MicroRNAs (miRNAs) are small non-coding RNAs 18-25 nucleotides in length that downregulate gene expression during various crucial cell processes such as apoptosis, differentiation and development. Changes in the expression profiles of miRNAs have been observed in a variety of human tumors, including colorectal cancer (CRC). Functional studies indicate that miRNAs act as tumor suppressors and oncogenes. These findings significantly extend Vogelstein's model of CRC pathogenesis and have shown great potential for miRNAs as a novel class of therapeutic targets. Several investigations have also described the ability of miRNA expression profiles to predict prognosis and response to selected treatments in CRC patients, and support diagnosis of CRC among cancer of unknown primary site. miRNAs' occurrence has been repeatedly observed also in serum and plasma, and miRNAs as novel minimally invasive biomarkers have indicated reasonable sensitivity for CRC detection and compare favorably with the fecal occult blood test. In this review, we summarize the knowledge regarding miRNAs' functioning in CRC while emphasizing their significance in pathogenetic signaling pathways and their potential to serve as disease biomarkers and novel therapeutic targets.  相似文献

MicroRNAs (miRNAs) are an abundant class of small non-protein-coding RNAs that function as negative gene regulators. They regulate diverse biological processes, and bioinformatic data indicates that each miRNA can control hundreds of gene targets, underscoring the potential influence of miRNAs on almost every genetic pathway. Recent evidence has shown that miRNA mutations or mis-expression correlate with various human cancers and indicates that miRNAs can function as tumour suppressors and oncogenes. miRNAs have been shown to repress the expression of important cancer-related genes and might prove useful in the diagnosis and treatment of cancer.  相似文献

MicroRNAs (miRNAs/miRs) are sensitive biomarkers and endogenous repressors of gene expression by decreasing mRNA stability and interfering with mRNA translation. Despite a number of investigations revealing the dysregulation of miRNA expression associated with cardiotoxicity induced by doxorubicin (Dox), perturbation of miRNAs directly resulting from Dox at early stage in cardiomyocytes and the target gene interaction remain largely unknown. In the present study, high-throughput deep-sequencing was used to analyze changes in global miRNA expression in H9c2 cardiomyocytes exposed to 5 µg/ml Dox for 0, 12 or 24 h. Compared with the 0-h time point, the expression levels of 386 unique miRNAs were altered. Based on miRNA expression and fold-change, the target genes of 76 selected miRNAs were further analyzed using gene interaction networks and pathway enrichment analysis. These miRNAs were involved in the regulation of different pathways, whose functions included apoptosis, cell proliferation, extracellular matrix remodeling, oxidative stress and lipid metabolism. These differentially expressed miRNAs included let-7 family, miR-29b-3p, miR-378-3/5p, miR-351-3p, miR-664-3p, miR-455-3p, miR-298-3p, miR-702-5p, miR-128-1-5p, miR-671 and miR-421-5p. The present data indicated that global wide miRNA profiling in Dox-induced cardiomyocytes may provide a novel mechanistic insight into understanding Dox-induced heart failure and cardiotoxicity, as well as novel biomarkers and therapeutic targets.  相似文献

原发性肝癌是世界范围内最常见和最具侵袭性的恶性肿瘤之一。miRNA是一系列调控基因转录后表达的非编码小RNA。研究发现,miRNA具有癌基因和抑癌基因的作用,在肿瘤的发生和发展中起着重要的作用。已有证据表明,原发性肝癌中存在一些异常表达的miRNA,它们多靶向于肝癌发生、发展相关的基因和信号通路,从而广泛调节肝癌细胞的增殖、分化、凋亡、侵袭和转移等重要的涉及癌细胞发生演变的病理过程。异常表达的miRNA及其功能靶标的发现不仅丰富了肝癌的发病机制,也为探索新的基因靶向治疗提供了可能; miRNA表达谱还与肝癌病理类型、恶性程度、分期、分级等临床病理过程密切相关,提示miRNA不仅可能用于肝癌诊断及个体化治疗,也可能作为判断预后的工具。本文就肝癌发生、发展、诊断、靶向治疗和预后中发挥重要作用的miRNA及其作用靶点作一综述。  相似文献

Malignant tumors are characterized by multiple genetic and epigenetic alterations, which include activation and overexpression of oncogenes and inactivation of tumor suppressor genes. Recently it has become apparent that apart from genetic and epigenetic changes in classical oncogenes and tumor suppressor genes, small 17?C25 nucleotides long non-coding microRNAs (miRNAs) regulate major cellular processes involved in tumor biology, such as cell proliferation, differentiation, apoptosis and metastasis. miRNAs bind through sequence-specific base pairing to the 3?? untranslated region (3??-UTR) of the mRNA leading to its translational repression or degradation, thereby negatively regulating protein expression. At least 506 different miRNAs have been identified (http://www.mirbase.org) and it is believed that the human genome encodes >1,000 miRNAs. The translation of ~30% of all human genes is regulated by miRNAs. Each miRNA targets on average 200 gene products and multiple miRNAs can bind to the 3??-UTR of a single mRNA. Evidence is increasing that the deregulation of miRNAs in malignant tumors is linked to altered expression and activity of the miRNA-biogenesis machinery.  相似文献

Hepatocellular carcinoma (HCC) is a primary malignancy of the liver of global importance. Recent studies of the expression and role of microRNA (miRNA) in HCC are providing new insights into disease pathogenesis. In addition, therapeutic efforts targeting specific miRNAs are being evaluated in animal models of HCC. The potential of miRNAs as biomarkers of disease or prognostic markers is being explored. Herein, we review studies of miRNA expression in human HCC, and discuss recent advances in knowledge about the involvement and role of selected miRNAs in disease pathogenesis, as biomarkers, or as therapeutic targets for HCC.  相似文献

Pancreatic adenocarcinoma and hepatocellular carcinoma are devastating human malignancies that are characterized by poor prognosis, late onset, and a lack of known biomarkers. New diagnostic and therapeutic molecular targets are desperately needed to develop novel and effective treatment strategies. MicroRNAs (miRNAs) are an emerging class of molecules with roles in various cellular processes, including growth, survival, and apoptosis. Most importantly, aberrant expression of miRNAs has been implicated in cancer pathogenesis. miRNA expression profiles of pancreatic adenocarcinoma and hepatocellular carcinoma indicate selective overexpression of oncogenic miRNAs and down-regulation of tumor suppressive miRNAs in these cancers. This review summarizes results from key studies conducted to characterize the miRNA expression profiles of pancreatic adenocarcinoma and hepatocellular carcinoma and describes the potential mechanisms by which some oncogenic or tumor suppressive miRNAs act. Furthermore, this review outlines novel therapeutic strategies for targeting miRNAs.  相似文献
