本文以沙田柚为材料,对其染色体组型及带型进行了观察分析。组型分析:染色体数目2n=18,根据染色体的相对长度分成大小染色体两种类型,前者包括1、2、3、4和5对,后者为6、7、8和9对,根据臂比,9对染色体能够被分成中部着丝点和近中着丝点染色体两种类型。即第5、7,9对为亚中部着丝点,其余为中部着丝点,第6对染色体上有随体;Giemsa带型:除第二对染色体只显中间带外,其余都显着丝点带,并在3、4、8对染色体短臂上和2、3、1对染色体长臂上均显端带,第2、3,6对同源染色体之间的C带显示杂合性。  相似文献

1.东方蝾螈(Cynops orientalis)、肥螈(Pachytriton brevipes)染色体数均为2n=24。按染色体相对长度、臂比、着丝点指数可将12对染色体分成三组。染色体1—4为第一组,均为中部着丝点染色体。染色体5—8为第二组,除染色体7为亚中部着丝点外,其余为中部着丝点染色体。染色体9—12为第三组,除染色体9为中部着丝点外,其余都为亚中部着丝点。东方蝾螈和肥螈染色体核型相似,但却各具独特的C-带带谱图式。 2.经间接免疫过氧化物酶染色后,在东方蝾螈和肥螈肠上皮细胞染色体的着丝点两侧和长、短臂的端粒部位都显示棕褐色的5-甲基胞嘧啶特异位置的深着色,染色体的长臂、短臂上则为浅棕色。 3.东方蝾螈和肥螈雌雄个体肠上皮细胞的染色体核型中均未见染色体的异型性配对。  相似文献

本文研究了八角莲Dysosma versipellis(Hance)M.Cheng和六角莲Dysomapleiantha((Hance)Woodson的核型。二者的染色体数目均为2n=12,由四对具中部着丝点染色体、一对具近中部着丝点染色体和一对具端部着丝点染色体组成,各有一对染色体具有次缢痕,八角莲的次缢痕在第3对染色体的长臂上,六角莲的次缢痕在第1对染色体的短臂上。二者均属较对称的“2A”核型。但它们在染色体相对长度的变异幅度和差值、臂比的变异幅度和差值以及最长与最短染色体的比值上均有微小的差异。结果表明二者有密切的亲缘关系。演化趋势是八角莲→六角莲。八角莲的核型为首次报道。  相似文献

采用常规的白细胞培养技术及BSGC-带技术分析了白豚的核型、C-带核型。结果是:白豚的染色体数目2n=44,其核型由12条中部着丝点染色体、18条亚中部着丝点染色体、4条亚端部着丝点染色体、8条端部着丝点染色体和2条性染色体所组成。染色体臂数(NF)雌性为76,雄性为75。C-带异染色质呈现出很深的着色区,主要分布在染色体臂上,着丝点区则几乎没有。C-带异染色质的量约为总染色质量的12%。这一结果表明白豚与海生豚类的核型、C-带核型有较明显的相似性。  相似文献

救荒野豌豆(Vicia sativa L.)是豆科野豌豆属植物,是一种优良的绿肥和饲料。根尖用0.005%秋水仙素预处理2小时,改良苯酚品红压片,供核型分析。分带用去壁低渗法制片,BSG技术处理。结果如下:染色体数2n=12。染色体的相对长度在10.64%—21.13%之间。臂比值在1.55—2.88之间。第1和3对为中部着丝点染色体,第2、4、5和6对为次中部着丝点染色体,第5对具随体。其核型公式为2n=12=4m+8sm(2SAT)。带型分析结果如下:第1对染色体具着丝点带和长、短臂上具末端带,第2对染色体除具着丝点带和长、短臂上末端带外,在长臂上还具中间带,第3对带型与第1对相同,第4、6对染色体长、短臂上都具末端带,第5对具着丝点带、末端带和次缢痕带。其带型公式为2n=12=4CT+2CTI_++4T+2CTN。与朱凤绥等人的报道结果有差异。  相似文献

本文研究了我国不同地区韭菜三个品种(尸州韭、青岛韭、银川韭)的核型。结果表明:三个品种的核型均为同源四倍体,其核型公式为2n=4x=32=28m+4st(SAT),第1—7号为具中部着丝点染色体,第8号为近端着丝点染色体并带有随体,但其中有二个染色体带有明显的随体,另二个染色体所带随体较难以分辨。三个不同品种之间,染色体全组总的长度、染色体相对长度、最长和最短的变异范围、臂比范围都存在一些差异,但并不显著。本实验不同品种韭菜的核型研究结果与文献记载相符,不同者为第8对带随体染色体为具中部着丝点。而有的作者测定为近端着丝点染色体。此外,对于带随体染色体类型的不同与随体显现的程度进行了初步讨论。  相似文献

彭泽鲫是一种两性型天然雌核发育群体。染色体组型分析结果表明:雌、雄鱼的染色体数目均为166。它们的核型组成是:32条中部着丝点染色体、40条亚中部着丝点染色体、18条亚端部着丝点染色体和76条端部着丝点染色体。臂数(NF)为238。本研究结果与其它雌核发育类型的鲫鱼有明显的差别。  相似文献

利用骨髓细胞蒸汽固定法制备染色体标本,研究了分布于河南新乡的花背蟾蜍(Bufo raddei)的核型、C-带和Ag-NORs。结果表明:河南产花背蟾蜍体细胞染色体数为22条,核型公式为2n=22(20m 2sm),全部为中部或亚中部着丝点染色体,NF=44。Ag-NORs具有多态现象。C-带核型显示22条染色体的着丝点均正染,No.1染色体近着丝粒处有不恒定插入型C-带,No.4染色体具有恒定近端部插入型C-带,随体部位被正染的仅占所观察细胞的15%。  相似文献

采用常规压片法对砂燕麦、西班牙燕麦和短燕麦3个二倍体燕麦种进行了核型研究。结果表明:砂燕麦染色体核型公式为2n=2x=14=10m+4sm(2SAT),具近中部和中部着丝点染色体,第4对染色体组的短臂上有1对随体,核不对称系数为68.17%;西班牙燕麦染色体核型公式为2n=2x=14=10m+4sm(2SAT),具近中部和中部着丝点染色体,第7对染色体短臂上有1对随体,核不对称系数为59.31%;短燕麦染色体核型公式为2n=2x=14=6m+4sm+4st(2SAT),具近端部、近中部和中部着丝点染色体,第6对染色体组的短臂上有1对随体,核不对称系数为63.91%。虽然3个燕麦种的核型均为2A,但它们的染色体形态有明显不同,比较认为砂燕麦相对进化,短燕麦次之,西班牙燕麦较原始。本研究对燕麦种质资源的核型分析及进化地位研究具有参考价值。  相似文献

The karyotypes of Hystrix coreana from eastern USSR and H. patula from USA were investigated by Giemsa C-banding. Both species are outbreeders and have 2n = 4x = 28. The karyotype of two plants of H. coreana has 10 metacentric, 6 submetacentric, 8 heterobrachial and 4 SAT chromosomes; two plants differed by having 12 metacentric, 4 submetacentric, 8 heterobrachial and 4 SAT-chromosomes, and 10 metacentric, 4 submetacentric, 9 heterobrachial and 5 SAT-chromosomes, respectively. The C-banding pattern had no or few inconspicuous intercalary bands, but conspicuous telomeric C-bands in one or both arms giving a high content of heterochromatin (16.3–18.2%). The chromosome complement of one plant of H. patula had 8 metacentric, 6 submetacentric, 8 heterobrachial and 6 SAT-chromosomes. The C-banding pattern had between 1 and 4 intercalary or centromeric bands and conspicuous telomeric bands on one or both arms giving a high content of constitutive heterochromatin (16.4%).  相似文献

A Giemsa C-banding technique was used to study the amount and location of constitutive heterochromatin in diploid (2n = 2x = 16) Medicago sativa ssp. falcata (L.) Arcangeli. Most accessions had the standard C-banding pattern with centromeric bands on all the chromosomes and a prominent heterochromatic band at the nucleolar organizer regions (NOR) of the satellited (SAT) chromosomes. However, we observed in various accessions that constitutive heterochromatic C-bands can exist at the telomeric ends of all the chromosomes. Interstitial bands occurred on the short arms of all chromosomes except for chromosome 3 and on the long arms of chromosomes 1, 2, 3, and 6, only. Rearranged chromosomes such as isochromosomes have been observed for the short arms of chromosomes 2 and 6. This is the first report on the existence of C-banding polymorphisms and the detection of putative isochromsomes in the chromosomes of diploid ssp. falcata which could have contributed to the variation observed in cultivated alfalfa.  相似文献

以Giemsa C带技术处理串叶松香草根尖细胞染色体(2n=14),全部着丝点及第5和第7对染色体短臂端部显稳定的C带,第6对染色体长臂有两条明显的居间带,其他居间带小而不稳定(重复率不高)。间期细胞核染色体呈Rable构型,其着丝点一极最多出现20个染色中心。统计分析表明,靠近着丝点的短臂端带区和居间带区异染色质有易与着丝点区异染色质融合的倾向。分裂中期Giemsa C带数目与间期染色中心数目存在数量对应关系。  相似文献

It has been proposed that the genera Zebrina and Setcreasea of the family Commelinaceae should be united and reunited, respectively, with the genus Tradescantia, mainly based on morphological studies. In the present study, karyotypes and Giemsa C-banding patterns in the root-tip cells of three Zebrina and two Setcreasea clones were analyzed, and were compared with those of a triploid Tradescantia clone. Z. pendula and Z. purpusii (both 2n = 24) were found to have similar karyotypes (4 M + 6 ST + 14 T; M = meta-, ST = subtelo-, T = telocentric chromosomes), while Z. pendula cv Quadricolor (2n = 23) had a unique karyotype (6 M + 5 ST + 11 T + 1 SA; SA = short acrocentric chromosome). The only clear difference between Z. pendula and Z. purpusii was that one and two subtelocentric chromosomes, respectively, had satellites at the short arms. Two clones of S. purpurea (2n = 24) had karyotypes (8 M + 8 M' + 8 SM; M' = nearly meta-, SM = submetacentric chromosomes) similar to each other. T. ohiensis (2n = 18) had a symmetric karyotype (9 M + 9 SM) consisting of larger chromosomes than S. purpurea. Many clear Giemsa C-bands were detected, in addition to centromeric bands in all chromosomes of all clones. Z. pendula and Z. purpusii commonly had single clear interstitial bands in eight telocentric chromosomes each, but they also had unique telomeric and other interstitial bands, respectively. Z. pendula cv Quadricolor had a unique banding pattern, i.e., satellite bands in the unique short chromosome, telomeric bands at the long arms of all metacentric chromosomes, and single interstitial bands in six telocentric chromosomes. Two clones of S. purpurea had telomeric bands at many chromosome arms and satellite bands in two nearly metacentric and one submetacentric chromosomes, but some differences were found between them. On the other hand, all the chromosomes of T. ohiensis had telomeric bands at both arms, and three submetacentric chromosomes had satellite bands. These result prove structural differentiation of chromosomes occurred among the clones, especially in Zebrina, and show that S. purpurea is relatively close to T. ohiensis, while Zebrina is obviously distant from the other two genera. Therefore, there remains a question cytologically at least for uniting Zebrina with Tradescantia.  相似文献

The C-banded karyotypes of three Allium cepa plants of different background are described. Identification of all chromosomes of Allium cepa is possible on the basis of complex telomeric and intercalary C-bands. A standard system of chromosome nomenclature is proposed. Infraspecific variation in heterochromatin amount per genome, number of intercalary bands per genome, relative area of telomeric bands, relative chromosome length, relative chromosome arm length and centromeric index are statistically analysed. Although extensive polymorphism in Allium cepa chromosomes is found especially with respect to the size of telomeric bands, the overall similarity of the karyotypes is obvious. The value of C-banding for biosystematics of cultivated plants related to Allium cepa and for their breeding is suggested.  相似文献

The karyotypes of two populations ofFestucopsis serpentini (2n = 2x = 14) endemic to Albania were investigated in detail by Giemsa C- and N-banding, AgNO3 staining, and in situ hybridization with an rDNA probe. The complements consisted of 14 large chromosomes, 10 metacentric and 4 SAT-chromosomes, a metacentric and a submetacentric pair. SAT-chromosomes from one population carried exclusively minute satellites, whereas SAT-chromosomes from another population also carried larger polymorphic satellites, suggesting a geographical differentiation. The existence of four chromosomes with nucleolus forming activity was established through AgNO3 staining; however, the rDNA probe additionally hybridized to intercalary positions in the short arms of two metacentric chromosomes revealing two inactive rDNA sites. C-banding patterns comprised from zero and up to four very small to larger, generally telomeric bands per chromosome giving low levels of constitutive heterochromatin. Similarities in chromosome morphology and C-banding patterns identified the homologous relationships of all chromosomes in one population, but of three pairs only in the other. Reliable identification of homologous chromosomes between plants was only possible for the SAT-chromosomes. A comparison between the C-banded karyotypes ofF. serpentini andPeridictyon sanctum supports their position in two genera.  相似文献

达乌尔黄鼠分布在我国北方及蒙古和苏联等区域,对牧草及农田危害甚大。有关达乌尔黄鼠的核型国内外已有报道(Lyapunova等,1970;蔡有余等,1985;马继霞等,1985)。签于其染色体的一些特征,达乌尔黄鼠有可能成为染色体工程及检测环境诱变剂等方面的实验材料。虽然苏联Lyapunova等(1978,1980)对黄鼠属某些种的G-带和C-带进行过比较研究,我国蔡有余等(1985)对达乌尔黄鼠的C-带和Ag-NOR进行了观察,但无法对其染色体进行逐个地准确识别,特别是对Χ染体色的正确识别。为此,我们对达乌尔黄鼠的显带染色体进行了较详细的研究。  相似文献

Detailed karyotypes of Lilium longiflorum and L. rubellum were constructed on the basis of chromosome arm lengths, C-banding, AgNO3 staining, and PI-DAPI banding, together with fluorescence in situ hybridisation (FISH) with the 5S and 45S rDNA sequences as probes. The C-banding patterns that were obtained with the standard BSG technique revealed only few minor bands on heterologous positions of the L. longiflorum and L. rubellum chromosomes. FISH of the 5S and 45S rDNA probes on L. longiflorum metaphase complements showed overlapping signals at proximal positions of the short arms of chromosomes 4 and 7, a single 5S rDNA signal on the secondary constriction of chromosome 3, and one 45S rDNA signal adjacent to the 5S rDNA signal on the subdistal part of the long arm of chromosome 3. In L. rubellum, we observed co-localisation of the 5S and 45S rDNA sequences on the short arm of chromosomes 2 and 4 and on the long arms of chromosomes 2 and 3, and two adjacent bands on chromosome 12. Silver staining (Ag-NOR) of the nucleoli and NORs in L. longiflorum and L. rubellum yielded a highly variable number of signals in interphase nuclei and only a few faint silver deposits on the NORs of mitotic metaphase chromosomes. In preparations stained with PI and DAPI, we observed both red- and blue-fluorescing bands at different positions on the L. longiflorum and L. rubellum chromosomes. The red-fluorescing or so-called reverse PI-DAPI bands always coincided with rDNA sites, whereas the blue-fluorescing DAPI bands corresponded to C-bands. Based on these techniques, we could identify most of chromosomes of the L. longiflorum and L. rubellum karyotypes.  相似文献
