with thyroid function. Further investigation is warranted to characterize the risks of PBDE exposures during pregnancy.  相似文献

This study examined human exposure to polybrominated diphenyl ethers (PBDEs) associated with fish consumption and inhalation in China. The median intake of sigma7 PBDEs via human milk was 48.2 ng/day for nursing infants (0-1 years old) (a range of 23.4-99.1 ng/day). For all other age groups, the median intake of sigma11 PBDEs via fish consumption was 1.7-12.9 ng/day with a range of 0.59-56.3 ng/g. Additionally, human exposure to PBDEs via inhalation was 2.7-9.2 ng/day (a range of 0.72-108 ng/day). The median total sigma11 PBDEs intakes for nursing infants (6874 and 7372 pg/ kg b.w./day for males and females, respectively) were much higher than other age groups (215-608 pg/kg b.w./ day). No significant difference in the total PBDEs intakes was found between males and females. Of the 11 PBDEs congeners, BDE-47 was predominant in the total intake for nursing infants with a mean contribution of 38%, whereas BDE-209 was the dominant congener of total intake for other age groups, varying from 44 to 61%. Currently, the PBDEs levels in Chinese consumer fish and the total intakes of PBDEs via fish consumption were at the lower end of the global range. Compared with similar studies in other countries, however, human exposure to PBDEs via inhalation in China was relatively high. Overall, estimated daily intake of total PBDEs in the Chinese population was far below the LOAEL. However, studies are needed to further understand the fate and impact of PBDEs as PBDE-containing products are still used widely in large quantities in China.  相似文献

The objective of this study was to determine the concentrations and compositions of polybrominated biphenyls (PBBs), polybrominated dibenzo-p-dioxins (PBDDs), and polybrominated dibenzofurans (PBDFs) as contaminants in the commercial polybrominated diphenyl ether (PBDE) mixtures DE-71, DE-79, and DE-83 and to ascertain the lot-to-lot variations in the proportions of these contaminants. Commercial PBDE mixtures tested in the present study contained both PBBs and PBDFs, as impurities, at concentrations in the range of several tens to several thousands of nanograms per gram. Concentrations of total PBDFs were greater than those of total PBBs in DE-79 and DE-83 mixtures. PBDDs were not detected at levels above the limit of detection. Profiles of PBB and PBDF congeners varied with the degree of bromination of the commercial PBDE mixtures (i.e., more highly brominated mixtures of PBDEs contained heavily brominated homologues of PBBs and PBDFs). On the basis of the production/ usage of commercial PBDE mixtures in 2001, potential global annual emissions of PBBs and PBDFs were calculated to be 40 and 2300 kg, respectively. Results of our study suggestthat PBDFs can also be formed during the production of commercial PBDE mixtures, in addition to their formation during pyrolysis of brominated flame retardants.  相似文献

Some ubiquitous pollutants of the aquatic environment, such as PCBs or other polyhalogenated aromatic hydrocarbons, may disrupt the thyroid hormone system. In a partial life cycle assay with zebrafish (Danio rerio), we studied the effects of the reference compound propylthiouracil (PTU) on reproduction, growth and development, histopathology of some target tissues, and plasma thyroid hormone levels. PTU induced a concentration-dependent increase of egg production with a concomitant decrease of mature oocyte size but had no effect on fertilization rate or hatching. In F1, serious dysmorphogenesis was found in 4 dph larvae at the highest PTU level tested (100 mg/L), and there was a dose-dependent decrease in body length and weight at 42 dph (significant at 100 mg/L PTU). At this time, there was also a decreased scale thickness, suggesting inhibited metamorphosis, detectable at 1 mg/L PTU and higher. PTU also induced activation of the thyroid follicles in a concentration-dependent way, in juveniles associated with hyperemia in the thyroid area, and depletion of liver glycogen. Effects in adults were associated with decreased circulating levels of the thyroid hormones T3 and T4. These observations indicate that disruption of the thyroid hormone system may affect the fitness of these aquatic organisms. The zebrafish model may contribute to the identification of thyroid hormone disrupting activity in water samples and also in the interpretation of histological observations in free-ranging fish species.  相似文献

The biotransformation of high bromosubstituted polybrominated diphenyl ethers (PBDEs) congeners contained in a commercial deca mixture (DeBDE) is of environmental concern because it might lead to the increase of toxic low brominated PBDEs in biota. A few studies have reported that freshwater fish dietary exposed to DeBDE or its main constituent, decabrominated PBDE congener (BDE-209), had their tissues enriched with PBDEs not initially present in fish or feed. In the present study, Atlantic tomcod (Microgadus tomcod) were intraperitoneally (IP) injected with DeBDE to assess hepatic concentration changes of PBDEs and methoxy polybrominated diphenyl ethers (MeO-PBDEs) in a marine fish species. Tomcod were also IP injected with polychlorinated biphenyl (PCB)-126 to evaluate the impact of cytochrome P4501A (CYP1A) induction on the biotransformation of injected PBDEs contained in DeBDE and PBDEs initially present in fish. Besides BDE-209, concentrations of BDE-203 and three other unidentified octabrominated PBDEs and the nonabrominated PBDEs (BDE-206, -207, and -208) were enriched in the liver of fish injected with DeBDE. All these PBDE congeners, essentially absent in control fish, were identified as impurities in DeBDE, and, thus, their presence could not be attributed exclusivelyto biotransformation. Despite a 4.3times increase of EROD activity in the liver of tomcod injected with both PCB-126 and DeBDE, compared to DeBDE alone, no further increases of PBDE hepatic concentrations were observed. However, depleted concentrations of BDE-17 (x 1.5) and 6-MeO-BDE-47 (x 1.4) were found in fish IP injected with DeBDE compared to control fish, likely due to activated hepatic metabolic enzymes other than CYP1A. Fish injected with PCB-126 showed an even more significant depletion of BDE-17 hepatic concentrations (x 3.5) than the one associated with the DeBDE treatment and a significantly lower proportion of fish with quantifiable concentrations of BDE-203. Thus, CYP1A inducers can promote the biotransformation of PBDEs in fish liver. This study shows that exposure of fish to DeBDE is expected to result in the enrichment of high brominated PBDEs in fish liver and that metabolic activities in fish can affect their PBDE bioaccumulation pattern and possibly the toxicity of PBDEs to fish.  相似文献

Polybrominated diphenyl ethers (PBDEs) have been widely used to flame-retard products common in homes and the workplace, and subsequently, they have become widely dispersed in the environment. Detailed compositional knowledge of these complex PBDE mixtures is crucial to a fuller understanding of their toxicological potencies and environmental fate due to selective congener biomagnification, degradation, and transport. Utilizing recenttechnical enhancements and newly available commercial standards, we developed a method capable of analyzing a larger suite of mono- through deca-BDEs. We then characterized the congener composition of six common technical flame-retardant mixtures: two penta-BDE products (DE-71 and Bromkal 70-5DE) two octa-BDE products (DE-79 and Bromkal 79-8DE) and two deca-BDE products (Saytex 102E and Bromkal 82-0DE). PBDEs were analyzed by gas chromatography/mass spectrometry (GC/MS). Structural conformations based on fragmentation patterns and molecular ions were established by electron-capture negative ionization (ECNI) and electron ionization (El). Sixty-four commercially available PBDE standards were chromato-graphed on two GC columns (DB-1HT and DB-5HT) and relative retention indexes (RRI) calculated. Thirty-nine PBDEs were identified in these products, 29 at concentrations >0.02% by weight. Of these, 12 previously unreported congeners have been confirmed as commercial mixture components. Four of these congeners were detected >0.02% w/w (BDE-144, -171, -180, and -201) and three (BDE-75, -184, and -194) at <0.02%. Five other congeners (four <0.02% by weight) were tentatively identified based on their molecular ion and ECNI fragmentation in the absence of corresponding analytical standards.  相似文献

The role of insect larvae and pupae as sources of organochlorine compounds (OCs) and polybromodiphenyl ethers (PBDEs) in freshwater food webs for high predators such as fish is evaluated. Trichoptera and diptera have been taken as organisms of choice for such comparison because they are common in benthic aquatic habitats and accumulate substantial amounts of these compounds. Hexachlorobenzene, hexachlorocyclohexanes,4,4'-DDE,4,4'-DDT, polychlorobiphenyls, and PBDEs have been measured. The results show a nonselective enrichment of OCs and PBDEs from larvae to pupae. These concentration increases may result from the weight loss of pupae during metamorphosis as a consequence of mainly protein carbon respiration and lack of feeding. Despite the lack of change in total amount, the concentration increases from larvae to pupae are very relevant for the pollutant ingestion of the higher predators. The intakes of OCs and PBDEs by trout are between 2- and 5-fold higher per calorie gained when predating on pupae than on larvae. Since pollutant concentration, energy reward, predation susceptibility, and duration of life stage are very different between these two insect stages, and none of them is irrelevant for the incorporation of OCs or PBDEs to higher levels, bioaccumulation food-web models should distinguish between the two sources.  相似文献

In this work, we investigated exposure levels, distribution patterns, and potential harmful impacts of polybrominated diphenyl ethers (PBDEs) on thyroid hormone activity in 26 children with congenital hypothyroidism and their mothers' pair and 12 normal control pairs. The average concentration of PBDEs in congenital hypothyroidism (median: 22.16 ng/g lipid) was higher than in normal controls (median: 14.76 ng/g lipid), but there was no statistical difference between the two groups. The BDE congeners were dominated by penta- to hepta-BDEs, but the greater brominated congeners (e.g., BDE 197, 196, 207, and 208) were relatively abundant in congenital hypothyroidism. BDE 138 was only observed in the congenital hypothyroidism cases. The maternal transfer and transport ratio of individual BDE congeners was shown for BDE 28 (0.588, p < 0.001), BDE 47 (0.564, p < 0.001), BDE 49 (0.712, p < 0.001) and BDE 119 (0.477, p = 0.002). The thyroid hormones were most obviously influenced by the internal exposure to PBDEs in normal mothers, showing a positive relationship with TSH (0.641 with BDE 154; 0.591 with BDE 153) and FT4 (0.584 with BDE 49; 0.572 with BDE 66) and a negative relationship with T3 (-0.577 with BDE 154) in the normal infants group. No significant correlations were observed in the congenital hypothyroidism cases.  相似文献

Juvenile lake trout (Salvelinus namaycush) were exposed to three dietary concentrations (0, approximately 2.5, and approximately 25 ng/g per BDE congener) of 13 BDE congeners (3-10 Br atoms) in the laboratory for 56 days, followed by 112 days of clean food, to examine bioaccumulation parameters and potential biochemical effects. The bioaccumulation of BDEs by the trout was highly influenced by biotransformation, via debromination, which resulted in bioaccumulation parameters that were much different than would be expected based on studies of chlorinated organic compounds (e.g., PCBs). Half-lives (t1/2's) for some BDE congeners (e.g., BDE-85 and -190) were much lower than expected based on their Kow, which was likely due to biotransformation, whereas t1/2's of other BDE congeners (e.g., BDE-66, -77, -153, and -154) were much longer than anticipated based on Kow. This was likely because the metabolites of BDE formed via debromination had the same chemical structure of these BDE congeners, which supplemented measured concentrations. The detection of three BDE congeners (an unknown penta, BDE-140, and an unknown hexa) in the fish that were not present in the food or in the control fish provide further evidence forthe debromination of BDEs. Half-lives of BDEs ranged from 38 +/- 9 to 346 +/- 173 days and biomagnification factors ranged from 1.6 (BDE-190) to 45.9 (BDE-100), but these bioaccumulation parameters need to be viewed with caution because they were highly influenced by debromination and relative abundance of individual BDEs that the fish were exposed to. CYP1A enzyme activity, measured as EROD, and free tri-iodothyronine (T3) concentrations in the plasma of lake trout varied significantly throughout the experiment but were not related to BDE exposure. In contrast, plasma levels of thyroxine levels (T4) were lower in both groups of PBDE-exposed fish compared with control fish after 56 days of exposure, and after 168 days in the high dose, suggesting that PBDEs may influence thyroid homeostasis at levels that are higher than what is normally found in the environment.  相似文献

为研究转苏云金芽胞杆菌(Bacillus thuringiensis,Bt)基因抗虫玉米长期食用对大鼠免疫的影响,以其亲本非转基因玉米饲料及商业饲料为对照,分别采用流式细胞术及液相芯片法检测喂养90 d后亲代及子代刚断乳大鼠外周血、脾脏免疫细胞分型及相关血清细胞因子的表达。结果发现,转Bt基因抗虫玉米饲料组亲代大鼠外周血和脾脏及子代脾脏各淋巴细胞亚群(CD4~+、CD8~+、TCRαβ~+、TCRγδ~+)的细胞数量比例及亲代血清中10种细胞因子表达水平均与非转基因玉米饲料组无显著差异(P0.05);子代外周血B淋巴细胞数量比例显著增高,巨噬细胞炎症蛋白-1α、单核细胞趋化因子-1β、白细胞介素-5表达水平明显降低(P0.05)。结果表明:转Bt基因抗虫玉米暴露90 d对亲代大鼠免疫系统影响不大;对子代刚断乳大鼠的细胞免疫及体液免疫有一定的影响。  相似文献

目的了解上海淡水水产品中多氯联苯(polychlorinated biphenyls,PCBs)和多溴联苯醚(poly brominated diphenyl ethers,PBDEs)的污染状况,并进行相关的人体健康风险评估。方法水产品经冷冻干燥后,经加速溶剂萃取(accelerated solvent extraction,ASE)提取、分散固相萃取(dispersive solid phase extraction,D-SPE)除脂、串联固相萃取(solid phase extraction,SPE)深度净化后,用气相色谱-串联质谱(gas chromatography-tandem mass spectrometry,GC-MS/MS)检测。结果 14类97件样品中,PCBs的平均检出率为97%,PBDEs的平均检出率为61%~71%。∑_(51)(PCBs)的平均浓度值为2588 pg/g ww,范围为328~12724 pg/g ww,中位值为2135 pg/g ww,∑_(39)(PBDEs)的平均值浓度为454 pg/g ww,范围为56~2210 pg/g ww,中位值为294 pg/g ww。结论目前上海居民食用淡水水产品不会因PBDEs和PCBs残留而对人体健康构成危险,但对于大量食用鱼和虾的婴幼儿(一般指2~3岁)应适当减少食用量。  相似文献

It has been hypothesized that a principal route of human exposure to polybrominated diphenyl ethers (PBDEs), used as flame retardants, is through fish consumption. Between 1999 and 2005 PBDE-47, -99, -100, -153, and -154 were analyzed in 3797 biological samples of 33 species of the Hudson River, New York. Approximately 98.4% of the samples contained PBDEs between 0.5 and 37 169 ng g(-1) lipid, with a median concentration of 772 ng g(-1) lipid. Yearly median sigmaPBDE concentrations fluctuated. Samples from river miles 112 and 153 contained higher sigmaPBDEs than those from other locations of the river. The 7-year median sigmaPBDE concentrations were the highest in large carnivorous fishes and the lowest in insects. The median abundance of congener PBDE-47 decreased from 80% to 63% with decreasing levels of sigmaPBDEs in the samples, while an increase from 2% to 23% was observed for PBDE-99. The median abundance of other congeners did not change with concentrations of sigmaPBDEs. Positive-, negative-, and no-correlation between sigmaPBDE concentrations and fish weight were observed for different species and for the same species from different locations of the river. The sources of PBDE contamination, diet, metabolic activity, and sediment chemistry might affect the levels of PBDEs in a fish.  相似文献

The purpose of this study was to determine the effects of perchlorate on thyroid function in mosquitofish. Adult mosquitofish were exposed to 0, 0.1, 1, 10, 100, and 1000 mg/L sodium perchlorate for 2, 10, and 30 d. Whole body thyroxin (T4) content and histological assessment of thyroid follicles (e.g., follicular epithelial height, hyperplasia, hypertrophy, and colloid depletion) were used to gauge alterations in thyroid function. Follicular epithelial cell height, hyperplasia, and hypertrophy increased with increasing perchlorate concentration, especially in fish exposed for 30 d, and these effects were statistically significantly different from control at concentrations as low as 0.1 mg/L (nominal concentration). The percent occurrence of follicles with depleted colloid decreased with increasing perchlorate concentration, which is contrary to what is expected with thyroid inhibition. There also was a decrease in whole body T4 concentration in fish exposed to perchlorate for 30 d, but clear dose-response relationships were less evident for whole body T4 than for histopathological endpoints. In conclusion, thyroid histopathology provides a sensitive biomarker for thyroid endocrine disruption at environmentally relevant concentrations of sodium perchlorate, and whole body T4 is a less sensitive indicator of perchlorate exposure than is histopathology.  相似文献

Novaluron, a novel chitin synthesis inhibitor (CSI), was tested against the various developmental stages of Tribolium castaneum (Herbst), at concentrations ranging from 0.1 to 100 ppm. It did not kill T. castaneum adults at these concentrations, but at 1.0 ppm it caused total mortality of third-instar larvae. Novaluron did not affect the number of eggs laid by T. castaneum adults that were exposed to treated wheat flour, but it totally inhibited their hatching after the third day of exposure at a concentration of 1 ppm. On the first day after infestation novaluron did not totally prevent hatching, even at the highest tested concentration of 100 ppm. The time needed to restore egg hatchability after adults were transferred to untreated flour depended on the concentration of novaluron used in the treatment. When the adults of T. castaneum were exposed to novaluron-treated whole wheat grains (at 1 ppm), similar effects to those of the treated flour at the same concentration were observed: egg hatching was drastically reduced. The effect of uptake via contact of adults with novaluron-treated surfaces was examined by exposing them to a mixture of untreated flour and 10% novaluron-treated sand at a concentration of 10 ppm, and only 1% of the eggs hatched after 18 days of exposure. When adults were removed from treated surfaces of Petri dishes to untreated flour, the hatching rate was again reduced by 100%. It was concluded that the contact penetration of novaluron into T. castaneum adults prevents the hatching of eggs subsequently laid.  相似文献

A rapid method for the determination of seven predominant polybrominated diphenyl ethers (PBDEs) in fish was developed. The analysis could be completed in 1 h. The PBDEs in the sample were extracted with fat by selective pressurized liquid extraction (SPLE) using acid alumina as a fat retainer. After concentration, the extract was directly injected into online gel permeation chromatography with gas chromatography-mass spectrometry (GPC-GC/MS) in negative chemical ionization (NCI) mode. Method detection limits of seven PBDEs were between 0.002 and 0.005 ng g?1 fat. Quantitative recoveries ranging from 81.9% to 110.3% were obtained by analysis of spiked fish. The relative standard deviations were less than 15% (n = 6). The method was applied to a certified reference material of fish; all results were in the reference ranges. It was shown to be a rapid and reliable alternative in the routine analysis for PBDEs.  相似文献

In this study, we used Callosobruchus maculatus as a model insect to assess the idea of whether or not environmental conditions experienced by parents subsequently moderate offspring fitness and competency. We observed that parents’ experiences (sex ratio of parents, restricted access to mates, and honey access) influenced their offspring’s features and did not significantly affect traits such as offspring sex ratio, larval duration, and longevity. However, not only offspring (F1) but also grand-offspring (F2) copulation behaviors, egg number, and hatching percentage were influenced by parents’ experiences. Hence, the offspring of parents living in unfavorable conditions had higher copulation success, higher male mating propensity, and a lower mating latency time in adulthood than the progeny of parents kept under natural or favorable conditions. Interestingly, the offspring of parents with favorable living conditions showed the least tendency to copulate immediately. Even grandparents’ experience could affect descendant behaviors and shorten copulation duration. Adjustments of the parents to their environment were inherited by offspring and grand-offspring to cope better with challenging circumstances and might make the organism more competent. Therefore, the parents’ living conditions have a remarkable impact on how their progeny behave in different situations.  相似文献

There is a pressing need for high throughput methods to assess potential effects of endocrine disrupting chemicals (EDCs). released into the environment. Currently our ability to identify effects in vitro exceeds that for in vivo monitoring. However, only in vivo analysis provides the full spectrum of physiological impacts exerted by a given chemical. With the aim of finding a physiological system compatible with automatic plate reading we tested the capacity of early embryonic stage Xenopus laevis tadpoles to monitor thyroid hormone (TH) disruption. Fluorescent transgenic X. laevis embryos bearing a TH/bZIP-eGFP construct, placed in 96 well plates, were used for a physiological-based screen for potential TH signaling disruptors. Using stage NF-45 embryos (time of thyroid gland formation) allowed rapid detection of chemical interference with both peripheral TR signaling and production of endogenous TH. Nanomolar concentrations of TH receptor agonists could be detected within 72 h. Moreover, when testing against a 5nM T3 challenge, the effects of inhibitors of TH production were revealed, including inhibitors of TH synthesis, (methimazole: 1 mM or sodium perchlorate: 3.56 microM), as well as antagonists acting at the receptor level (NH3: 2 microM) and a deiodinase inhibitor (iopanoic acid: 10 microM). Finally, we show that the thyroid disrupting activities of BPA (10 microM) and TBBPA (1 microM) can also be detected in this rapid screening protocol. Finally, this noninvasive technology using an automatic reading system shows low variability (around 5%) and permits detection of subtle changes in signaling by EDCs that either inhibit or activate TH signaling in vivo.  相似文献
