【目的】探究铜绿微囊藻(Microcystis aeruginosa)藻际微生物群落组成以及与溶藻细菌侧孢短芽孢杆菌(Brevibacillus laterosporus)Bl-zj共培养对藻际微生物群落的影响。【方法】将侧孢短芽孢杆菌Bl-zj与铜绿微囊藻共培养4 d后(BL组),收集样品,以单独培养的铜绿微囊藻(BG组)为对照,提取样品总基因组DNA后进行16S rDNA扩增、构建文库、测序及生物信息学分析,比较BL组与BG组间藻际微生物群落的差异。【结果】测序平均获得高质量序列85390条,946个OUT(Operational Taxonomic Unit),BL组特有OTU 339个。物种组成分析显示,BG组微生物群落主要以微囊藻属(Microcystis)为主,其余相对丰度由高到低分别为假单胞菌属(Pseudomonas)、中慢生根瘤菌属(Mesorhizobium)、短芽孢杆菌属(Brevibacillus)和红杆菌属(Rhodobacter)。BL组微生物群落主要以短芽孢杆菌属(Brevibacillus)为主。BL组的微生物多样性相较于BG组显著下降(P<0.05)。经预测,BG组微生物功能集中在能量代谢与外源生物降解与代谢,BL组微生物功能集中在碳水化合物代谢和氨基酸代谢功能。【结论】侧孢短芽孢杆菌Bl-zj与铜绿微囊藻共培养,打破了铜绿微囊藻藻际微生物群落的平衡,与Bl-zj的溶藻进程有关。  相似文献

Cyanobacterial blooms in eutrophic freshwater systems are a worldwide problem, creating adverse effects for many aquatic organisms by producing toxic microcystins and deteriorating water quality. In this study, microcystins (MCs) in Microcystis aeruginosa, and Daphnia magna exposed to M. aeruginosa, were analyzed by HPLC-MS, and the effects of M. aeruginosa on D. magna were investigated. When D. magna was exposed to M. aeruginosa for more than 2 h, Microcystin-LR (MC-LR) was detected. When exposed to 1.5 × 106, 3 × 106, 0.75 × 107, and 1.5 × 107 cell/mL of M. aeruginosa for 96 h, average survival of D. magna for treatments were 23.33%, 33.33%, 13.33%, 16.67%, respectively, which were significantly lower than the average 100% survival in the control group (P < 0.05). The adverse effects of M. aeruginosa on body length, time for the first brood, brood numbers, gross fecundity, lifespan, and population growth of D. magna were density-dependent. These results suggest that the occurrence of M. aeruginosa blooms could strongly inhibit the population growth of D. magna through depression of survival, individual growth and gross fecundity. In the most serious situations, M. aeruginosa blooms could undermine the food web by eliminating filter-feeding zooplankton, which would destroy the ecological balance of aquaculture water bodies.  相似文献

【目的】研究异养鞭毛虫(Paraphysomonas sp.)在不同密度铜绿微囊藻(Microcystis aeruginosa)中的生长状况,探讨其对铜绿微囊藻的摄食及抑制作用。【方法】实验共设6个铜绿微囊藻密度,分别是0、250×10~4、500×10~4、750×10~4、1 000×10~4和2 000×10~4cells/mL,研究异养鞭毛虫与微囊藻的种群数量变化。【结果】异养鞭毛虫主要行异养生活。在铜绿微囊藻密度为250×10~4~2 000×10~4 cells/mL时,异养鞭毛虫对铜绿微囊藻的抑制率为98.00%~99.69%。单个鞭毛虫对微囊藻的摄食率为13~49 cells/d。异养鞭毛虫的密度和生长率随铜绿微囊藻密度的升高而增加,在铜绿微囊藻密度为500×10~4~1000×10~4cells/mL时,异养鞭毛虫密度为21×10~4~24×10~4cells/mL、生长率为0.6~0.74 d-1。但是,铜绿微囊藻密度过高也不利于异养鞭毛虫的种群增长,在2 000×10~4 cells/mL密度时,异养鞭毛虫的生长速率显著低于其它密度组。【结论】异养鞭毛虫能有效抑制铜绿微囊藻种群增长,其生长的最适微囊藻密度为500×10~4~1 000×10~4 cells/mL。  相似文献

Colony morphology is important for Microcystis to sustain a competitive advantage in eutrophic lakes. The mechanism of colony formation in Microcystis is currently unclear. Extracellular polysaccharide (EPS) has been reported to play an important role in cell aggregate formation of some phytoplankton. Microcystis aeruginosa was cultivated under varied abiotic conditions, including different nutrient, light, and temperature conditions, to investigate their effects on EPS production and morphological change. The results show that nutrient concentration and light intensity have great effects on EPS production in M. aeruginosa. There was a considerable increase in EPS production after M. aeruginosa was cultivated in adjusted culture conditions similar to those present in the field (28.9mg C/L, 1.98mg N/L, 0.65mg P/L, light intensity:100 μmol/(m2s)). These results indicate that abiotic factors might be one of the triggers for colony formation in Microcystis.  相似文献

In transgenic process, a foreign gene can be integrated in the host genome in two directions, which may influence its expression. In order to study the effects of insertal orientation, the gfp reporter gene was inserted in the isiAB locus of Synechococcus sp.PCC7942 in different directions, and the GFP expression levels and the growth of the transgenic algae were compared. It was showed that the gfp gene could express in each direction, and no significant difference was detected on algal growth and GFP expression levels between the two recombinant algae.  相似文献

The effects of a diet containing Hanseniaspora opuntiae C21 on growth and digestive enzyme activity were estimated in juvenile Apostichopus japonicus. Groups of sea cucumbers were fed diets containing H. opuntiae C21 at 0(control), 10 4, 10 5, and 10 6 CFU(colony-forming units)/g feed. Results showed that after 45 d the specific growth rate(SGR) of sea cucumbers fed a C21-supplemented diet at 10 4 CFU/g feed was significantly higher than that of the control( P 0.05). Intestinal trypsin and lipase activities were significantly enhanced by C21 administration at 10 4 and 10 5 CFU/g feed compared with the control( P 0.05). After feeding for 23–42 d, C21 was demonstrated by denaturing gradient gel electrophoresis to be present in the intestine of sea cucumbers. In addition, after feeding the C21-supplemented diets for 15 d, the sea cucumbers were switched to an unsupplemented diet and C21 was confirmed to be capable of colonizing the intestine for at least 31 d after cessation of feeding. In conclusion, C21 was shown to successfully colonize the intestine of juvenile A. japonicus via dietary supplementation, and improve growth and digestive enzyme activity.  相似文献

This study examined the effects of salinity on embryonic development, survival, and growth of the Hong Kong oyster Crassostrea hongkongensis. The embryos, larvae, and juveniles of C. hongkongensis were held in artificial seawater at three different salinities (low, 15; medium, 23; and high, 30) to determine the optimum hatchery and nursery conditions for mass production of the seeds. Results showed that the percentage production of straight-hinged larvae from fertilized eggs was significantly lower at the high salinity than at the low- and medium-salinities (P < 0.05). The survival rates of larvae and juveniles differed significantly among the three salinity trials, with the highest survival rate observed at the low salinity (P < 0.05). The shell height of larvae was significantly larger at the low salinity than at the high and medium salinities from days 9 to 15 (P < 0.05), whereas that of juveniles was significantly larger at the low salinity than at the high and medium salinities on day 70 (P < 0.05). These results indicate that the larvae and juveniles of C. hongkongensis are tolerant to a wide range of salinities (15 to 30), but show better growth and survival at relatively low salinities. Thus, it is recommended to use relatively low salinities in hatchery and nursery systems for improved yields of C. hongkongensis.  相似文献

Prepared in this experiment were six groups of diets, i.e. VC0, VC1, VC2, VC3, VC4 and VC5 with the contents of vitamin C (VCmg(100g)-1 diet) of 0, 100, 200, 400, 800, and 1200 respectively. It was found that vitamin C increased the concentrations of immunoglobulin-like (IgG-like, IgA-like and IgM-like) substances in the serum of Penaeus chinensis after a feeding period of 3 weeks. The differences among groups were significant (P< 0.01), but there was no difference in the contents of complement3-1ike and complement4-1ike substances in the serum (P >0.05). Phenoloxidase (PO) activity in the serum of VC3 group shrimps was higher than that of VC0 and other groups, but no significant difference was observed between VC0 group and other groups. Furthermore, bactericidal activity of the serum to Vibrio parahaemolyticus in shrimps fed with the VC1 diet was higher than that in the other groups (P< 0.01), while no difference was demonstrated among all groups for the bactericidal activity to Vibrio alginolyticus (P >0.05). It is, therefore, suggested that vitamin C (100-400 mg(100 g) -1 diets) could be used as an immunostimulant of P. chinensis.  相似文献

This experiment on the effects of fishes (Pelteobagrus fulvidraco, Channa argus, Cyprinus carpio) on growth and survival of juvenile Chinese mitten crab,Eriocheir sinensis was conducted in cages placed in Bao'an Lake. T-tests showed that effects of fishes on the carapace width growth were not significant as compared with the control (no fish). But the effects of these fishes on crab survival were negative. The causes of negative effects of fishes on crab survival were simply discussed on the basis of food habits. The effects of environmental Na+/K+ ratio on the gill ion-transport enzyme activity, plasma osmolality and growth of Cynoglossus semilaevis juveniles were investigated. The results showed that, plasma osmolality was similar among flounder adapted to different Na+/K+ ratios of saline groundwaters (P>0.05), while the growth, gill Na+, K+-ATPase and HCO3 -ATPase activities were affected by Na+/K+ ratio significantly (P<0.05). The gill Na+, K+-ATPase activity reached its maximum on day 3, then decreased gradually from day 3 to day 9 and remained constant from day 9 to day 15. The peak values of gill Na+, K+-ATPase activity were detected on day 3 for all Na+/K+ ratios of saline groundwaters, then the enzyme activities descended, and on day 9 the enzyme activities achieved steady state, and the gill HCO3 -ATPase activity increased rapidly and achieved steady state after one day. During steady state, the gill Na+, K+-ATPase and HCO3 -ATPase activity of Na+/K+ ratios 20 and 40 treatments were significantly higher than those in the control group (Na+/K+ ratio 27.5), while there were no significant differences between the Na+/K+ ratio 30 treatment and the control group; the gill Na+, K+-ATPase activity of Na+/K+ ratio 20 and 40 treatments were significantly higher than that for ratio 30 treatment, but there were no significant differences of gill HCO3 -ATPase activity among these treatments. At the end of the 15-day experiment, the weight gain (%) and specific growth rate (SGR) of flounders maintained in seawater were significantly higher than those in groundwaters; significant differences also occurred among the treatments; Na+/K+ ratio 30 treatment had the highest values (33.7% and 1.94 respectively), which were significantly higher than those under Na+/K+ ratios 20 and 40 treatments. Therefore, for the saline groundwater used in this experiment, it is suggested that the Na+/K+ ratio be adjusted to approximately 30, i.e., as close to that of natural seawater as possible in the culture of flounder.  相似文献

The protease activity in digestive tract of young turbot Scophthalmus maximum was studied, and the optimal pH, temperature and NaCl concentration were determined for different portions of the fish's internal organs. The optimal activity in the fish's stomach was at pH of 2.2, while that in the intestinal extracts was within the alkaline range from 9.5 to 10.0. In hepatopancreas, the optimal pH was in low alkalinity at 8.5. The optimal reaction temperature was above 40℃ in stomach, intestine and hepatopancreas. With increasing temperature, the pH value increased in stomach, while in the intestine, an opposite tendency was observed due to combined effect of pH and temperature. NaCl concentration showed inhibitory impact on protein digestion in hepatopancreas. The main protease for protein digestion in turbot seemed to be pepsin. Moreover, the maximum protease activity in different segments of intestine existed in the hindgut.  相似文献

Pyropia haitanensis,a commercially important species,was cultured at two CO2 concentrations(390×10- 6 and 700×10-6(parts per million)) and at low and high nutrient levels,to explore the effect of elevated CO2 on the species under nutrient enrichment. Results show that in CO2-enriched thalli,relative growth rate(RGR) was enhanced under nutrient enrichment. Elevated CO2 decreased phycobiliprotein(PB) contents,but increased the contents of soluble carbohydrates. Nutrient enrichment increased the contents of chlorophyll a(Chl a) and PB,while soluble carbohydrate content decreased. CO2 enrichment enhanced the relative maximum electronic transport rate and light saturation point. In nutrient-enriched thalli the activity of nitrate reductase(NRA) increased under elevated CO2. An instantaneous pH change in seawater(from 8.1 to 9.6) resulted in reduction of NRA,and the thalli grown under both elevated CO2 and nutrient enrichment exhibited less pronounced reduction than in algae grown at the ambient CO2. The thermal optima of NRA under elevated CO2 and/or nutrient enrichment shifted to a lower temperature(10–15°C) compared to that in ambient conditions(20°C). We propose that accelerated photosynthesis could result in growth increment. Nassimilation remained high in acidified seawater and refl ected increased temperature sensitivity in response to elevated CO2 and eutrophication.  相似文献

We evaluated the effects of lithium on the survival,growth,and reproduction of D.tibetana in the laboratory.The safe concentration value was 69.3 mg/L.The time to first brood was significant lower for individuals reared in 5 and 10 mg/L(24.4 and 24.0 d,respectively) compared with individuals reared in20,40,or 60 mg/L.Females reared in 5 mg/L lithium produced a mean of 16.5 neonates/brood and had the highest number of broods(3.0±1.95).The rate of egg production(a),the intrinsic rate of increase(r_m),net reproduction rate(R_0),and finite rate of increase(λ) were highest for D.tibetana reared at 5 and 10 mg/L lithium,and the duration of development was shorter than for the remaining groups.The results indicated that rearing in 5-10 mg/L lithium can accelerate the growth and reproduction of D.tibetana.  相似文献

Effect of salinity on survival,feeding behavior and growth of juvenile swimming crab P ortunus trituberculatus was investigated under 5 salinity levels of 5,10,20,30 and 40. The results show that the crab juveniles fed 2 or 3 times at the salinity 20 and 30,each lasted for about 25 minutes,for a total feeding time of 73.2±22.65 minutes per day. At these salinities,there were significantly higher in the frequency of feeding and in total feeding time than those at lower salinities of 5 and 10. All crab juveniles moulted when reared at a salinity of 20 during the 5 days duration of the experiment,which is significantly higher than those at other salinities. All juveniles survived at salinity 20,and the survivorship was not significantly different from that at 30,but was signif icantly higher than those at other salinities. The crab juveniles reared at a salinity of 20 had the highest value of food ration of 0.190 8±0.011 3 g/g BW,average body weight gain of 0.796±0.128 g,gain rate of 87%–96%,and food conversion ratio of 1.20±0.09. There was no significant difference in the values found between 20 and 30 but these values were significantly lower than that at the other salinities( P 0.05). Highest activities of digestive enzymes(Amylase,Protease,Lipase) and lowest activities of protective enzymes(SOD,PO,CAT) were also obtained on crab juveniles reared at salinity of 20.  相似文献

Three color variants of the sea cucumber,Apostichopus japonicus are recognized,the red one is highly valued in the market. When the red variant is cultured in ponds in China,its body color changes from red to celadon in 3–6 months. The effects of water depth and substrate color on the growth and body color of this animal were investigated. Juveniles of red A. japonicus were cultured in cages suspended at a range of water depths(20,50,100,150 and 200 cm). The specific growth rate of red sea cucumbers was significantly higher in animals cultured at deeper water layers compared with those grown at shallowers. Body weights were greatest for sea cucumbers cultured at a depth of 150 cm and their survival rates were highest at a depth of 200 cm. A scale to evaluate the color of red sea cucumbers(R value) was developed using a Pantone standard color card. All stocked animals in the 9-month trial retained a red color,however the red body color was much more intense in sea cucumbers cultured at shallower depths,while animals suspended in deeper layers became pale. In a separate trial,A. japonicus were cultured in suspended cages with seven different colored substrates. Substrate color had a significant effect on the growth and body-color of red A. japonicus. The yield were greatest for A. japonicus cultured on a yellow substrate,followed by green white orange red black and blue. All sea cucumbers in the 7-month trial retained a red color,although the red was most intense(highest R value) in animals cultured on a blue substrate and pale(lowest R value) for animals cultured on a green substrate.  相似文献
