Summary The introgression of wildfire (races 0 and 1) and angular leaf spot (ALS) resistance from N. rustica var. Brasilea into N. tabacum has proved economically useful in Zimbabwe although the mode of inheritance of, and genetic relationships between the resistance are unknown. This study was undertaken to (1) examine the mode of inheritance of the resistance to races 0 and 1 of wildfire, and ALS, (2) determine the genetic relationship between the resistances and (3) establish whether the N. rustica-derived wildfire race 0 resistance is allelic to that obtained from N. longiflora. Inheritance was examined under greenhouse and field conditions by studying disease reactions in the parental, F1, F2 and backcross generations derived from crosses of three susceptible lines to a resistant line Nr-7. Three-point backcrosses to the susceptible parent were examined for linkage and segregating generations from a cross of Nr-7 to Burley 21 which carries the N. longiflora race 0 resistance were used to test for allelism. In general, we observed that all resistances are determined by a single dominant gene although some incosistent ratios were obtained likely due to misclassification of disease reactions and erratic transmission. All resistances showed linkage although pleiotropism cannot be ruled out. Allelism tests demonstrated that the N. rustica race 0 resistance is not allelic to that obtained from N. longiflora. Our findings are examined in relation to the efficacy of indirect selection for resistance.  相似文献

Summary First- and second-cycle doubled haploids of burley tobacco (Nicotiana tabacum L.) were examined to ascertain the significance of source genotype on the performance of anther-derived material and to determine the magnitude of changes from the haploid-production and chromosome-doubling stages. The anther-derived lines evaluated in this study did not exhibit siginficant yield reductions. However, significant interactions between the cycle comparison and the source genotypes indicated that source genotype can affect the degree of vigor reduction observed. Significant variation was also observed among the second-cycle doubled haploids examined in this study. Partitioning of this variation indicates that most of it is due to alteration present at the haploid-production step instead of the chromosome-doubling step. In general, the variation present in the second-cycle doubled haploids was distributed around the values observed for the source first-cycle doubled haploids. Even though unexpected variation is often observed in anther-derived materials, these results suggest that reduced vigor is not intrinsically associated with androgenetic lines of burley tobacco. The most plausible explanations for the variation and vigor losses in anther-derived lines propose that natural phenomena, given the opportunity for expression via anther culture, are responsible. Spontaneous variants present in the gametic pool of inbreds or first-cycle doubled haploids offer one possible explanation for the performance observed in anther-derived lines of tobacco.Contribution from the Department of Agronomy, University of Kentucky, Lexington, KY 40546-0091. Summary The genetic consequences of anther culture and chromosome-doubling techniques on burley tobacco (Nicotiana tabacum L.) were examined in this study. Three diploid populations, obtained from a burley tobacco inbred by conventional and anther-culture techniques, were compared. The first population consisted of 50 conventionally-selfed lines; the second population was made up of 35 doubled-haploid lines obtained from individual haploid plants by in vitro techniques (IVDH); and the third population consisted of 20 doubled-haploid lines whose chromosome complements had been doubled with colchicine (CDH). Comparisons of doubled-haploid lines with sexually-derived lines revealed significant differences for yield, maturity, leaf length, and alkaloid content. Yield reductions in the doubled-haploid populations averaged 8.5%. Significant differences observed between the IVDH and CDH populations indicate that the reported deleterious effects of colchicine contributed to the vigor reduction of doubled haploids. The anther derived lines in this study exhibited greater variation than did the sexual materials. This variation could provide useful variation for a breeding program. Variation exhibited by the sexual progeny of the highly inbred line, Kentucky 16, suggests that the differences among anther-derived materials are at least partially due to natural phenomena.Contribution from the department of Agronomy, University of Kentucky, Lexington, KY 40546-0091. Summary Anthers from Nicotiana tabacum L., cv. Wisconsin-38, were treated with various concentrations of Ethylmethane-sulphonate (EMS) during different application times.Low doses and short application times showed increases in the number of anther producing plantlets, average number of plantlets per anther, and early plantlet production. The mutagen treatment increased the variance of protein electrophoresis fractions and chlorophyll content of the plantlets.  相似文献

Summary Variation in the capacity to regenerate shoots and morphogenetic potential were investigated by cyclic culture and regeneration of Nicotiana tabacum var. BEL W3. Explants were excised from different shoots formed by the same callus line. Differences in the organogenetic in vitro response among regenerated plantlets was observed in two subsequent regenerate generations. A progressive decrease of the regenerative capacity was observed across callus line generations.  相似文献

Summary Angular leaf spot (Isariopsis griseola Sacc.) is a serious disease of French bean in the hills of India and 40 to 70 per cent of the green pods are damaged and rendered unmarketable. Crosses were made between PLB 257, (Phaseolus coccineus L.), a red flowering pole tope, resistant to angular leaf spot, and Contender (Phaseolus vulgaris L.), a highly susceptible commercial cultivar. Studies of the F1, F2, and F3 progenies indicated that PLB 257, carries a recessive gene imparting resistance to angular leaf spot.  相似文献

Summary Seven bean lines (Phaseolus vulgaris L.) with differential resistance or susceptibility to race 2 of halo blight (Pseudomonas syringae pv. phaseolicola) and a necrosis-inducing isolate of bean common mosaic virus were inoculated with one or both pathogens in combination, to determine the feasibility of dual screening to identify resistance to both pathogens simultaneously. Dual screening yielded the same results as separate screenings. Neither pathogen affected the disease expression of the other. Summary over 13000 CIAT bean accessions were evaluated for their reactions to the anthracnose (Colletotrichum lindemuthianum) and angular leaf spot (Isariopsis griseola) pathogens over a 3 yr period. Among these accessions, 156 were resistant to all races of the anthracnose pathogen collected from Popayán, Colombia. Thirty were resistant to numerous races obtained from other parts of the world, including Europe. Although many of these new resistant sources originated in Mexico and Central America, they are quite diverse for geographic origin, plant type, seed color and seed size. In addition, more than 50 of the 156 lines were also resistant to isolates of I. griseola with diverse sources of origin throughout Colombia.  相似文献

Summary Agronomic field performance traits and leaf chemical characteristics of gynogenic doubled haploids (GDH) and androgenic double haploids (ADH) were compared with selfed lines of a burley tobacco cultivar to determine which type of doubled haploids was superior. Summary Breeding tobacco resistant to CMV using Holmes' resistant line was initiated in 1966. The resistance was controlled by five genes. N from N. glutinosa, rm1 and rm2 from Ambalema and t1 and t2 from T.I. 245. The genotype of Holmes' resistant line was assumed to be NNrm1rm1rm2rm2t1t1t2t2, which showed a few-small-late (FSL) lesion reaction to TMV. This reaction facilitates the selection of CMV resistant plants in a segregating population.Hicks Broadleaf was crossed to Holmes' resistant line and backcrossed to the susceptible varieties Vam-fen-Hicks, Kutsaga 51 and T.T.4. Two resistant lines, 418 and 470, were selected and tested for their CMV resistance, yield and agronomic characters in 1976–1979. The % reduction of infection rate when compared with a check was 41.8% and 52.7% respectively. The two lines outyielded the check and had slightly higher value/kg leaf tobacco and lower % midrib. They were also taller and later maturing. Line 470 showed slight leaf-wilting in the early growth stage, which disappeared after topping.Smoking tests showed that the two lines had better scores than check T.T.5. The nicotine and tar contents in the cigarette smoke stream proved that the leaf quality was acceptable. The lines 418 and 470 were registered as T.T.6 and T.T.7.  相似文献

The existence of genetic variability for angular leaf spot (ALS) resistance in the common bean germplasm allows the development of breeding lines resistant to this disease. The BAT 332 line is an important resistance source to common bean ALS. In this work we determined the inheritance pattern and identified RAPD markers linked to a resistance gene present in BAT 332. Populations F1, F2,BCs and BCr derived from crosses between BAT 332 and cultivar Rudá were used. Rudá is a commercial cultivar with carioca type grains and susceptible to ALS. The resistance of BAT 332 to race 61.41 of the pathogen was confirmed. Segregation analysis of the plants indicated that a single dominant gene confers resistance. For identification of RAPD markers linked to the resistance gene, bulk segregant analysis (BSA) was used. Two RAPD markers,OPAA07950 and OPAO12950, linked in coupling phase at 5.10 and 5.83 cM of this gene, respectively, were identified. These molecular markers are important for common bean breeders and geneticists as source of genetic information and for marker assisted selection in breeding programs. Summary Five cytoplasmic male sterile lines of Nicotiana tabacum homozygous for a dominant rootless mutation [Rac-] were used as female parents in crosses with three male fertile varieties. Androgenetic haploids were selected by their ability to form plantlets with a normal root system, whereas hybrid plantlets failed to grow. High frequencies of androgenetic haploid plants were obtained with N. debneyi cytoplasm, and with one paternal genotype. Chromosome doubling was performed using three different methods (pollination, in vitro culture, acenaphthen) and cytoplasmic male sterile doubled haploid plants have been obtained. The results demonstrate the efficiency of this method for a rapid cytoplasm transfer in Nicotiana tabacum; the integrity of the cytoplasm is conserved. For the first time, we possess a counter selectable marker universally usable for Nicotiana tabacum and closely related species.  相似文献

Common beans (Phaseolus vulgaris) are separated into two distinct groups: Andean and Middle American. We identified CAL 143 as the first Andean bean with resistance to angular leaf spot disease caused by Phaeoisariopsis griseola. Angular leaf spot is the most widespread and economically important bean disease in southern and eastern Africa, and it is especially severe on the extensively grown Andean beans. Cal 143 was resistant in Malawi, South Africa, Tanzania, and Zambia, but it was susceptible in Uganda. This was attributed to the presence of races of P. griseola in Uganda not present in the other countries. We identified two additional Andean bean lines, AND 277 and AND 279, with resistance to angular leaf spot in Malawi. We also characterized the virulence diversity of 15 isolates of P. griseola from southern and eastern Africa into nine different races. Five of six isolates from Malawi and two of seven from Uganda, obtained from large-seeded Andean beans, were characterized into four different races considered Andean. These were compatible only or mostly with large-seeded Andean cultivars. The other eight isolates from Uganda, Malawi, and the Democratic Republic of Congo, obtained from a small- or medium-seeded Middle American beans, were characterized into five different Middle American races. These were compatible with Middle American and Andean cultivars. CAL 143 was resistant or intermediate under greenhouse conditions to all but one of the same 15 isolates from southern and eastern Africa, but it was susceptible to an isolate from Uganda obtained from a medium-seeded Middle American bean. 采用大田试验研究了苹果酸、油酸和腐殖酸对烤烟品质指标及产值效果的影响。结果表明, 3种有机酸能明显提高烟叶中P素、K素、总糖、还原糖含量、钾氯比和糖碱比值, 以及糠醛、苯甲醛、茄酮、巨豆三烯酮和新植二烯等主要香气成分的含量, 改善烟草的香吃味。同时不同程度地降低烟叶中N素、淀粉、蛋白质、烟碱和氯离子的含量, 从而协调和平衡烟叶中的化学成分, 改善烟叶的品质。其中, 苹果酸的效果最佳, 腐殖酸和油酸次之。苹果酸可明显提高烤烟的均价和上中等烟的比例, 腐殖酸可明显提高烤烟的产量、产值及上中等烟的比例。  相似文献

Summary Three triticale lines, Siskiyou, M2A-Beagle, and OK 77842 have been reported to possess resistance to bacterial leaf streak caused by Xanthomonas campestris, pv. translucens (Xct.). The three resistant lines were crossed to susceptible lines and crossed with each other. F2, BC1-F1, BC2-F1 plants were inoculated with a mixture of two Xct strains. The segregation data indicate the presence of a single dominant gene in each of the three resistant lines to bacterial leaf streak. 以烤烟品种NC89为试材, 通过天然含硒和低硒土施硒处理的盆栽试验,对烤烟苗期含硒量和根际-根外土壤硒的存在形态进行了分析。结果表明,烤烟苗期根系含硒量高于地上部,并随天然含硒和施硒量的增加而增加,且与施硒量呈极显著的正相关。天然含硒土中,残渣态硒(Res-Se)占2/3以上,而低硒土施硒下,KH2PO4-Se所占比例最大。无论天然含硒土还是施硒低硒土,水溶性硒(H2SO4-Se)所占比例均不到1%,而根际土壤总硒、KH2PO4-Se和NH3·H2O-Se的含量均高于根外土。土壤施硒处理的NH3·H2O-Se占总硒的比例高于根外土壤,而Res-Se所占比例低于根外土。由此可见,烤烟能够富集土壤中的硒,天然含硒土和低硒土施硒的硒形态分布不同,施硒土壤硒的有效性高于天然含硒土,烤烟根际硒的有效性高于根外土,烤烟根际通过促进有机态硒的形成而减少土壤矿物对硒的固定来提高土壤施硒的有效性。  相似文献

Halo-blight is an important worldwide bacterial disease of common bean (Phaseolus vulgaris L.) caused by Pseudomonas syringae pv. phaseolicola. Nine races of the pathogen and five race-specific resistance genes have been previously described. However, a quantitative response to this pathogen has also been described. The objective of this study was to identify halo-blight resistance loci linked to molecular markers that could be used in resistance breeding. Chromosomal regions related to race 5 halo-blight resistance were localized on a genetic map of RAPD and AFLP molecular markers and constructed by the analysis of a “Jules” × “Canela” F2 progeny. “Jules” shows quantitative resistance to halo-blight and “Canela” is a very appreciated but susceptible Spanish bean landrace. Two QTL for resistance to halo-blight were mapped in two linkage groups. There were four large groups, with 14–22 molecular markers each, five with 4–8 markers each, and three with 2 or 3 markers each.  相似文献

Summary The inheritance of resistance to coffee berry disease (CBD) has been studied by applying a preselection test to F2 progenies of a half diallel cross between 11 coffee varieties with different degrees of resistance and to sets of parental, F1, F2, B11 and B12 generations of crosses between resistant and susceptible varieties. True resistance to CBD appears to be controlled by major genes on three different loci. The highly resistant variety Rume Sudan carries the dominant R- and the recessive K-genes. The non-allelic interaction between these two genes is of a duplicate nature. The R-locus has multiple alleles with R 1R1alleles present in Rume Sudan and the somewhat less effective R 2R2alleles in a variety like Pretoria, which also has the K-gene. The moderately resistant variety K7 carries only the recessive K-gene. The arabica-like variety Hibrido de Timor (a natural interspecific arabica x robusta hybrid) carries one gene for CBD resistance on the T-locus with intermediate gene action. It probably inherited this gene from its robusta parent. There is circumstantial evidence that the resistance to CBD is of a stable nature, but it is advisable to accumulate in one genotype as many resistance genes as possible by combining in the breeding programme the resistance of Rume Sudan with that of Hibrido de Timor.  相似文献
