石斛内生细菌DEB-2对5种辣椒病原真菌的抑制作用   总被引:3,自引:2,他引:1  
为了探明石斛内生细菌DEB-2菌株对贵州辣椒病原真菌的抑制作用,采用生长速率法测定了菌株DEB-2发酵液对辣椒黑斑病病原菌Alternaria alternate、辣椒疫病病原菌Phytophthora capsici、辣椒黑点炭疽病病原菌Colletotrichum capsici、辣椒红色炭疽病病原菌Colletotrichum gloesporioide和辣椒早疫病病原菌Alternaria solani的毒力。结果显示,内生菌DEB-2菌株能显著抑制5种辣椒病原真菌菌丝生长和孢子萌发,菌丝尖端均有不同程度变粗、膨大、崩解或者菌丝分枝增多的现象;分生孢子及芽管产生畸形。DEB-2菌株发酵液对5种病原菌菌丝生长的抑制中浓度EC50值分别是28.5、76.2、80.7、79.9、72.0 μL/mL;对辣椒疫病病原菌和早疫病病原菌分生孢子萌发的EC50分别为73.5 μL/mL和68.7 μL/mL。研究表明,DEB-2菌株能有效地同时抑制5种辣椒病原真菌菌丝生长和孢子萌发,对该5种辣椒真菌病原菌具有一定的生防潜力,可作为生防菌进行开发和应用。  相似文献

为评价牛至精油及其2种主要成分香芹酚和百里酚在植物真菌病害防治方面的应用前景,采用菌丝生长速率法和孢子萌发法测定3种物质对16种植物病原真菌菌丝生长和孢子萌发的抑制活性,并采用黄瓜子叶法测定其对黄瓜灰霉病的防治效果。结果表明,牛至精油在500 mg/L浓度处理下可完全抑制番茄早疫病菌Alternaria solani、黄瓜灰霉病菌Botrytis cinerea和小麦根腐病菌Bipolaris sorokiniana等12种植物病原真菌的菌丝生长,对棉花黄萎病菌Verticillium dahliae的菌丝生长抑制率也达到97.66%。牛至精油对这13种病原真菌的EC50在83.09~236.58 mg/L之间,香芹酚和百里酚对这13种病原真菌的毒力均高于牛至精油,其中香芹酚对黄瓜灰霉病菌、油菜菌核病菌Sclerotinia sclerotiorum、小麦全蚀病菌Gaeumannomyces graminis、棉花黄萎病菌和小麦茎基腐病菌Fusarium pseudograminearum菌丝生长的毒力较强,EC50分别为9.09、22.45、24.28、28.40和28.80 mg/L;百里酚对黄瓜灰霉病菌和小麦全蚀病菌菌丝生长的毒力较强,EC50分别为21.32 mg/L和27.08 mg/L。香芹酚和百里酚对棉花黄萎病菌、玉米大斑病菌Setosphaeria turcica、苦瓜枯萎病菌Fusarium oxysporum f.sp.momordicae、黄瓜灰霉病菌和黄瓜炭疽病菌Colletotrichum orbiculare的孢子萌发亦有较强毒力,其EC50为3.78~289.07 mg/L。1 000 mg/L牛至精油对黄瓜灰霉病的预防效果与400 mg/L嘧霉胺悬浮剂的预防效果无显著差异;500 mg/L百里酚对黄瓜灰霉病的预防效果和治疗效果均与400 mg/L嘧霉胺悬浮剂的效果相当,而香芹酚的治疗效果显著优于400 mg/L嘧霉胺悬浮剂。表明牛至精油及其主要成分香芹酚和百里酚的抑菌谱广且活性强,具有作为植物源杀菌剂的开发应用潜力。  相似文献

本文对美国进境大豆及其中混杂的豆杆上发现的拟茎点属(Phomopsis sp.)进行了形态学和分子生物学鉴定。无论从形态学还是从分子生物学角度都支持将该检测种鉴定为Phomopsis longicolla。该种病原真菌是在大豆上发生的危险性病原真菌,国内过去未见报道。  相似文献

为明确大豆抗大豆花叶病毒(soybean mosaic virus,SMV)SC4株系的基因位点RSC4(resistance to SMV strain SC4,RSC4)的候选基因Glyma.14G204700在大豆不同抗性品种中的序列结构特征和保守结构域,以齐黄1号、科丰1号、大白麻和南农1138-2共4个大豆品种为材料,通过基因克隆获得大豆Glyma.14G204700基因的cDNA全长序列,并采用生物信息学方法分析其序列特征和编码蛋白的理化特性及结构特征。结果表明,大豆Glyma.14G204700基因在4个大豆品种中的cDNA全长为4 719~4 776 bp,编码1 295~1 307个氨基酸,预测蛋白的分子量为148.38~149.33 kD,等电点为5.53~5.61,均为具较强亲水性的非分泌蛋白。Glyma.14G204700蛋白含有植物抗病基因家族蛋白的保守功能域——核苷酸结合域和富含亮氨酸重复结构域。该蛋白的二级结构主要由α-螺旋、无规则卷曲、延伸链和β-折叠组成,所占比例分别为59.45%~61.08%、28.65%~30.15%、8.11%~8.88%和1.61%~2.16%。启动子序列分析发现该基因含有脱落酸、低温及干旱等多种逆境胁迫响应元件。表明大豆RSC4抗病候选基因Glyma.14G204700在大豆不同抗性品种中具有不同的等位变异,优异等位变异的鉴定和开发可为抗SMV大豆育种提供基础材料。  相似文献

通过对辽宁省多个地区的调查,发现此病害发生普遍,尤其是二年生的五味子茎基腐病发生严重。五味子茎基腐病在5月上旬开始出现,6月初为发生盛期。本文对五味子茎基腐病进行了症状描述,并对不同地点采集的病样分别进行木质部和韧皮部病原物的分离、纯化。对分离得到的菌株按照柯赫氏法则进行致病性测定并对病原菌进行鉴定,结果表明,此病害可由木贼镰刀菌(Fusarium equiseti)、茄腐镰刀菌(F.solani)、尖孢镰刀菌(F.oxysporum)和半裸镰刀菌(F.semitectum)4种镰刀菌属真菌引起。针对此病害的发生特点,提出了相应的防治措施。  相似文献

为明确引起福建省大豆炭疽病的病原菌种类,2018—2019年从福建省福州、三明、莆田、泉州和南平市采集具有大豆炭疽病症状的大豆豆荚,采用组织分离法分离病原菌,结合形态学特征和多基因系统发育分析对病原菌进行鉴定。结果表明,从采集的豆荚样本中共分离获得152株炭疽菌菌株,经形态学初步鉴定分属于3种类型,从3种类型的病原菌中分别选取3株代表菌株进行多基因系统发育分析,这9株代表菌株分别与Colletotrichum plurivorum、平头炭疽菌C.truncatum和胶孢炭疽菌C.gloeosporioides聚在一起。形态学和多基因系统发育分析结果表明,引起福建省大豆炭疽病的病原菌有3种,分别为C.plurivorum、平头炭疽菌和胶孢炭疽菌,其占比分别为10.53%、50.00%、39.47%。致病性测定结果表明,C.plurivorum、平头炭疽菌和胶孢炭疽菌对大豆均有致病作用,但不同的菌株致病力存在差异。表明平头炭疽菌是引起福建省大豆炭疽病的主要病原菌,而C.plurivorum为引起我国大豆炭疽病的新纪录种。  相似文献

采用真核生物通用引物ITS1/ITS4,对来自江西省吉安县2个车前草穗枯病菌菌株进行rDNA-ITS区段的PCR扩增,对扩增产物进行测序,结果获得2个长度均为579 nt且序列完全一致的rDNA-ITS序列,该序列与GenBank中当归间座壳菌(Diaporthe angelicae)及其无性态拟茎点霉菌(Phomopsis subordinaria)对应序列的同源性高达99.0%,根据以rDNA-ITS序列同源性大小划分种类的标准,将车前草穗枯病菌鉴定为当归间座壳菌。  相似文献

ABSTRACT Diaporthe/Phomopsis helianthi causes brown stem canker of sunflower (Helianthus annuus) and is responsible for considerable yield loss. This species shows considerable variation for morphological characters, growth, and pathogenicity. Molecular variability of two sample groups was assessed with amplified fragment length polymorphism (AFLP) markers. Isolates of the first sample were collected from infected sunflower tissues from the main regions in France where the crop is grown, whereas isolates from the second sample came from stems within a single field of sunflower. A soybean strain was taken as an outgroup for AFLP analyses. Within sample one, the greatest genetic distance among isolates was 0.97, whereas it was 0.44 within sample two isolates. For the whole of France, the average genetic distance was 0.68, whereas in the one field it was 0.12. Nei's genetic diversity indices were 0.20 and 0.06 for France and for one field, respectively. The greatest genetic distance was found between isolates from the most northern crops. The greatest genetic distance between D. helianthi isolates and the strain isolated from soybean was similar to that observed for D. helianthi isolates from different geographical areas. The problems in defining the genus Phomopsis are discussed. It is shown that internal transcribed spacer sequencing could be a useful criteria for Diaporthe/Phomopsis species determination. The considerable genetic variability of the pathogen could lead to the occurrence of new strains that could be more aggressive or more resistant to chemical control.  相似文献

BACKGROUND

The fungus Diaporthe eres was isolated from a fungal pathogen‐infected leaf of Hedera helix (English ivy) exhibiting necrosis. It is hypothesized that the causative fungus produces phytotoxins as evidenced by necrotic lesions on the leaves.

RESULTS

The fungus was isolated and grown in Czapek Dox broth culture medium and potato dextrose broth culture medium and identified as Diaporthe eres. The ethyl acetate extracts of the culture broths were phytotoxic to lettuce (Lactuca sativa) and bentgrass (Agrostis stolonifera). 3,4‐Dihydro‐8‐hydroxy‐3,5‐dimethylisocoumarin ( 1 ) and tyrosol ( 2 ) were isolated and identified as the phytotoxic constituents. Six analogs of 3,4‐dihydro‐isocoumarin were synthesized and shown to be phytotoxic. The synthesized 3,4‐dihydro‐8‐hydroxy‐3,7‐dimethylisocoumarin and 3,4‐dihydro‐8‐hydroxy‐3,3,7‐trimethylisocoumarin were two‐ to three‐fold more phytotoxic than the naturally occurring 1 in a Lemna paucicostata growth bioassay.

CONCLUSION

Synthesis and herbicidal activities of the several new analogs of 1 are reported for the first time. These promising molecules should be used as templates for synthesis and testing of more analogs. © 2017 Society of Chemical Industry  相似文献

To obtain a better understanding of the onset and the progress of epidemics caused by Diaporthe adunca on Plantago lanceolata we investigated the role of the asexual phase of the fungus not only in the transmission of the disease from season to season, but also in the spread of the fungus within the same season. Splash dispersal of conidia during rain episodes proved to be an effective method for the spread of the fungus. Conidial production by pycnidia present on scapes left on the ground during winter was delayed in comparison to that of pycnidia produced on greenhouse infected scapes, indicating that the fungus overwinters with immature pycnidia. Latent infections occurred in 8.3% of the seedlings which emerged from seeds originating from infected spikes. A scenario for the transmission and spread of the fungus is proposed.  相似文献

向日葵茎溃疡病菌(Diaporthe helianthi)是我国进境植物检疫性病原真菌。本研究针对D.helianthi的cal基因保守序列设计特异性引物,建立该病菌的重组酶聚合酶扩增检测技术(RPA)方法,并对其特异性、灵敏度及适用性进行评价。结果显示,建立的RPA方法特异性强,只有3个D.helianthi样品能够检测到234 bp的目的片段;方法灵敏度达到0.1 ng/μL;从模拟带菌的种子中也能够成功检测到目标扩增片段。本研究建立D.helianthi的RPA检测方法具有较高的特异性和灵敏度,能够直接应用于种子带菌检测,适用于口岸进境向日葵籽的现场快速检测。  相似文献

An outbreak of dieback on plum trees at the Institute of Horticultural Research, East Mailing, in the summer of 1983 resembled a disease prevalent in Britain earlier this century attributed to Diaporthe perniciosa. Numerous isolates of this fungus were obtained from diseased tissues but an attempt to reproduce the disease by inoculation was unsuccessful.
