氮离子注入选育耐酸性α-淀粉酶产生菌诱变效应的研究

利用低能(30keV)氮离子注入中温中性α-淀粉酶产生菌枯草芽孢杆菌BF7658,研究对其产耐酸性α-淀粉酶的诱变效应.结果表明,BF7658菌株存活率曲线是典型的"马鞍型"剂量-效应曲线,在"马鞍型"区域对应注入剂量50×1014～100×1014 ions/cm2下具有较高的突变率.通过诱变筛选到一株最适作用pH为5.0,较出发菌株的最适作用pH偏低一个单位的突变株,编号TUST743.该突变株产酸性α-淀粉酶的酶活为343U/mL.该酶在pH4.5和4.0条件下的相对酶活为61%和38%,说明此突变株所产酸性α-淀粉酶对低pH有较好的耐受性.经连续传代实验,表明该菌株遗传性质稳定.

克拉维酸高产菌的选育

在克拉维酸高产菌的选育过程中,应用解除终产物结构类似物作为选择压力,能有效地除去低效价菌株,提高高产菌株的检出率,较传统筛选法优越.试验表明,以带棒链霉菌Strep tomycesclavuligerusCCRC11518(效价为382.4×10-6g/mL)为出发菌,经紫外线诱变,筛选到一株抗20×10-3g/mL的舒巴坦钠突变株my51,my51菌株产克拉维酸的效价达到834.7×10-6g/mL,比出发菌提高了1.18倍.该菌株连续传代7代,克拉维酸的效价保持稳定.     

链霉素抗性突变理性筛选链霉素高产菌株

以灰色链霉菌(Streptomyces griseus) CICC11002为出发菌株,采用紫外诱变并结合链霉素抗性筛选法育种,以期获得高产菌株.灰色链霉菌孢子悬液经90％紫外致死剂量诱变后,在含有链霉素最小抑制浓度(0.8μg/mL)的培养基平板上,分离得到105株链霉素抗性突变菌.其中链霉素产量高于出发菌株的有19株,正突变率高达18.1％,同时获得了产链霉素能力约为出发菌株2倍的突变株SM-55.     

利福平及杆菌肽抗性突变对黄单胞菌产胶率的影响

黄单胞菌（Xanthomonas Campestris．X．C．）的利福平及杆菌肽抗性突变与产黄原胶（xanthan gum）的能力正相关。从出发菌株X．C．NK-01中筛选自发突变的利福平抗性（rifampicin—resistent．Rif）和抗菌肽抗性（bacitracin—resistent，Baci）菌株，然后通过28℃，200r／min的250ml摇瓶发酵试验，筛选到两株Rifr及两株Bacir菌株。其产量分别比出发菌株提高了20％和30％，黄原胶粘度分别提高了20％-30％。分析了利福平和杆菌肽抗性突变菌株产胶能力提高的可能生化因素。     

沼泽红假单胞菌高产辅酶Q10菌株的诱变选育研究

以沼泽红假单胞菌为出发菌株,通过紫外诱变和罗红霉素抗性筛选,以期获得辅酶Q10高产菌株.试验结果表明,紫外照射时间60-70s时可能达到最大正向突变概率.通过紫外诱变、罗红霉素初筛和发酵复筛,获得一株沼泽红假单胞菌突变株R-1,辅酶Q10产量达到0.021g/L,较出发菌株产量（0.018g/L）提高16％.经过6次传代培养,突变菌株产辅酶Q10产量稳定,可用于进一步研究.     

纤维素降解菌的筛选与诱变育种

采用羧甲基纤维素(CMC)平板初筛和50℃培养,筛选到一株纤维素酶活力较高的耐高温霉菌MY菌株,其CMC培养基最适pH为6.0,培养温度为40℃,最佳产酶时间为5d.以MY菌株为出发菌株,分别采用紫外线和硫酸二乙酯诱变,以透明圈直径与菌落直径的比值(HC值)提高30%以上或菌落形态发生明显变异为筛选指标,共筛选到97株突变株;通过测定粗酶液CMC酶活力,从中筛选出29株CMC酶活力提高30%以上的菌株;结合突变菌株的传代稳定性实验,最后筛选得到1株性能优良的霉菌MY004突变株,其CMC酶活力比原始菌株MY提高了80.6%.     

He-Ne激光对红曲霉ZL307的诱变育种

以红曲霉ZL307为出发菌株,利用氦-氖（He-Ne）激光对其单孢子悬液进行诱变育种．通过计算存活率和正突变率,确定激光诱变育种的处理条件为电流强度20mA,照射时间50min．通过平板初筛和摇瓶发酵进行复筛,选育出突变株红曲霉ZZ307．相对于出发菌株,实验不仅缩短了菌种的培养时间,而且产色素色价达到100．39U／mL（提高了34％）,色价达到峰值时间提前1d．进行10代遗传稳定性实验,突变菌株仍具有良好的遗传稳定性．     

2-脱氧葡萄糖抗性黑曲霉高产柠檬酸突变株的选育

以黑曲霉 W3为出发菌株 ,通过 60 Co-γ射线诱变 ,在以甘油为碳源的基本培养基上 ,选育出2 -脱氧葡萄糖 (2 - DG)抗性突变株。以玉米粉为原料的柠檬酸发酵的结果表明 ,产酸高于出发菌株(8.4 8g/ 10 0 m L )的突变株约占 13.5% ,其中突变株 KI- 5产酸比 W3菌株提高 18%。     

链霉菌702在林可霉素致死剂量下UV诱变研究

在林可霉素致死剂量下UV诱变筛选链霉菌702产抗细菌活性物质高产菌种,试验结果表明,紫外作用20 s对林可霉素致死突变率达到20.77%,抗药性致死突变菌株与该菌产抗细菌活性物质的高产菌株的正突变率达25%,效价提高20%以上达10%,从突变菌株中摇瓶筛选到42-3473菌株,摇瓶发酵生物效价达2062μg/mL,比诱变出发菌株生物效价提高了70%。     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
