Several plant products are known to exhibit immense medicinal value against human diseases. Our earlier studies showed that dry stem crude extract (DSCE) ofTinospora cordifolia contained a polyclonal B cell mitogen, G1-4A. DSCE as well as G1-4A also enhanced immune response in mice. In order to explore the possibility of using G1-4A/PPI (partially purified immunomodulator) to modulate radiation induced immunosuppression, the antioxidant effect of PPI from this plant was examined against reactive oxygen and nitrogen species (ROS/RNS), generated by photosensitization/peroxynitrite. Levels of lipid peroxidation products, superoxide dismutase (SOD) and catalase in liver/spleen homogenate from mouse were monitored. Photosensitization induced significant increase in thiobarbituric acid reactive substances (TBARS) in liver. The activities of SOD and catalase were reduced considerably. PPI, present during photosensitisation, prevented lipid peroxidation and restored the activities of both the enzymes. Likewise, oxidative damage induced by peroxynitrite was inhibited by PPI. The degradation of proteins due to photosensitization as assessed by SDS-PAGE was effectively reduced by simultaneous treatment with PPI during photosensitization. Selective inhibitors of ROS like mannitol, SOD, sodium azide and antioxidants, GSH and vitamin C brought about significant inhibition of formation of TBARS suggesting possible involvement of O2 ,OH and1O2. Photosensitization in deuterated buffer enhanced formation of TBARS thus indicating generation of1O2. Thus, the action of PPI may be against oxidative damage through Type I and II photosensitization mechanisms. Therefore, the immunomodulator fromTinospora cordifolia may also be beneficial as an antioxidant.  相似文献

The total phenolic and flavonoid content, phenolic composition, and in vitro antioxidant capacity of ethanolic extracts of Ximenia parviflora Benth. var. parviflora fruits collected at Zinaparo, Michoacan (in central Mexico) were determined. Fruit extracts present a high scavenging activity of 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis[3-ethylbenzothiazoline-6-sulfonic acid] radicals (71.49?±?0.11% and 85.00?±?1.29% inhibition, respectively). The four phenolic compounds identified in fruit extracts by high-performance liquid chromatography with diode array detection were gallic acid, chlorogenic acid, caffeic acid, and quercetin. X. parviflora fruits may be used as a starting material for the extraction of high value antioxidant phenolic compounds with potential applications in the pharmaceutical and dietary supplement industries.  相似文献

Proteins present within the cell layer and those released in the cell medium from in vitro cultured normal human dermal fibroblasts were separated and characterized in terms of their isoelectric point and molecular weight, by two-dimensional (2-D) gel electrophoresis. All spots in the synthetic gel were firstly analyzed by the Melanie 3 software and compared with those of breast cancer cells, colorectal epithelial cells, HL60, lymphoma cells, and platelets, already available on-line. From the identification of 144 spots from both the cell layer and the medium, we were able to recognize 89 different proteins, since a certain number of spots represented different isoforms of the same molecule. Identifications were performed by matching with on-line 2-D databases, and by matrix assisted laser-desorption/ionization-time of flight-mass spectrometry (MALDI-TOF-MS), in order to confirm the identification by matching, or to identify new proteins. The procedure we used allows (i) to design a highly reproducible reference map of the proteome of adult human normal fibroblasts in culture, (ii) to evaluate protein species produced in the cell layer as well as those released in the culture medium, and (iii) to compare data from gel matching with those obtained by MS. This work represents an essential step for a better knowledge of mesenchymal cells, given the widespread use of this cell type in both clinical and experimental investigations.  相似文献

Ageing process in cells is associated with oxidative stress. Ultraviolet A produces reactive oxygen species responsible for accumulation of DNA and cellular damage. After the evaluation of antioxidant enzyme activities and oxidative stress markers at the basal state, we have studied the responses to UVA stress of coetaneous fibroblasts, isolated from different male donors (2-88 years, n=23) in terms of cytotoxicity, genotoxicity and DNA repair capacities. For this purpose, we have determined level of DNA damage using the comet assay (single strand breaks and alkali-labile sites) and the cell cycle distribution after a 5 J/cm2 irradiation. No differences with age were observed for antioxidant enzyme activities and oxidative stress markers. DNA strand breaks after UVA irradiation (5-20 J/cm2), was found to be age-dependent. DNA repair was slow and also significantly affected by ageing. The cell cycle distribution analysis showed that high repair correlated with high proliferative capacities at basal level. Twenty-four hours after the stress, fraction of young fibroblasts blocked in G1 phase was significantly increased whereas significant modifications concerned the G2-M phase for adult and older fibroblasts. These results indicate an age-dependent decline in the DNA repair capacities correlated with modifications of the cell cycle parameters.  相似文献

An antioxidant activity of indapamide (IDP) and its metabolite (OH-IDP) is demonstrated in this study. Both IDP and OH-IDP were found to scavenge 1,1-diphenyl-2-picryl-hydrazyl free radical. The scavenging effect of OH-IDP was stronger than that of IDP. Lipid peroxidation of rat liver microsomes initiated by reduced nicotinamide adenine dinucleotide phosphate (NADPH) and adenosine diphosphate (ADP)-Fe3+ was inhibited by IDP and OH-IDP with IC50 values of about 6 and 2 microM, respectively. The lipid peroxidation in human erythrocyte membrane, induced by 2,2'-azobis-(2-amidinopropane dihydrochloride) treatment, was also inhibited by 10 microM IDP. The antioxidant capacity of OH-IDP was at almost the same level as that of alpha-tocopherol, tested for comparison. The present data show that IDP and OH-IDP at micromolar concentrations are able to trap the free radicals involved in the lipid peroxidation.  相似文献

Diverse signaling pathways have been proposed to regulate store-operated calcium entry (SOCE) in a wide variety of cell types. However, it still needs to be determined if all of these known pathways operate in a single cell type. In this study, we examined involvement of various signaling molecules in SOCE using human fibroblast cells (HSWP). Bradykinin (BK)-stimulated Ca2+ entry, previously shown to be via SOCE, is enhanced by the addition of vanadate, an inhibitor of tyrosine phosphatases. Furthermore, SOCE is regulated by cytochrome P-450, as demonstrated by the fact that the products of cytochrome P-450 activity (14,15 EET) stimulated SOCE while econazole, an inhibitor of cytochrome P450, suppressed BK-stimulated Ca2+ entry. In contrast, Ca2+ entry was unaffected by the guanylate cyclase inhibitor LY83583, or the membrane permeant cyclic GMP analog 8-bromo-cyclic GMP (8-Br-cGMP). Neither nitric oxide donors nor phorbol esters affected BK-stimulated Ca2+ entry. SOCE in HSWP cells is primarily regulated by tyrosine phosphorylation and the cytochrome P-450 pathway, but not by cyclic GMP, nitric oxide, or protein kinase C. Thus, multiple pathways do operate in a single cell type leading to the activation of Ca2+ entry and some of these signaling pathways are more prominently involved in regulating calcium entry in different cell types.  相似文献

Many lines of evidence indicate that the immortalization step is critical for the neoplastic transformation of normal human cells. Once normal human cells have been immortalized, they are relatively easily transformed into neoplastic cells. In order to understand these phenomena, patterns of protein phosphorylation in proliferating normal human fibroblast cell strains and their immortalized cell lines were compared by using two-dimensional polyacrylamide gel electrophoresis. It was found that the expression and phosphorylation levels of the human heat shock protein 27 (HSP27) were predominantly downregulated in the immortalized cells compared with those in their normal counterparts. In the normal cells, HSP27 expression and phosphorylation were markedly increased by physiological and nonphysiological stresses, such as serum addition, treatment with a carcinogenic agent like 4-nitroquinoline-1-oxide, and a high osmotic pressure. This may be a normal defense against acute changes of cellular environment and cytotoxic effects. However, these stresses had no effects on the expression and phosphorylation of HSP27 in the immortalized cells. These results suggest that an abnormal regulation of HSP27 expression and phosphorylation may be one of the reasons for easy neoplastic transformation of the immortalized cells by the treatment with carcinogenic agents.  相似文献

The culture of cells in a microbioreactor can be highly beneficial for cell biology studies and tissue engineering applications. The present work provides new insights into the relationship between cell growth, cell morphology, perfusion rate, and design parameters in microchannel bioreactors. We demonstrate the long-term culture of mammalian (human foreskin fibroblasts, HFF) cells in a microbioreactor under constant perfusion in a straightforward simple manner. A perfusion system was used to culture human cells for more than two weeks in a plain microchannel (130 microm x 1 mm x 2 cm). At static conditions and at high flow rates (>0.3 ml h(-1)), the cells did not grow in the microchannel for more than a few days. For low flow rates (<0.2 ml h(-1)), the cells grew well and a confluent layer was obtained. We show that the culture of cells in microchannels under perfusion, even at low rates, affects cell growth kinetics as well as cell morphology. The oxygen level in the microchannel was evaluated using a mass transport model and the maximum cell density measured in the microchannel at steady state. The maximum shear stress, which corresponds to the maximum flow rate used for long term culture, was 20 mPa, which is significantly lower than the shear stress cells may endure under physiological conditions. The effect of channel size and cell type on long term cell culture were also examined and were found to be significant. The presented results demonstrate the importance of understanding the relationship between design parameters and cell behavior in microscale culture system, which vary from physiological and traditional culture conditions.  相似文献

Numerous original alkaloids are present in tubers of Arisarum vulgare Targ. a species belonging to the Araceae family known in Morocco for its toxicity. Some previous works deal with the activity of these natural compounds as R-irniine. As the enantioselective total synthesis of irniine has been realised, the R- and S-forms were obtained and this last one could be tested. Thus, a study of the cytotoxicity and the electrophysiological activity of S-irniine was carried out on human MRC-5 fibroblasts. A cytotoxic effect of S-irniine at 40 microg/mL was detected on MRC-5 fibroblasts. An electrophysiological study was also carried out on the MRC-5 cells by using the patch-clamp technique and no effect of this compound at this concentration on the outward potassium current of MRC-5 fibroblasts was observed. Thus, this study showed, as it was for R-irniine, that the cytotoxicity of S-irniine was not explained by an effect on the potassium currents.  相似文献

Abstract

L-Ascorbic acid (AA), known as vitamin C, can form browning products by a non-enzymatic process during storage and the browning products cause deterioration of agricultural products. In the browning reaction, a red pigment, 2,2´-nitrilodi-2(2´)-deoxy-L-ascorbic acid ammonium salt (NDA), is generated from AA via L-scorbamic acid (SCA) as an intermediate. However, the biological activities of SCA and NDA have not yet been clarified. In this study, we assayed the antioxidant activities of SCA and NDA using ABTS radical cation and their neurite outgrowth-enhancing activities in PC12 cells. SCA showed stronger radical-scavenging activity than that of AA, while NDA hardly showed any activity. SCA and NDA enhanced the neurite outgrowth induced by dibutyryl cyclic AMP after their incorporation into cells in the same manner as that of AA. The results indicated that SCA has antioxidant activity and that SCA and NDA have neurite outgrowth-enhancing activity.  相似文献

Interfacial tension and interfacial adsorption parameters for benzyloctadecyldimethyl ammonium chloride (BODMAC) in three organic diluents were determined and interpreted. The interfacial activity of BODMAC is affected by the type of the organic diluent and the composition of the aqueous phase. The general order of interfacial activity of BODMAC is n-heptane (5% isobutanol) > carbon tetrachloride > chloroform. The effectiveness of adsorption of BODMAC is not only dependent on the organic diluent, but also on the inorganic electrolytes in the aqueous phase.  相似文献

Antioxidant activity of hydroxystilbene derivatives in homogeneous solution   总被引:3,自引:0,他引:3  
The antioxidant activity of the cis and trans isomers of several analogues of resveratrol and pterostilbene has been investigated, especially with regard to the effect of the stereochemistry about the olefinic double bond. The antioxidant power of these compounds was estimated by measuring the rate constants for their reactions with peroxyl radicals and, with two of them, the bond dissociation enthalpy (BDE) of the phenolic O-H bond which is cleaved in the inhibition reaction. The present data show that in homogeneous solution the various hydroxystilbenes investigated behave as mild antioxidants with the notable exceptions of the trans isomer of 4 and 6, whose activities are only slightly lower than that of alpha-tocopherol (vitamin E). The rate constants of the inhibition reaction show that the antioxidant activity of the cis-hydroxystilbene is in all the examined cases worse, by a factor ranging between 2 and 6, than that of the corresponding trans isomers. This lower reactivity depends on enthalpy factors as it can be inferred by the experimental values of the O-H bond dissociation enthalpy in the two geometric isomers of 3',5'-di-tert-butyl-4'-hydroxy-3,5-dimethoxystilbene showing that the strength of the O-H bond in the cis isomer is larger by 1.8 kcal/mol. DFT calculations provide a rationalization of this result, indicating that, although the cis geometry implies a destabilization with respect to the trans species of both phenoxyl radical and parent hydroxystilbene, the destabilization of the radical is larger because the folding of the structure strongly reduces the delocalization of the unpaired electron on the styryl group. A comparison of these results with previously reported data on the proapoptotic activity of these stilbenoids suggests that these two properties are not correlated.  相似文献
