NLRP3炎症小体是由NOD样受体(NOD-like receptor, NLR) NLRP3、接头蛋白ASC和胱冬肽酶-1(Caspase-1)所形成的多聚蛋白复合体,能够感受来自病原微生物的病原相关分子模式(pathogen-associated molecular patterns, PAMPs)和胞内自身危险信号-危险相关分子模式(danger-associated molecular patterns, DAMPs),促进细胞因子IL-1β和IL-18的成熟和分泌、引起细胞焦亡,从而在多种生理、病理过程中发挥重要作用. NLRP3炎症小体是目前研究最深入的炎症小体,其表达水平和活化强度与多种疾病的发生、发展密切相关,如感染性疾病、痛风、Ⅱ型糖尿病、动脉粥样硬化、阿尔兹海默症及癌症等.因此,阐明NLRP3炎症小体活化的调控机制,对于揭示这些疾病发生、发展的机理,寻找免疫调节治疗的新途径具有重要意义.本文详细介绍了NLRP3炎症小体的负向调控机制.  相似文献

【目的】牛病毒性腹泻病毒(bovine viral diarrhea virus, BVDV)是引起牛病毒性腹泻-黏膜病的关键病毒。BVDV的结构蛋白Erns可在病毒感染的初期削弱宿主的免疫防御,引发牛群炎症反应。核苷酸寡聚化结构域样受体(nucleotide-binding oligomerization domain, NOD)热蛋白结构域相关蛋白3 (NLRP3)炎症小体是NOD样受体(NOD-like receptor, NLRs)家族重要成员,调控炎症性疾病的发生发展,同时激活的NLRP3炎症小体能够引起宿主细胞焦亡,进而诱发级联放大的炎症反应。但BVDV Erns蛋白在BVDV感染诱发炎症反应的分子机制尚不清楚。【方法】为进一步探索Erns蛋白对BVDV感染激活NLRP3炎症小体诱发细胞焦亡的影响,构建了BVDV Erns蛋白的真核表达质粒pCMV-HA-Erns,过表达BVDV Erns蛋白,检测BVDV感染细胞中NLRP3炎症小体组分[半胱氨酸蛋白酶(caspase-1)、凋亡相关斑点样蛋白(apoptosis-associated speck-like protein, ASC)和NLRP3]、IL-1β的mRNA转录水平和蛋白表达水平,以及细胞死亡调节蛋白(gasdermin D, GSDMD)的基因表达和蛋白剪切情况,并通过扫描电镜观察牛睾丸(bovine testis, BT)细胞膜成孔及BT细胞内容物释放情况,以分析Erns蛋白诱导BT细胞产生细胞焦亡。【结果】Erns蛋白能够显著引起NLRP3炎症小体活化进而激活caspase-1,活化的caspase-1一方面切割GSDMD,形成有活性的GSDMD-N端并在BT细胞膜形成孔洞,释放内容物,诱导BT细胞发生细胞焦亡;另一方面活化的caspase-1切割pro-IL-1β,形成有活性的IL-1β,并释放到BT细胞外,引起BT细胞上清中IL-1β水平上升。【结论】系统解析了BVDV Erns蛋白激活NLRP3炎症小体介导细胞焦亡的产生,对疫苗及治疗药物的研制具有重要指导意义。  相似文献

炎症小体(Inflammasome)是细胞内识别危险信号的多蛋白复合体,是固有免疫的重要组成部分,NOD样受体家族含热蛋白结构域蛋白3炎症小体(NLRP3)是目前研究最多的一种炎症小体。真菌感染中,NLRP3炎症小体通路募集半胱天冬蛋白酶的前体半胱氨酸天冬氨酸蛋白酶1(pro-caspase-1)自身剪切活化,活化后的半胱天冬蛋白酶(Caspase-1),通过对促炎因子IL-1β(interleukin-1β, IL-1β)和IL-18(interleukin-18, IL-18)的激活,引起宿主的炎症反应,在宿主免疫应答中发挥了重要作用。  相似文献

NLRP3炎症小体作为固有免疫系统的重要组成部分,在2型糖尿病的发病过程中起着重要作用,而白细胞介素1β(IL-1β)是介导其发挥作用的关键因子。核糖体蛋白质合成、嘌呤受体P2X7、活性氧敏感的硫氧还原蛋白相互作用蛋白(TXNIP)与NLRP3炎症小体激活密切相关。肥胖时胰岛素作用的靶组织中NLRP3、IL-1β表达均增高,其介导的炎症反应在胰岛β细胞功能障碍、凋亡以及胰岛素抵抗发生过程中起关键作用。NLRP3炎症小体被多种途径激活,从而上调胰岛和脂肪组织中IL-1β的表达,促进胰岛β细胞凋亡及胰岛素抵抗的发生发展,导致糖尿病的产生。  相似文献

炎症小体在炎症相关疾病的发生发展中发挥重要作用,其中NLRP3炎症小体能够被多种病原相关分子模式(pathogen-associated molecular patterns,PAMPs)和损伤相关分子模式(damage-associated molecular patterns,DAMPs)激活,进而活化caspase-1,释放成熟形式的IL-1β和IL-18,引起机体的炎症反应,并参与多种疾病的发生发展,包括2型糖尿病、痛风、动脉粥样硬化、神经退行性疾病、肿瘤、炎症性肠病等。因此,研究NLRP3炎症小体的作用机制不仅有助于加深对炎症性疾病发生发展的认识,也为寻找此类疾病的潜在治疗靶点提供了新的思路。就NLRP3炎症小体在炎症相关疾病中的研究进展作一综述。  相似文献

阿尔兹海默症(Alzheimer’s disease, AD)是常见的神经退行性疾病,严重影响患者的生存质量。目前尚无有效的针对性治疗措施。AD发病机制复杂,是环境、遗传和年龄影响因素共同作用的结果。大脑中β-淀粉样蛋白(β-amyloid, Aβ)沉积、微管相关蛋白tau过度磷酸化形成的神经纤维缠结及神经元丢失是AD典型病理特征,大量研究证明,Aβ、tau蛋白聚集诱导核苷酸结合寡聚化结构域样受体含pyrin结构域蛋白3(nucleotide-binding oligomerization domain-like receptor pyrin domain-containing 3,NLRP3)炎症小体活化是AD炎性机制的核心环节,且以其和上下游分子为靶点的抑制剂和化合物治疗在细胞和动物模型中均发挥神经保护作用,改善空间记忆功能障碍,但临床疗效和安全性仍待研究。因此,抑制NLRP3炎症小体的活化可能是AD的潜在治疗靶点。本文以NLRP3炎症小体的活化机制和影响因素及与AD关系进行综述,并总结以NLRP3炎症小体为靶点的AD治疗药物,以期为AD等NLRP3炎症小体相关疾病提供新的治疗方向...  相似文献

目的观察牙龈卟啉单胞菌感染通过激活含NLR家族PYRIN域蛋白3(NLRP3)小体诱导人牙周膜细胞(hPDLCs)炎症反应及凋亡的效应。方法取健康前磨牙样本并分离培养hPDLCs,分为牙龈卟啉单胞菌感染的感染组和常规处理的对照组,检测细胞中NLRP3小体[NLRP3、凋亡相关斑点样蛋白(ASC)、含半胱氨酸的天冬氨酸蛋白水解酶(Caspase)-1]、凋亡基因[自杀相关因子(Fas)、Fas配体(FasL)、B淋巴细胞瘤-2基因(Bcl-2)、Bcl-2相关x蛋白(Bax)、Caspase-3]的表达量及培养基中炎症细胞因子[白细胞介素(IL)-1β、IL-18、肿瘤坏死因子-α(TNF-α)]的含量。结果感染组hPDLCs中NLRP3、ASC、Caspase-1、Fas、FasL、Bax、Caspase-3的表达量及培养基中IL-1β、IL-18、TNF-α的含量明显高于对照组,细胞中Bcl-2的表达量明显低于对照组。结论牙龈卟啉单胞菌感染能够诱导hPDLCs的炎症反应及凋亡且该作用与NLRP3小体的激活有关。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献
