Spatial distribution, diversity, and composition of bacterial communities within the shallow sub-seafloor at the deep-sea hydrothermal field of the Suiyo Seamount, Izu-Bonin Arc, Western Pacific Ocean, were investigated. Fluids were sampled from four boreholes in this area. Each borehole was located near or away from active vents, the distance ranging 2–40 m from active vents. In addition, fluids discharging from a natural vent and ambient seawater were sampled in this area. We extracted DNA from each sample, amplified bacterial 16S rRNA genes by PCR, cloned the PCR products and sequenced. The total number of clones analyzed was 348. Most of the detected phylotypes were affiliated with the phylum Proteobacteria, of which the detection frequency in each clone library ranged from 84.6% to 100%. The bacterial community diversity and composition were different between hydrothermal fluids and seawater, between fluids from the boreholes and the vent, and even among fluids from each borehole. The relative abundances of the phylotypes related to Thiomicrospira, Methylobacterium and Sphingomonas were significantly different among fluids from each borehole. The phylotypes related to Thiomicrospira and Alcanivorax were detected in all of the boreholes and vent samples. Our findings provide insights into bacterial communities in the shallow sub-seafloor environments at active deep-sea hydrothermal vent fields. 相似文献

A sediment sample was collected from a deep-sea hydrothermal vent field located at a depth of 2 951 m on the Southwest Indian Ridge. Phylogenetic analyses were performed on the prokaryotic community using polymerase chain reaction(PCR) amplification of the 16 S rRNA and nifH genes. Within the Archaea, the dominant clones were from marine benthic group E(MBGE) and marine group I(MGI) belonging to the phyla Euryarchaeota and Thaumarchaeota, respectively. More than half of the bacterial clones belonged to the Proteobacteria, and most fell within the Gammaproteobacteria. No epsilonproteobacterial sequence was observed. Additional phyla were detected including the Actinobacteria, Bacteroidetes, Planctomycetes, Acidobacteria, Nitrospirae, Chloroflexi, Chlorobi, Chlamydiae, Verrucomicrobia, and candidate divisions OD1, OP11, WS3 and TM6, confirming their existence in hydrothermal vent environments. The detection of nifH gene suggests that biological nitrogen fixation may occur in the hydrothermal vent field of the Southwest Indian Ridge. Phylogenetic analysis indicated that only Clusters I and III NifH were present. This is consistent with the phylogenetic analysis of the microbial 16 S rRNA genes, indicating that Bacteria play the main role in nitrogen fixation in this hydrothermal vent environment. 相似文献

以青岛、威海水域夏冬两季表层沉积物中的基因组DNA为模板,扩增沉积物样品中细菌的16S rDNA 片段,并构建其相应文库.进行16S rDNA序列分析,并与数据库中的序列进行比对.分析结果表明:2个站位沉积物样品细菌分别属于11个主要细菌门类,包括:变形菌门(Proteobacteria)、浮霉菌门(Plancomycene)、芽单胞菌门(Gemmatimonadetes)、酸杆菌门(Acidobacteria)、硝化螺菌门(Nitrospirae)、疣微菌门(Verrucomicrobial)、拟杆菌门(Bacteroidetes)、绿屈挠菌门(Chloroflexi)、厚壁菌门(Firmicutes)、高G+C含量革兰氏阳性菌放线菌门(Actinobacteria)和低G+C含量革兰氏阳性菌梭菌门(Clostridiales).其中变形菌门是主要类群,且γ-变形菌纲(γ-Proteobacteria)占优势.与QD2站位相比,QD1站位沉积物细菌多样性随季节变化不大. 相似文献

对我国沿海地区10个菲律宾蛤仔野生群体线粒体16S rRNA和COI基因部分序列进行测序,分别得到了长度为473bp和632bp的片段。结果表明,16S rRNA 193条序列A+T平均含量为66.6%,共检测到21个变异位点,193个个体具有22种单倍型;COI基因183条序列A+T平均含量为64.8%,共检测到126个变异位点,183个个体具有67种单倍型。基于群体间遗传距离利用Mega5.1软件构建10个群体的NJ 树,聚类结果表明,大连群体和荣成群体聚为一支,其余8个群体聚为一支。AMOVA分析表明,大连群体和荣成群体间分化不显著,而荣成、大连群体与其余8个群体间的分化达到极显著水平(P<0.01),说明我国沿海的菲律宾蛤仔野生群体存在一定的遗传分化。 相似文献

研究海水环境中Q235碳钢锈层中微生物群落的多样性。应用16SrDNA基因文库技术,分别对实海全浸一年的Q235碳钢内外锈层中的微生物进行基因文库的构建和分析。通过克隆测序构建了含有64个OTUs的细菌16SrRNA基因文库,结果表明锈层中微生物种类丰富,主要分布于13个已知的菌门,包括变形菌门;拟杆菌门;硅藻门;酸杆菌门;浮霉菌门;硝化螺旋菌门;厚壁菌门;绿弯菌门;疣微菌门;绿菌门;放线菌门;红藻门以及螺旋体门。内外锈层的优势菌均为变形细菌门,分别占文库序列的46%和53%,其中α-变形菌纲又为门中优势类群,但在内外锈层中还存在明显的系统发育学分歧。 相似文献

为分析凡纳滨对虾(Litopenaeus vannamei)海水养殖过程中水体细菌群落多样性变化,本研究采用Illumina测序技术对福建漳浦县凡纳滨对虾海水养殖中后期水体样品进行16S rRNA基因高通量测序,同时进行水质参数测定。结果表明,养殖过程中,氨氮(NH4+-N)、化学需氧量(COD)值波动较大,其他水质参数值波动较小。检测到的细菌种群归属于19个门、35个纲、80个目、135个科、254个属,说明该养殖水体中细菌群落具有高度的多样性。变形菌门和拟杆菌门是养殖期间主要优势类群,不同养殖时期细菌组成存在一定的波动,但没有明显的变化规律。蓝细菌在第67天、115天表现出高丰度,且其丰度在养殖期间变化较大(比例变化为0.7%~63.9%)。此外,益生菌和潜在致病菌丰度在养殖期间很低。冗余分析(Redundancy analysis,RDA)表明,对细菌群落影响最大的水质因子是化学需氧量与硝酸盐浓度比值(COD/NO3-)和COD;降低溶氧可以增加细菌丰富度。总之,本研究初步揭示了对虾养殖中后期水体微生物的演替规律,发现水质因子对养殖水体微生物组成具有显著影响,研究结果为凡纳滨对虾健康养殖提供一定的参考意义。 相似文献

The dietary importance of marsh vascular plants (primarilySalicornia virginica), algae and upland particulate inputs to macro-invertebrate consumers was studied in Carpinteria Salt Marsh, southern California, using stable carbon and nitrogen isotope ratios. This marsh is predominantly a marine or hypersaline system and succulents are the most common vascular plant species. Of invertebrates collected from the vegetated marsh, tidal flats and channels, only detritivores from the vegetated marsh (Traskorchestia traskiana,Melampus olivaceus) had isotope values (δ13C=−20‰) that suggested some use ofSalicornia-derived carbon.T. traskianacultured in the laboratory on decomposingS. virginicaor blue-green micro-algal mat had distinctive isotopic signatures, reflecting the capability of this consumer to assimilate carbon and nitrogen derived from these sources. The δ13C values (generally −16‰ to −15‰) of species from tidal flats and channels (e.g.Cerithidea californica,Protothaca staminea,Mytilus galloprovincialis,Neotrypaea californiensis) were most similar to values for benthic algae and phytoplankton. Specimens ofM. galloprovincialisalong a gradient of presumed increase in marine influence had similar isotope values, suggesting little contribution to diet from upland runoff. The present results differ most noticeably from published values in the13C enrichment of suspension-feeders, suggesting the use of resuspended13C-enriched benthic microalgae in tidal channels by these consumers, and in the13C depletion and15N enrichment of plants and consumers along a portion of the marsh boundary receiving inputs of nutrient-enriched perched groundwater. In general, the isotopic composition of macro-invertebrates indicated the incorporation of algal production rather than ofS. virginicaor upland sources into the marsh food web. 相似文献

Soil core samples fromRhizophora mangleL. andAvicennia schauerianaStapf & Leech forests from south-eastern Brazil were analysed for their total organic matter content and their sugars, amino acid and amino sugars composition. Organic carbon and nitrogen contents were higher inAvicenniathan inRhizophorasoils. The contribution of sugars and amino acids to the total organic carbon pool was constant with depth inRhizophorasoils whereas inAvicenniasoils it increased. Spectral distribution of sugars and amino acids showed a dominance of Ca-affine monomers, particularly acidic amino acids, and the sugar arabinose. Biogeochemical indicators derived from ratios of individual sugar and amino acid monomers confirm previous studies which showed that organic matter in both soils is mainly of mangrove origin. The results further suggest accumulation of organic matter inRhizophorasoils and a continuing degradation of organic matter inAvicenniasoils. The latter may thus release more nutrients to adjacent ecosystems thanRhizophorasoils. 相似文献

34S of sinking material from Otsuchi Bay was measured at the center and rocky shore of the bay. At the center of the bay δ
34S was high (18∼21‰) in the material collected from April to November. However, δ
34S was low (9∼14‰) in the material collected from December to March. The increase in δ
34S in April was attributed to an increase in phytoplankton biomass because marine phytoplanktonic δ
34S is high. When δ
34S of sinking material was low, input of riverine material or bottom sediment resuspension were considered as the probable
causes, because their δ
34S is low. Marine sulfur was always high (more than 70%) at both stations. The difference between the δ
34S of sinking material collected from the different sampling stations indicates that marine macroalgae contribute to sinking
material near the shore when phytoplankton is scarce. In conclusion, the relative influence of different material sources
to sinking materials could be successfully estimated using δ
