A study was made of the behavior of the phosphoglucomutase (PGluM) and creatine phosphokinase (CPK) in the liver tissue of guinea pigs after chronic digitalis intoxication. It was found that the latter causes a statistically significant decrease of both enzymatic activities. The results obtained by the authors with this investigation, together with those of previous researchers, show that the toxic effect of digitalis on the enzymatic activities is not limited to the reported effect on the ATPase but also involves, at least at the doses and under the conditions used, other enzymatic activities.  相似文献

To determine the role played by the kidney in the metabolism and excretion of plasma adenosine 3′,5′-monophosphate (cAMP) we have studied the fate of this nucleotide (0.01–1.0mM) when it is perfused in a recirculating medium through the isolated rat kidney. cAMP was rapidly taken up and degraded by the kidney, the rate of its disappearance from the perfusate being at least twice its rate of excretion in the urine. Nevertheless, the cAMP excretory rate exceeded the filtration rate by 1.5 to 2 fold, and thus net secretion (transtubular transport) was demonstrated. The rates of filtration, perfusate clearance, and degradation of cAMP were proportional to its perfusate concentration. Methyl xanthines (caffeine and aminophylline) at 10mM, and probenecid at 0.9mM abolished transtubular transport of cAMP and greatly retarded disappearance of the nucleotide from the perfusate. It is concluded that there is a ready penetration of cAMP into renal cells from peritubular capillaries. Depending on the perfusate concentration of cAMP, transtubular transport may or may not exceed the simultaneous intra-renal breakdown of the compound. A low rate of cAMP excretion in the urine may accompany a considerably higher rate of cAMP clearance from the perfusate by the kidney.  相似文献

The incubation of [5,6-3H]prostaglandin E1 ([3H]PGE1) with guinea pig kidney cortex microsomes in the presence of NADPH in an atmosphere of air, resulted in chromatographically polar metabolites. The incubation products were treated with base which converted PGE1 derivatives into PGB1 derivatives, with a λmax = 278 nm and the products were analyzed by TLC and high pressure-liquid chromatography (HPLC). Based on UV absorption, mobility on TLC and retention time in HPLC, as compared with authentic compounds, it was concluded that the two polar UV-absorbing peaks in HPLC represented 19-hydroxy-PGB1 (19-OH-PGB1) and 20-hydroxy-PGB1 (20-OH-PGB1). Further identification of the metabolites was obtained by derivatizing the incubation products as methyl esters and t-butyldimethylsilyl ethers, followed by co-injection with similarly derivatized authentic compounds in HPLC and gas chromatography. Finally, the derivatized metabolites were identified by comparing their mass fragmentation with that of similarly derivatized authentic compounds. There was an absolute requirement for NADPH, and NADH did not significantly support the hydroxylation of PGE1. Inhibitors of microsomal monooxygenase (SKF 525A, metyrapone, and cytochrome c) inhibited the hydroxylation of PGE1 by kidney cortex microsomes. By contrast, carbon monoxide at a CO:O2 ratio of 5:1 did not inhibit the hydroxylation of PGE1, pointing to a low or lack of CO sensitivity of the hydroxylation of PGE1. The addition of PGE1 or laurate to guinea pig kidney cortex microsomes elicited Type I spectral changes. The spectral dissociation constant (Ks) for PGE1 was 2.4 × 10?4m. The kinetic constants for 19- and 20-hydroxylations of PGE1 were determined. The KM values for the 19- and 20-hydroxylation pathways were found to be identical, being 3.3 × 10?4m, suggesting that the same enzyme is involved in both hydroxylations; however, the Vmax values for 19-hydroxylation and 20-hydroxylation of PGE1 were 50 nmol/hr and 20.8 nmol/hr respectively. These results demonstrate that PGE1 is a substrate for the kidney cortex microsomal monooxygenase. The similarities and differences of the kidney monooxygenase in the guinea pig with that in the rat are discussed.  相似文献

We have isolated a new hematoside from guinea pig kidney. Like the usual hematoside (II3NeuAc LacCer), isolated from human erythrocytes, this new hematoside contained glucose, galactose, and N-acetylneuraminic acid in an equimolar proportion. By thin-layer chromatography (TLC), however, it migrated faster than the usual hematoside. After mild alkaline hydrolysis the TLC mobility of this ganglioside became identical to that of the usual hematoside. The sialic acid in this ganglioside was susceptible to Clostridial neuraminidase. Based on TLC mobility and the results of periodate oxidation, the sialic acid of the new hematoside was identified as 9-O-acetyl-N-acetylneuraminic acid. Therefore, the structure of this new hematoside is 9-O-Ac-NeuAc alpha 2 leads to 3Gal beta 1 leads to 4GLc beta 1 leads to 1'Cer.  相似文献

We treated pregnant guinea pigs on Day 50 of gestation with 10 mg testosterone propionate (TP), obtaining fetuses 2, 4, 8, or 18 h later as well as after 5 days of treatment. In a second group of pregnant guinea pigs, dihydrotestosterone propionate (DHTP), estradiol benzoate (E2B), progesterone (P), or cortisol was given 2 h before obtaining fetuses. Although TP treatment elevated fetal serum T (p less than 0.05), brain cytosolic androgen receptor (ARc) content was unchanged in fetuses of either sex. In female fetuses, nuclear androgen receptors (ARn) increased 10-fold in medial-basal hypothalamus (MBH) and preoptic area (POA) at 2 and 4 h (respectively) after treatment, while fetal male ARn content was unchanged. Maternal injection of other steroids (E2B, P, or cortisol, but not DHTP) significantly increased these hormones in the fetus 2 h later (p less than 0.05). Only androgens affected fetal androgen receptor (AR) content. While TP increased ARn in female MBH, DHTP decreased ARc in fetal anterior pituitary of both sexes. In this latter case, a metabolite of DHT may mediate the effects. We conclude that T crosses the guinea pig placenta and activates ARn in POA and MBH of female fetuses; male ARn appear to be maximally occupied by endogenous T. Steroids of other classes do not induce AR responses in fetal guinea pig brain. These AR changes may represent an initial cellular mechanism in brain sexual differentiation.  相似文献

The cellular response to Leishmania major (L. major) is usually evaluated in vivo by the delayed-type-hypersensitivity (DTH) test using leishmanin. Leishmanin can give false-positive reactions in areas where there is a background of leishmaniasis. In a previous study, it was shown that a 56 kDa antigen purified from promastigote and culture supernatant of L. major induce strong DTH reactions in sensitized guinea pigs. In this study, the species-specificity of this antigen was further investigated. Three groups of guinea pigs were sensitized with L. major, L. tropica, and L. infantum and both flanks of sensitized animal were injected intradermally with purified 56 kDa antigen or soluble leishmania antigen (SLA). The extent of indurations were measured after 24, 48, and 72 h. In animals which were sensitized with three species of leishmania, only those immunized with L. major showed skin reactions to purified antigen by an increase in skin thickness. Since complex antigen mixtures such as SLA and leishmanin show cross-reactivity and can be non-specific, the result obtained here suggest that 56 kDa antigen may be a useful diagnostic tool for species specific diagnosis in field studies of leishmaniasis.  相似文献

Distribution of platelet activating factor (PAF) receptor was examined in the guinea pig kidney. Northern blot analysis showed a single band electrophoresed just below the 28S rRNA, and the mRNA was richest in the cortex with lesser amounts in the outer and then inner medulla. Scatchard analysis of membrane fraction using [3H]WEB 2086, a specific PAF receptor antagonist, revealed a single binding site with Bmax of 522, 228, 58 fmol/mg protein for the cortex, outer medulla and inner medulla, respectively. Kd values were in the same order of magnitude (10(-8) M). These results indicate the presence of a single class of PAF receptor in the guinea pig kidney which is most abundant in the cortex.  相似文献

The subcellular distribution of alanine: glyoxylate aminotransferase 1 in guinea pig and rabbit kidneys was examined by centrifugation in a sucrose density gradient. The enzyme was located in the peroxisomes of guinea pig kidney and cross-reactive with the antibody against rat liver alanine: glyoxylate aminotransferase 1. This is the first report on the presence of the enzyme in the peroxisomes of mammalian kidney. The enzyme was found to be located in the mitochondria but not in the peroxisomes in rabbit kidney.  相似文献

Starting from the ultrastructural differences observed between the juxtaglomerular apparatus of the rat and that--only very rarely analysed--of the guinea pig kidney, the authors supposed that enzymhistochemical differences might also exist. This hypothesis could be confirmed by the results obtained in the Macula densa- and the Goormaghtigh-cells (Tab. 1 and 2). Indeed, in both places the Isocitrate dehydrogenase- and the Malate dehydrogenase-activities are stronger in the guinea pig than in the rat. On the other hand, the Glucose-6-phosphate and the 6-Phosphogluconate dehydrogenases show clearly weaker activities in the guinea pig Macula-cells compared with those of the rat, where these two enzymes give by far the strongest reaction. Metabolic differences might be related to the described diversities of the enzyme equipment.  相似文献
