Components of the ethylene signal perception and transduction pathway (ethylene signaling pathway, ESP) were studied in respect to their involvement in regulation of UV-B-induced changes in levels of polyamines in plants Arabidopsis thaliana (L.) Heynh. Experiments were performed on 15-day old wild type (WT) plants, the mutant etr1-1 with impaired ethylene reception, and the ethylene-insensitive mutant ctr1-1 with constitutively activated ESP. The plants were cultivated aseptically. It was found that exogenous ethylene or an inhibitor of its action 1-methylcyclopropen (1-MCP), which blocks ethylene receptors did not affect the polyamine content in leaf rosettes of plants, which had not been subjected to UV-B stress. A day after UV-B irradiation at intermediate (9 kJ/m2) or high doses (18 kJ/m2), the putrescine levels increased, respectively, 6.4 and 3.0 times in WT, 4.5 and 3.2 times in etr1-1, and 5.5 and 4.7 in ctr1-1. Pretreatment with ethylene (1 μL/L) for 24 h reduced the putrescine accumulation along with the loss in spermidine and spermine pools in WT plants and, to a lesser extent, in etr1-1 mutant. Treatment with 1-MCP (50 nL/L, 3 h before and 24 h after the irradiation) enhanced plant sensitivity to UV-B, putrescine accumulation, as well as spermidine and spermine consumption in WT and, to a lesser degree, in etr1-1. The mutant ctr1-1 was insensitive to both ethylene and 1-MCP. The results show that the activation of ESP by ethylene increases plant resistance to UV-B because the irradiation stimulates accumulation of putrescine, which converts to spermidine and spermine functioning as ROS traps.  相似文献

Temperature fluctuation profoundly affects the plant growth and development. In this study, we show that ethylene receptor ETR1 is involved in regulating leaf petiole elongation mediated by higher temperatures (at 32 °C in this study). ETR1 loss-of-function mutant etr1-7 cannot elongate the leaf petiole at 32 °C as much as wild-type seedlings (WT). Overexpression of ETR1 in etr1-7 not only fully rescued the deficient in petiole elongation under higher temperature conditions but also caused longer petiole length under normal temperature conditions (22 °C). Plants with different mutant ETR1 alleles including etr1-7 etr1-1, and etr1-9 but not etr1-3 impair the petiole elongation mediated by elevated temperature. RNA-Seq analysis showed that hundreds of genes induced by elevated temperature in WT were not differentially expressed in etr1-7. Gene ontology enrichment analysis reveals that the molecular functions of these genes primarily relate to photosynthesis and protein degradation. Furthermore, genes involved in regulating organ elongation (such as BRI1-EMS-SUPPRESSOR 1, BES1), are significantly up-regulated in WT rather than in etr1-7 after the treatment of higher temperature. The results from this study suggest ETR1 is involved in regulating Arabidopsis response to elevated ambient temperature in both molecular and morphological levels.  相似文献

Diverse functions of ethylene in plants may depend on its ability to interact with other hormones. We studied the participation of ethylene in the regulation of accumulation and metabolism of cytokinins comparing ethylene-insensitive mutant plants of arabidopsis (Arabidopsis thaliana [L.] Heynh., etr1-1) with the plants of original ecotype Columbia (Col-0). Because cytokinins can regulate growth of both leaves and roots, we determined the weights of these organs and the ratio between them. The content of zeatin and its riboside in the roots of etr1-1 plants was two times greater than in Col-0 plants, which could be accounted for by inhibition of conversion of these forms of cytokinins into 9-N-glucosides. In the leaves of mutant plants, expression of IPT3 gene responsible for the synthesis of cytokinins was more intense than in Col-0 plants, which could also contribute to a rise in the content of cytokinins. In this case, the weight of roots in etr1-1 mutants was lower than in the plants of original ecotype. Because high concentrations of cytokinins can inhibit root growth, suppression of accumulation of their biomass in mutant plants may be related to a greater content of cytokinins therein. The obtained results suggest that ethylene can suppress accumulation of cytokinins and, thereby, maintain redistribution of biomass in favor of the roots, which is important for plant adaptation to a shortage of water and ions.  相似文献

The responses of 14-day-old Arabidopsis thaliana (L.) Heynh. plants to UV-B irradiation (280–320 nm) and ABA treatment were investigated. Wild-type plants as well as ethylene-insensitive etr1-1 and ctr1-1 mutants were used. Theetr1-1 mutant considerably differed from the ctr1-1 one in the fresh weight production after UV-B treatment (29.5 kJ/m2). The irradiated etr1-1 plants fell well behind the nonirradiated ones during the first two days after stress, but by the 8th day, their weight attained 70% of control plant weight. In contrast, Ctr1-1 mutant weight comprised 70% of control level after two days of stress but, by the 8th day, it was only 56% of the weight of control plants. In wild-type and ctr1-1 plants, ABA, in the 8 × 10–6 to 2 × 10–4 M concentration range, increased the difference between the weights of nonirradiated and irradiated plants, but in etr1-1 plants, ABA decreased this difference. The etr1-1, ctr1-1, and wild-type plants were very similar in the dynamics of ethylene evolution after UV-B treatment (7.4 kJ/m2). In wild-type, etr1-1, and ctr1-1 plants, ABA, in a concentration-dependent manner, inhibited UV-B-induced ethylene evolution to the same extent. The results obtained show that ABA exerted an opposite effect on UV-B-dependent growth in the plants with active (wild type and ctr1-1) and blocked (etr1-1) ethylene signal pathway, whereas the inhibition of ethylene synthesis by ABA was not related to ethylene signal transmission.  相似文献

The effects of the salt stress (200 mM NaCl) and exogenous jasmonic acid (JA) on levels of osmolytes and flavonoids in leaves of four-week-old Arabidopsis thaliana L. plants of the wild-type (WT) Columbia-0 (Col-0) and the mutant jin1 (jasmonate insensitive 1) with impaired jasmonate signaling were studied. The increase in proline content caused by the salt stress was higher in the Col-0 plants than in the mutant jin1. This difference was especially marked if the plants had been pretreated with exogenous 0.1 μM JA. The sugar content increased in response to the salt stress in the JA-treated WT plants but decreased in the jin1 mutant. Treatment with JA of the WT plants but not mutant defective in jasmonate signaling also enhanced the levels of anthocyanins and flavonoids absorbed in UV-B range in leaves. The presence of JA increased salinity resistance of the Col-0 plants, since the accumulation of lipid peroxidation products and growth inhibition caused by NaCl were less pronounced. Under salt stress, JA almost did not render a positive effect on the jin1 plants. It is concluded that the protein JIN1/MYC2 is involved in control of protective systems under salt stress.  相似文献

The effects of blue light (BL) and jasmonic acid (JA) on morphogenesis of Arabidopsis thaliana (L.) Heynh seedlings of genotypes Col and Ler and their mutants, namely, axr1-3 and jar1-1 mutants resistant to IAA and JA, respectively, and a CRY1 photoreceptor-deficient mutant hy4 were studied. Both 1 μM JA and BL exposure retarded hypocotyl growth of Ler, Col, and jar1-1 seedlings, whereas JA had no effect on hypocotyl growth of axr1-3, but the suppression of hypocotyl growth of this mutant by BL was even more noticeable than that of Ler, Col, and jar1-1. JA and BL applied simultaneously inhibited hypocotyl growth of axr1-3 and especially of Ler, Col, and jar1-1 more than either of factors applied separately. The hy4 mutant did not respond to BL, whereas JA stimulated its hypocotyl growth. JA did not change the cotyledon size of Col, axr1-3, and jar1-1 and reduced the cotyledon size of Ler and hy4. BL enhanced the cotyledon growth of all wild-type and mutant plants used in the study. The cotyledon sizes of all plants except Ler were also increased when JA and BL were applied together. Some of the growth responses correlated with the endogenous IAA and ABA contents. Thus, for example, the hypocotyl and cotyledon growth retardation of Ler seedlings in the presence of JA correlated with a reduced level of free IAA and a considerable increase in the free ABA level in plants grown both in darkness and in BL. Under other growth conditions, no correlation between the endogenous IAA and ABA levels and A. thaliana seedling growth was noted. The interaction between the signal transduction pathways triggered by BL and JA at the early stages of arabidopsis morphogenesis is discussed on the basis of Col, Ler, axr1-3, and jar1-1 hypocotyl growth responses.  相似文献

Flowers of the complex orchid hybrid Burrageara ‘Stefan Isler Lava Flow’ had been shown previously to react sensitively to ethylene. Via Agrobacterium tumefaciens, the mutant ethylene receptor ETHYLENE RESPONSE 1 (etr1-1) from Arabidopsis thaliana under the control of the flower-specific promoter FLOWER BINDING PROTEIN 1 (fbp1) from Petunia hybrida was transferred to Burrageara. One single-copy event was analyzed in this study aiming to investigate the expression of the fbp1::etr1-1 transgene in different plant and flower organs by quantitative RT-PCR and the reaction of flowers and inflorescences to ethylene. It was shown that the heterologous promoter led to high expression levels in the perianth of the orchid flowers compared to low levels in leaves and roots. The expression shift to the first whorl (sepals) described here corresponds to extended expression of endogenous B class MADS box homeotic genes in orchids in general. The transgenic plants grew and developed similar to the wild-type plants, except for slightly faster rooting in vitro and smaller flowers. Flower longevity was improved by 7 days in 10 ppm ethylene. Moreover, bud drop starting at day 5 of incubation of inflorescences in 10 ppm ethylene in the wild-type was efficiently prevented for at least 19 days in the fbp1::etr1-1 transgenic plants. The function of the tissue-specific promoter fbp1 and the mutant receptor etr1-1 was shown for the first time in a monocotyledonous plant.  相似文献

The effects of treatment with ethylene (0.01–100 μl/l) on ABA and polyamine contents and treatment with ABA on ethylene synthesis, polyamines content, and the resistance to UV-B radiation of two-week-old Arabidopsis thaliana (L.) Heynh, Columbia ecotype plants grown u?er sterile conditions were studied. Ethylene stimulated the accumulation of polyamines only at concentrations of 0.1–10 μl/l, which could activate ABA synthesis. Treatment with ABA (50–5000 μM, 1 μl per plant) decreased the UV-B-induced ethylene synthesis and a spermine and spermidine loss, increasing the content of putrescine, the precursor of these polyamines. ABA inhibited fresh weight accumulation in irradiated and nonirradiated plants but prevented them from severe damage and death at the high (18 kJ/m2) and lethal (27 kJ/m2) UV-B dose, respectively. The data obtained demonstrated a mutual regulation of ethylene and ABA syntheses and the participation of these hormones in the control of the polyamine level during adaptation of A. thaliana to UV-B stress.  相似文献

ERFs are downstream component in ethylene signaling pathway and involved in plant’s abiotic stress response. The specific role of ERFs under stress and the molecular mechanism underlying the signaling cross talk still need to be elucidated. This study describes the isolation and characterization of ZmERF1 promoter. There were many cis-regulatory elements related to stress responses in the ZmERF1 promoter sequence. ZmERF1 could be highly induced by ABA and ethylene treatment in maize, suggesting that it might be at the crossroads of multiple hormone signaling pathways. Furthermore, ZmERF1 transgenic Arabidopsis lines (35S::ZmERF1) showed higher salt-tolerant, drought- and heat resistance. Consistently, tolerance-related genes were up-regulated in 35S::ZmERF1 lines compared with the WT plants in Arabidopsis. Overall, ZmERF1 might play an important role in plant resistance to a coercive environment by mediating various physiological processes via ethylene and ABA signaling pathways.  相似文献

The roles of ethylene (ET) or salicylic acid (SA) in plant response to low temperature (LT, 5 °C) have been implicated. However, the combined effect of ET- and SA-signaling on plant growth and metabolism under LT remains to be evaluated. In this study, we comparatively analyzed the response of Arabidopsis ethylene insensitive (ein) 2-1 (an ET insensitive mutant), nonexprressor of pathogenesis relative (npr)1-1 (an SA insensitive mutant) and double mutant ein2-1/npr1-1 plants to LT. The results show that a LT of 5 °C induced plant growth retardation to a less degree in ein2-1, an intermediate degree in npr1-1, but a much larger in ein2-1/npr1-1 compared to the wild-type (WT) plants. The LT susceptibility of the ein2-1/npr1-1 plants was correlated to a lower net photosynthetic rate and proline content, and a higher content of H2O2 and malondialdehyde and electrolyte leakage relative to the WT plants. Lower activities of superoxide dismutase, peroxidase, and catalase, as well as a lower glutathione content and a ratio of its reduced form to its oxidized form were also observed in the double mutant plants as compared with the WT plants. However, at normal conditions (23 °C), all the tested physiological and biochemical parameters were comparable between the ein2-1/npr1-1 and WT plants, and plant growth was even better in the double mutant than in the WT plants. On the contrary, most of the above-mentioned parameters were advantageous in the ein2-1 and npr1-1 plants over the WT plants under the LT conditions. These data suggest that a parallel function or physiological redundancy of nonexpressor of pathogenesis relative 1 and ethylene insensitive 2 existed in the Arabidopsis plant response to the LT. On the other hand, an interaction between ET- and SA-signaling occurred during this process.  相似文献

Casein kinase II (CK2), an evolutionarily well-conserved Ser/Thr kinase, plays critical roles in all higher organisms including plants. CKB1 is a regulatory subunit beta of CK2. In this study, homozygous T-DNA mutants (ckb1-1 and ckb1-2) and over-expression plants (35S:CKB1-1, 35S:CKB1-2) of Arabidopsis thaliana were studied to understand the role of CKB1 in abiotic stress and gibberellic acid (GA) signaling. Histochemical staining showed that although CKB1 was expressed in all organs, it had a relatively higher expression in conducting tissues. The ckb1 mutants showed reduced sensitivity to abscisic acid (ABA) during seed germination and seedling growth. The increased stomatal aperture, leaf water loss and proline accumulation were observed in ckb1 mutants. In contrast, the ckb1 mutant had increased sensitivity to polyaluminum chloride during seed germination and hypocotyl elongation. We obtained opposite results in over-expression plants. The expression levels of a number of genes in the ABA and GA regulatory network had changed. This study demonstrates that CKB1 is an ABA signaling-related gene, which subsequently influences GA metabolism, and may play a positive role in ABA signaling.  相似文献

UV-B acclimation effects and UV-B damage repair induced by a 632.8-nm He-Ne laser were investigated in Arabidopsis thaliana plants in response to supplementary UV-B stress. There was an increasing trend in growth parameters in the combination-treated plants with He-Ne laser and UV-B light compared to those stressed with enhanced UV-B light alone during different developmental stages of plants. The photosynthetic efficiency (Pn) and survival rates of seedlings were significantly higher in the combination treatments than UV-B stress alone. The expression of UVR8, phytochrome B (PhyB), and their mediated signal responsive genes such as COP1, HY5, and CHS were also significantly upregulated in plants with the laser irradiation compared with other groups without the laser. Levels of flavonol accumulation in leaves and capsule yield of He-Ne laser-treated plants were increased. The phyB-9 mutants were more sensitive to enhanced UV-B stress and had no obvious improvements in plant phenotypic development and physiological damage caused by enhanced UV-B stress after He-Ne laser irradiation. Our results suggested that UVR8 and its mediated signaling pathway via interaction with COP1 can be induced by He-Ne laser, and these processes were dependent on cytoplasmic PhyB levels in plant cells, which might be one of the most important mechanisms of He-Ne laser on UV-B protection and UV-B damage repair. These current data have also elucidated that the biostimulatory effects of He-Ne laser on Arabidopsis thaliana plants would happen not only during the early growth stage but also during the entire late developmental stage.  相似文献

Despite the paraquat-resistant mutants that have been reported in plants, this study identified a novel A. thaliana mutant (pqr2) from an XVE inducible activation library based on its resistance to 2 μM paraquat. The pqr2 mutant exhibited a termination mutation in the exon of AT1G31830/PAR1/PQR2, encoded a polyamine uptake transporter AtPUT2/PAR1/PQR2. The PQR2 mutation could largely reduce superoxide accumulation and cell death in the pqr2 plants under paraquat treatment. Moreover, compared with wild type, the pqr2 mutant exhibited much reduced tolerance to putrescine, a classic polyamine compound, which confirmed that PQR2 encoded a defective polyamine transporter. Notably, co-treated with ABA and paraquat, both pqr2 mutant and wild type exhibited a lethal phenotype from seed germination, but the wild type like pqr2 mutant, could remain paraquat-resistance while co-treated with high dosage of Na2WO4, an ABA synthesis inhibitor. Gene expression analysis suggested that ABA signaling should widely regulate paraquat-responsive genes distinctively in wild type and pqr2 mutant. Hence, this study has for the first time reported about ABA negative effect on paraquat-resistance in A. thaliana, providing insight into the ABA signaling involved in the oxidative stress responses induced by paraquat in plants.  相似文献

Late embryogenesis abundant (LEA) proteins are closely related to abiotic stress tolerance of plants. In the present study, we identified a novel Em-like gene from lettuce, termed LsEm1, which could be classified into group 1 LEA proteins, and shared high homology with Cynara cardunculus Em protein. The LsEm1 protein contained three different 20-mer conserved elements (C-element, N-element, and M-element) in the C-termini, N-termini, and middle-region, respectively. The LsEm1 mRNAs were accumulated in all examined tissues during the flowering and mature stages, with a little accumulation in the roots and leaves during the seedling stage. Furthermore, the LsEm1 gene was also expressed in response to salt, dehydration, abscisic acid (ABA), and cold stresses in young seedlings. The LsEm1 protein could effectively reduce damage to the lactate dehydrogenase (LDH) and protect LDH activity under desiccation and salt treatments. The Escherichia coli cells overexpressing the LsEm1 gene showed a growth advantage over the control under drought and salt stresses. Moreover, LsEm1-overexpressing rice seeds were relatively sensitive to exogenously applied ABA, suggesting that the LsEm1 gene might depend on an ABA signaling pathway in response to environmental stresses. The transgenic rice plants overexpressing the LsEm1 gene showed higher tolerance to drought and salt stresses than did wild-type (WT) plants on the basis of the germination performances, higher survival rates, higher chlorophyll content, more accumulation of soluble sugar, lower relative electrolyte leakage, and higher superoxide dismutase activity under stress conditions. The LsEm1-overexpressing rice lines also showed less yield loss compared with WT rice under stress conditions. Furthermore, the LsEm1 gene had a positive effect on the expression of the OsCDPK9, OsCDPK13, OsCDPK15, OsCDPK25, and rab21 (rab16a) genes in transgenic rice under drought and salt stress conditions, implying that overexpression of these genes may be involved in the enhanced drought and salt tolerance of transgenic rice. Thus, this work paves the way for improvement in tolerance of crops by genetic engineering breeding.  相似文献

Ethylene response factors (ERFs) are widespread in plants, which are widely involved in plant response to biotic and abiotic stress. In this research, a soybean gene, GmERF9, was identified and the function was characterized. The results showed that GmERF9 contained a typical AP2/ERF binding domain and a putative nuclear localization signal sequence. The real-time fluorescence quantitative PCR (qPCR) revealed that the expression of GmERF9 could be induced by ethylene (ET), abscisic acid (ABA), drought, salt and cold stresses. GmERF9 protein could specifically bind to the GCC-box and activate the expression of the reporter gene in the yeast cells and tobacco leaves. Overexpression of GmERF9 enhanced the expression of pathogenesis-related (PR) genes, including PR1, PR2, Osmotin (PR5), and SAR8.2. Also, the overexpression of GmERF9 increased the accumulation of proline and soluble carbohydrate, and decreased the accumulation of malondialdehyde under drought and cold stresses in the transgenic tobacco compared to the wild type (WT) tobacco, which indicated that GmERF9 enhanced the tolerance to drought and cold stresses in the transgenic tobacco. In summary, the function of GmERF9 is involved in the response to environmental stresses for plants, which can be used as a candidate gene for genetic engineering of crops.  相似文献

Background

Plant phospholipase D (PLD), which can hydrolyze membrane phospholipids to produce phosphatidic acid (PA), a secondary signaling molecule, has been proposed to function in diverse plant stress responses. Both PLD and PA play key roles in plant growth, development, and cellular processes. PLD was suggested to mediate the regulation of stomatal movements by abscisic acid (ABA) as a response to water deficit. In this research, we characterized the roles of the cucumber phospholipase D alpha gene (CsPLDα, GenBank accession number EF363796) in the growth and tolerance of transgenic tobacco (Nicotiana tabacum) to drought stress.

Results

The CsPLDα overexpression in tobacco lines correlated with the ABA synthesis and metabolism, regulated the rapid stomatal closure in drought stress, and reduced the water loss. The NtNCED1 expression levels in the transgenic lines and wild type (WT) were sharply up-regulated after 16?days of drought stress compared with those before treatment, and the expression level in the transgenic lines was significantly higher than that in the WT. The NtAOG expression level evidently improved after 8 and 16?days compared with that at 0?day of treatment and was significantly lower in the transgenic lines than in the WT. The ABA content in the transgenic lines was significantly higher than that in the WT. The CsPLDα overexpression could increase the osmolyte content and reduce the ion leakage. The proline, soluble sugar, and soluble protein contents significantly increased. By contrast, the electrolytic leakage and malondialdehyde accumulation in leaves significantly decreased. The shoot and root fresh and dry weights of the overexpression lines significantly increased. These results indicated that a significant correlation between CsPLDα overexpression and improved resistance to water deficit.

Conclusions

The plants with overexpressed CsPLDα exhibited lower water loss, higher leaf relative water content, and heavier fresh and dry matter accumulation than the WT. We proposed that CsPLDα was involved in the ABA-dependent pathway in mediating the stomatal closure and preventing the elevation of intracellular solute potential.
