The potential of using Rhodotorula glutinis alone or in combination with salicylic acid (SA) for the control of postharvest Rhizopus rot of strawberries, and their effects on enzyme activities of fruits were investigated. The combination of R. glutinis (1 × 108 CFU ml−1) with SA (100 μg ml−1) resulted in a significant reduction in the disease incidence and lesion diameter of Rhizopus rot on the strawberry fruits at 20 °C and 4 °C, and more so than with SA or yeast alone. SA at the concentration of 100–1000 μg ml−1 significantly inhibited spore germination of Rhizopus stolonifer. About 100 μg ml−1 of SA did not inhibit the growth of the antagonistic yeast, and could significantly increase the population growth of R. glutinis in strawberry wounds at 20 °C. SA, combined with R. glutinis, increased the activity of strawberry host defence enzymes (POD) and cell wall lytic enzymes (β-1,3-glucanase).  相似文献

The biocontrol activity of Rhodotorula glutinis on green mold decay of oranges caused by Penicillium digitatum was investigated in vitro and in vivo. Significant control was achieved with a washed cell suspension and an unwashed cell culture mixture of R. glutinis. Treatment of wounds with autoclaved cell cultures or cell-free culture filtrate did not prevent decay. The protection provided by the washed yeast cells was dose-dependent. The higher the concentration of R. glutinis, the better the effect of the biocontrol capacity. At concentrations of yeast of 1×109 colony-forming units per milliliter or higher and pathogen spore suspensions of 5×104 spores per milliliter, green mold was almost inhibited after 4-days incubation at 20 °C. The interval between the pathogen inoculation and the antagonist application significantly influenced the biocontrol ability. The biocontrol efficacy of R. glutinis applied before the pathogen was better than that of applied after the pathogen. Surprisingly, R. glutinis was also effective in controlling green mold at low temperature (4 °C). Rapid colonization of the yeast in wounds was observed during the first 3 days at 20 °C, and remained stable after 5-days incubation. On fruits stored at 4 °C, even after 21 days, the population of R. glutinis in wounded fruits was more than 1,600-fold of what it was just prior to storage. In the test on potato dextrose agar plates, agar disks of R. glutinis nutrient yeast dextrose agar cultures placed on PDA plates seeded with pathogens did not inhibit the growth of P. digitatum. Spore germination of pathogens in potato dextrose broth was greatly controlled in the presence of living cell suspensions.  相似文献

The efficacy of Rhodotorula mucilaginosa against postharvest gray mold, blue mold and natural decay development of apples and the possible mechanisms involved were investigated. The decay incidence and lesion diameter of gray mold and blue mold of apples treated by R. mucilaginosa were significantly reduced compared with the control fruits, and the higher concentration of R. mucilaginosa, the better the efficacy of the biocontrol. R. mucilaginosa also significantly reduced the natural decay development of apples following storage at 20 °C for 35 days or at 4 °C for 45 days followed by 20 °C for 15 days. Germination and survival of spores of Penicillium expansum and Botrytis cinerea were markedly inhibited by R. mucilaginosa in an in vitro test. Rapid colonization of the yeast in apple wounds was observed whether stored at 20 °C or 4 °C. In apples, the activities of peroxidase (POD) and polyphenoloxidase (PPO) were significantly induced and lipid peroxidation (malondialdehyde (MDA) content) was highly inhibited by R. mucilaginosa treatment compared with those of the control fruits. All these results indicated that R. mucilaginosa has great potential for development of commercial formulations to control postharvest pathogens on fruits. Its modes of action were based on competition for space and nutrients with pathogens, inducement of activities of defense-related enzymes such as POD, PPO and inhibition of lipid peroxidation (MDA content) of apples, so as to enhance the resistance and delay the ripening and senescence of apples.  相似文献

Rhodotorula glutinis IFO1125 acquired increased aluminum (Al) resistance from 50 μM to more than 5 mM by repetitive culturing with stepwise increases in Al concentration at pH 4.0. In our previous study we performed differential display to find that three genes (RgFET3, RgGET3, and RgCMK) encoding proteins homologous to Saccharomyces cerevisiae FET3p, GET3p, and CMK1p and CMK2p, respectively, were up-regulated in the Al-resistant cells. In this study we cloned these genes and found they were indeed up-regulated in Al-resistant strains. The cloned genes were introduced into S. cerevisiae and corresponding mutants to test their relevance to Al resistance. The introduction of RgFET3 and RgGET3 conferred Al resistance to the host, but that of RgCMK did not. Green fluorescent protein (GFP)-tagged RgFet3p was localized at the cell periphery in the host. GFP-tagged RgGet3p formed more punctate bodies in the host under Al stress than in the absence of Al. Different growth responses to Fe (III), Cu (II), Ca ions, and cyclosporin A in the wild type and resistant cells of R. glutinis suggested the involvement and possible links of the three genes in Al resistance.  相似文献

The influence of chitin on the efficacy of Rhodotorula glutinis in controlling postharvest grey mould of strawberries and the possible mechanisms involved were investigated. The results showed that the antagonistic activity of R. glutinis harvested either from the culture media of NYDB amended with chitin at 0.5% or from the culture media in which chitin was the sole carbon source (NYCB) was improved greatly compared with that without chitin. The application of R. glutinis cultivated in the culture media of the chitin-supplement (0.5%) induced higher β-1,3-glucanase activity and reduced more MDA content of strawberries compared with that R. glutinis cultivated in the NYDB. Moreover, the control efficacy of the cell-free filtrate of the chitin-supplement culture media (0.5%) and NYCB were higher than that of cell-free culture filtrates of NYDB in 2 days incubation, with the associated high level of chitinase activity.  相似文献

The effects of preharvest methyl jasmonate (MJ) application on fruit quality, antioxidant activity and flavonoid content in blackberries (Rubus sp.) were determined. Anticancer activity against human lung A549 cells and HL-60 leukemia cells was also evaluated. Three blackberry cultivars (Chester Thornless, Hull Thornless and Triple Crown) were used in these experiments. Blackberries treated with MJ (0.01 and 0.1 mM) had higher soluble solids content, and lower titratable acids than untreated fruit as well as enhanced content of flavonoids and increased antioxidant capacity. Extracts of treated fruit showed enhanced inhibition of A549 cell and HL-60 cell proliferation and induced the apoptosis of HL-60 cells. Cultivar Hull Thornless had higher soluble solids and lower titratable acids compared to cv. Chester Thornless and Triple Crown. On the basis of fresh weight of fruit, Hull Thornless also had significantly higher anthocyanin, total phenolic content, antioxidant and antiproliferation activity than other two cultivars.  相似文献

The feasibility of 0.2 g l−1 benzo-thiadiazole-7-carbothioic acid S-methyl ester (BTH) to improve the efficacy of Pichia membranefaciens in controlling postharvest blue mould decay in peach fruit was investigated. Our results showed that biocontrol activity of P. membranefaciens against blue mould caused by Penicillium expansum in peach fruit could be enhanced by addition of 0.2 g l−1 BTH. The combination of P. membranefaciens and BTH resulted in a more effective control of blue mould than individual treatment of P. membranefaciens or BTH alone. The combined treatment had a synergistic effect on the induction of superoxide dismutase, catalase, ascorbate peroxidase, chitinase and β-1,3-glucanase activities, which induced stronger disease resistance in fruit than BTH or yeast alone, and resulted in a lower lesion diameter and disease incidence of blue mould decay in peaches. Furthermore, the combined treatment did not impair the quality parameters including fruit firmness and contents of total soluble solids, titratable acidity and vitamin C of peach fruit after 6 days of storage at 20 °C. These results suggested that the use of BTH may be an effective method to improve the biological activity of P. membranefaciens.  相似文献

We investigated the effects of marine yeast Rhodosporidium paludigenum in combination with a food additive, carboxymethylcellulose sodium (CMC-Na), on prevention of postharvest decay and food quality of Chinese winter jujubes. R. paludigenum (1 × 108 cells/ml) combined with CMC-Na (0.3%) significantly increased the inhibition of black rot on jujubes at 25 °C when compared with R. paludigenum-alone treatment (5.8% vs. 20%, p < 0.05). The combination also reduced natural rot from 86% (control) to 56%. The combination caused transient changes in enzyme activities or contents of some oxidation reactive markers such as peroxidase (POD), superoxide dismutase (SOD), and malondialdehyde (MDA) of jujubes. The combination had no significant effect on the food qualities such as colour (chroma and hue angle), total soluble solid (TSS) and titratable acidity (TA) of the fruit. While enhancing these effects, CMC-Na did not affect the survival of R. paludigenum in nutrient yeast dextrose agar (NYDA) culture. Thus, we conclude that the combination of R. paludigenum and CMC-Na is a promising formulation to control postharvest decay of Chinese winter jujubes.  相似文献

The effect of nitric oxide fumigation on phenolic metabolism of harvested Chinese winter jujube in relation to the fruit quality was investigated. The fruits were fumigated for 3 h with NO (0, 10, 20 and 30 μl/l) then stored at 22 °C and RH 95%. Changes in color, phenol and anthocyanin levels in pericarp, activities of the related enzymes, and total soluble solids and vitamin C from mesocarp were measured. The results showed that treatment with 20 μl/l NO significantly slowed the increase in red index, inhibited changes of polyphenol oxidase (PPO) and phenylalanine ammonia lyase (PAL) activities, maintained a low total anthocyanin content and a high total phenol content, and delayed the increase of soluble solids and decrease of vitamin C. Treatment with NO solution at less than 1 μmol/l exhibited inhibitory effects on in vitro PPO and PAL activities in a dose-dependent manner.  相似文献

Sardinian abbamele is a typical product obtained from the honey recuperation from combs (traditional procedure) or by concentration of the honey diluted in water (industrial procedure). Seven abbamele samples were obtained to study the volatiles’ composition, the presence of honey marker compounds and their relationship with the production procedures. The long thermal treatment applied in abbamele production caused very high (1007.0–4405.8 mg/kg) HMF content (HPLC-DAD), while glucose and fructose amounts were quite similar to the honey ones (HPLC-RI). Total antioxidant activity (FRAP assay) of the samples ranged between 13.3 and 71.2 mmol Fe2+/kg, while antiradical activity (DPPH assay) ranged between 3.8 and 23.3 mmol TEAC/kg. Such high antioxidant values were linearly correlated with total phenol amount (1297.8–4469.5 mg GAE/kg) determined by Folin–Ciocalteau method. Thermally derived furan derivatives and terpenes were abundant among the headspace volatiles (HS-SPME), particularly limonene (0.5–76.0%) that probably originated from citrus rinds’ addition during abbamele production. GC and GC–MS analyses of USE isolates revealed HMF predominance as well as the honey marker compounds (if/when existing) such as methyl syringate (up to 49.2%), marker of Asphodelus microcarpus honey. High isophorone percentage (up to 30.9%) determined by HS-SPME followed by minor percentage of 4-ketoisophorone and norisoprenoids in one sample indicated Arbutus unedo L. honey use in the production. HPLC-DAD analysis confirmed the presence of specific honey markers: two samples showed high methyl syringate concentrations (150.4–120.1 mg/kg) while homogentisic acid and other specific markers of A. unedo honey were found in one sample. The compared GC–MS and HPLC-DAD data proved to be useful to obtain information about the use of specific honey in the production and to verify citrus addition.  相似文献
