Two silages were prepared from the first-cut sward of timothy-meadow fescue and wilted to a dry matter (DM) content of 300 g kg-1. One was ensiled with the addition of a formic-acid-based additive (4 litres formic acid (FA) per tonne) and the other with the addition of a bacterial inoculant (LAB) at a rate of 5×106 colony forming units g-1. Both silages were well preserved, but the extent of fermentation was greater in LAB-silage than in FA-silage as indicated by a lower concentration of water soluble carbohydrates (68 vs 177 g kg-1 DM) and a higher concentration of lactic acid (147 vs 32 g kg-1 DM). Four Ayrshire cows were used in a 4×4 Latin square experiment with 21-day periods to study the effects of silage fermentation and postruminal casein supplementation on silage intake, nutrient supply and milk production. The four treatments were FA-silage without casein (FA-0), LAB-silage without casein (LAB-0), FA-silage with casein (FA-C) and LAB-silage with casein (LAB-C). Both silages were given ad libitum with 8 kg day-1 of barley without or with 400 g day-1 of casein infused into the duodenum. Organic matter digestibility was lower (0·723 vs 0·753; P<0·01) for FA-silage than for LAB-silage. Cows offered FA-silage had a higher molar proportion of acetate and a lower proportion of propionate in ruminal fluid than cows offered LAB-silage. Microbial protein synthesis estimated from the output of purine derivatives in urine was greater (288 vs 260 g N day-1; P<0·05) for cows given FA-silage compared with LAB-silage. Feeding LAB-treated silage tended (P<0·10) to decrease silage DM intake compared with FA-treated silage (10·61 vs 11·77 kg DM day-1). Silage composition did not affect significantly milk yield or milk composition. Casein infusion increased milk yield (25·1 vs 27·1 kg day-1; P<0·05), milk protein content (32·4 vs 33·8 g kg-1; P<0·05) and protein yield (808 vs 905 g day-1; P<0·01). Four cows were used in a 4×4 Latin square design with a 2×2 factorial arrangement of treatments to examine the effects of the extent of silage fermentation and post-ruminal casein infusion on the arterial concentrations and mammary arterial–venous (A–V) difference of energy-yielding nutrients and plasma amino acids. Of the two silages one was preserved with formic acid (FA-silage) and the other with an inoculant of lactic acid bacteria (LAB-silage). Both silages were given ad libitum with 8 kg day-1 of barley, without or with duodenal casein infusing (400 g day-1). Blood was sampled from the coccygeal vessel and milk vein. FA-silage less fermented than LAB-silage as indicated by higher concentration (g kg-1 dry matter) of water soluble carbohydrates (178 vs 68) and lower concentration of lactic acid (32 vs 147). Plasma concentration of glucose was lower (P<0·05) and concentrations of essential amino acids (P<0·001) and blood β-hydroxybutyrate (BHB) (P<0·01) were higher with FA-silage than with LAB-silage. There was a strong linear relationship between arterial concentration and mammary A–V difference of blood metabolites, except BHB, and amino acids. The relationship between arterial concentration and mammary A–V difference of glucose was different between the silages. Mammary extraction rates of glucose, BHB, histidine, leucine and lysine were affected by their arterial concentrations. The interaction between silage fermentation and casein infusion was significant (P<0·05) in arterial concentration of branched-chain amino acids, the increase being greater with FA-silage than with LAB-silage. The results suggest that the supply of glucose may limit milk synthesis in cows given restrictively fermented grass silage and barley, whereas the supply of amino acids may be more limiting with extensively fermented silages. It is further suggested that the mammary gland has a capacity to adapt it's efficiency of nutrient uptake according to the supply and mammary demand of nutrients. © 1997 SCI.  相似文献

This study measured the effects of including 5% NaOH-treated corn straw (T-CS) as a substitute for 15% wheat hay in the control total mixed ration (TMR) of lactating cows on performance, digestibility, and rumen microbial profile. Two groups of 21 cows each, similar in initial performance, were fed individually 1 of the 2 TMR examined. Voluntary dry matter intake of cows fed the control TMR was 4.3% higher than that of the T-CS cows, but in vivo dry matter and organic matter digestibilities of both groups were similar. Crude protein digestibility was higher in the control cows but digestibility of neutral detergent fiber polysaccharides (cellulose and hemicelluloses) was higher in the T-CS TMR. This was followed by 4.6% reduction in rumination time of the T-CS group. A slightly higher milk yield was observed in the control cows compared with the T-CS group; however, milk fat and milk protein content were higher in cows fed the T-CS TMR. This was reflected in 1.3% increase in energy-corrected milk yield and 5.34% increase in production efficiency (energy-corrected milk yield/intake) of the T-CS cows compared with the control. Welfare of the cows, as assessed by length of daily recumbence time, was improved by feeding the T-CS TMR relative to the control group. As a whole, the rumen bacterial community was significantly modulated in the T-CS group in the experimental period compared with the preexperimental period, whereas the bacterial community of the control group remained unchanged during this period. Out of the 8 bacterial species that were quantified using real-time PCR, a notable decrease in cellulolytic bacteria was observed in the T-CS group, as well as an increase in lactic acid-utilizing bacteria. These results illustrate the effect of T-CS on the composition of rumen microbiota, which may play a role in improving the performance of the lactating cow.  相似文献

A leafy corn hybrid was compared to a grain corn hybrid as silage and high moisture grain to evaluate dry matter intake, milk yield, and milk composition. Sixteen multiparous Holstein cows averaging 97 DIM were used in a feeding trial based on 4 x 4 Latin squares with 21-d periods. Each of four diets contained (dry basis) 8% chopped hay, 42% corn silage, 11% high moisture corn grain, 10% whole, fuzzy cottonseed, and 29% protein concentrate. One diet used leafy corn as both high moisture grain and silage. A second diet contained grain corn hybrid (control) as both high moisture grain and silage. A third diet contained leafy corn for high moisture grain and control corn for silage and the fourth diet used control corn for high moisture grain and leafy corn for silage. Cows fed diets containing leafy silage produced more milk and milk protein and ate more DM than cows fed control silage. The corn hybrid used for high moisture grain did not influence milk yield or composition. Dry matter intake was greater for cows fed the diet containing both leafy high moisture grain and leafy silage than for cows fed both control high moisture grain and control silage, but milk yield and composition were not different. When fed as silage, the leafy corn hybrid used in this experiment supported greater DMI as well as higher milk and protein yields when compared to the grain corn hybrid.  相似文献

Our objective was to investigate the effect of i.v. dextrose as an adjunct therapy to oral propylene glycol on the resolution of hyperketonemia (HYK; blood β-hydroxybutyrate ≥1.2 mmol/L), disease incidence, and early lactation milk yield. Cows (n = 1,249) between 3 and 16 d in milk (DIM) from 4 New York dairy farms were screened once weekly for HYK for 2 wk. Those with HYK and no previous history of retained placenta, metritis, or HYK were randomly assigned to 1 of 3 treatment groups: 300 mL of oral 100% propylene glycol for 3 d (PG3); 300 mL of oral 100% propylene glycol for 3 d plus 500 mL i.v. 50% dextrose on d 1 (PG3D1); or 300 mL of oral 100% propylene glycol for 3 d plus 500 mL i.v. 50% dextrose on all 3 d (PG3D3). Cows with a blood β-hydroxybutyrate <1.2 mmol/L at initial screening were re-screened the following week and randomly assigned to the above treatment groups if blood β-hydroxybutyrate was ≥1.2 mmol/L. Cows were assessed for post-treatment HYK resolution 1 and 2 wk after initial HYK diagnosis. We collected farm-diagnosed occurrence of adverse events (sold, died, metritis, displaced abomasum, or ketosis) during the first 60 DIM and milk yield data from the first 10 wk of lactation from herd management software. We used mixed-effects multivariable Poisson regression models to assess the risk of post-treatment HYK resolution at 1 and 2 wk following initial HYK diagnosis and adverse event occurrence among treated cows. We used repeated-measures ANOVA to assess differences in average daily milk yield between treatments. The overall HYK incidence was 30.1% (n = 373). Sixty-four percent of cows (n = 237) were assigned to a treatment group in the first week (3 to 9 DIM), and 36% (n = 136) assigned the second week (10 to 16 DIM). The incidence of 1 or more adverse events during the first 60 DIM was 9.4% (n = 35). We found no effect of treatment on risk of post-treatment HYK resolution at wk 1 (PG3 56.9%, PG3D1 45.0%, PG3D3 50.0%) or wk 2 (PG3 60.0%, PG3D1 52.1%, PG3D3 59.5%) following initial diagnosis, or for risk of adverse event occurrence (PG3 7.4%, PG3D1 8.0%, PG3D3 12.6%). Average daily milk yield (mean ± SE) was similar between treatment groups (PG3: 42.7 ± 0.6 kg/d, PG3D1: 42.4 ± 0.6 kg/d, PG3D3: 42.6 ± 0.6 kg/d). The addition of dextrose for 1 or 3 d provided no improvement in resolution of ketosis assessed once weekly, reduction in adverse events during the first 60 d of lactation, or a difference in average daily milk yield during the first 10 wk of lactation.  相似文献

以3,5-二叔丁基水杨醛和乙二胺(摩尔比2∶1)为原料合成了N,N’-二3,5 -二叔丁基水杨醛基乙二胺(salen配体),salen配体与Cu( OAC)2·H20的乙醇溶液混合回流得到Cu - salen配合物.对重结晶提纯后的salen配体及配合物进行了红外、紫外、核磁等表征.Cu - salen配合物作为大豆油环氧化反应催化剂,以叔丁基过氧化氢(TBHP)为氧源,最佳反应条件为:60℃下反应8h,催化剂用量为大豆油质量的2%,得到最佳环氧化效果,其环氧产物转化率为52.5%,选择性为59.6%.超声波辅助下,环氧值由2.2%提高到4.0%,环氧值提高了45%;转化率提高至71.4%,选择性提高到63.5%;反应时间由8h缩短至0.5h,催化剂用量减少为大豆油质量的0.5%.  相似文献
