Soy protein consumption has been linked to reduction in hypercholesterolemia, a risk for coronary heart disease. However, to what extent soy protein itself or its non-nutritive components, e.g., isoflavones and saponins, exert this cholesterol-lowering effect requires further investigation. To evaluate the effect of the protein component alone on lipid variables, ethanol-extracted, isoflavone-depleted soy protein isolate (SPe) was studied in ovarian hormone-deficient hamsters. Forty-eight 6-month-old female Golden Syrian hamsters were either sham-operated or ovariectomized and fed casein-based or SPe-based diets for 70 d. Ovariectomy, but not protein source, significantly (P < 0.05) increased serum phospholipids and total, non-high density lipoprotein, free and esterified cholesterol concentrations. Serum HDL cholesterol concentrations were not altered with either treatment. No significant differences were observed in liver total lipids or liver total cholesterol among the groups. Soy protein isolate, however, lowered serum triglyceride concentrations in both sham-operated and ovariectomized hamsters. These findings confirm the ovariectomized hamster as a model of postmenopausal hypercholesterolemia. The results are consistent with earlier observations that isoflavones or other nonprotein components, perhaps in combination with soy protein, play an important role in exerting this hypocholesterolemic effect. Further studies are needed to investigate whether isolated nonprotein components of soy would be able to prevent the ovarian hormone deficiency-associated rise in serum cholesterol regardless of dietary protein source.  相似文献

The effect of experimental overnutrition via high-protein cafeteria feeding on atherogenesis and adiposity were determined in groups of congenic adult lean and corpulent LA/N-cp (corpulent) rats derived from the Koletsky strain, and fed Purina Laboratory Chow or a high-protein cafeteria diet consisting of the Purina Chow plus cooked eggs and red meat for 8 weeks. High protein cafeteria feeding resulted in more marked increases in body weight and adiposity in corpulent than in lean rats. Oral glucose tolerance was similarly impaired in corpulent rats before and after cafeteria feeding. Serum cholesterol concentrations were greater in corpulent than in lean rats initially, and increased 2-fold following the cafeteria feeding. In contrast, HDL cholesterol increased proportionately to the total cholesterol concentrations in cafeteria fed lean but not corpulent rats, resulting in highly significant increases in the VLDL+LDL: HDL and Total: HDL cholesterol ratios in the cafeteria-fed corpulent group. These results indicate that the corpulent phenotype of the LA/N-cp rat strain may be predisposed to atherogenesis in association with an exaggeration of their obesity when fed atherogenic high-animal-protein cafeteria diets for a prolonged period of time.  相似文献

The effects of supplementation of methionine to a 20% soy protein isolate diet on serum level of cholesterol. 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase activity, cholesterol 7 alpha-hydroxylase activity, and biliary and fecal steroids in rats with or without receiving polychlorinated biphenyls (PCB) were investigated. Supplementation of methionine and PCB did not affect the growth. Serum level of cholesterol was higher in rats fed PCB than in controls. In rats fed PCB, addition of methionine elevated serum level of cholesterol synergistically. The activity of HMG-CoA reductase was higher in rats fed the methionine-supplemented diet than in those fed the unsupplemented diet when PCB was included in the diets. Cholesterol 7 alpha-hydroxylase activity (nmol/h.100 g body weight) was higher in rats fed PCB than in controls. Biliary secretion of bile acids was higher in rats fed PCB than in controls. On the other hand, fecal excretion of bile acids decreased in PCB-treated rats, but total steroids were not affected by PCB. In rats fed PCB, the addition of methionine did not alter cholesterol 7 alpha-hydroxylase activity and biliary and fecal steroid output. The data suggest that the increase in serum level of cholesterol due to dietary addition of methionine together with PCB would be mediated through the stimulation of hepatic synthesis of cholesterol.  相似文献

目的观察大豆异黄酮对高脂模型大鼠血清胆固醇浓度及其肝脏胆固醇代谢关键酶基因表达的影响。方法 9周龄SD雄性大鼠8只喂饲AIN93M饲料(阴性对照组),18只喂饲高脂饲料建立高胆固醇模型。14 d后将高胆固醇大鼠随机分为异黄酮组(每天灌胃360 mg/kg BW大豆异黄酮)和高脂对照组,喂饲高脂饲料至实验结束。30 d后测血脂,实时定量聚合酶链式反应(polymerase chain reaction,PCR)测定肝脏羟甲戊二酰辅酶A还原酶(3-hydroxy-3-methylglutaryl-Coenzyme A reductase,Hmgcr)、胆固醇7α-羟化酶(cytochrome P450,family 7,subfamily a,polypeptide 1,Cyp7a1)、低密度脂蛋白受体(low density lipoprotein receptor,Ldlr)mRNA表达量。结果异黄酮组大鼠血清总胆固醇(total cholesterol,TC)和低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDLC)浓度分别比高脂对照组降低了23.7%(q=2.79,P=0.010)和23.2%(q=2.63,P=0.015);动物肝脏的Hmgcr和Cyp7a1 mRNA表达量均出现下降,但差异无统计学意义(均有P〉0.05)。结论大豆异黄酮可明显降低高胆固醇大鼠血清TC和LDLC浓度,有可能影响肝脏胆固醇代谢。  相似文献

The effects of supplementing 8% casein or 10% soy protein isolate (SPI) diets with graded levels of oligo-L-methionine (a mixture of hexa- and heptapeptides, OM) or L-methionine (Met) were studied in rats to determine the reason for the difference in nutritional quality between proteins and corresponding amino acid mixtures. As the OM concentration of the casein-based diet was increased from 0.02% to 0.6%, maximum weight gain was attained at 0.2%, and the growth-promoting activity of OM was comparable to Met at all the corresponding levels tested. Liver fat began to accumulate when supplemental Met reached a level of 0.08% of the casein diet, but OM addition did not produce a fatty liver at dietary levels of less than 0.3%. When SPI was used as the dietary protein source, the effect of supplemental OM was significantly less than that of Met. Digestibility of OM (assessed by incremental portal plasma Met concentration) was measured 30 min after feeding the casein or SPI diet supplemented with 3% OM using rats fasted for 24 h. Plasma Met concentration was greatly increased in rats fed the casein plus OM diet compared with that of rats fed the SPI + OM diet. Similarly, the 30-min portal Met concentration significantly increased in response to the casein + OM diet compared with the SPI + OM diet regardless of the prefed proteins (25% casein and 25% SPI for 2 wk).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

This study was conducted to investigate the effects of a buckwheat protein product (BWP) on plasma cholesterol, gallbladder bile composition and fecal steroid excretion in hamsters fed diets with 5 g/kg cholesterol. Diets also contained 200 g/kg of casein, soy protein isolate (SPI) or BWP as protein sources. After 2 wk, plasma and liver concentrations of cholesterol in the hamsters fed BWP were significantly lower than those in the hamsters fed casein and SPI. Rats were fed purified diets which provided 24% protein from casein (C), soy protein isolate (SPI), or egg white (EW) for 18 mo. Groups of rats were killed at 3, 6, 12 and 18 mo; the pancreata were removed and examined histologically for occurrence of atypical nodules. The weight, protein, DNA, trypsin and chymotrypsin concentrations of the pancreas at each period were measured. Over the entire experimental period, body weight did not differ among groups. Pancreatic weight, protein and trypsin activity were highest in the EW group, followed by the SPI group, and lowest in the C group. Chymotrypsin activity was significantly higher in the EW and SPI groups than the C group. DNA content did not differ significantly among groups over the entire experimental period, although it was elevated in the SPI or EW groups compared to the C groups at some of the time periods. Only one microscopic nodule was observed in all of the animals; it was found at 3 mo in the pancreas from an animal fed EW. Overall, the results suggest that the elevation in enzyme activity and pancreatic weight associated with long-term consumption of EW and SPI did not result in development of pancreatic lesions in rats.  相似文献

Diseases and injuries impose oxidative stress on different organs and systems of the body. This study was conducted to investigate the hepatoprotective effects of soy protein isolate against acute liver toxicity induced by dimethylnitrosamine (DMNA). Forty-eight Sprague Dawley rats were randomly divided into three groups. Groups A and B consumed a casein containing diet and the Group C received a soy protein isolate (SPI) containing diet for 18 days. Group A was then given an intra-peritoneal saline injection and continued on a casein diet for another 4 days before being killed. Each animal from groups B and C was given a single intra-peritoneal injection of DMNA (30 mg/kg) on the 18th day of the study. All groups continued their diets for 4 days before their sacrifice. The serum ALT decreased and albumin increased significantly in rats fed 20 % SPI containing diet (P < 0.05). Histological results showed that SPI improved DMNA-induced alteration in the liver structure. Morphological and biochemical data suggest that soy protein isolate containing diet decreased DMNA-induced liver damage.  相似文献

The effect of L-methionine supplementation on the utilization of a soy protein isolate (SPI) was evaluated by short-term nitrogen balance studies in young women. Thirteen female students were given SPI in an initial period and SPI supplemented with 1% methionine in a second period immediately after menstruation as the sole source of protein. After one day on protein-free diet, each subject received conventional low-protein diet for three days, and then low protein, semisynthetic diet containing 0.5 g/kg/day (seven subjects) or 0.3 g/kg/day (six subjects) of SPI or SPI supplemented with methionine for seven days. The energy intake was approximately of a maintenance level of 36.5 +/- 3.8 kcal/kg/day. The mean N balances of the subjects at an intake level of 0.5 g/kg/day in the SPI period and methionine supplemented period were -6.2 +/- 12.6 mg N/kg and -9.8 +/- 9.8 mg N/kg, respectively, while their N balances at an intake level of 0.3 g/kg/day were -17.8 +/- 7.2 mg N/kg in the SPI period and -15.5 +/- 3.0 mg N/kg in the methionine supplemented period. There was no significant difference between the values in the SPI and methionine supplemented periods at both levels of protein intake. Blood analyses were carried out before and after the SPI period and after the period of methionine supplementation. The urinary creatinine and urea excretions during these periods were not markedly affected.  相似文献

OBJECTIVE: This study examined the effects of a ratio of soy protein to animal protein on bone metabolism of rats. METHODS: Experimental groups were a high soy protein group (200 g of soy protein and 0 g of casein per kilogram of diet; HSoy), a middle soy protein group (100 g of soy protein and 100 g of casein per kilogram of diet; MSoy), a low soy protein group (50 g of soy protein and 150 g of casein per kilogram of diet; LSoy), and a no soy protein group (0 g of soy protein and 200 g of casein per kilogram of diet; NSoy). Calcium excretion and retention, biochemical parametrically related calcium metabolism, and bone mineral density were measured. Statistical analysis was performed with SAS software. RESULTS: Urinary excretion of calcium was significantly high in the LSoy and NSoy groups, and there was no difference in absorption of calcium across experimental groups. Calcium retention was significantly higher in the HSoy and MSoy groups than in the LSoy and NSoy groups. Experimental groups showed no differences in the activity of alkaline phosphatase. The casein group (NSoy) showed a remarkably lower degree of serum osteocalcin concentration. The concentration of deoxypyridinoline in urine showed an increasing tendency, i.e., HSoy and MSoy < LSoy < NSoy, indicating that at a higher degree of animal protein/soy proteins, its concentration increases. Wet weight of the femur appeared to be significantly greater in the MSoy and LSoy groups than in the NSoy group. Ash content of the femur and bone density were highest in the MSoy group, with the ratio of 1:1 between soy protein and animal protein. CONCLUSIONS: This study indicated that deoxypyridinoline concentration was lower and the density of osteocalcin was higher in the MSoy group than in the NSoy group, and that calcium retention was high and bone mineral density was the highest in the MSoy group. The ratio of soy to animal protein that seemed to have the most positively significant effect on calcium metabolism was 1:1.  相似文献

Utilization and requirement of soy protein isolate (SPI) and SPI-rice combination were examined in twenty-five female students. After 1 day on protein-free diet, each subject received a low-protein diet for 10 days. The protein sources were SPI for ten subjects and SPI-rice combination, in which the ratio of two proteins was 6:4, for fifteen subjects. The nitrogen intakes were about 45, 65 and 85 mg/kg in both the two series of experiments. Energy intake was at an approximate maintenance level of 36.1 +/- 3.0 kcal/kg. Apparent nitrogen balance improved with increase in nitrogen intake. The regression equations between nitrogen intake (X, mg/kg) and balance (Y, mg/kg) are shown in the following: SPI: Y = 0.411 X - 40.8 (n = 10, r = 0.812) SPI and rice protein: Y = 0.392 X -32.7 (n = 15, r = 0.739) From the above equations, the maintenance intakes of SPI and SPI-rice combination for an apparent nitrogen equilibrium were calculated to be 99 and 83 mg N/kg, respectively. Digestibilities were 98.2 +/- 5.0% for SPI and 93.1 +/- 6.1% for SPI-rice combination. The NPUs of SPI at intake levels of 40, 60 and 80 mg N/kg were 47 +/- 24 (n = 4), 49 (n = 2) and 44 +/- 3 (n = 4), respectively. The NPUs of SPI and rice mixed protein at intake levels of 45, 70 and 90 mg N/kg were 67 +/- 13 (n = 5), 51 +/- 7 (n = 5) and 54 +/- 12 (n = 5), respectively. It was concluded from the present study that both SPI and the SPI-rice combination had a high nutritive efficiency comparable with that of egg protein.  相似文献

ObjectiveThe aim of the present study was to evaluate the effect of a soy-protein diet on plasma lipid levels of renal transplant recipients with moderate hypercholesterolemia.DesignDietary intervention case-control observational study.SettingRenal transplantation outpatient clinic.PatientsFifteen stable patients who had renal transplantation (serum creatinine < 2 mg/dL) with moderate hypercholesterolemia (low-density lipoprotein [LDL] cholesterol > 140 mg/dL).InterventionAfter a baseline dietary interview, dietary counseling was given individually with the goal of substituting 25 g of animal protein with 25 g of soy protein for a 5-week period, using commercially available soy foods, according to each patient’s own preference.Main outcome measuresBefore and after the soy-diet period, plasma lipid profiles including total, LDL, and high-density lipoprotein (HDL) cholesterol, triglycerides, apolipoprotein A1 and B were determined. Protein catabolic rate was assumed as a measure of dietary protein intake.ResultsTwo patients dropped out. After the soy diet, total cholesterol (254 ± 22 to 231 ± 31 mg/dL, P < .05) and LDL cholesterol (165 ± 20 versus 143 ± 20 mg/dL, P < .01) decreased significantly. No significant changes were observed regarding HDL cholesterol and triglycerides. Dietary protein intake did not differ at baseline (73.2 ± 22.9 g/day) and during the soy diet (72.6 ± 15.6 g/day), when the reported actual soy protein intake resulted 26 ± 8 g/day.ConclusionsThis study shows that soy proteins given as part of the daily protein intake have beneficial effects on serum LDL cholesterol levels of renal transplant recipients with moderate hypercholesterolemia. The influence of soy protein isolate (SPI) in the diet on whole-body retention of extrinsically radiolabeled iron from test meals containing or not containing SPI was evaluated in marginally iron-deficient weanling rats. In experiment 1 SPI was compared with casein in a 2 X 2 factorial design: diets and test meals were either SPI-based or casein-based. Diets were fed for 13 days prior to the test meal and for 7 days subsequent to the test meal. Rats fed the SPI-based diet retained less iron from test meals than did rats fed the casein-based diet (66.1 vs. 74.8%, P less than 0.01). Experiment 2 showed that an SPI-based diet fed during the final 4 days of a 14-day pre-test meal period and subsequent to the test meal led to less iron retention compared to a casein-based diet. In addition to the observed diet effect, experiment 1 showed that iron retention was less from an SPI-based test meal than from a casein-based test meal, confirming previous reports of adverse effects of SPI on iron retention. The present experiments show that SPI can adversely affect from retention in two ways: by its presence in the diet before and after a test meal, and by its presence in a test meal.  相似文献
