[reaction: see text] In this paper we describe an Al-promoted, Co-mediated O-->C rearrangement reaction of cyclic enol ethers. This process delivers functionalized cyclohexanones with good to excellent levels of diastereocontrol, whereby the product stereochemistry is dependent on the E/Z-stereochemistry of the starting enol ether. The rearrangement process also permits access to highly substituted alpha-spirocyclic cyclohexanones as well as cyclopentanones. The latter rearrangement appears to proceed via an unusual 5-(enolendo)-exo-trig cyclization process.  相似文献

The thermal reaction of chiral 2-phenyl-4-(p-X-phenyl)-5-(p-Y-phenyl)-4-tert-butyldimethylsilylperoxy-4H-isoimidazoles (5b: X = CF3, Y = OMe; 5c: X = CF3, Y = F) was carried out in DMSO. The chiral 2-phenyl-5-(p-X-phenyl)-5-(p-Y-phenyl)-5H-imidazol-4-ones (4b: X = CF3, Y = OMe; 4c: X = CF3, Y = F) were quantitatively obtained in 50-60% enantiomer excess (ee). The mechanism for the reaction was proved to be stereoselective 1,5-phenyl migration. Although the sigmatropic 1,5-phenyl migration should be thermally allowed according to the Woodward-Hoffmann rule, the migration actually includes a stepwise process.  相似文献

The potential energy surface of the rearrangement of 3-vinylmethylenecyclobutane to 4-methylenecyclohexene has been studied computationally using density functional theory (B3LYP) and complete active space ab initio methods (CASSCF and CASPT2). The parent reaction is nonconcerted and occurs through several parallel diradical pathways. Transition structures and diradical intermediates are highly comparable in energy, with no deep potential energy well on the potential energy surface. In the substituted system, stereoelectronic effects of the trialkylsiloxy group regulate torquoselectivity in the bond-breaking processes and this, combined with low barriers to cyclization, leads to a stepwise Cope rearrangement that is, nevertheless, stereoselective.  相似文献

A highly stereoselective Saucy-Marbet rearrangement using chiral ynamides and propargyl alcohols is described here. This rearrangement can be catalyzed by para-nitrobenzenesulfonic acid and leads to high diastereoselectivities for a range of different chiral propargyl alcohols and ynamides in a stereochemically intriguing matched, mismatched or indifferent manner. The stereoselective Saucy-Marbet rearrangement of ynamides provides an excellent entry to highly substituted chiral homo allenyl alcohols.  相似文献

A series of 1,3-dioxanyl vinyl acetals were readily synthesized from the corresponding dioxanone by a reduction and in situ acylation followed by Petasis olefination. A collection of structurally complex and chemically diverse small molecules is a useful tool to explore cell circuitry. In this article, we report the split-pool synthesis of more than 3000 spirooxindoles on high capacity macrobeads. The key reaction to assemble the spirooxindole core stereoselectively is a Lewis acid variant of the Williams' three-component coupling. After formation, the skeleton was elaborated using Sonogashira couplings, amide forming reactions, and N-acylations of gamma-lactams. The final library was analyzed by sampling individual macrobeads and by using binomial confidence limits. It was determined that at least 82% of the library compounds should have better than 80% purity. To demonstrate the utility of our discovery process, a high-throughput chemical genetic modifier screen was performed using stock solutions of the resultant products. A number of positives were identified as enhancers of the cellular actions of latrunculin B, an actin polymerization inhibitor. Through resynthesis, we confirmed one of the positives and demonstrated that, in yeast cells, it has an EC50 in the sub-micromolar range.  相似文献
