The emergence of a new environmentally caused variant of Creutzfeldt-Jakob disease (vCJD), the result of food-born infection by the causative agent of bovine spongiform encephalopathy (BSE), has stimulated research on a practical diagnostic screening test. The immunocompetitive capillary electrophoresis (ICCE) assay has been reported to detect disease-specific, proteinase-resistant prion protein (PrPres) in the blood of scrapie-infected sheep. We have applied this method to blood from CJD-infected chimpanzees and humans. The threshold of detection achieved with our ICCE was 0.6 nM of synthetic peptide corresponding to the prion protein (PrP) C-terminus, and 2 nM of recombinant human PrP at the optimized conditions. However, the test was unable to distinguish between extracts of leucocytes from healthy and CJD-infected chimpanzees, and from healthy human donors and patients affected with various forms of CJD. Thus, the ICCE assay as presently performed is not suitable for use as a screening test in human transmissible spongiform encephalopathies (TSEs).  相似文献

Since the implementation of ISO/IEC 17025 in 2002, all accredited laboratories (at the least) need to establish traceability in all their tests and calibration methods. Traceabilty, though well understood in the calibration field (through an unbroken chain of comparisons to the International System of Units —SI), is less straight forward and not so well understood in the testing laboratories. Traceability in analytical and biological testing is found through the use of reference materials, and the validated steps of a test method. The European Commission has adopted a large number of emergency measures to prevent the spread of the Bovine spongiform encephalopathy (BSE) in the Member States, and among them, a European Regulation prohibits the feeding to ruminants of animal protein and animal feed containing such protein. Aim of this work has been to propose the thermal analysis as a new rapid and sensitive screening tool for a preliminary determination of possible contamination in products destined to bovine growth, since the possibility to screen the samples by a rapid and sensitive tool could reduce the time of analysis required by the actual methodology and could allow the systematic surveillance of bovine feeding, with the official EU methods applied only to positive samples.  相似文献

 Proficiency testing is a means of assessing the ability of laboratories to competently perform specific tests and/or measurements. It supplements a laboratory's own internal quality control procedure by providing an additional external audit of their testing capability and provides laboratories with a sound basis for continuous improvement. It is also a means towards achieving comparability of measurement between laboratories. Participation is one of the few ways in which a laboratory can compare its performance with that of other laboratories. Good performance in proficiency testing schemes provides independent evidence and hence reassurance to the laboratory and its clients that its procedures, test methods and other laboratory operations are under control. For test results to have any credibility, they must be traceable to a standard of measurement, preferably in terms of SI units, and must be accompanied by a statement of uncertainty. Analytical chemists are coming to realise that this is just as true in their field as it is for physical measurements, and applies equally to proficiency testing results and laboratory test reports. Recent approaches toward ensuring the quality and comparability of proficiency testing schemes and the means of evaluating proficiency test results are described. These have led to the drafting of guidelines and subsequently to the development of international requirements for the competence of scheme providers. In laboratory medicine meaningful measurements are essential for diagnosis, risk assessment, treatment and monitoring of patients. Thus methods applied in diagnostic measurements must be accurate, precise, specific and comparable among laboratories. Inadequate or incorrect analytical performance has consequences for the patients, the clinicians, and the health care system. One key element of metrology is the traceability of a measurement result to the SI system ensuring comparable results. This principle is described in the ISO/TC 212/WG2 N65 prEN 17511 Standard. In addition to the principles of metrology, the clinical usefulness, the diagnostic needs, and the biological and disease associated variations in patients' specimens have to be considered when the analytical biases for diagnostic purposes are defined. It must be the general goal of diagnostic laboratories to produce results that are true and comparable worldwide. The recent European in vitro diagnostic (IVD) Directive 98/79 EC follows the above mentioned standard of the International Organization for Standardization (ISO) and the European Committee for Standardization (CEN) requesting its application for all IVD reagents used within the European Union. This new European legislation will have a worldwide impact on manufacturers and clinical laboratories and will be implemented in 2003. It states that "traceability of values assigned to calibrators and/or control materials must be assured through available reference measurement procedures and/or available reference materials of a higher order". Thus a worldwide reference system needs to be established by collaboration and mutual recognition between the United States National Institute of Standards and Technology (NIST), European Metrology Institutes (EUROMET), regulatory bodies (e.g. United States Food and Drug Administration, FDA) the IVD industry and professional organizations (e.g. International Federation of Clinical Chemistry and Laboratory Medicine, IFCC). In June 2002, in Paris, representatives of international and regional organizations and institutions decided to form the "Joint Committee on Traceability for Laboratory Medicine" (JCTLM), which will support industry in registration and licensing of the "CE" label to test systems conforming to the IVD Directive.Presented at the International ILAC/IAF Conference on Accreditation in Global Trade, 23–25 September 2002, Berlin, Germany  相似文献

It is now over two years since ISO/IEC 17025 General requirements for the competence of testing and calibration laboratories was published. The standard places increased emphasis on the demonstration of traceability of measurements made by laboratories. In the areas of chemical and biological metrology, the introduction of this standard has brought new challenges for laboratories to grapple with. This paper will examine the requirements regarding traceability for chemical and biological measurements, with specific reference to the use of reference materials by laboratories. This will be explored from the perspective of both accreditation bodies and a laboratory which is both a user and producer of certified reference materials. Moreover, the paper will describe mechanisms that are being used to improve the use of reference materials by accredited laboratories and hence the traceability of measurements. An analytical method is described for detection of endogenous disease-associated prion protein in the buffy coat fraction from the blood of sheep infected with scrapie. The method has been improved and evaluated for its performance in the preclinical diagnosis of ovine transmissible spongiform encephalopathies. The test system uses a protocol for sample preparation that includes extraction and concentration and a test method that uses a liquid-phase competitive immunoassay for prion protein. Antibodies directed to a peptide sequence at the C-terminus of the prion protein (PrP) and a fluorescein-labeled peptide conjugate are used in the assay. Free zone capillary electrophoresis with laser-induced fluorescence for detection is used to separate the antibody-bound fluorescently labeled peptide and free labeled peptide. In this assay, the PrP competes with the fluorescently labeled peptide for limited antibody binding sites, which results in a reduction of the peak representing the immunocomplex of the antibody bound to the fluorescently labeled peptide. When blood samples from scrapie-infected sheep aged 7-12 months and of the scrapie-susceptible PrP genotypes VRQ/VRQ and VRQ/ARQ were analyzed, the abnormal PrP was found in blood samples. These results correlated with the post-mortem diagnosis of scrapie. The sheep were preclinical and appeared normal at the time of testing but later died with clinical disease approximately 12 months after testing. In older animals, and those with clinical signs, a smaller percentage of animals tested positive. This study has demonstrated that this technology can be used as a sensitive, rapid preclinical test to detect the disease-associated PrP in the blood of scrapie-infected sheep. Improvements in the extraction protocol and capillary electrophoresis conditions will enhance the robustness of this test.  相似文献

 Research and development activities are carried out by various types of laboratories that are not the typical testing and calibration laboratories for which the ISO/IEC 17025 is the quality assurance implementation reference. In this paper, such laboratories engaged in R&D activities are classified and different approaches they can adopt with a view to implementing a quality system that are suited to their characteristics and the type of work they conduct are proposed. These approaches take account of existing standards for the certification/accreditation of laboratories and of guides on quality assurance for non-routine analytical laboratories.  Since 1989 testing laboratories in Europe have had the possibility to confirm their competence in carrying out certain testing procedures by means of accreditation. Over the years an independent European Accreditation system has been developed, which many member states of the European Union have joined. The existing accreditation organizations joined to form the unified organization EA (European Cooperation for Accreditation) in 1997. Members of this organization are, inter alia, one representative of the accreditation authorities of each of the member states. The German accreditation system is different to the European system in that it is split. Here, we differentiate between the sector established by law and that which is not. Both sectors, however, pursue the same goal. Because of this double certification of competence and the resulting multiple reviews, German testing laboratories face a considerably greater load in comparison to their European colleagues. Several authorities are already working on the removal of this split accreditation system. The efforts must, however, be concentrated and brought into line. Efforts by the DAP and DACH for flexible accreditation and the creation of a standardized registration authority for testing laboratories in the field of workplace measures are examples of how the German accreditation system could develop in the future.  相似文献

Summary A well established totally automated testing procedure for characterization of RP materials is presented. The test is suited to the determination of hydrophobic and silanophilic interactions, as well as trace metal impurities on silica surfaces. it includes measurements of stationary phase stability and shows column performance at two different pH conditions. The very different parameters examined readily reveal batch to batch reproducibility and are therefore suitable for laboratories dealing with quality control and validation of columns.  There is currently no national program for recognition of United States environmental laboratories. This situation should change sometime in 1998 as environmental testing laboratories should be able to apply for accreditation in the United States via a system developed by the National Environmental Laboratory Accreditation Conference (NELAC). NELAC is an organization whose purpose is to establish performance standards for accrediting environmental laboratories. Laboratories to be accredited will be required to demonstrate qualifications of testing personnel, pass on-site inspections, perform satisfactorily on proficiency test samples, and meet specified quality assurance standards.  相似文献

A novel botanical dietary supplement, formulated as a chewable tablet containing a defined mixture of Souroubea spp. vine and Platanus spp. Bark, was tested as a canine anxiolytic for thunderstorm noise-induced stress (noise aversion). The tablet contained five highly stable triterpenes and delivered 10 mg of the active ingredient betulinic acid (BA) for an intended 1 mg/kg dose in a 10 kg dog. BA in tablets was stable for 30 months in storage at 23 °C. Efficacy of the tablets in reducing anxiety in dogs was assessed in a blinded, placebo-controlled study by recording changes in blood cortisol levels and measures of behavioral activity in response to recorded intermittent thunder. Sixty beagles were assigned into groups receiving: placebo, 0.5×, 1×, 2×, and 4× dose, or the positive control (diazepam), for five days. Reduction in anxiety measures was partially dose-dependent and the 1× dose was effective in reducing inactivity time (p = 0.0111) or increased activity time (p = 0.0299) compared with placebo, indicating a decrease in anxiety response. Cortisol measures also showed a dose-dependent reduction in cortisol in dogs treated with the test tablet.  相似文献

 In 1995, a laboratory proficiency test was carried out in Germany with labelled sediment which was analyzed by 40 participants (operators of nuclear power plants, officially authorized institutions of the German Federal States, and laboratories of research centers). Preparation and certification of the labelled material were described in detail. The deviations of the laboratory means from the corresponding certified reference values of ten radionuclides were determined. Outliers of type 2 were recognized by means of the Grubbs test and eliminated. The consistency of laboratory means and reference values was tested by a modified t test. To describe the reliability of the measurements performed, a radioanalytical quality index which has been defined elsewhere was used. By use of this index, the radioanalytical quality of laboratory means and the means of laboratory means could be evaluated and a very realistic picture of the "state-of-the-practice" of the participating laboratories obtained. Historically, due to the size and nature of the instrumentation, highly skilled laboratory professionals performed clinical testing in centralized laboratories. Today’s clinicians demand realtime test data at the point of care. This has led to a new generation of compact, portable instruments permitting ”laboratory” testing to be performed at or near the patient’s bedside by nonlaboratory workers who are unfamiliar with testing practices. Poorly controlled testing processes leading to poor quality test results are an insidious problem facing point of care testing today. Manufacturers are addressing this issue through instrument design. Providers of clinical test results, regardless of location, working with manufacturers and regulators must create and manage complete test systems that eliminate or minimize sources of error. The National Committee for Clinical Laboratory Standards (NCCLS) in its EP18 guideline, ”Quality management for unit-use testing,” has developed a quality management system approach specifically for test devices used for point of care testing (POCT). Simply stated, EP18 utilizes a ”sources of error” matrix to identify and address potential errors that can impact the test result. The key is the quality systems approach where all stakeholders – professionals, manufacturers and regulators – collaboratively seek ways to manage errors and ensure quality. We illustrate the use of one quality systems approach, EP18, as a means to advance the quality of test results at point of care. Issues of current interest to certified reference material producers are addressed. Alternative strategies for certification of matrix reference materials are discussed and the benefits of adopting a flexible, cost-effective approach are described. The difficulty of undertaking homogeneity testing where certification is to be carried out with definitive techniques capable of providing very small measurement uncertainty is discussed. Methodology is described which combines conventional screening of the candidate material for homogeneity with an additional, precise assessment of homogeneity based on isotope dilution mass spectrometry measurements. A systematic procedure for evaluating the commutability (horizontal traceability or scope) of matrix reference materials has been evaluated and shows that in some circumstances matrix effects may be less pervasive than is generally believed. This offers the possibility, especially for trace analysis applications, of more efficient use of existing reference materials without compromising measurement reliability. Vertical traceability of matrix reference material data is of growing interest but is difficult to achieve with present interlaboratory certification exercises. A modification is described which attempts to address this issue. A computer program is described, which calculates the induced activities of isotopes after irradiation in a known neutron field, thermal and epithermal neutron fluxes from the measured induced activities and from nuclear data of 2–4 monitor nuclides as well as the element concentrations in samples irradiated together with the monitors. The program was developed for operation in Windows 3.1 (or higher). The application of the program for neutron activation analysis allows to simplify the experimental procedure and to reduce the time. The program was tested by measuring different types of standard reference materials at the FRJ-2 (Research Centre, Jülich, Germany) and Triga (University Mainz, Germany) reactors. Comparison of neutron flux parameters calculated by this program with those calculated by a VAX program developed at the Research Centre, Jülich was done. The results of testing seem to be satisfactory.  相似文献

 The necessity for analytical quality assurance is primarily a feature of the analytical process itself. With the full establishment of the EU domestic market, it is also becoming a legal necessity for an increasing number of analytical laboratories. The requirements which laboratories will need to fulfil are stipulated in DIN EN 45 001. Accredited testing laboratories must in fact provide evidence that they work solely in accordance with this standard. National and EU commissions, which are legislative authorities, tend therefore to specify analytical methods, e.g. in the form of regulations or appendices thereto, intended to ensure that results from different laboratories will be comparable and hence will stand up in a court of law. The analytical quality assurance system (AQS), introduced by the Baden-Württemberg Ministry for the Environment in 1984, obliges laboratories to regularly participate in collaborative studies and thereby demonstrate their ability to provide suitably accurate analyses. This alone, however, does not sufficiently demonstrate the competence of a laboratory. Only personal appraisal of the laboratory by an auditor, together with the successful analysis of a sample provided by the same and performed under his observation, can provide proof of the competence of the laboratory. From an analytical point of view, the competence of a laboratory must be regarded as the decisive factor. Conclusions The new GPO-PAP method for the determination of triglycerides is a suitable test for routine laboratories, for manual testing as well as for automated systems. The main characteristics of this method are: 1. No sample blank measurement; 2. Excellent reproducibility; 3. Linearity range up to 8.0 mmol triglycerides/l; 4. Very good correlation to the enzymatic UV-method; 5. Applicability to mechanized systems.

As of August 2001, 38 laboratory accreditation bodies of the International Laboratory Accreditation Cooperation (ILAC) have signed the multi-lateral, mutual recognition arrangement (the ”ILAC Arrangement”) to promote the acceptance of accredited test and calibration data. This Arrangement provides significant technical underpinning to international trade. Until now, there has been no international mutual recognition agreement in laboratory accreditation, which has been a hindrance for some types of international trade. The key to the Arrangement is the developing global network of accredited testing and calibration laboratories that are assessed and recognised as being competent by ILAC Arrangement signatory accreditation bodies. The signatories have, in turn, been peer-reviewed and shown to meet ILAC’s criteria for competence. Now that the ILAC Arrangement is in place, governments can take advantage of it to further develop or enhance trade agreements. The ultimate aim is increased use and acceptance by industry, as well as government, of the results from accredited laboratories, including results from laboratories in other countries. In this way, the free-trade goal of ”a product tested once and accepted everywhere” can be realised.  相似文献
