为了解金钗石斛乙醇提取物正丁醇部位的化学成分,通过硅胶、凝胶、反相硅胶和高效液相等各种色谱技术进行分离纯化得到3个化合物,并根据核磁共振、质谱等波谱手段确定其结构分别为2,11-epoxy-11,13-dihydroxypicrotoxano-3(15)- lactone (1)、2,4,6-三甲氧基苯-1-O-β-d-葡萄糖苷 (2)和3-methylbutan-1-ol β-d-glucopyranoside (3)。其中化合物1是一个新的木防己毒烷型倍半萜,该类化合物是金钗石斛中具有代表性的倍半萜类型。所有化合物均没有α-葡萄糖苷酶和酪氨酸酶抑制活性,同时化合物1也没有神经保护活性。 相似文献

三峡区域药用植物拐芹的根中富含倍半萜类抗溃疡活性成分没药烷吉酮,为可开发和利用的中草药资源。本文对该化合物进行了提取分离、结构修饰和初步的构效关系研究,从拐芹根茎中提取并分离了没药烷吉酮,通过选择性还原、缩合和加成反应制备了四个没药烷吉酮氨基甲酰腙衍生物。用核磁共振波谱、质谱、红外和元素分析等方法确证了其结构,并测试了其体外对H~+/K~+-ATP酶的抑制活性和细胞毒活性。没药烷吉酮还原衍生物2及4-氯苯基取代的氨基甲酰腙衍生物4d较阳性对照药物奥美拉唑具有更好的体外抗溃疡活性(IC5024μmol/L)。本文探明了没药烷吉酮衍生物的结构对体外H~+/K~+-ATP酶抑制活性的影响,为消化性溃疡的治疗提供了新型倍半萜类候选药物。 相似文献

The major sesquiterpenes in the foliage of Dacrydium cupressinum are α-longipinene, longifolene, longibornyl acetate, caryophyllene, caryophyllene oxide, humulene, α- and β-selinene, β- and δ-elemene, aromadendrene and the rare 9βH-caryophyllene. Sesquiterpene levels vary greatly from tree to tree. As this variation is largely independent of environmental factors, genetic control is proposed. Longifolene and α-longipinene levels are closely correlated, as are those of caryophyllene and humulene. The biosynthetic implications of these correlations are discussed. 相似文献

Artemisia douglasiana afforded, in addition to known compounds, two new C14-acetylenes, five longipinene derivatives, three nerolidol derivatives, a lactone and a ketone with a new carbon skeleton and lavendulol-2-methylbutyrate. The structures were elucidated by spectroscopic methods and some chemical transformations. The configurations of several oxo longipinene-7, 9-di- and 7, 8, 9-triesters isolated previously were corrected. The biogenesis of the new lactones is discussed briefly. 相似文献

The biosynthesis of sesquiterpene hydrocarbons was studied in maritime pine (Pinus pinaster) needles by incorporation of 14CO2, [1-14C] acetate and [2-14C] mevalonate. It was shown that the mechanisms of sesquiterpene biosynthesis are different according to the applied tracer. 微生物来源的syrbactins属于十二元内酰胺短肽类化合物,包括丁香霉素(syringolins)、滑杆菌素(glidobactins)、cepafungins和luminmycins等。其中,syringolins、glidobactins、luminmycins等几个化合物生物合成基因簇已被克隆、测序和异源表达。研究发现它们的十二元内酰胺环骨架都是由非核糖体肽合成酶(NRPS)-聚酮合酶(PKS)复合体采用模块化组装的方式,将系列底物组装而成。这类化合物因具有优异的蛋白酶体抑制活性而受到广泛关注。本文从syrbactins的分子结构、生物合成、作用机理等几个方面进行综述,介绍了近年来syrbactins的研究进展。 相似文献

The yeast Saccharomyces cerevisiae was chosen as a microbial host for heterologous biosynthesis of three different plant sesquiterpenes, namely valencene, cubebol, and patchoulol. The volatility and low solubility of the sesquiterpenes were major practical problems for quantification of the excreted sesquiterpenes. In situ separation of sesquiterpenes in a two-phase fermentation using dodecane as the secondary phase was therefore performed in order to enable quantitative evaluation of different strains. In order to enhance the availability of the precursor for synthesis of sesquiterpenes, farnesyl diphosphate (FPP), the ERG9 gene which is responsible for conversion of FPP to squalene was downregulated by replacing the native ERG9 promoter with the regulatable MET3 promoter combined with addition of 2 mM methionine to the medium. This strategy led to a reduced ergosterol content of the cells and accumulation of FPP derived compounds like target sesquiterpenes and farnesol. Adjustment of the methionine level during fermentations prevented relieving MET3 promoter repression and resulted in further improved sesquiterpene production. Thus, the final titer of patchoulol and farnesol in the ERG9 downregulated strain reached 16.9 and 20.2 mg/L, respectively. The results obtained in this study revealed the great potential of yeast as a cell factory for production of sesquiterpenes. 相似文献
