Chronic use of high oxytocin (OT) dosages can cause a reduced response to endogenous OT. In this study the OT dosages used in the milking practice of 82 dairy cow farms were recorded. The OT dosages per cow used were high, especially when injected i.m. (23 ± 2 IU) compared with i.v. (7 ± 1 IU). In addition, the minimum OT dosages needed to obtain normal milk removal in cows with disturbed milk ejection were investigated. Seventeen cows routinely treated with OT during milking (group T) and 17 cows without previous OT treatment were used (group C). After cessation of spontaneous milk flow, both T and C groups were injected i.v. with a low dosage of OT (0.2 or 0.5 IU/cow). The time from injection until cessation of the OT-induced milk flow was recorded (response phase). The response phase and the amounts of removed milk by effect of the OT injection increased with increasing OT dosage. Values for 0.2 and 0.5 IU/cow of OT injected i.v. were (response phase and amount of milk removed) 198 ± 27 and 302 ± 18 s and 3.4 ± 0.7 kg and 6.5 ± 1.3 kg, respectively, for the C group, and 157 ± 15 and 221 ± 16 s and 3.2 ± 0.5 and 5.5 ± 1.0 kg, respectively, for the T group. Within 20 min of the OT injection, plasma concentrations returned to basal levels. The threshold OT concentration at cessation of milk flow after injection of 0.2 or 0.5 IU/cow of OT was calculated based on the OT plasma half-life. The threshold increased with increasing dosages of OT and was higher in group T (8 ± 1 and 14 ± 1 pg/mL for 0.2 and 0.5 IU/cow, respectively) than in group C (7 ± 1 and 11 ± 1 pg/mL for 0.2 and 0.5 IU/cow, respectively). In conclusion, desensitization of the udder toward OT occurs when the udder is exposed to elevated OT plasma concentrations, both short-term during the actual milking and long-term due to chronic high-dosage OT treatment. However, low-dosage OT treatments to induce normal milk removal can minimize the observed side effects.  相似文献

Endosulphan residues were determined in milk samples collected from various locations of plains of Uttarakhand covering Tarai and Kumaon regions. Residues were extracted from milk by liquid–liquid partition followed by clean‐up by alumina column chromatography. High‐performance liquid chromatography (HPLC) was used for the detection and quantification of residues. Of the total 170 milk samples collected from different species, 1.17% samples showed residues of endosulphan‐alpha; 2.35% endosulphan β and 4.7% milk samples showed endosulphan‐sulphate residues with mean residual concentrations of 0.244, 0.566 and 0.265 μg/mL, respectively. About 6.47% of milk samples showed endosulphan residues above the maximum residue limit (MRL) of 0.1 mg/kg.  相似文献

In experiment I, the effect of i.m. oxytocin (OT) injection (50 IU) on OT blood pattern was tested. Blood samples from 6 cows were collected for 2 h after OT injection either with or without milking. To test the effect of i.m. OT injection (50 IU) on milk ejection efficiency, intramammary pressure (IMP) was measured in 13 cows (experiment II). Milk ejection was induced by manual teat stimulation. After IMP increased and reached a plateau, OT was injected. In experiment III, the effect of chronic OT treatment on mammary gland sensitivity and endogenous OT release was tested. For 19 d, cows were i.m. injected at each milking with 50 IU OT (n = 13) or 5 mL of NaCl 0.9% (n = 14) 1 min before the start of udder preparation. To test mammary gland sensitivity, IMP recording was performed after a long (11 h, 7 OT cows, and 7 NaCl cows) and a short (3 h, 6 OT cows, and 7 NaCl cows) milking interval at d -1 and d 18. To test the effect of withdrawal of chronic i.m. treatment on OT release, blood sample collection was performed during evening milking at d 0 and d 19. Intramuscular oxytocin injection (experiment I) caused elevated OT blood levels observed at least for 2 h and showed an even more pronounced effect when milking was also performed. Intramuscular OT injection after teat stimulation (experiment II) caused additional milk ejection but only in 6 out of 13 cows. Withdrawal of chronic OT treatment (experiment III) did not reduce OT release during milking. However, ejection time was prolonged during OT infusion after a long milking interval. Ejection pressure tended to be lower after a short milking interval. It seems that the reduction of spontaneous milk removal after chronic OT treatment was due to reduced contractibility of myoepithelial cells in the mammary gland at a physiological range of OT concentrations.  相似文献

In this survey, 150 samples of raw milk, white cheese and ice cream from three different dairy‐processing plants in Ankara were analysed to find out if they were contaminated with Escherichia coli, Staphylococcus aureus or Bacillus cereus. The highest contamination percentages were found in raw milk samples as follows: B. cereus (90%), E. coli (74%) and S. aureus (56%) followed by cheese (70% B. cereus, 60% E. coli, and 48% S. aureus) and ice cream (56% E. coli, 36% S. aureus and 20% B. cereus). The survey showed that 2% of cheese samples were contaminated with E. coli O157. It was also found that the numbers of S. aureus and E. coli in raw milk, cheese and ice cream samples exceeded the numbers permitted under the Turkish Food Codex (TFC). The number of B. cereus in raw milk, cheese and ice cream samples was lower than the limit given in the TFC standards. The study also showed that E. coli and S. aureus exhibit resistance to ampicillin, penicillin, tetracycline, erythromycin, gentamicin and trimethoprim/sulfamethoxazole. Escherichia coli isolates also showed resistance to chloramphenicol and ciprofloxacin but none of them exhibited resistance to cefotaxime. All S. aureus isolates were found to be susceptible to cefotaxime, chloramphenicol, and ciprofloxacin. Bacillus cereus isolates were found to be resistant to ampicillin, penicillin and trimethoprim/sulfamethoxazole and sensitive to cefotaxime, chloramphenicol, ciprofloxacin erythromycin, gentamicin and tetracycline.  相似文献

The purpose of this study was to evaluate the incidence and characterisation of aerobic spore‐forming bacteria originating from dairy milk in Tunisia. The distribution of Bacillus species in raw milk, pasteurised milk and UHT milk were 47.5%, 27.5% and 25%, respectively. Seven Bacillus species, including Bacillus pumilus (10%), Bacillus subtilis (12.5%), Brevibacillus brevis (10%), Bacillus cereus (22.5%), Bacillus sphaericus (7.5%), Bacillus licheniformis (12.5%) and Bacillus sporothermodurans (25%) were identified in different milk samples. Bacillus cereus was predominant in raw and pasteurised milk. Although B. sporothermodurans was the predominant sporogenous micro‐organism in UHT milk, B. cereus, B. sphaericus and B. licheniformis were also present. This study showed that there is a high degree of diversity, both phenotypic and genotypic, among Bacillus isolates from Tunisian milk and the persistence of spoilage risk in UHT milk.  相似文献

This study aims to detect aflatoxins (AFs) in dairy cow feed, milk and milk products using a high-performance liquid chromatography coupled with fluorescence detection (HPLC-FLD) method. All the validation parameters met the method performance criteria of the European Union. The samples comprised 76 dairy cow feeds and 205 milk and milk products (including yoghurt and yoghurt-based beverage, ayran). AFs were present in 26.3% of the feed samples. Two feed samples exceeded the maximum limit (ML) of 5 µg kg?1 for AFB1 as established by the EU. Nineteen milk samples (21.1%) contained aflatoxin M1 (AFM1) of which three exceeded the EU ML of 0.05 µg l?1. In addition, only two yoghurt samples and one ayran sample contained AFM1, but the levels were lower than the EU ML.  相似文献

The current study was based on evaluation of the therapeutic potential (antimicrobial and antidiabetic) of a synbiotic fermented milk product developed by optimising levels of the prebiotic ingredients (fructooligosaccharide and isomaltooligosaccharide). The maximum growth rate (0.27 ± 0.21a cfu/mL) of Lactobacillus acidophilus ATCC® 4357? was observed with 2.345–2.445% of fructooligosaccharide and 2.53–2.62% of isomaltooligosaccharide. Significant antimicrobial potential of the synbiotic fermented milk was observed against Escherichia coli and Staphylococcus aureus due to the addition of prebiotics. Similarly, the final product also showed 62.9%, 71.5% and 57.0% reduction in blood glucose, urea and creatinine levels, respectively, of diabetic rabbits, when supplemented with 6% of the synbiotic fermented milk. A synbiotic fermented milk product with significant therapeutic potential could be developed using fructooligosaccharide and isomaltooligosaccharide as prebiotic ingredients.  相似文献

Ewe milk samples from different ovine dairy farms from the Castilla-La Mancha region of Spain were collected from bulk tanks to estimate the occurrence of antibiotic residues in raw and heated (82 degrees C, 10 min) milk by the Delvotest SP test. The month of collection, somatic cell counts, and bacteriology were analyzed and examined by means of a logistic regression model. The screening of a total of 2686 raw milk samples showed 1.7% "positive" and 2.1% "doubtful" results, which decreased after heating treatment to 1.3% and 0.4%, respectively. "Positive" and "doubtful" samples were identified by penicillinase and p-aminobenzoic acid solutions, and the majority of them corresponded to antimicrobials different than beta-lactams or sulfonamides. By applying a logistic regression model, a significant effect of month of collection and bacteriology was observed in the initial screening and after the heat treatment. The highest percentages of "positive plus doubtful" results were observed in late summer-early autumn. A slight peak was also observed in spring in raw milk samples. Bacteriology was positively correlated with "positive plus doubtful" results.  相似文献

To improve the detection of ovulation and early diagnosis of pregnancy on dairy farms, a disposable electrochemical immunosensor was developed using indirect competitive immunoassays and screen‐printed carbon electrodes. After optimisation of the assay conditions, the linear range was from 0.16 to 50 ng/mL. The limit of detection was 0.16 ng/mL. There was no cross‐reactivity with testosterone, 17‐β‐estradiol and estriol. The intra‐assay coefficient of variation was less than 13% and interassay coefficient of variation was < 9%. The percentage recovery was from 95.8% to 115.7%. Stability lasts 4 days at 4 °C. The results of preliminary application trials indicate that the method could have practical potential.  相似文献

建立了乳与乳制品中双氰胺残留的高效液相色谱测定方法.采用水-乙腈提取样品、氨基色谱柱分离、紫外检测器检测、外标法定量测定双氰胺残留.该方法的检出限为牛奶0.01mg/kg,乳粉为0.05 mg/kg(均为质量分数),线性范围为0.25.0 mg/L,并确定了回归方程,相关系-R2=0.9998,样品加标回收率在90%~8%,相对标准偏差为1.2%~2.3%.该法具有样品处理简单方便,灵敏度高,分析时间短等优点,可以满足乳与乳制品中双氰胺残留的测定.  相似文献

Four different methods of teat preparation during milking in an automatic milking system were studied in 2 experiments on Red Holstein/German Fleckvieh cross-breed cows. Milking routines used were milking: 1) without premilking teat preparation; 2) with one cleaning cycle (58 to 60 s) with cold (13 to 15 degrees C) water; 3) with one cleaning cycle with warm water (30 to 32 degrees C); or 4) with 2 cleaning cycles (122 s) with warm water. In experiment 1, milking characteristics were evaluated and milking routines were randomly assigned to 62 cows during 3 measuring periods of 24 h each. In experiment 2, 10 randomly selected cows were assigned to the same milking routines during 4 d and blood samples for oxytocin (OT) determination were taken during milking in addition to milk flow recording. Milk production, peak flow rate, total, and quarter milk yields showed no differences among treatments. Premilking preparation with cold water compared with warm water showed no differences in OT release, milk yield, peak flow rate, main milking time, average flow rate, or time until main milk flow. Baseline OT concentrations were consistently low. At the start of teat cup attachment without premilking teat preparation OT concentrations remained on the basal level but were elevated in all other treatments. By 30 s from the start of milking, OT concentrations were markedly increased in all treatments and were no longer different between treatments. In conclusion, the teat cleaning device used in the automatic milking system, either with warm or cold water, was suitable to induce milk ejection in cows before the start of milking.  相似文献

The incidence of allergy to seafood, and in particular to molluscs is second only to that of nuts. To protect consumers, the regulators of food products insisted on identifying molluscs as allergens. The aim was to develop quantitative assay for the presence Mytilus species in processed food products. The chosen platform was real‐time PCR (qPCR) targeting either the gene encoding mitochondrial cytochrome C oxidase I or the nuclear gene encoding β‐actin. Recombinant plasmids containing each of target regions were used as a reference for quantification purposes. Limit of detection (LOD) and of quantification (LOQ) were determined. Spiked food samples containing 50–500 μg g?1 of Mytilus chilensis were analysed both by qPCR and by ELISA. The former assay gave a positive outcome over this range, whereas the latter was sensitive down to a concentration of 125 μg g?1.  相似文献

In this study, some probiotic characteristics such as antimicrobial activity, vancomycin resistance, growth ability at pH, resistance to bile salts, bile salt deconjugation and hydrophobicity of 30 Enterococcus faecium and Enterococcus durans strains isolated and identified from raw milk and various dairy products were investigated. According to the study results, antimicrobial activity profiling, pH and bile salt resistance and bile salt deconjugation ability of Enterococcus strains varied depending on the species and strains and all the strains showed resistance to the tested bile salt concentrations. It was concluded that the E. faecium and E. durans strains tested showed probiotic characteristics and have the potential to be used in food production.  相似文献

Waste milk (WM) is a common source of feed for preweaned calves in US dairy farms. However, limited information is available about characteristics of this product, including concentration of drug residues and potential hazards from antibiotic-resistant bacteria present in the milk. The aims of this cross-sectional study were to (1) identify and measure the concentration of antimicrobial residues in raw WM samples on dairy farms in the Central Valley of California, (2) survey farm management practices for factors associated with the occurrence of specific antimicrobial residues in raw WM, (3) characterize the antimicrobial resistance patterns of E. coli cultured from raw WM samples, and (4) evaluate the potential association between WM quality parameter and risk of identifying drug residues in milk. A single raw bulk tank WM sample was collected from dairy farms located in California's Central Valley (n = 25). A questionnaire was used to collect information about farm management practices. Waste milk samples were analyzed for a multidrug residue panel using liquid chromatography–tandem mass spectrometry. Bacteria were cultured and antimicrobial resistance was tested using standard techniques; milk quality parameters (fat, protein, lactose, solids-not-fat, somatic cell count, coliform count, and standard plate count) were also measured. Of the 25 samples collected, 15 (60%) contained detectable concentrations of at least 1 antimicrobial. Of the drug residue–positive samples, 44% (11/25) and 16% (4/25) had detectable concentrations of β-lactams and tetracycline, respectively. The most prevalent drug residues were ceftiofur (n = 7, 28%), oxytetracycline (n = 4, 16%), and cephapirin (n = 3, 12%). No significant associations were identified between farm characteristics or management practices and presence of drug residues in WM. In this study, 20% of farms did not pasteurize WM before feeding to calves. Two of the 10 Escherichia coli isolated from WM samples were multidrug resistant. Streptococcus spp. (n = 21, 84%) was the most common genus cultured from WM samples, followed by Staphylococcus spp. (n = 20, 80%) and E.coli (n = 10, 40%). Mycoplasma spp. was cultured from 2 WM samples (n = 2, 8%). The presence of drug residues in WM at concentrations that increase selection of resistant bacteria indicates the need for additional studies targeting on-farm milk treatments to degrade drug residues before feeding to calves. The presence of multidrug-resistant E. coli in WM urges the need for on-farm practices that reduce calf exposure to resistant bacteria, such as pasteurization.  相似文献

The formation and dispersion of biofilms in the dairy industry is a problem because it increases cross‐contamination, affecting the shelf life of the products and their safety. The objective of this study was to evaluate the influence of different dairy substrates (cows’ milk and whey protein) on the formation and dispersion of Enterococcus faecalis, Listeria monocytogenes, Staphylococcus aureus and Bacillus cereus in two biofilm systems (mono‐species and multi‐species) on stainless steel at 25 °C. The dominant behaviour of E. faecalis occurred in most of the tests on mono‐species and multi‐species biofilms. A greater dispersion of biofilm cells was observed in skimmed milk.  相似文献

During 2016–2017, 156 samples of fresh milk samples were collected from local markets of Karachi, Pakistan and analysed for aflatoxin M1 (AFM1) contamination using ELISA technique. AFM1 was detected in 143 (91.7%) samples, ranged from 20 to 3090 ng L?1 with a mean level of 346.2 ng L?1. In 125 (80.1%) samples, the AFM1 contamination was greater than the maximum limit (ML = 50 ng L?1) set by EU. However, in 51 (32.7%) samples, the AFM1 level was higher than the ML of 500 ng L?1 as assigned by the USA. Statistical analysis showed that the AFM1 level in milk samples from summer was significantly (p < 0.05) higher than that obtained in winter. It was concluded that the AFM1 levels in the tested samples appear to be a serious public health problem. Therefore, immediate measures should be taken and re-evaluation done for the procedures for farming, transportation, refrigeration, and storage for the control of AFM1 level in milk samples.  相似文献

A study of two adjacent dairy farms, one using conventional confined herd management and the other organic management, revealed significant differences in the fatty acid composition of the milk. Compared with conventional milk, organic milk had higher levels of conjugated linoleic acid (CLA) and α‐linolenic acid (the major omega‐3 fatty acid in milk), and less stearic and linoleic acid (the major omega‐6 fatty acid in milk) during the spring–summer grazing season. When discarding geography and weather as variables, organic milk appears to yield more CLA and α‐linolenic acid, which should be beneficial to health.  相似文献

建立超高效液相色谱-串联质谱(UPLC-MS/MS)法测定乳与乳制品中氯霉素、甲砜霉素和氟苯尼考的检测方法。样品经乙腈及乙酸乙酯提取、正己烷脱脂、C18柱净化后,通过超高效液相色谱-质谱联用仪进行检测,内标法定量。各标准溶液在0.1~10μg/L范围内呈线性相关(r=0.999),不同基质样品中添加回收率为75%~114%,检出限为0.1μg/kg。结果表明,该方法是一种准确测定乳与乳制品中氯霉素、甲砜霉素和氟苯尼考的检测方法。  相似文献

5-Hydroxy-flunixin concentrations in milk samples were quantified by two commercially available screening assays – CHARM® and enzyme-linked immunoabsorbant assay (ELISA) – to determine whether any concentrations could be detected above the tolerance limit of 2 ng g?1 from different regions in the United States. Milk samples came from large tanker trucks hauling milk to processing plants, and had already been screened for antibiotics. Positive results for flunixin residues based on a screening assay were confirmed by ultra-HPLC with mass spectrometric detection. Of the 500 milk samples analysed in this study, one sample was found to have a 5-hydroxy-flunixin concentration greater than the tolerance limit. The results of this study indicate that flunixin residues in milk are possible. Regulatory agencies should be aware that such residues can occur, and should consider incorporating or expanding flunixin screening tests as part of routine drug monitoring in milk. Larger studies are needed to determine the true prevalence of flunixin residues in milk from other regions in the United States as well as different countries.  相似文献
