用离体兔胸主动脉条研究东莨菪碱(scopolamine,Scop)扩张血管作用的可能机理。Scop与维拉帕米对CaCl2和Scop对KCI所致兔胸主动脉条收缩的量-效曲线右移,最大反应降低,呈现非竞争性拮抗,其pD'2分别为3.4,5.8和3.4。两药能明显抑制NE的依赖内Ca(2+)性收缩,Scop对NE的依赖外Ca(2+)性收缩几无影响,表明Scop可能主要作用于电压依赖性的钙通道,阻断Ca(2+)内流。  相似文献

肝性脑病大鼠海马CA1区锥体细胞氨基酸激活电流改变   总被引:1,自引:0,他引:1  
探讨氨基酸受体门控性离子通道电流改变在肝性脑病(HE)发生中的作用。方法用快速加药装置将50μmol/Lγ-氨基丁酸(GABA)或1mmol/LN-甲基-D-天冬氨酸(NMDA)+10μmol/L甘氨酸(Gly)加于急性分离的大白鼠海马CA1区锥体细胞周围,用全细胞膜片钳技术记录GABA或NMDA激活的电流。结果HE大鼠海马CA1区锥体细胞GABA激活的内向电流的强度为413.3±436.8pA(31例),比对照大鼠(233.4±179.7pA,28例)有较大增强(t=2.1049,P<0.05)。并观察到HE大鼠GABA激活电流的出现频率较对照大鼠高。HE大鼠NMDA激活的内向电流的强度为408.5±317.2pA(37例),比对照大鼠(882.6±896.5pA,26例)弱(t=2.5851,P<0.05)。两组大鼠NMDA激活电流的出现频率无明显差异。结论氨基酸受体门控性离子通道电流改变在肝性脑病发生中起一定作用。  相似文献

研究萘甲异喹(NI)对离体豚鼠回肠平滑肌的作用。NI(10、30μmol/L)可明显抑制离体豚鼠回肠的自发性收缩,能拮抗组胺、乙酰胆碱和5-羟色胺对回肠的收缩效应,表现为非竞争性拮抗,其pD'2分别为5.36±0.49、4.74±0.32和4.81±0.51。NI(10、30μmol/L)尚能抑制CaCl2和BaCl2引起的离体豚鼠回肠收缩,并可抑制组胺引起肠组织内Ca2+浓度升高。结果提示:NI能抑制肠平滑肌细胞外Ca2+内流及细胞内Ca2+释放引起的平滑肌收缩,可能是一非特异性钙拮抗剂。  相似文献

目的探讨非胰岛素依赖型糖尿病(NIDDM)患者红细胞膜Na+-K+-ATPase及Ca2+-AT-Pase活性改变的影响因素。方法测定77例NIDDM患者和50例正常人血糖、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDLC)、脂质过氧化物(LPO)、谷胱甘肽(GSH)、糖化血红蛋白(HbAlc)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSHPX)、红细胞膜Na+-K+-AT-Pase和Ca2+-ATPase活性。t检验、直线相关分析和多元逐步回归分析。结果NIDDM组血糖、HbAlc、TC、TG、TC/HDLC、LPO显著高于对照组(P<0.01),HDLC、GSH、SOD、GSHPX、红细胞膜Na+-K+-ATPase和Ca2+-ATPase活性低于对照组(除GSH的P<0.05外,其他P<0.01);不同HbAlc、TC/HDLC、GSH、LPO、TC组的红细胞膜Na+-K+-ATPase活性有显著性差别。红细胞膜Na+-K+-ATPase活性=12.80+0.27(GSH)-0.12(LPO)-0.09(TC/HDLC),红细胞胞膜Ca2+-ATPase活性=108.20+1.  相似文献

目的 研究乙型肝炎病毒x( H Bx) 蛋白对细胞周期影响,探讨 H Bx 蛋白致细胞周期紊乱的分子机理。方法 构建 H Bx 基因真核表达载体p C E P4 X,电转染入 Hep G2 细胞( X 细胞) ,以p C E P4空载体为对照( X0 细胞) ,潮霉素选择培养,克隆扩增转染细胞,经无血清培养同步化后,流式细胞仪检测细胞周期变化;细胞组化和 Western blot 检测增殖细胞核抗原( P C N A) 和p21 C I P1/ W A F1 表达变化。结果 流式细胞仪检测示对照 X0 细胞70 % 进入 G0 G1 期,而转 X 细胞仅56 % 进入 G0 G1 期,与含血清培养的亲本细胞 Hep G2 几乎相同。p21 C I P1/ W A F1 D 表达在 X0 细胞阳性率为(86 ±8) % , X 细胞为(16 ±6) % 。 P C N A 表达在 X0 细胞阳性率为(9 ±5) % , X 细胞为(86 ±3) % 。结论  H Bx 有推进细胞周期作用,并使细胞生长对血清依赖减少。 H Bx 可抑制细胞周期抑制蛋白p21 C I P1/ W A F1 表达,同时增强细胞 D N A 合成。  相似文献

选择26例原发性高血压(EH)伴Ⅱ型糖尿病(NIDDM),其中糖尿病肾病(DN)12例,加用卡托普利37.5~112.5mg/日口服治疗,共4周.结果:收缩压(SBP)/舒张庄(DBP)由21.9±2.3/13.5±0.5kp_a降为19.3±1.5/12.2±0.8Kp_a(P<0.01),空腹血糖/餐后2小时血糖由10.7±4.5/13.7±3.9mmol/L减为6.5±1.6/10.2±2.7mmol/L(P<0.01);24小时尿蛋白由1.9±0.3g减为0.6±0.3g(P<0.01).表明该药在降压的同时确有改善糖代谢、降低尿蛋白排泄的作用。  相似文献

反义寡脱氧核苷酸对人肝癌裸鼠模型生长及转移的影响   总被引:3,自引:0,他引:3  
目的 观察反义H-ras寡脱氧核苷酸(ODN)对高表达蛋白p21H- ras的人肝癌高转移裸鼠模型LCI-D20细胞生长、凋亡及转移潜能的影响。方法原代培养LCI-D20细胞,经10μmol/L浓度反义ODN处理后,以1.5×10细胞数分别接种于14只裸鼠皮下缝制皮囊内:6只接种反义H-rasODN处理细胞,4只接种H-ras非特异性反义ODN处理细胞,余4只接种未作处理的对照细胞。结果反义H-ras对LCI-D20细胞增殖具有显著抑制作用(t=31.529,P<0.01),反义ODN处理细胞,S期比率(36.0±1.4)显著低于对照组(58.5±0.9,t=13.519,P<0.01),而G1/G0期比率(56.7%±1.1%)高于对照组(37.4±0.7,t=14.802,P<0.01);反义H-rasODN处理细胞的凋亡发生率(34.0%±4.5%)显著高于对照组(2.5%±1.2%,t=13.434,P<0.01);反义H-ras处理对LCI-D20细胞在接种动物体内的生长具有显著延迟抑制作用(t=3.509,t=3.452,P<0.01);接种动物的肺转移率,反义H-rasODN处理组显著低于对照组。  相似文献

取家兔16只,冠状动脉左室支结扎后均分为血液稀释组(HD)和正常红细胞压积(Hct)组(NG)。另取兔8只仅行开胸手术作为对照组(CG)。观察血液稀释对心肌梗塞兔循环。肾上腺素诱发室性心律失常剂量(E/V)及梗塞区大小的影响。结果Hct:HD31.3±2.7%,NG38.1±2.62%,CG39.1±1.6%。冠状动脉结扎后至半小时内三组HR,MAP,CVP及心肌耗氧量均明显增加。HD的增加达1h(P<0.05)。(EIV:CO23.3±4.5μg·kg-1,HD18.0±2.95μg·kg-1(P>0.05),NG,6.86±1.7μg·kg-1,差异显著(P<0.05)。心肌梗塞区占全心重HD为6.4%,NG8.1%。(P>0.05)。证实血液稀释有提高心肌应激性,成少室性心律失常,改善心肌微循环和减小梗塞范围的功效。  相似文献

内源性高甘油三酯血症(HTG)常伴血浆HDL-C含量下降及HDL组成异常。为了研究HTG患者血浆HDL亚类的改变,作者建立了密度梯度聚丙烯酰胺梯度凝胶电泳法(GGE),对25例HTG患者(血浆TG>2.26mmol/L,TC<6.72mmol/L)及25例年龄、性别匹配的血脂正常者(血浆TG<1.69mmol/L,TC<5.17mmol/L)的HDL亚类——HDL2a、HDL2b、HDL3a、HDL3b和HDL3c进行了分析,同时对血浆及HDL中脂质及载脂蛋白(apo)AⅠ、AⅡ、B100、CⅡ、CⅢ及E进行了测定。结果发现,HTG患者空腹血浆TG、apoB100、apoCⅡ、apcCⅢ及apoE显著增加(P<0.001),HDL-C及apoAⅠ显著降低(P<0.001及P<0.05);患者血浆HDL组成异常,HDL中TG含量显著增加(P<0.001),TC、apoAⅠ、apoCⅡ、apoCⅢ及apoE含量下降(P<0.01);患者血浆HDL亚类含量改变,HDL2b显著降低(P<0.02),HDL2a及HDL3a增加(P<0.05),且随血浆TG含量的升高而升高,HDL3b及HDL3c则未见改变  相似文献

观察了27例高血压病(PHT)、19例冠心病(CHD)患者和40例正常对照(NC)者血浆脂蛋白(a)(Lp(a))的水平,并分析了各组Lp(a)与其它脂类的相关性。结果显示,PHT组Lp(a)水平为240.5±122.4mg/L,CHD组为220.7±126.5mg/L,两组均显著高于NC组的166.5±76.1mg/L(分别为P<0.01和P<0.05,血浆Lp(a)与甘油三酯TG)、总胆固醇(TC)、载脂蛋白Al(Apoal)、载脂蛋白B(ApoB)和AooAl/ApoB的比值之间均无显著相关性(P<0.05)。表明:Lp(a)可通过干扰脂质代谢和纤溶过程而发挥其致动脉粥样硬化(AS)作用,它是CHD的独立危险因子。Lp(a)可能在PHT的发生与发展过程中也起一定作用。  相似文献

Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimets were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ml, P <0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.  相似文献

Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer's was infused from portal veins to donated livers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.  相似文献

Shock wave lithotripsy (SWL) is a treatment of choice for upper urinary stones. However, this procedure is inappropriate for obese patients because the focus is often unable to reach the target owing to the limited focal distance in shock wave source. Although treating such patients in a blast path may increase the application length of shock wave source, it's difficult to find this path on the lithotripter monitor. Excess production of reactive oxygen species(ROS)of mitochondrion mediated by hyperglycemia is the common pathogenesis of angiopathic complications of diabetes.TCM holds that the damp from the dysfunction of spleen.kidney and liver is the causative factor of complications of diabetes.This is similar to the mechanism of Ros resulting in angiopathic complications of diabetes.When the angiopathic complications of type II diabetes mellitus(T2DM)are difierentiated as caused by turbid damp in TCM can be explained as ROS.Since the obstruction of pathogenic damp in channels and collaterals is said to be the main pathogenesis,the treating principle should be dissolving the damp to remove the obstruction.  相似文献

Objective To observe blood pressure change with age in salt-sensitive teenagers whose salt sensitivity were determined by repeated testing.Methods Salt sensitivity was determined through intravenous infusion of normal saline combined with volume-depletion by oral diuretic furosemide in 55 teenagers. After five years, salt sensitivity was re-examined and subject blood pressure was followed up. Blood pressure changes in salt-sensitive teenagers were compared to that of non-salt sensitive teenagers over five years.Results After 5 years, the repetition rate of salt sensitivity determined by intravenous saline loading is 92.7%. In teenagers with salt sensitivity on the baseline, both the systolic blood pressure increments and increment rates were much higher than non-salt sensitive teenagers (12.7±12.1 mmHg vs. 2.8±5.2 mmHg, P< 0.01; 12.2%± 12.0% vs. 2.5% ±4.4%, P< 0.001,respectively). There was a similar trend for diastolic blood pressure (8.4 ± 6.4 mmHg vs. 3.7 ± 6.4 mmHg, P = 0.052; 13.2% ±10.6 % vs. 6.8%± 10.1%, P = 0.053, respectively).Conclusions Salt sensitivity determined by intravenous saline loading showed good reproducibility. Blood pressure increments with age were much higher in salt-sensitive teenagers than non-salt sensitive teenagers, especially in terms of systolic blood pressure.  相似文献

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献
