目的 检测CD4+CD25+FoxP3+ 调节性T细胞(Treg)在类风湿关节炎(RA)患者外周血和关节液中的变化,探讨该细胞亚群在RA免疫耐受失衡中的作用。方法 采用流式细胞术检测活动期RA患者关节液、外周血和健康对照外周血CD4+CD25+FoxP3+ Treg细胞比例;实时定量聚合酶链反应(Real-time PCR)检测外周血和关节液单个核细胞FoxP3基因的表达。分析外周血CD4+CD25+FoxP3+Treg细胞比例与病情活动指标的相关性。结果 活动期RA患者关节液CD4+CD25+FoxP3+ Treg细胞比例(7.56%±2.89%)高于外周血(1.84%±0.97%)和健康对照外周血(2.19%±0.54%),差异均有统计学意义(P<0.01);RA患者抗环瓜氨酸肽(CCP)抗体(ACPA)阴性组外周血CD4+CD25+FoxP3+Treg细胞水平(2.85%±0.87%)高于ACPA阳性组(1.40%±0.63%,P<0.01)。活动期RA患者关节液单个核细胞FoxP3基因表达水平高于RA患者外周血和健康对照外周血(P<0.01);活动期RA患者外周血CD4+CD25+FoxP3+ Treg细胞比例与疾病活动指标无相关性(P>0. 05)。结论 RA患者关节液CD4+CD25+FoxP3+ Treg细胞的变化更能反映RA炎症部位免疫耐受失衡机制。  相似文献

[目的]研究白附子混悬液对荷瘤小鼠免疫功能的调节作用,探讨其抗肿瘤作用的免疫学机制。[方法]采用H22荷瘤小鼠模型为研究对象,用酶联免疫吸附实验(ELISA)法检测小鼠血清中白介素-2(IL-2)和白介素-4(IL-4)水平;用流式细胞术(FACS)法分析对CD4+,CD8+T细胞的调节作用。[结果]白附子混悬液能够上调荷瘤小鼠血清IL-2细胞因子的表达,降低IL-4水平,可以使CD4+T细胞表达升高,CD8+T细胞表达降低。[结论]白附子混悬液能够通过调节荷瘤机体异常免疫功能状态而发挥抗肿瘤作用。  相似文献

目的:观察他克莫司软膏联合308nm准分子激光治疗颜面部白癜风疗效。方法:试验组,给予0.03%他克莫司软膏外抹,1日1次,同时予308 nm准分子激光照射患处,1周2~3次照射;对照组0.03%他克莫司软膏外抹,1日1次,疗程均为8~10周。结果:两组有效率分别为79.75%和57.14%(P<0.05),痊愈率分别为24.05%和12.86%(P<0.05),差异均具有统计学意义,试验组疗效优于对照组。结论:他克莫司软膏联合308 nm准分子激光治疗颜面部白癜风疗效确切,不良反应少。  相似文献

目的观察外用0.1%他克莫司软膏联合308准分子激光局部照射治疗稳定期白癜风的临床疗效及不良反应。方法将112例白癜风患者148片皮损随机分为2组,每组74片,治疗组外用0.1%他克莫司软膏联合308准分子激光局部照射;对照组单独给予308准分子激光局部照射,每周2次,治疗16周以后开始进行疗效评估并记录不良反应。结果治疗组有效率为87.0%,对照组有效率为67.5%,2组比较差异有统计学意义(P〈0.05)。2组患者出现局部红斑、瘙痒的概率很低,症状轻微。结论 308准分子激光局部照射治疗白癜风疗效好,不良反应少,联合0.1%他克莫司软膏可进一步提高疗效。  相似文献

目的 探讨外周血CD4+/CD8+在2型糖尿病合并蛋白尿(albuminuria,AU)发生过程中的变化及临床意义。方法收集2020 年3 月~2021年6月于徐州医科大学附属医院内分泌科住院的2型糖尿病患者595例,根据尿微量白蛋白/肌酐比值(urinary albumin to creatinine ratio,UACR)分为非AU组(n=329)和AU组(n=266),另选取同期体检的健康人群作对照组(NC组,n=154),收集研究对象的一般资料及实验室指标,探讨CD4+/CD8+与2型糖尿病合并AU的关系。结果 与NC组、非AU组比较,AU组的CD4+/CD8+显著降低(P<0.05)。2型糖尿病合并蛋白尿患者UACR与CD4+/CD8+呈显著负相关(P<0.001)。Logistic回归分析显示,糖尿病病程、收缩压、肌酐、超敏C反应蛋白水平升高、CD4+/CD8+降低可能是2型糖尿病合并AU的危险因素(OR=0.519,95% CI:0.359~0.750,P<0.001)。结论 2型糖尿病合并AU患者外周血CD4+/CD8+降低可能在蛋白尿的发生、发展中起重要作用。  相似文献

目的探讨单纯激光与联合卡泊三醇及他克莫司软膏对局限型白癜风患者治疗疗效。方法132例局限型白癜风患者随机分为对照组(n=66)和观察组(n=66),对照组患者给予308nm准分子激光照射治疗,观察组患者在对照组的基础上给予他克莫司软膏联合卡泊三醇软膏,连续用药3个月后比较临床疗效。结果观察组总有效率高于对照组(P<0.05);观察组第2疗程和第3疗程复色率高于对照组(P<0.05);观察组复发率低于对照组(P<0.05)。结论他克莫司软膏联合卡泊三醇软膏治疗局限型白癜风安全、有效,对皮肤刺激少,联合308nm准分子激光照射效果更为显著。  相似文献

目的 探讨卵巢癌患者外周血中CD4+CD25+及CD4+CD25+Foxp3+调节性T细胞的表达,及其与转化生长因子β1(TGF-β1)水平的相关性。方法 采用流式细胞术检测24例上皮性卵巢癌患者外周血中CD4+CD25+及CD4+CD25+Foxp3+调节性T细胞的表达,收集每例患者对应的血浆,ELISA法检测卵巢癌患者血浆中TGF-β1浓度,并与20例卵巢良性肿瘤及20例健康体检者做比较研究。结果 与卵巢良性疾病及健康对照组相比,卵巢癌患者CD4+CD25+、CD4+CD25+Foxp3+调节性T细胞及TGF-β1的表达水平明显增高(均P<0.01);III/IV期卵巢癌患者CD4+CD25+、CD4+CD25+Foxp3+调节性T细胞及TGF-β1的表达水平明显高于I/II期患者(P<0.05);Pearson相关性分析结果显示,卵巢癌患者外周血CD4+CD25+及CD4+CD25+Foxp3+调节性T细胞水平与血浆TGF-β1水平均呈现正相关关系(r=0.656, P < 0.05; r=0. 574, P < 0.05)。结论 卵巢癌患者外周血高表达CD4+调节T细胞,且与血浆TGF-β1呈正相关。检测外周血CD4+调节T细胞与TGF-β1水平对于判断卵巢癌病期有重要意义。TGF-β1水平增高可能参与调节CD4+调节T细胞对卵巢癌机体的免疫负调节机制。  相似文献

目的: 探讨他克莫司软膏联合308 nm准分子激光治疗白癜风的临床疗效。方法: 将80例白癜风患者随机分为对照组和观察组各40例,对照组给予308 nm准分子激光进行治疗,观察组在对照组的基础上加0.1%他克莫司软膏外用,治疗结束后比较2组的临床疗效。结果: 对照组和观察组患者的临床有效率分别为72.5%和92.5%,差异有统计学意义(P<0.05)。结论: 他克莫司软膏联合308 nm准分子激光治疗白癜风的疗效显著,不良反应少。  相似文献

Summary To determine the CD30 expression on peripheral blood T lymphocyte subsets in patients with hemorrhagic fever with renal syndrome (HFRS) and its clinical implications, double immunofluorescence technique and flow cytometry were used. There was no significant difference among the severe group, mild-moderate group and normal control group in the CD+CD30 T lymphocyte subset. While the CD4 +CD30 T cells of HFRS patients were increased and the difference between severe group and mild-moderate group or normal control group were very significant (P<0.01) and the difference between the mild-moderate group and normal control group was also significant (P<0.05). The CD8 +CD30 T cells were increased while the CD8 +CD30 - T cells decreased obviously in HFRS patients, and the differences among three groups in both subsets were very significant (P<0.01). The results showed that the humoral immunity and cellular immunity are overactive in HFRS patients during acute phase. The loss of balance between T lymphocyte subsets may play an important role in the pathophysiology of HFRS and is closely correlated with the severity of the HFRS. Summary Concentrations of K+, Na+ and Mg++ were measured in plasma (K+ e, Na+ e, Mg++ e) and in erythrocytes (K+ i, Na+ i, Mg++ i) in patients with digitalis toxicity before and after administration of Mg. K+ concentration in 24-hour urine was also measured. 70 persons were divided into 5 groups: (1) 11 normal persons as controls; (2) 19 patients with congestive heart failure, in whom Mg++ i and Mg++ e concentrations were found to be lower than that of the control group; (3) 16 patients with digitalis toxicity treated by Mg++, whose K+ i concentration increased much quicker, and Na+ i and Na+ i/K+ i decreased remarkably in comparison to non-Mg treatment group; (4) 12 patients with digitalis toxicity not treated by Mg++: in these the above-mentioned changes were not significant; (5) 12 patients with arrhythmia of other causes treated by Mg++: whose K+ i also increased significantly on the 3rd day after treatment. K+ concentration in 24-hour urine was much lower in Mg treatment group than in non-Mg treatment group. Besides, acute digitalis toxicity was observed in experimental dogs. The concentration of K+ i recovered much quicker in Mg treatment group than in control group. These findings indicate that hypomagnesemia may induce digitalis toxicity. In this paper the possible mechanism of the therapeutic effect of Mg in the treatment of digitalis toxicity is discussed.  相似文献

Summary In order to investigate mitogenic effect of Entamoeba histolytica extracts (EHE) on T cell subpopulations, monoclonal antibodies of the OKT system were used to separate T inducer/helper (OKT4 +) and T suppressor/cytotoxic (OKT8 +) cells. Only OKT4 + cells were found to respond to EHE stimulation as indicated by incorporation of3HTdR into cultured cells. Determination of interleukin-2 (IL-2) activities revealed that the response of OKT4 + cells to EHE was due to the action of IL-2.  相似文献

The expression of CD8+CD25+FoxP3+ regulatory T cells(CD8+Tregs) in the peripheral blood of patients with stable chronic obstructive pulmonary disease(COPD),and the effect of muscarinic cholinergic receptor antagonist tiotropium bromide on the expression of CD8+Tregs were investigated.Twenty-three patients with moderate to severe stable COPD were enrolled in this study.All patients inhaled tiotropium bromide(18 μg daily) for 3 months.Before and after inhalation of tiotropium bromide,peripheral blood samples were collected from the patients,and T cells were labeled by three-color labeled monoclonal antibodies.Flow cytometry was used to detect the quantity and percentage of CD8+T cells,CD8+CD25+T cells,CD8+Tregs,CD4+T cells,CD4+CD25+T cells and CD4+CD25+FoxP3+ regulatory T cells(CD4+Tregs) respectively.The percentage of CD4+T cells was increased from(27.82±2.18)% to(35.53±1.3)%(t=3.20,P=0.004) in the peripheral blood of patients with stable COPD after inhalation of tiotropium bromide for 3 months,that of CD4+CD25+T cells was decreased from(10.03 ±1.42)% to(4.21 ±0.65)%(t=3.78,P=0.001),and that of CD8+Tregs was increased from(8.41 ±1.68)% to(21.34 ±4.20)%(t=2.72,P=0.013).At baseline,CD8+T cells,CD8+CD25+T cells and CD4+Tregs were detectable in the peripheral blood,but no significant changes were observed after treatment.Linear correlation analysis revealed that the difference before and after treatment in CD4+T cells and CD4+CD25+T cells was negatively correlated with the ratio of change in CD8+Tregs before and after treatment(r=-0.61,P=0.013;r=-0.72,P=0.001 respectively).In the peripheral blood of patients with stable COPD,there was the expression of CD8+Tregs and CD4+Tregs.Muscarinic receptor antagonist,tiotropium bromide,can promote the amplification of CD4+T cells,inhibit the expression of CD25+T cells,and enhance the expression of CD8+Tregs.CD8+Tregs and CD4+Tregs can be used as new indicators to understand the immune status of patients.They are helpful in judging the treatment efficacy and disease immunophenotype.  相似文献

Summary Potential effect of the interaction of CD2 with its ligand LFA-3 on B cell responses in vitro was studied to evaluate the action of CD2-LFA-3 in immune responses. It was found that: 1) addition of autologous erythrocytes to uniractional mononuclear cells resulted in enhancement of PWM-induced lymphocyte proliferation, and had no significant influence on non-mitogen stimulation; 2) autologous erythrocytes potentiated the synthesis of immunoglo bulins; and 3) costimulating effects of autologous RBC can be depressed by preincubation of PBMNC with anti-CD2 monoclonal antibody (WuTll). The results suggest that preincubation of mononuclear cells with anti-CD2 McAb has a down-regulating effects on B cell responses and the interaction of CD2 with its ligand LFA-3 expressed by RBC is closely related to immune responses.  相似文献

目的探讨孟鲁司特联合舒利迭治疗重度慢性阻塞性肺疾病(COPD)急性加重对免疫功能的影响。方法将113例重度COPD急性加重期患者,随机分为孟鲁司特联合舒利迭治疗组57例,单纯舒利迭治疗组56例,治疗14 d后,评价所有患者治疗前、后T细胞亚群CD3+、CD4+及CD4+/CD8+的变化。结果治疗组CD3+、CD4+、CD4+/CD8+指标差异有统计学意义(P<0.05),而对照组无明显变化,药物冶疗安全性好。结论孟鲁司特钠联合舒利迭治疗中重度COPD急性加重期,能有效改善患者免疫功能,提高患者免疫力。  相似文献

Objective: To study immune function of children viral myocarditis and to evaluate the clinical effect of Shuanghuanglian Powder (SHLP) by injection.Methods: The 62 patients of viral myocarditis were divided into two groups randomly, the SHLP group (n = 32) treated with conventional therapy plus SHLP and the conventional treatment group (n = 30) with conventional therapy alone. Their serum antibody of Coxsackie virus group B (COXB-IgM), T-lymphocyte subsets including CD3 +, CD4 +, CD8 + and CD4 +/CD8 + were determined with ELISA and indirect immunofluorescent assay.Results: COXB-IgM was positive in 39 of the 62 patients, which was significantly different with those of normal controls (P< 0.001). Patients’ serum level of CD4 + cells and CD4 +/CD8 + ratio decreased while CD8 + increased. After treatment, the recovery of symptoms, signs and immune function in patients of the SHLP group were significantly better than those in patients treated with conventional treatment alone (P<0.01).Conclusion: Immunoregulatory disturbance is involved in children with viral myocarditis and SHLP is an effective drug in the treatment of children viral myocarditis.  相似文献

目的 探讨在体外人乳牙牙髓干细胞(stem cells from human exfoliated deciduous teeth, SHED)及其联合IL-6抗体对干燥综合征患者(Sjgren syndrome, SS)外周血活化的CD4+T细胞的免疫抑制效应。方法 培养并鉴定SHED;流式细胞术检测健康者和SS患者外周血CD4+T细胞中Th1、Th2、Th17、Treg及Tfh的比例;将健康者和SS患者PBMC各自单独培养或与SHED共培养,ELISA法检测培养上清液中IL-6浓度,在各组中添加或不添加IL-6抗体,72h后流式细胞术检测CD4+T细胞增殖情况。结果 SS患者外周血CD4+T细胞中Th17及Tfh比例较健康者升高(P<0.05);健康者与SS患者共培养组的Th17比例显著降低(P<0.001),SS患者共培养组的Treg比例有所升高(P<0.05);在SS患者共培养组中添加IL-6抗体,可显著降低CD4+T细胞中Th2的比例(P<0.05)。结论 SHED可抑制SS患者CD4+T细胞的增殖,降低SS患者CD4+T细胞中Th17的比例且增加Treg的比例;IL-6抗体可降低与SHED共培养的SS患者PBMC中CD4+T细胞Th2亚型比例。  相似文献

[目的]观察维胃方对胃溃疡大鼠胃黏膜髓过氧化物酶(MPO)和H+-K+-ATP酶的影响,探讨维胃方治疗胃黏膜损伤的可能机制。[方法]采用乙酸烧灼法,建立胃溃疡模型,用比色法测定胃黏膜MPO和H+-K+-ATP酶活性,并观察胃黏膜组织病理学变化。[结果]从胃黏膜大体标本、切片观察,维胃方中剂量组疗效最佳。与自愈组和模型组比较,维胃方各组胃黏膜MPO活性显著降低(P<0.01或P<0.05),其中以维胃方中剂量组值最小。维胃方各剂量组的H+-K+-ATP酶活性均高于自愈组(P<0.01或P<0.05),接近正常组(P>0.05),表明维胃方能恢复其活性。[结论]维胃方对大鼠实验性胃溃疡具有明显的保护作用,其机制可能是通过降低大鼠胃黏膜MPO活性,进而减轻炎症介质对胃黏膜的损伤。同时其能修复已损伤的胃黏膜,故能恢复H+-K+-ATP酶活性。  相似文献
