考察了有机酸对西索米星产生菌的菌体生长和产物合成的影响 .不同的有机酸对西索米星产生菌的菌体生长有不同程度的抑制作用 ,在产物合成阶段添加苹果酸、柠檬酸或延胡索酸有助于菌体的比产物合成能力 (Qp)的提高 ,这可能与发酵过程中胞内ALP的活力和TCA循环受到调节有关 .  相似文献

卡那霉素发酵过程中菌体生长和产物合成符合GadenⅢ类过程.卡那霉素发酵过程对数生长期最大比生长速率为0.0759h-1,最高呼吸强度可达11.0mmol·m-3·s-1;产物合成期比生长速率为0.009h-1,平均呼吸强度为7.0mmol·m-3·s-1,产物最大比合成速率为3.37×10-4g·g-1·h-1.建立了变容体系中卡那霉素产物合成期动力学模型,对模型参数进行了估计,模型拟合偏差为9.23%.该模型可用于带中间补料的卡那霉素变容发酵过程的优化控制.  相似文献

克雷伯氏菌(Klebsiella pneumoniae)分批发酵生产1,3-丙二醇(1,3-PD),根据菌体生长、产物生成和补料可将发酵过程分成4个阶段.不同阶段控制不同的底物浓度会对菌体生长和产物生成产生不同的影响.研究发现,菌体生长阶段(阶段Ⅱ)底物浓度控制在6.0 g/L最适合菌体生长,产物主要生成阶段(阶段Ⅲ)底物浓度控制在18.9 g/L最适合产物生成,菌浓可达7.83 g/L,最终1,3-PD的浓度为68.5 g/L,摩尔转化率为61%,生产强度为2.21 g/(L·h).  相似文献

研究了不同类型国产阳离子交换树脂对水溶液中西索米星的静态交换吸附性能.考察了各种因素对吸附分离的影响.结果表明:大孔型弱酸性D1树脂对西索米星的吸附容量达20.92×104μg.mL-1;用氨水解吸,解吸率达96.2%.弱酸树脂吸附西索米星的最佳pH值为7.8.较大的溶液浓度和适中的搅拌速度有利于弱酸树脂对西索米星的吸附.  相似文献

pH对大肠杆菌L-天冬酰胺酶的产生有很大的影响.通过对不同缓冲体系试验和对摇瓶发酵过程pH变化的分析,了解摇瓶发酵过程中pH对L-天冬酰胺酶合成的影响.采用流加磷酸自控L-天冬酰胺酶发酵过程的pH,可延长发酵周期,有效地防止菌体的过早自溶,大大促进L-天冬酰胺酶的合成,较分批发酵提高产酶83%,达到110 u/mL.发酵动力学特性分析表明,该发酵方式L-天冬酰胺酶的合成与菌体生长不相关,控制比生长速率μ为0.15~0.30 h-1,均有利于L-天冬酰胺酶的合成,可使比产酶速率的最大值Qpmax达到3 922 u/(g h).  相似文献

采用发酵过程参数相关分析理论,分析了毕赤酵母在不同发酵阶段利用不同碳源时的生理代谢参数变化特征。同时,研究了不同比生长速率下重组毕赤酵母表达猪胰岛素前体补料分批发酵过程,建立蛋白表达阶段的细胞生长非结构模型,结果表明:比生长速率和甲醇浓度符合Monod方程,最大比生长速率(μmax)为0.101h-1,基质的半饱和常数(Ks)为0.252g/L,细胞得率系数YX/S=0.386g/g,细胞维持系数m=0.011g/(g·h)。当比生长速率为0.016h-1时猪胰岛素前体(PIP)最大比形成速率(qp,max)达0.098mg/(g·h)。该模型能较好预测毕赤酵母表达阶段细胞生长和PIP表达的发酵趋势,用其控制发酵过程,目标蛋白PIP产量为优化前的1.5倍,达0.976g/L。  相似文献

庆大霉素生物合成过程中的genP基因和西索米星生物合成过程中的sisI基因属同工异源酶基因,均能催化绛红糖胺3′,4′-双脱羟基反应。本研究借助组合生物合成,探索绛红小单孢菌中的genP基因能否在依纽小单孢菌中表达。首先构建genP基因替换sisI基因的同源重组质粒pKPI4,并经接合转移将其导入依纽小单孢菌TS388中,最后再通过影印筛选及PCR鉴定获得genP基因替换sisI基因的工程菌PTI886。将野生型菌株TS388、sisI基因缺失突变株TI1102(TS388△sisI)及工程菌PTI886在同等条件下进行发酵,提取其代谢产物并经薄层层析及质谱分析。结果表明,突变株TI1102不能合成西索米星,而工程菌PTI886仍能继续合成西索米星,且代谢产物组分与野生型菌株TS388相比无明显变化,说明genP基因能在依纽小单孢菌中表达。本研究揭示了同工异源酶基因异源表达的可行性,可为今后不同微生物异源基因组合的研究提供借鉴。  相似文献

研究肠膜明串珠菌CICC-21725发酵产右旋糖酐过程。根据菌体生长量、蔗糖浓度和右旋糖酐产率与发酵时间的关系,采用Logistic Law方程和Luedeking-Piret方程,建立肠膜明串珠菌CICC-21725菌体生长动力学模型、产物合成动力学模型和底物消耗动力学模型。运用Matlab 7.0软件对实验数据拟合,获得模型参数。用红外光谱和核磁氢谱、碳谱对菌体发酵产物进行结构表征。结果表明:所建立的动力学模型的预测值与实验值吻合较好,拟合模型的相关系数分别为0.994 2、0.996 9和0.991 2,模型能较好的预测肠膜明串珠菌CICC-21725发酵产右旋糖酐的动力学过程。肠膜明串珠菌CICC-21725发酵过程中右旋糖酐的合成属于生长偶联型。红外光谱及核磁共振光谱结果表明肠膜明串珠菌的发酵产物是由为α-(1,6)糖苷键连接的直链α型右旋糖酐。  相似文献

以黑曲霉C71为出发菌株,分批发酵生产木聚糖酶,在发酵过程中每隔8h取样,测定菌体细胞浓度、总糖与还原糖含量和木聚糖酶活力,探讨了发酵过程中菌体生长、产物合成及基质消耗的特性.基于Logistic和Luedeking-Piret方程,建立了描述黑曲霉C71分批发酵过程的菌体生长动力学模型、产物合成动力学模型、基质消耗动力学模型.结果表明,模型计算值与实验数据拟合良好,基本反映了黑曲霉C71分批发酵过程的动力学特征.黑曲霉C71分批发酵过程中菌体生长与产物合成属于部分生长偶联型.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
