We characterized alterations in the ability of concussive brain injury (CBI) models to perform a water-finding task and examined effects of (+)-eburnamenine-14-carboxylic acid (2-nitroxyethyl) ester (VA-045), a novel apovincaminic acid derivative, on post-traumatic impairments in latent learning and memory processes. Two types of CBI-induced amnesia (retrograde and anterograde) were produced by means of post- or preacquisition head impact using a simple weight-drop device. Profound impairments of latent learning and memory processes related to retention and retrieval were observed in the CBI mice. In the CBI-induced retrograde amnesia model, VA-045 (0.5-4.0 mg/kg) significantly ameliorated impairments of latent learning and retention in both the retention test and the retest. In the CBI-induced anterograde amnesia model, the protective effects of the compound on impairments in latent learning and retention or retrieval were significant in both the retention test and the retest. These results suggested that VA-045 may be a novel cognitive enhancer for attenuating or protecting against the learning and memory dysfunction associated with CBI.  相似文献

Controversy surrounding dissociative identity disorder (DID) has focused on conflicting findings regarding the validity and nature of interidentity amnesia, illustrating the need for objective methods of examining amnesia that can discriminate between explicit and implicit memory transfer. In the present study, the authors used a cross-modal manipulation designed to mitigate implicit memory effects. Explicit memory transfer between identities was examined in 7 DID participants and 34 matched control participants. After words were presented to one identity auditorily, the authors tested another identity for memory of those words in the visual modality using an exclusion paradigm. Despite self-reported interidentity amnesia, memory for experimental stimuli transferred between identities. DID patients showed no superior ability to compartmentalize information, as would be expected with interidentity amnesia. The cross-modal nature of the test makes it unlikely that memory transfer was implicit. These findings demonstrate that subjective reports of interidentity amnesia are not necessarily corroborated by objective tests of explicit memory transfer. Recent developments in the functional and neural bases of several aspects of memory are described including long term cortical memory storage, the transition from immediate to permanent memory mediated by medial temporal structures, working memory, memory retrieval, and implicit memory. These are linked to current data on the nature of anterograde and retrograde amnesia in the degenerative diseases, and also to issues in the clinical diagnosis of memory impairments. Understanding the bases of memory can inform the diagnosis of memory impairments in degenerative diseases, and the patterns of impairment seen in the degenerative diseases can help contribute to knowledge of the mechanisms of normal memory.  相似文献

There have been few studies of psychogenic amnesia based on a cognitive or neuropsychological framework. In the present study, a patient with acute onset of profound psychogenic retrograde amnesia was examined. Although her performance on neuropsychological tasks revealed intact anterograde memory, language functioning, visuospatial and constructional skills, and mental speed and flexibility, she displayed severe impairments on a variety of retrograde memory tasks. Furthermore, initial observations revealed inconsistencies between the patient's recall of semantic knowledge on direct questioning and her ability to demonstrate the use of this knowledge on indirect tasks. To test this formally, we devised an indirect remote knowledge task to examine a possible dissociation between explicit and implicit memory. Two healthy subjects matched for age, gender, education, occupation, and estimated IQ were also tested. As predicted, the findings demonstrate implicit knowledge despite impaired explicit recall for the same material.  相似文献

We present an overview of two of our ongoing projects relating processes in the hippocampus to memory. We are trying to understand why retrograde amnesia occurs after damage to the hippocampus. Our experiments establish the generality of several new retrograde amnesia phenomena that are at odds with the consensus view of the role of the hippocampus in memory. We show in many memory tasks that complete damage to the hippocampus produces retrograde amnesia that is equivalent for recent and remote memories. Retrograde amnesia affects a much wider range of memory tasks than anterograde amnesia. Normal hippocampal processes can interfere with retention of a long-term memory stored outside the hippocampus. We conclude that the hippocampus competes with nonhippocampal systems during memory encoding and retrieval. Finally, we outline a project to understand and manipulate adult hippocampal neurogenesis in order to repair damaged hippocampal circuitry to recover lost cognitive functions. A male patient suffering from a generalized functional retrograde amnesia, for all autobiographical information, was admitted to a psychiatric inpatient facility. Psychological testing indicated possible sexual assault. Following five hypnotic sessions, the patient regained his memory, and a violent sexual attack by two males was validated. The amnesia is discussed in terms of the constructs of implicit versus explicit memory, and reasons why a male rape victim might suffer such an extreme reaction. Twenty patients (mean age 64 years) with a previous episode of transient global amnesia (TGA) were examined to assess the functioning of objective memory (by using the Randt Memory Test), the metamemory capacities (Sehulster Memory Scale), the residual level of retrograde amnesia (Questionnaire of Remote Events), and the level of depression (Geriatric Depression Scale). Patients with residual retrograde amnesia scored significantly lower than non-amnesic ones on indices of both short-term and long-term memory, and for one of three main metamemory components, namely self-rating of memory functioning through comparison with memory functioning of peers (Set3). Age, time interval from TGA attack and TGA duration did not prove to influence memory and metamemory scores. Retrograde amnesia and depression were rather substantially associated (1/5), and this association was found to negatively influence nearly all memory and metamemory scores. Depression level showed a positive correlation with short-term memory functioning in non-amnesics. The different pattern and strength of the relationships between metamemory components and objective memory dimensions observed in amnesics and non-amnesics indicate that metamemory evaluations are more closely related to memory functioning in amnesics than in non-amnesics.  相似文献

There is a popular hypothesis that performance on implicit and explicit memory tasks reflects 2 distinct memory systems. Explicit memory is said to store those experiences that can be consciously recollected, and implicit memory is said to store experiences and affect subsequent behavior but to be unavailable to conscious awareness. Although this division based on awareness is a useful taxonomy for memory tasks, the authors review the evidence that the unconscious character of implicit memory does not necessitate that it be treated as a separate system of human memory. They also argue that some implicit and explicit memory tasks share the same memory representations and that the important distinction is whether the task (implicit or explicit) requires the formation of a new association. The authors review and critique dissociations from the behavioral, amnesia, and neuroimaging literatures that have been advanced in support of separate explicit and implicit memory systems by highlighting contradictory evidence and by illustrating how the data can be accounted for using a simple computational memory model that assumes the same memory representation for those disparate tasks. A 54 year old patient of average intelligence with a severe and enduring loss of old autobiographical memories after herpes simplex type 1 infection is described. She was tested with a comprehensive neuropsychological battery two years after the infection. Special emphasis was laid on examining different aspects of retrograde memory. The neurological examination involved MRI and SPECT. Brain damage was found mainly in the right temporofrontal region, but minor left sided damage to this region seems possible. The patient was in the normal or slightly subnormal range for all tested anterograde memory functions, but manifested severe retrograde memory deficits with respect to episodic old memories and more moderate deficits in tests of general knowledge (semantic old memories). It is concluded that the ecphory of old autobiographical memories relies heavily on an activation of the right lateral temporofrontal junction area, but that probably only some complementary left hemispheric damage to these regions will lead to major and persistent retrograde amnesia. Alternatively, the disconnection between major prefrontal and posterior cortical regions may provide a basis for retrograde amnesia.  相似文献

Existing public events tests for retrograde amnesia quickly become out of date and require frequent restandardization. The initial development of the Oscar Test is reported, which requires Ss to match the titles of Academy Award winning movies with the years in which they were made, a procedure that updates itself automatically every year. Data from 25 normal Ss and 7 patients with severe anterograde amnesia are presented. As a group, the patients demonstrated significant impairment, misdating movies by an average of 14 yrs. Several individual amnesic patients were much more impaired and produced profiles similar to those found on other remote memory tests. Performance on the Oscar Test tended to parallel performance on the Autobiographical Memory Interview in revealing mild, time-limited retrograde amnesia in some patients and severe, temporally extensive retrograde amnesia in others. Three major types of amnesia, i.e. selective deficit of memory formation without disturbance of immediate memory have been recognized clinically. The hippocampal type is characterized by relatively short retrograde amnesia and preserved insight into his own amnesic trouble. Persistent amnesia is always associated with bilateral lesion. The diencephalon type usually shows longer retrograde amnesia and poor insight. Unilateral lesion is usually modal in deficit, i.e. verbal in the left and non-verbal in the right, and lacks clear evidence of retrograde amnesia. Responsible locus within the thalamus is still controversial. Our experience point to the mammilothalamic tract. The basal forebrain type is characterized by disorganized and confused recollection of the memorized items. Cued recall is fair compared to voluntary recall. Two special types of amnesic syndrome, i.e. transient global amnesia and isolated focal retrograde amnesia are also important clinical entities, although their etiologies and responsible lesions are not clear yet.  相似文献

This article attempts a synthesis of the range of disorders that have been subsumed under the rubric of retrograde amnesia. At a functional level, it is possible to make distinctions between various forms of retrograde amnesia, including a distinction between episodic amnesia for personally experienced events and semantic retrograde amnesia for components of knowledge, such as those relating to people and events. At an anatomical level, discrete lesions to limbic-diencephalic structures usually result in a limited degree of retrograde amnesia. Marked episodic or marked semantic retrograde amnesia is usually associated with significant involvement of cortical and neocortical structures. Retrograde amnesia is a functionally heterogeneous rather than a unitary phenomenon. Discontinuities and dissociations found in published studies point to the potential fractionation of retrograde amnesia into component disorders, each with its own neural profile. We studied 11 patients with transient global amnesia (TGA) and ten patients with functional retrograde amnesia (FRA). Patients with TGA had a uniform clinical picture: a severe, relatively isolated amnesic syndrome that started suddenly, persisted for 4-12 h, and then gradually improved to essentially normal over the next 12-24 h. During the episode, the patients had severe anterograde amnesia for verbal and non-verbal material and retrograde amnesia that typically covered at least two decades. Thirty hours to 42 days after the episode, the patients had recovered completely and performed normally on tests of anterograde and retrograde amnesia. By contrast, patients with FRA had a sudden onset of memory problems that were characterized by severe retrograde amnesia without associated anterograde amnesia and with a clinical presentation that otherwise varied considerably. The episodes persisted from several weeks to more than two years, and some of the patients had not recovered at the time of our last contact with them. The uniform clinical picture of TGA and the variable clinical picture of FRA presumably reflect their respective neurologic ('organic') and psychogenic ('non-organic') aetiologies.  相似文献

We describe a 44-year-old patient who had a transient attack of autobiographic amnesia. When assessed during the attack, her learning abilities were normal, with no sign of anterograde amnesia. In the remote memory domain, she showed a striking dissociation between a detailed knowledge of public events and famous people and a complete loss of autobiographic information. During the attack, EEG recorded bilateral frontotemporal slow waves and single-photon emission CT (SPECT) showed hypoperfusion in the right temporal and parietal lobes; no abnormalities were detected when both EEG and SPECT were repeated 1 week later. This case provides evidence for an organic etiology for the episode and supports the hypothesis that autobiographic memory is independent of other forms of retrograde memory.  相似文献

Dissociation of classification and recognition in amnesia is widely taken to imply 2 functional systems: an implicit procedural-learning system that is spared in amnesia and an explicit episodic-learning system that is compromised. We argue that both tasks reflect the global similarity of probes to memory. In classification, subjects sort unstudied grammatical exemplars from lures, whereas in recognition, they sort studied grammatical exemplars from lures. Hence, global similarity is necessarily greater in recognition than in classification. Moreover, a grammatical exemplar's similarity to studied exemplars is a nonlinear function of the integrity of the data in memory. Assuming that data integrity is better for control subjects than for subjects with amnesia, the nonlinear relation combined with the advantage for recognition over classification predicts the dissociation of recognition and classification. To illustrate the dissociation of recognition and classification in healthy undergraduates, we manipulated study time to vary the integrity of the data in memory and brought the dissociation under experimental control. We argue that the dissociation reflects a general cost in memory rather than a selective impairment of separate procedural and episodic systems. To examine the status of conceptual memory processes in amnesia, a conceptual memory task with implicit or explicit task instructions was given to amnesic and control groups. After studying a list of category exemplars, participants saw category labels and were asked to generate as many exemplars as possible (an implicit memory task) or to generate exemplars that had been in the prior study list (an explicit memory task). After incidental deep or shallow encoding of exemplars, amnesic patients showed normal implicit memory performance (priming), a normal levels-of-processing effect on priming, and impaired explicit memory performance. After intentional encoding of exemplars, amnesic patients showed impaired implicit and explicit memory performance. Results suggest that although amnesic patients can show impairments on implicit and explicit conceptual memory tasks, their deficit does not generalize to all conceptual memory tasks. In anterograde amnesia (AA), memory loss is obtained for events that occur subsequent to the traumatic insult. But because the effects of an anterograde agent or treatment usually last for minutes or even hours after the nominal training event, processing of information may be altered during the postacquisition period as well as during acquisition. Since posttraining manipulations are themselves capable of modulating memory or inducing retrograde amnesia, the memory loss due to a putative anterograde treatment may instead represent retrograde processes. The present experiment examined this source of confounding by using an amnestic treatment that can be quickly reversed after training to remove postacquisition effects. The presence of amnesia would isolate anterograde contributions as the source of loss. For induction of AA, 56 male albino Sprague-Dawley rats were trained while at reduced body temperature (29°C). A rapid rewarming procedure was introduced for some immediately after training to ensure that the hypothermic state did not extend into the postacquisition period. Other Ss were rapidly rewarmed 1 hr after training to control for any effects of the rewarming manipulation. Both groups showed severe AA that was indistinguishable from that obtained in the gradually rewarmed controls. Data provide an empirical example of an anterograde-induced memory deficit that is independent of retrograde influences. The gamma aminobutyric acid-A (GABAA) agonist, muscimol, the glutamate N-methyl-D-aspartate (NMDA) receptor antagonist, D-2-amino-5-phosphonopentanoic acid (AP5), and the inhibitor of the extracellularly regulated kinases (ERKs), UO 126, cause retrograde amnesia when administered to the hippocampus. In the present study, the authors found that they all cause retrograde amnesia for 1-trial inhibitory avoidance, not only when infused into the dorsal CA1 region of the hippocampus, but also when infused into the basolateral amygdala or the entorhinal, parietal, and posterior cingulate cortices. The posttraining time course of the effect of each drug was, however, quite different across brain structures. Thus, in all of them, NMDA receptors and the ERK pathway are indispensable for memory consolidation, and GABAA receptor activation inhibits memory consolidation: but in each case, their influence is interwoven differently. The 11-year longitudinal study of a right-handed male patient, L. C., who suffered from a severe amnesic syndrome following a softening in the right thalamus, is reported. Memory impairment involving retrograde and long-term anterograde memory, both verbal and spatial, persisted without modification. Investigation revealed some residual implicit learning ability. Positron emission tomography studies in the resting state displayed a bilateral hypometabolism of the mesial frontal lobes. Evidence suggests that a lesion confined to the thalamus may not on its own account for severe amnesia; that involvement of other structures is necessary for severe amnesia to appear; and that a functional investigation should always be included in cases of small thalamic lesions before drawing conclusions about the structures responsible for a given deficit. Stress blocks hippocampal primed-burst potentiation, a low threshold form of long-term potentiation, thereby suggesting that stress should also impair hippocampal-dependent memory. Therefore, the effects of stress on working (hippocampal-dependent) and reference (hippocampal-independent) memory were evaluated. Rats foraged for food in seven arms of a 14-arm radial maze. After they ate the food in four of the seven baited arms, they were placed in an unfamiliar environment (stress) for a 4-hr delay. At the end of the delay they were returned to the maze to locate the food in the 3 remaining baited arms. Stress impaired only working memory. Stress interfered with the retrieval of previously stored information (retrograde amnesia), but did not produce anterograde amnesia. Stress appears to induce a transient disruption of hippocampal function, which is revealed behaviorally as retrograde amnesia and physiologically as a blockade of synaptic plasticity.
