The virulence and transmissibility of viruses are highly associated with their binding specificity to the host cell receptor. In influenza, this initial event of viral pathogenesis is mediated by a glycoprotein known as hemagglutinin (HA). In the present study we constructed homology models of the chain A of hemagglutinin (HA1) of 2009 swine influenza strain. The modeled proteins were compared with atomic coordinates of 1918 (Spanish flu strain) and 1930 HA1 (swine influenza strain). HA1 of recent swine influenza strain showed 84.83% and 93.14% homology with the same versions of 1918 and 1930 strains, respectively. Discrepancies in multiple sequence alignment particularly at the ligand-binding residues notified its receptor specificity to α-2,6 sialic acids in 1918 and 2009 viral strains in contrast to α-2,3 sialic acids as found in 1930 swine flu strain. This implicated the relatively closer relationship of 2009 strain with 1918 strain rather than swine origin strain of 1930. Similarly, the spatial orientations of receptor-binding residues, located in 190-helix, 130-loop and 220-loop, were found more aligned in 1918 and 2009 (RMSD 0.98 Å) than in 1930 and 2009 (RMSD 1.06 Å) strains HA1. More similarities were established between both human origin influenza viruses (1918 and 2009 strains) by the receptor-binding cavity architecture and the orientation of protease cleavage site (Arg327). Briefly, the present finding is expected to show molecular discrepancies and congruencies among the recent and past pandemic influenza strains and may also potentially illustrate the drug targets to rein the infection at earlier stages.  相似文献

To gain insight into the possible origin of the hemagglutinin of 2009 outbreak, we performed its comparative analysis with hemagglutinin of influenza viral strains from 2005 to 2008 and the past pandemics of 1977, 1968, 1957 and 1918. This insilico analysis showed a maximum sequence similarity between 2009 and 1918 pandemics. Primary structure analysis, antigenic and glycosylation site analyses revealed that this protein has evolved from 1918 pandemic. Phylogenetic analysis of HA amino acid sequence of 2009 influenza A(H1N1) viruses indicated that this virus possesses a distinctive evolutionary trait with 1918 influenza A virus. Although the disordered sequences are different among all the isolates, the disordered positions and sequences between 2009 and 1918 isolates show a greater similarity. Thus these analyses contribute to the evidence of the evolution of 2009 pandemic from 1918 influenza pandemic. This is the first computational evolutionary analysis of HA protein of 2009 H1N1 pandemic.  相似文献

The impact of the 1918 influenza pandemic on human fertility has been subject to significant scholarly debate. The current study characterizes the inter-temporal association between excess deaths during the pandemic and the subsequent birth deficit by identifying the length of time between these two phenomena using cross-correlations of monthly death and birth data from Taiwan from 1906 to 1943. The analysis demonstrates a strong and negative correlation between deaths (d) at time t and births (b) at time t + 9 (rdb(9) = –0.68, p < .0001). In other words, a significant drop in births was observed nine months after pandemic mortality peaked. The findings suggest that the 1918 influenza pandemic impacted subsequent births primarily through the mechanism of reduced conceptions and embryonic loss during the first month of pregnancy rather than through late-first-trimester embryonic loss.  相似文献

Plant pathogens secrete effector molecules during host invasion to promote colonization. However, some of these effectors become recognized by host receptors to mount a defence response and establish immunity. Recently, a novel resistance was identified in wild tomato, mediated by the single dominant V2 locus, to control strains of the soil-borne vascular wilt fungus Verticillium dahliae that belong to race 2. With comparative genomics of race 2 strains and resistance-breaking race 3 strains, we identified the avirulence effector that activates V2 resistance, termed Av2. We identified 277 kb of race 2-specific sequence comprising only two genes encoding predicted secreted proteins that are expressed during tomato colonization. Subsequent functional analysis based on genetic complementation into race 3 isolates and targeted deletion from the race 1 isolate JR2 and race 2 isolate TO22 confirmed that one of the two candidates encodes the avirulence effector Av2 that is recognized in V2 tomato plants. Two Av2 allelic variants were identified that encode Av2 variants that differ by a single acid. Thus far, a role in virulence could not be demonstrated for either of the two variants.  相似文献

In this paper we bring together the literatures on frame analysis, the meaning of race and campus racial climate to analyse the race frames – lenses through which individuals understand the role of race in society – held by white students attending elite US universities. For most, the elite university experience coincides with a strengthening or emergence of the diversity frame, which emphasizes the positive benefits of cultural diversity. Still, many also hold a colour-blind frame, which sees race groups as equivalent and racial identities as insignificant. We highlight the ambivalence that these divergent frames create for student perspectives on affirmative action and interracial contact on campus. Our findings demonstrate the mutability of race frames. We also highlight the impact that institutions may have on individuals' race frames. The paper is based on in-depth interviews with forty-seven US-born white undergraduates attending Brown University and Harvard University.  相似文献

The results of the first detailed screening of a resistance to Bremia lactucae in naturally growing populations of Lactuca saligna are presented here. In total, 146 accessions from 25 populations of L. saligna originating in Israel (N = 136), France (N = 8), Jordan (N = 1) and Turkey (N = 1) were tested at seedling stage for their resistance to 10 highly virulent isolates (races) of B. lactucae from Lactuca sativa (DEG2, Bl:5, Bl:15, Bl:16, Bl:17, Bl:18, Bl:21, Bl:22, Bl:24 and Bl:25). Our study strongly supports the suggestion that L. saligna is indeed generally highly resistant to B. lactucae. However, our results provide evidence that at least at a seedling stage L. saligna may not be a non‐host plant for B. lactucae, as was hypothesised for approximately the last 30 years. Some accessions expressed a differential (i.e. race‐specific) response, which accords with other recently published data for this Lactuca species. Furthermore, some geographical differences in race‐specific resistance were observed, too. Tests performed at an adult‐plant stage, however, did not prove race‐specificity of the respective accessions. To summarise, what is behind the race‐specific character of the responses observed at a seedling stage is still uncertain, as is its comparability with the race‐specific resistance of some other Lactuca species such as L. sativa or L. serriola. The presence of plant stage‐dependent resistance, governed by a combined effect of different quantitative trait loci in young and adult plants of L. saligna, is discussed.  相似文献

Annual coral mortality events due to increased atmospheric heat may occur regularly from the middle of the century and are considered apocalyptic for coral reefs. In the Arabian/Persian Gulf, this situation has already occurred and population dynamics of four widespread corals (Acropora downingi, Porites harrisoni, Dipsastrea pallida, Cyphastrea micropthalma) were examined across the first‐ever occurrence of four back‐to‐back mass mortality events (2009–2012). Mortality was driven by diseases in 2009, bleaching and subsequent diseases in 2010/2011/2012. 2009 reduced P. harrisoni cover and size, the other events increasingly reduced overall cover (2009: ?10%; 2010: ?20%; 2011: ?20%; 2012: ?15%) and affected all examined species. Regeneration was only observed after the first disturbance. P. harrisoni and A. downingi severely declined from 2010 due to bleaching and subsequent white syndromes, while D. pallida and P. daedalea declined from 2011 due to bleaching and black‐band disease. C. microphthalma cover was not affected. In all species, most large corals were lost while fission due to partial tissue mortality bolstered small size classes. This general shrinkage led to a decrease of coral cover and a dramatic reduction of fecundity. Transition matrices for disturbed and undisturbed conditions were evaluated as Life Table Response Experiment and showed that C. microphthalma changed the least in size‐class dynamics and fecundity, suggesting they were ‘winners’. In an ordered ‘degradation cascade’, impacts decreased from the most common to the least common species, leading to step‐wise removal of previously dominant species. A potentially permanent shift from high‐ to low‐coral cover with different coral community and size structure can be expected due to the demographic dynamics resultant from the disturbances. Similarities to degradation of other Caribbean and Pacific reefs are discussed. As comparable environmental conditions and mortality patterns must be expected worldwide, demographic collapse of many other coral populations may soon be widespread.  相似文献

The pathogen was first observed in the Württemberg area, where races 1 and 4 were identified using a set of differential lines. Later, commercial fields near Baden were found to be infected by race 5, which is the first occurrence of that race outside of North America. With the discovery of race 5, there are now eight races of the sunflower downy mildew fungus that have been found in Europe. The sunflower cultivars most frequently grown in Germany were investigated for resistance to race 1, 4 and 5; while all were resistant to race 1, none were resistant to either race 4 or 5.  相似文献

Background

During the 2009 influenza pandemic, individuals over the age of 60 had the lowest incidence of infection with approximately 25% of these people having pre-existing, cross-reactive antibodies to novel 2009 H1N1 influenza isolates. It was proposed that older people had pre-existing antibodies induced by previous 1918-like virus infection(s) that cross-reacted to novel H1N1 strains.

Methodology/Principal Findings

Using antisera collected from a cohort of individuals collected before the second wave of novel H1N1 infections, only a minority of individuals with 1918 influenza specific antibodies also demonstrated hemagglutination-inhibition activity against the novel H1N1 influenza. In this study, we examined human antisera collected from individuals that ranged between the ages of 1 month and 90 years to determine the profile of seropositive influenza immunity to viruses representing H1N1 antigenic eras over the past 100 years. Even though HAI titers to novel 2009 H1N1 and the 1918 H1N1 influenza viruses were positively associated, the association was far from perfect, particularly for the older and younger age groups.

Conclusions/Significance

Therefore, there may be a complex set of immune responses that are retained in people infected with seasonal H1N1 that can contribute to the reduced rates of H1N1 influenza infection in older populations.  相似文献

The 2009 pandemic influenza A (H1N1) virus exhibits hemagglutinin protein sequence homology with the 1918 pandemic influenza virus. We found that human monoclonal antibodies recognized the Sa antigenic site on the head domains of both 1918 and 2009 hemagglutinins, a site that is hypervariable due to immune selection. These antibodies exhibited high potency against the 2009 virus in vitro, and one exerted a marked therapeutic effect in vivo.In March and April of 2009, an outbreak of a swine-origin novel H1N1 influenza A virus began in Mexico (2). As of 29 November 2009, worldwide, more than 207 countries and overseas territories or communities have reported laboratory-confirmed cases of 2009 A (H1N1), including at least 8,768 deaths. The hemagglutinin (HA) gene of the 2009 A (H1N1) strain has been present in classical swine H1N1 viruses that have circulated in pigs at least since the discovery by Shope in the 1930s (Table 10, 11, 16). In contrast, the HA of human H1N1 influenza viruses circulating from 1918 to 1957 and from 1977 to present drifted progressively away from the 1918 virus HA (Table (Table1d)1d) (8, 14). Elderly subjects born prior to 1918 were found to have serum neutralizing antibody titers to the A/California/04/2009 (CA04) virus (5, 6).

A crown, foot and fruit rot of watermelon has been observed in most of the watermelon production areas in Tunisia. A survey conducted from 2000 to 2001 allowed the isolation of 291 isolates which were identified as Fusarium solani. These isolates were identified as F. solani f. sp. cucurbitae (Fsc) and races 1 and 2 characterized on the basis of pathogenicity tests on watermelon seedlings and muskmelon fruits. These results were confirmed by counts of the number of septa in the macroconidia. About 271 isolates were identified as Fsc race 1, 12 isolates were identified as Fsc race 2 and eight isolates were not pathogenic. Race 1 is widely distributed in watermelon production areas in Tunisia and race 2 has a lower incidence but it is present in the north, the middle and southern Tunisian watermelon cropping areas. Additionally, a study to compare the virulence of 122 isolates of Fsc race 1 showed different degrees of virulence among them. This is the first report of Fsc races 1 and 2 in Tunisia.  相似文献

Adaptation to early training and racing (i.e. precocity), which is highly variable in racing Thoroughbreds, has implications for the selection and training of horses. We hypothesised that precocity in Thoroughbred racehorses is heritable. Age at first sprint training session (work day), age at first race and age at best race were used as phenotypes to quantify precocity. Using high‐density SNP array data, additive SNP heritability () was estimated to be 0.17, 0.14 and 0.17 for the three traits respectively. In genome‐wide association studies (GWAS) for age at first race and age at best race, a 1.98‐Mb region on equine chromosome 18 (ECA18) was identified. The most significant association was with the myostatin (MSTN) g.66493737C>T SNP (= 5.46 × 10?12 and = 1.89 × 10?14 respectively). In addition, two SNPs on ECA1 (g.37770220G>A and g.37770305T>C) within the first intron of the serotonin receptor gene HTR7 were significantly associated with age at first race and age at best race. Although no significant associations were identified for age at first work day, the MSTN:g.66493737C>T SNP was among the top 20 SNPs in the GWAS (= 3.98 × 10?5). Here we have identified variants with potential roles in early adaptation to training. Although there was an overlap in genes associated with precocity and distance aptitude (i.e. MSTN), the HTR7 variants were more strongly associated with precocity than with distance. Because HTR7 is closely related to the HTR1A gene, previously implicated in tractability in young Thoroughbreds, this suggests that behavioural traits may influence precocity.  相似文献

The biocontrol activities of cells and cell-free extracts of Streptomyces griseus was tested against Fusarium oxysporum f.sp. cubense tropical race 4 (FOC race 4) in a sterile soil environment. They were first formulated in sodium alginate, kaolin clay and in alginate–kaolin combination, prior to introducing into sterile soil inoculated with 6 log10 cfu FOC race 4 g?1 soil. Results revealed that bioformulated cells of S. griseus, irrespective of the materials used, were generally more effective in inhibiting growth of FOC race 4 when compared to non-formulated cells of S. griseus. Kaolin was the most suitable inert material as formulation of S. griseus with kaolin effectively suppressed FOC race 4, with only 5.40 log10 cfu g?1 of FOC race 4 recovered after 20 days. Kaolin formulations also allowed good cell recovery post-formulation. Alginate was less desirable as poorer control was demonstrated, with 6.12 and 6.16 log10 cfu g?1 of FOC race 4 recovered from soils treated with alginate only and alginate–kaolin formulated S. griseus, respectively. Bioformulations did not benefit cell-free extracts at all. Our study suggests formulation of cells of S. griseus is more beneficial than cell-free extracts and kaolin is the preferred material for formulation.  相似文献

The distributions of invasive Neogobius species were investigated in the Slovak section of the River Danube from Bratislava downstream to the village of Chl'aba. During October 2004, the main channel of the Danube was sampled, including by‐pass, head‐race and tail‐race canals of the Gab?íkovo dam, backwaters and the lower‐most sections of the tributaries Malý Dunaj, Hron, Váh and Ipel’. Three Neogobius species already documented in Slovakia were captured (monkey goby Neogobius fluviatilis, bighead goby N. kessleri, round goby N. melanostomus), with the latter two species being found in almost all stretches of the Slovak Danube. Monkey goby had a most limited distribution, and no racer goby N. gymnotrachelus were observed. The abundance of particular Neogobius species appeared to depend on the character of the shoreline habitat, and a possible association between larger towns and the abundance of bighead and round gobies requires further investigation.  相似文献

In the postgenome era, the analysis of entire subproteomes in correlation with their function has emerged due to high throughput technologies. Early approaches have been initiated to identify novel components of the circadian system. For example, in the marine dinoflagellate Lingulodinium polyedra, a chronobiological proteome assay was performed, which resulted in the identification of already known circadian expressed proteins as well as novel temporal controlled proteins involved in metabolic pathways. In the green alga Chlamydomonas reinhardtii, two circadian expressed proteins (a protein disulfide isomerase and a tetratricopeptide repeat protein) were identified by functional proteomics. Also, the first hints of temporal control within chloroplast proteins of Arabidopsis thaliana were identified by proteome analysis.  相似文献
