Spinach chloroplasts and Clostridium butyricum cells were immobilized in 2% agar gel. Crude ferredoxin isolated from spinach and benzyl viologen were used as electron carriers. The optimum pH for both NADP reduction by immobilized chloroplasts and for hydrogen evolution by immobilized Cl. butyricum was 8.0. The optimum temperature was between 25 and 30°C for NADP reduction by immobilized chloroplasts, and 37°C for hydrogen evolution by immobilized cells. The total amount of hydrogen evolved in 6 h was 41 μmol/mg Chl for the immobilized chloroplast-benzyl viologen-immobilized Cl. butyricum system, and 11 μmol/mg Chl for the immobilized chloroplast-ferredoxin-Cl. butyricum system. The systems evolved only a trace amount of hydrogen when dichlorophenyldimethylurea was added. The immobilized chloroplast-benzyl viologen-immobilized Cl. butyricum system evolved hydrogen continuously for 6 h, and immobilized Cl. butyricum retained the initial hydrogenase activity. However, the photoreduction activity of chloroplasts decreased to 30% of the initial activity after 6 h of reaction.  相似文献

Chlorella vulgaris MSU 01 strain isolated from the sediment of the pond is able to produce molecular hydrogen in a clean way. To relate the dynamic coupling between the cultural conditions and biological responses, an original lab scale set up has been developed for hydrogen production. Different sources like mannitol, glucose, alanine, citric acid, aspartic acid, l-alanine, l-cysteine, sodium succinate and sodium pyruvate were used for algal media optimization. Corn stalk, from 1 to 5 g/L was tested for the effective algal growth and hydrogen production. The cell concentration of 1.6-19 g/L dry cell weight (DCW) was found at the 10th day. The kinetic parameters involved in the hydrogen production at 4 g/L corn stalk using the algal inoculum (50 mL) in the bioreactor volume (500 mL) was found to be with the hydrogen production potential (Ps) of 7.784 mL and production yield of (Pr) 5.534 mL respectively. The growth profile of the algal biomass at the above mentioned condition expressed the logistic model with R2 0.9988. The final pH of the broth was increased from 7.0 to 8.5-8.7. The anaerobic fermentation by C. vulgaris MSU 01 strain involved in the conversion process of complex carbon source has increased the H2 evolution rate and higher butyrate concentration in the fermentate.  相似文献

Immobilized chloroplasts and Clostridium butyricum were employed for a photochemical energy conversion system. Spinach chloroplasts were immobilized in 2% agar gel. The optimum temperature of immobilized chloroplasts was 30°C. The maximum activity was obtained in the phosphate buffer solution (pH 8.0) containing 8μM of ferredoxin under an N2 bubbling condition. Hydrogen was evolved under illumination by immobilized chloroplasts and C. butyricum. Hydrogen produced by this system was applied to a hydrogen-oxygen fuel cell. Photoinduced current was obtained from this photochemical energy conversion system. A photocurrent of 0.4?1.5 mA was continuously obtained for 4 h. The conversion ratio from hydrogen to current was 80?100%.  相似文献

We compared predation rates and behaviours of sea stars (Asterias vulgaris and Leptasterias polaris) and crabs (Cancer irroratus and Hyas araneus) preying on juvenile sea scallops (Placopecten magellanicus, 25-35 mm shell height) in laboratory. These predatory species co-occur with sea scallops on the sea bed of the Gulf of St. Lawrence, Canada, and limit scallop survival in seeding operations. We also examined, under controlled conditions, the effect of tethering scallops on predator-prey interactions. Predation rates, time budgets and encounter behaviours observed for A. vulgaris and C. irroratus preying on free (untethered) scallops were comparable to previous studies. C. irroratus were more effective predators as they consumed 3.1 scallops predator− 1 day− 1, although they spent only 0.9% of their time searching for prey. A. vulgaris consumed 0.9 scallops predator− 1 day− 1 and spent 7.6% of their time searching. Sea stars L. polaris had a lower predation rate (0.02 scallop predator− 1 day− 1) than A. vulgaris. The frequent avoidance behaviour of L. polaris and its low ability to capture scallops support the notion that scallops are not a main component of this sea star's diet. Crabs H. araneus had similar predation rates (1.3 scallops predator− 1 day− 1) and behaviours to C. irroratus, although the probability of consumption upon capture was affected by relatively high numbers of rejections and post-capture escapes of scallops. As expected, the tethering procedure increased predation rate of L. polaris (about 19 times higher), but surprisingly did not significantly affect that of A. vulgaris. Examination of behaviours indicated that A. vulgaris offered tethered scallops tended to have a higher probability of capture, but spent less time searching for prey (possibly because satiation was reached) than A. vulgaris offered free scallops. Predation rates and behaviours of both crab species were not affected by tethering, since encounter rate was the primary determinant of crab-scallop interactions. Identification and quantification of behaviours underlying the predation process allowed us to mathematically model predator-related mortality for the four predator species.  相似文献

An amino acid uptake system specific for glycine, alanine, serine and proline was induced by glucose in Chlorella vulgaris. The uptake system translocated the zwitterionic form of the amino acid. There was more than 100-fold accumulation which indicated a coupling to metabolic energy. The depolarization of the membrane potential during proline uptake and the sensitivity of its uptake rate to the membrane potential point to coupling with an ion flow. Inhibitors of plasmalemma-bound H+-ATPase inhibit proline uptake. These data are interpreted to mean that proline is taken up as a proton symport. In some Chlorella strains the proline-coupled H+ uptake could be measured with electrodes, but not in Chlorella vulgaris. There is evidence that the transport of amino acids rapidly stimulates the proton-translocating ATPase of Chlorella vulgaris, so that the proline-coupled proton uptake is immediately neutralized.  相似文献

Clostridium butyricum is one of the commonly used species for fermentative hydrogen production. While producing H2, it can produce acids (lactic, acetic and butyric acids) and CO2, as well as a small amount of ethanol. It has been proposed that elimination of competing pathways, such as the butyrate formation pathway, should increase H2 yields in Clostridium species. However, the application of this strategy has been hindered by the unavailability of genetic tools for these organisms. In this study, we successfully transferred a plasmid (pMTL007) to C. butyricum by inter-specific conjugation with Escherichia coli and disrupted hbd, the gene encoding β-hydroxybutyryl-CoA dehydrogenase in C. butyricum. Fermentation data showed that inactivation of hbd in C. butyricum eliminated the butyrate formation pathway, resulting in a significant increase in ethanol production and an obvious decrease in H2 yield compared with the wild type strain. However, under low partial pressure of H2, the hbd-deficient strain showed increased H2 production with the simultaneous decrease of ethanol production, indicating that H2 production by C. butyricum may compete for NADH with the ethanol formation pathway. Together with the discovery of a potential bifurcating hydrogenase, this study extends our understanding of the mechanism of H2 production by C. butyricum.  相似文献

The taxonomy and evolutionary species boundaries in a global collection of Cercospora isolates from Beta vulgaris was investigated based on sequences of six loci. Species boundaries were assessed using concatenated multi-locus phylogenies, Generalized Mixed Yule Coalescent (GMYC), Poisson Tree Processes (PTP), and Bayes factor delimitation (BFD) framework. Cercospora beticola was confirmed as the primary cause of Cercospora leaf spot (CLS) on B. vulgaris. Cercospora apii, C. cf. flagellaris, Cercospora sp. G, and C. zebrina were also identified in association with CLS on B. vulgaris. Cercospora apii and C. cf. flagellaris were pathogenic to table beet but Cercospora sp. G and C. zebrina did not cause disease. Genealogical concordance phylogenetic species recognition, GMYC and PTP methods failed to differentiate C. apii and C. beticola as separate species. On the other hand, multi-species coalescent analysis based on BFD supported separation of C. apii and C. beticola into distinct species; and provided evidence of evolutionary independent lineages within C. beticola. Extensive intra- and intergenic recombination, incomplete lineage sorting and dominance of clonal reproduction complicate evolutionary species recognition in the genus Cercospora. The results warrant morphological and phylogenetic studies to disentangle cryptic speciation within C. beticola.  相似文献

Cathespin L-like proteases (CPLs), characterized from a wide range of helminths, are significant in helminth biology. For example, in Caenorhabditiselegans CPL is essential for embryogenesis. Here, we report a cathepsin L-like gene from three species of strongyles that parasitize the horse, and describe the isolation of a cpl gene (Sv-cpl-1) from Strongylusvulgaris, the first such from equine strongyles. It encodes a protein of 354 amino acids with high similarity to other parasitic Strongylida (90-91%), and C.elegans CPL-1 (87%), a member of the same Clade. As S.vulgariscpl-1 rescued the embryonic lethal phenotype of the C.eleganscpl-1 mutant, these genes may be orthologues, sharing the same function in each species. Targeting Sv-CPL-1 might enable novel control strategies by decreasing parasite development and transmission.  相似文献

All transhydrogenases appear to have three components: dI, which binds NAD(H), and dIII, which binds NADP(H), protrude from the membrane, and dII spans the membrane. However, the polypeptide composition of the enzymes varies amongst species. The transhydrogenases of Mycobacterium tuberculosis and of Rhodospirillum rubrum have three polypeptides. Sequence analysis indicates that an ancestral three-polypeptide enzyme evolved into transhydrogenases with either two polypeptides (such as the Escherichia coli enzyme) or one polypeptide (such as the mitochondrial enzyme). The fusion steps in each case probably led to the development of an additional transmembrane helix. A hybrid transhydrogenase was constructed from the dI component of the M. tuberculosis enzyme and the dII and dIII components of the R. rubrum enzyme. The hybrid catalyses cyclic transhydrogenation but not the proton-translocating, reverse reaction. This shows that nucleotide-binding/release at the NAD(H) site, and hydride transfer, are fully functional but that events associated with NADP(H) binding/release are compromised. It is concluded that sequence mismatch in the hybrid prevents a conformational change between dI and dIII which is essential for the step accompanying proton translocation.  相似文献

Homogenization of Phaseolus vulgaris leaves at acid pH results in the evolution of hexanal, cis-3- and trans-2-hexenal. With cell-free extracts of leaves, linoleic and linolenic acids are enzymically converted to their hydroperoxides (predominantly the 13-hydroperoxide isomers) and to hexanal or hexenal respectively. Activity was highest in young, dark-green leaves and was stimulated by Triton X-100. Oleic acid is not a substrate for these reactions. Both 9- and 13-hydroperoxides were cleaved to carbonyl fragments and are proposed as intermediates in the formation of volatile aldehydes and non-volatile ω-oxoacids in P. vulgaris leaves. Aquilegia paui is a narrow endemic species from the north-eastern Iberian Peninsula. It has a reduced number of populations in the limestone mountain massif of Els Ports. Genetic diversity was evaluated through allozyme electrophoresis in this rare columbine, together with its widespread congener Aquilegia vulgaris. Low levels of genetic diversity were detected in both taxa, especially for A. paui. Genetic identity between the two columbines was particularly high (I = 0.990), and the allele pool of A. paui is a subset of that of its widespread congener (with the exception of a low-frequency allele that was only detected in one population of A. paui). A. paui may have recently evolved from nearby populations of A. vulgaris. Speciation of this taxon, which may still be ongoing, seems to be driven by ecological differentiation, as generally occurred for the European columbines as well as in many Mediterranean endemic species.  相似文献

Since the first isolation of type E botulinum toxin-producing Clostridium butyricum from two infant botulism cases in Italy in 1984, this peculiar microorganism has been implicated in different forms of botulism worldwide. By applying particular pulsed-field gel electrophoresis run conditions, we were able to show for the first time that ten neurotoxigenic C. butyricum type E strains originated from Italy and China have linear megaplasmids in their genomes. At least four different megaplasmid sizes were identified among the ten neurotoxigenic C. butyricum type E strains. Each isolate displayed a single sized megaplasmid that was shown to possess a linear structure by ATP-dependent exonuclease digestion. Some of the neurotoxigenic C. butyricum type E strains possessed additional smaller circular plasmids. In order to investigate the genetic content of the newly identified megaplasmids, selected gene probes were designed and used in Southern hybridization experiments. Our results revealed that the type E botulinum neurotoxin gene was chromosome-located in all neurotoxigenic C. butyricum type E strains. Similar results were obtained with the 16S rRNA, the tetracycline tet(P) and the lincomycin resistance protein lmrB gene probes. A specific mobA gene probe only hybridized to the smaller plasmids of the Italian C. butyricum type E strains. Of note, a ß-lactamase gene probe hybridized to the megaplasmids of eight neurotoxigenic C. butyricum type E strains, of which seven from clinical sources and the remaining one from a food implicated in foodborne botulism, whereas this ß-lactam antibiotic resistance gene was absent form the megaplasmids of the two soil strains examined. The widespread occurrence among C. butyricum type E strains associated to human disease of linear megaplasmids harboring an antibiotic resistance gene strongly suggests that the megaplasmids could have played an important role in the emergence of C. butyricum type E as a human pathogen.  相似文献

A study was made with a modulated oxygen electrode of the effect of variations of oxygen concentration on photosynthetic oxygen evolution from algal cells. When Chlorella vulgaris is examined with a modulated 650 nm light at 22°C, both the oxygen yield and the phase lag between the modulated oxygen signal and the light modulations have virtually constant values between 800 and 120 ergs · cm?1 · s?1 if the bathing medium is in equilibrium with the air. Similar results are obtained at 32°C between 1600 and 120 ergs · cm?2 · s?1. Under anerobic conditions both the oxygen yield and the phase lag decrease if the light intensity is lowered below about 500 ergs · cm?2 · s?1 at 22°C or about 1000 ergs · cm?2 · s?1 at 32°C. A modulated 706 nm beam also gives rise to these phenomena but only at significantly lower rates of oxygen evolution. The cells of Anacystis nidulans and Porphyridium cruentum appear to react in the same way to anaerobic conditions as C. vulgaris. An examination of possible mechanisms to explain these results was performed using a computer simulation of photosynthetic electron transport. The simulation suggests that a backflow of electrons from a redox pool between the Photosystems to the rate-limiting reaction between Photosystem II and the water-splitting act can cause a decrease in oxygen yield and phase lag. If the pool between the Photosystems is in a very reduced state a significant cyclic flow is expected, whereas if the pool is largely oxidized little or no cyclic flow should occur. It is shown that the effects of 706 nm illumination and removal of oxygen can be interpreted in accordance with these proposals. Since a partial inhibition of oxygen evolution by 3-(3.4-dichlorophenyl)-1,1-dimethylurea (10?8 M) magnifies the decreases in oxygen yield and phase lag, it is proposed that the pool which cycles back electrons is in front of the site of 3-(3,4-dichlorophenyl)-1,1-dimethylurea inhibition and is probably the initial electron acceptor pool after Photosystem II.  相似文献

Cladophora glomerata (L.) Kütz. and Enteromorpha ahlneriana Bliding are morphologically similar filamentous green algae that are dominants in the upper littoral zone of the brackish Baltic Sea. As these two species co-exist in a continuously fluctuating environment, we hypothesised that they may have different strategies to cope with oxidative stress. This was tested in laboratory experiments through stressing the algae by high irradiance (600 μmol photons PAR m−2 s−1) at two different temperatures (15 and 26 °C) in a closed system. Thus, oxidative stress was created by high irradiance (photo-oxidative stress) and/or carbon depletion. The extent of lipid oxidative damage, antioxidant enzyme activities and the amount of hydrogen peroxide excreted by the algae to the surrounding seawater medium were measured. The results suggest that the two species have different strategies: the annual C. glomerata could be classified as a more stress-tolerant species and the ephemeral E. ahlneriana as a more stress-susceptible species. Low temperature in combination with high irradiance created less lipid oxidative damage in C. glomerata than in E. ahlneriana, which was probably related to the higher regular activities of the hydrogen peroxide scavenging enzymes catalase and ascorbate peroxidase in C. glomerata, whereas in E. ahlneriana high activities of these enzymes were only obtained after the induction of oxidative stress. Superoxide dismutase activities were similar in both species, but the mechanisms to remove the hydrogen peroxide produced by the action of this enzyme were different: more through scavenging enzymes in C. glomerata and more through excretion to the seawater medium in E. ahlneriana. The high excretion of hydrogen peroxide, possibly in combination with brominated volatile halocarbons, by E. ahlneriana may have a negative effect on epiphytes and may partly explain why this alga is usually remarkably devoid of epiphytes and grazers compared to C. glomerata.  相似文献

An 11S seed globulin has been isolated from Phaseolus aureus and P. vulgaris by zonal isoelectric precipitation and the MWs of the constituent subunits determined. The protein of P. vulgaris occurs in the protein body fraction and its chemical composition, including the N-terminal amino acids and amino acid composition has been determined. The similarity between the 11S globulin of the two Phaseolus spp. and legumin from other leguines is discussed.  相似文献

Type E botulinum toxin (BoNT/E)-producing Clostridium butyricum strains isolated from botulism cases or soil specimens in Italy and China were analyzed by using nucleotide sequencing of the bont/E gene, random amplified polymorphic DNA (RAPD) assay, pulsed-field gel electrophoresis (PFGE), and Southern blot hybridization for the bont/E gene. Nucleotide sequences of the bont/E genes of 11 Chinese isolates and of the Italian strain BL 6340 were determined. The nucleotide sequences of the bont/E genes of 11 C. butyricum isolates from China were identical. The deduced amino acid sequence of BoNT/E from the Chinese isolates showed 95.0 and 96.9% identity with those of BoNT/E from C. butyricum BL 6340 and Clostridium botulinum type E, respectively. The BoNT/E-producing C. butyricum strains were divided into the following three clusters based on the results of RAPD assay, PFGE profiles of genomic DNA digested with SmaI or XhoI, and Southern blot hybridization: strains associated with infant botulism in Italy, strains associated with food-borne botulism in China, and isolates from soil specimens of the Weishan lake area in China. A DNA probe for the bont/E gene hybridized with the nondigested chromosomal DNA of all toxigenic strains tested, indicating chromosomal localization of the bont/E gene in C. butyricum. The present results suggest that BoNT/E-producing C. butyricum is clonally distributed over a vast area.  相似文献

Anaerobic hydrogenase activity in whole cells and cell-free preparations of H2-induced Anacystis was studied both manometrically and spectrophotometrically in presence of physiological and artificial electron acceptors.2. Up to 90% of the activity measured in crude extracts were recovered in the chlorophyll-containing membrane fraction after centrifugation (144 000 × g, 3 h).3. Reduction of methyl viologen, diquat, ferredoxin, nitrite and NADP by the membranes was light dependent while oxidants of more positive redox potential were reduced also in the dark.4. Evolution of H2 by the membranes was obtained with dithionite and with reduced methyl viologen; the reaction was stimulated by detergents.5. Both uptake and evolution of H2 were sensitive to O2, CO, and thiol-blocking agents. The H2-dependent reductions were inhibited also by the plastoquinone antagonist dibromothymoquinone, while the ferredoxin inhibitor disalicylidenepropanediamine affected the photoreduction of nitrite and NADP only. 3-(3,4-Dichlorophenyl)-1,1-dimethylurea did not inhibit any one of the H2-dependent reactions.6. The results present evidence for a membrane-bound ‘photoreduction’ hydrogenase in H2-induced Anacystis. Lectin variability within Phaseolus coccineus is revealed by non-denatured electrophoretic patterns and immunological labelling of total seed protein extracts, showing that the different cultivars and wild varieties studied can be classified into three main categories according to the number of isolectins (three, two or one) present in each extract. Attempts in the purification of these isolectins were performed on three different affinity systems in which ligands were thyroglobulin (known to purify the P. vulgaris isolectins), pig red cell membrane ghosts (stroma) or antibodies against the P. vulgaris cv. Contender E2L2 isolectin. The P. coccineus isolectins exhibit varied affinities towards thyroglobulin and stroma, the cathodic and anodic (pH 4.5) isolectins being respectively retained by the two systems, whereas the antibody affinity system is the only one able to purify the totality of the isolectins present in an extract.  相似文献

The common seastars Leptasterias polaris and Asterias vulgaris show competitive interactions in shallow subtidal communities in the northern Gulf of St. Lawrence, particularly during summer when aggregations of the two seastars forage on mussel beds at 1-2 m in depth. We examined interactions between the two seastars in a different situation, in a mussel bed at 6 m in depth (a rare situation in this region). In the deeper mussel bed, seastars were three times more abundant than in the shallower beds, and the mussels were larger. The deeper bed disappeared rapidly due to the intense predation. Although decreased prey abundance should have favored interference interactions, we did not detect either partitioning of mussels by size or avoidance of A. vulgaris by L. polaris as previously reported when mussels are in short supply in shallower water. The lack of an avoidance behavior by L. polaris, together with the higher proportion of L. polaris than A. vulgaris that were feeding, suggests that in this situation, the dominance of A. vulgaris (observed in shallower water) is attenuated, or that L. polaris may dominate.  相似文献
