The effect of anionic (sodium dodecyl sulphate or SDS) and cationic (cetyltrimethylammonium bromide or CTAB) surfactants on the stability of binary bacterial coaggregates comprising Acinetobacter johnsonii S35 and Oligotropha carboxidovorans S23 (both sewage sludge isolates) was studied and compared with that on the complex sewage sludge flocs. Both SDS and CTAB enhanced the bacterial coaggregation at their lower concentrations of 0.2 and 0.07 mg ml(-1), respectively. However, complete deflocculation of coaggregates was observed at 1 mg ml(-1) SDS and 0.3 mg l(-1) CTAB concentrations. Further, sewage sludge flocs did not deflocculate in the presence of CTAB, although a concentration-dependent deflocculation was observed in the presence of SDS. A. johnsonii S35 and O. carboxidovorans S23 cells were separately pretreated (prior to coaggregation) with the surfactants. In spite of the partial (complete) loss of viability during SDS (CTAB) pretreatment, washed cells still retained hydrophobic character and displayed significant coaggregation (aggregation index ranging from 84% to 97% in comparison to 96% in the case of non-treated cells), demonstrating reversibility of the surfactant induced deflocculation. Further, when exposed to lower concentration of surfactants (0.2 mg ml(-1) SDS), coaggregates were more resistant (76% viability) as compared to the individual partner (S35: 52%; S23: 39% viability). Since the coaggregates are stable and provide protection from surfactants at lower concentrations (those normally expected in the sewage treatment plants), their presence as well as a sustained role in the sewage sludge bioflocculation is evident.  相似文献

Whole cell protein extracts from strains of the currently recognized genomic species of Acinetobacter, together with those from a range of isolates of several genomic species identified using the Biolog system and obtained from a biological nutrient-removal activated sludge plant were analysed by SDS-PAGE. The dendrograms obtained after numerical analysis for the known genomic species generally supported the taxonomic relationships suggested from earlier DNA–DNA hybridisation data. In some cases the activated sludge isolates identified to genomic species level clustered closely with the corresponding genomic species reference strains, although isolates 5 and 8/9 were scattered throughout the dendrogram. Considerable variations were seen in the protein patterns of the 27 different environmental isolates of genomic species 7 that were analysed. Cells containing polyphosphate 71 micrograms P (mg protein)-1 and no poly-beta-hydroxybutyrate showed metachromatic granules but no lipid granules; cells containing poly-beta-hydroxybutyrate (15% of dry weight) showed fluorescence lipid granules but no metachromatic granules; whereas cells containing both polyphosphate and poly-beta-hydroxybutyrate showed both types of granules. These observations, together with a critical review of the literature, show a clear distinction between metachromatic (or volutin) granules and lipid granules.  相似文献

The bacterial community of a biological phosphate removal (BPR) activated sludge process was studied and compared to that of a non-BPR process treating the same municipal waste water. Bacterial isolates from the BPR process, as characterized by whole cell fatty acids, belonged to more than twenty genera, with Micrococcus, Staphylococcus and Acidovorax scoring highest. Acinetobacter spp represented 4% of cultured bacteria, ≤3% as estimated by fluorescence in situ hybridization, and well under 10% on the basis of the proportion of ubiquinone Q9 in the sludge. The mole proportions of ubiquinones, Q8 : Q10 : Q9 in the sludge were maintained fairly stable at approximately 9:4:1. The spectra of the isolated strains and the proportions of ubiquinones in the processes (BPR vs non-BPR) were otherwise similar, but a significant number of isolates related to actinomycetes were obtained from the BPR sludge only. The BPR process did not enrich Acinetobacter. Pure cultures of Acinetobacter isolated from the sludge stained for polyphosphate, but Acinetobacter cells responding to the ACA probe in native sludge from the BPR process did not. Instead, the bulk of the polyphosphate in the BPR sludge was located in a distinct morphotype of large, coccoid, highly clustered cells. Filamentous Chloroflexi species are often present in activated sludge wastewater treatment plants in relatively low numbers, although bulking incidences caused by Chloroflexi filaments have been observed. A new species-specific gene probe for FISH was designed and using phylum-, subdivision-, morphotype 1851- and species-specific gene probes, the abundance of Chloroflexi filaments were monitored in samples from 126 industrial wastewater treatment plants from five European countries. Chloroflexi filaments were present in 50% of the samples, although in low quantities. In most treatment plants the filaments could only be identified with phylum or subdivision probes, indicating the presence of great undescribed biodiversity. The ecophysiology of various Chloroflexi filaments was investigated by a suite of in situ methods. The experiments revealed that Chloroflexi constituted a specialized group of filamentous bacteria only active under aerobic conditions consuming primarily carbohydrates. Many exo-enzymes were excreted, e.g. chitinase, glucuronidase and galactosidase, suggesting growth on complex polysaccharides. The surface of Chloroflexi filaments appeared to be hydrophilic compared to other filaments present. These results are generally supported by physiological studies of two new isolates. Based on the results obtained in this study, the potential role of filamentous Chloroflexi species in activated sludge is discussed.  相似文献

Acinetobacter junii strain BB1A, a novel metal-tolerant bacterium, produced biofilm in the presence of added ions such as Ni(2+), AsO(2)(-), Cd(2+) and Hg(2+) on surfaces such as glass and polystyrene. Generation of a metal-sensitive and adhesion-deficient mutant by transposition of Tn5-mob in the A. junii genome has putatively confirmed the association of metal tolerance with the production of biofilm. The requirement of a critical cell density for biofilm formation and presence of acyl-homoserine lactone-like autoinducer molecules in the cell-free supernatant indicated the phenomenon of quorum sensing. Addition of a natural quorum-sensing inhibitor (garlic extract) or synthetic quorum-sensing inhibitor (4-nitro-pyridine oxide) significantly inhibited cell growth and biofilm formation in the presence of metal/metalloid ions.  相似文献

Activated carbons were prepared from sewage sludge by chemical activation. Pyrolusite was added as a catalyst during activation and carbonization. The influence of the mineral addition on the properties of the activated carbons produced was evaluated. The results show that activated carbons from pyrolusite-supplemented sewage sludge had up to a 75% higher BET surface area and up to a 66% increase in mesoporosity over ordinary sludge-based activated carbons. Batch adsorption experiments applying the prepared adsorbents to synthetic dye wastewater treatment yielded adsorption data well fitted to the Langmuir isotherm. The adsorbents from pyrolusite-supplemented sludges performed better in dye removal than those without mineral addition, with the carbon from pyrolusite-augmented sludge T2 presenting a significant increase in maximum adsorption capacity of 50 mg/g. The properties of the adsorbents were improved during pyrolusite-catalyzed pyrolysis via enhancement of mesopore production, thus the mesopore channels may provide fast mass transfer for large molecules like dyes.  相似文献

The aim of this work is to evaluate the adsorption performances of activated carbon derived from sewage sludge (ACSS) for gaseous formaldehyde removal compared with three commercial activated carbons (CACs) using self-designing adsorption and distillation system. Formaldehyde desorption of the activated carbons for regeneration was also studied using thermogravimetric (TG) analysis. The porous structure and surface characteristics were studied using N2 adsorption and desorption isotherms, scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR). The results show that ACSS has excellent adsorption performance, which is overall superior to the CACs. Adsorption theory indicates that the ACSS outperforms the CACs due to its appropriate porous structure and surface chemistry characteristics for formaldehyde adsorption. The TG analysis of desorption shows that the optimum temperature to regenerate ACSS is 75 °C, which is affordable and economical for recycling.  相似文献

We investigated ammonia-oxidizing bacteria in activated sludge collected from 12 sewage treatment systems, whose ammonia removal and treatment processes differed, during three different seasons. We used real-time PCR quantification to reveal total bacterial numbers and total ammonia oxidizer numbers, and used specific PCR followed by denaturing gel gradient electrophoresis, cloning, and sequencing of 16S rRNA genes to analyze ammonia-oxidizing bacterial communities. Total bacterial numbers and total ammonia oxidizer numbers were in the range of 1.6 x 10(12) - 2.4 x 10(13) and 1.0 x 10(9) - 9.2 x 10(10)cellsl(-1), respectively. Seasonal variation was observed in the total ammonia oxidizer numbers, but not in the ammonia-oxidizing bacterial communities. Members of the Nitrosomonas oligotropha cluster were found in all samples, and most sequences within this cluster grouped within two of the four sequence types identified. Members of the clusters of Nitrosomonas europaea-Nitrosococcus mobilis, Nitrosomonas cryotolerans, and unknown Nitrosomonas, occurred solely in one anaerobic/anoxic/aerobic (A2O) system. Members of the Nitrosomonas communis cluster occurred almost exclusively in association with A2O and anaerobic/aerobic systems. Solid residence time mainly influenced the total numbers of ammonia-oxidizing bacteria, whereas dissolved oxygen concentration primarily affected the ammonia-oxidizing activity per ammonia oxidizer cell.  相似文献

Polyphosphate-degrading enzymes were studied in Acinetobacter spp. and activated sludge. Polyphosphate: AMP phosphotransferase activity in Acinetobacter strain 210A decreased with increasing growth rates. The activity of this enzyme in cell extracts of Acinetobacter strain 210A was maximal at a pH of 8.5 and a temperature of 40 degrees C and was stimulated by (NH4)2SO4. The Km for AMP was 0.6 mM, and the Vmax was 60 nmol/min per mg of protein. Cell extracts of this strain also contained polyphosphatase, which was able to degrade native polyphosphate and synthetic magnesium polyphosphate and was strongly stimulated by 300 to 400 mM NH4Cl. A positive correlation was found between polyphosphate:AMP phosphotransferase activity, adenylate kinase activity, and phosphorus accumulation in six Acinetobacter strains. Significant activities of polyphosphate kinase were detected only in strain P, which contained no polyphosphate:AMP phosphotransferase. In samples of activated sludge from different plants, the activity of adenylate kinase correlated well with the ability of the sludge to remove phosphate biologically from wastewater.  相似文献

Most of 200 Activated Sludge Plant in Iran are overloaded and as a result, their efficiency is low. In this work, a pilot plant is manufactured and put into operation in one of the wastewater treatment plants in the west of Tehran. Instead of conventional activated sludge, a membrane bioreactor and an upflow anaerobic sludge blanket reactor used as a pretreatment unit in this pilot. For the sake of data accuracy and precision, an enriched municipal wastewater was opted as an influent to the pilot. Based on the attained result, the optimum retention time in this system was 4h, and the overall COD removal efficiency was 98%. As a whole, the application of this retrofit would increase the plant's capacity by a factor of 5 and reducing the excess sludge by a factor of 10. The sludge volume index in the anaerobic reactor was about 12 after granulation occurred.  相似文献

The use of different proportions of rape straw and grass as amendments in the composting of dewatered sewage sludge from a municipal wastewater treatment plant was tested in a two-stage system (first stage, an aerated bioreactor and second stage, a periodically turned windrow). The composition of feedstock affected the temperature and organic matter degradation in the bioreactor and the formation of humic substances, especially humic acids (HA), during compost maturation in the windrow. The total HA content (the sum of labile and stable HA) increased according to first-order kinetics, whereas labile HA content was constant and did not exceed 12% of total HA. Δlog K of 1.0–1.1 indicated that HA was of R-type, indicating a low degree of humification. Temperature during composting was the main factor affecting polymerization of fulvic acids to HA and confirmed the value of the degree of polymerization, which increased only when thermophilic conditions were obtained.  相似文献

A conceptual model describing the relative roles of sorption and precipitation processes for metals in solid-solution suspensions is presented. The model performance is demonstrated using experimental data on sorption and precipitation of metals in samples of activated sludge mixed liquor. Based on the experimental results presented here, it appears that, at total metal and mixed liquor suspended solids concentrations and pH values generally encountered in full-scale municipal (or combined municipal/industrial) activated sludge systems, metals are primarily removed by sorption processes.  相似文献

The effects of different cultivation periods and growth media on the cell surface hydrophobicity and coaggregation ability of Acinetobacter johnsonii S35 with Oligotropha carboxidovorans S23 (both sewage sludge isolates) were examined. It was observed that when cultivated in polypeptone medium, A. johnsonii S35 shows high hydrophobicity and strong coaggregation (98% aggregation index) with O. carboxidovorans S23, which remain unaltered after a 1-day, 3-day or 7-day cultivation period. When A. johnsonii S35 was cultivated with different carbon substrates, large coaggregates and a high aggregation index were observed using acetate-grown cells (96%), followed by ethanol-(83%) and vegetable oil-grown cells (72%). Coaggregates were small and narrow in ethanol-grown cells and still smaller in case of the vegetable oil-grown cells. Although no direct correlation could be observed, more hydrophobic cells showed a higher aggregation index. None of the culture supernatants of A. johnsonii S35 showed emulsification of hexadecane or a significant ability to aggregate O. carboxidovorans S23 cells, indicating that the coaggregation factor of A. johnsonii S35 is cell-bound. These results demonstrate that the coaggregation ability of A. johnsonii S35 is constitutive and is not lost under diverse cultivation conditions.  相似文献

不动杆菌属细菌分布广泛,作为重要的石油烃降解者,在乳化和降解石油烃、降低石油烃生物毒性等方面有重要作用。本文概述了不动杆菌属细菌对烷烃、芳香烃等石油烃组分的降解,总结了该属细菌中已发现的烷烃氧化酶和芳香烃氧化酶,综述了该属细菌所分泌的表面活性剂的类型和乳化机理,讨论了固定化对该属细菌降解石油烃的影响,展望了该属细菌降解石油烃的应用前景。基于此,作者认为探索不动杆菌属细菌降解石油烃的详细机理和途径、发现关键酶、寻找遗传工具、构建基因工程菌、发掘环境友好的固定化材料,应是未来的研究重点及热点。  相似文献

通过特异引物扩增环境中氨氧化细菌16S rDNAV2保守区域,将该片段克隆到T-easy载体上,PCR产物经测序和定量PCR扩增体系鉴定,证实PCR扩增产物为氨氧化细菌16S rDNA保守序列,以含该序列的重组质粒作为定量PCR监测氨氧化细菌数量的DNA标准品。用荧光定量PCR技术比较了五氯酚(PCP)对好氧颗粒污泥和活性污泥中氨氧化细菌数量的影响。结果表明,不加PCP的反应器中,好氧颗粒污泥和活性污泥中氨氧化细菌的数量分别为4.28×107±5.44×106cells/(g干污泥)和2.51×109±8.61×108cells/(g干污泥)。随着PCP浓度的增加(0~50mg/L),PCP对氨氧化细菌数量的影响不大(P>0.05),而且,污泥中氨氧化细菌的数量与氨氮的去除率无直接的正相关关系(P>0.05),PCP主要是抑制氨氧化细菌的代谢活性导致污泥氨氮去除效率降低。  相似文献
