用人,雄性Wistar大鼠和雄性莱亨鸡的肝细胞,或食管上皮细胞与V_(79)细胞混合培养,观察甲基苄基亚硝胺(MBN)经肝细胞或食管上皮细胞代谢后,能否产生活性代谢产物,从而诱导V_(79)细胞突变及其它有关的生物学反应。研究结果初步表明,大鼠和鸡肝细胞可以明显代谢MBN产生活性代谢产物,诱导混合培养的V_(79)细胞SCE和微核增高以及6-TG抗性突变。人胎儿肝细胞也可以代谢这种亚硝胺并诱导V_(79)细胞6-TG抗性突变,但对SCE和微核诱导明显偏低。说明这三种肝细胞代谢MBN在性质上是比较近似,可能在代谢激活程度上有一定差异。在食管上皮细胞和V_(79)细胞混合培养的实验中,大鼠食管上皮可以明显代谢MBN诱导V_(79)细胞SCE和微核的增高及6-TG抗性突变。鸡食管上皮细胞未见有明显的代谢MBN引起V_(79)细胞突变的作用。26只雄性莱亨鸡喂以MBN实验最长者达975天,总剂量为1674毫克,结果未见一例鸡发生食管癌。结果表明鸡的食管上皮可能对某些挥发性亚硝胺的致癌作用是不敏感的,目前没有证据支持在林县高发区中所发现的鸡咽食管癌是由于已找到的挥发性亚硝胺引起的。人食管上皮细胞具有一定的代谢MBN产生有致突变作用的代谢产物的能力,然而人与人之间有明显的个体差异。和大鼠相比,人食管代谢MBN诱导V_(79)细胞突变的能力明显较低,有关亚硝胺在高发区人食管癌发生中的作用还有待进一步深入研究。  相似文献

向日葵籽,日本市场上已出现用向日葵籽提取物制成的抗氧化剂。该抗氧化剂的主要成份与绿原酸属于同一类型,在低浓度条件下,仍具有很强的抗氧化性能。但是,在有酶存在的情况下以及在弱碱条件下,其活性就会降低,效果受到影响。将向日葵籽提取物与维生素C给合起来,制成抗氧化剂,效果较好,其中维生素C占40%,向日葵籽提取物占20%,其它成份有乳糖和柠檬酸。向日葵籽提取物中的活性物质可以抑制维生素C的分解。此种抗氧化剂可以用在水产品中,起到防止褪色作用;用在饮料中防止风味变化,放在蔬菜罐头中防止褪色等。绿茶提取物:绿…  相似文献

Peroxiredoxin V(Prx V)是过氧化物酶peroxiredoxins家族中的一员,在神经细胞中含量丰富,具有通过清除细胞内活性氧(reactive oxygen species,ROS)和过氧亚硝酸盐抑制氧化应激诱导的细胞凋亡的作用。过量的一氧化氮(nitric oxide,NO)具有较强的神经毒性,可引起小胶质细胞炎性反应,诱导神经细胞凋亡从而引发神经退行性疾病,而且可诱导神经小胶质细胞Prx V的表达,参与小胶质细胞的活性调控过程。但是,NO诱导的海马神经细胞凋亡过程中Prx V的作用尚不清楚。该研究利用硝普化钠(sodium nitroprusside dihydrate,SNP)作为NO供体,检测了NO诱导的HT22小鼠海马神经细胞的凋亡及对Prx V蛋白表达的影响。结果显示,SNP诱导的HT22细胞凋亡呈现时间、浓度依赖性;并特异性地抑制了Prx V的表达,致使细胞内ROS水平升高,激活线粒体依赖的经典凋亡途径,导致HT22细胞的凋亡。该研究结果揭示,NO通过抑制细胞内Prx V的表达导致细胞内ROS水平升高,最终诱导HT22细胞发生凋亡的机制,为保护NO诱导的神经细胞凋亡提供了新的理论依据。  相似文献

将重组创伤弧菌溶细胞素A(recombinant Vibrio vulnificus hemolysin,rVvhA)第201位和第289位的缬氨酸定点突变为甘氨酸,并表达rVvhAval201gly,val289gly突变蛋白。检测突变蛋白与未突变蛋白的溶血活性、对胞内钙离子浓度、钾离子外流及对细胞损伤的影响。结果显示,与rVvhAval201gly,val289gly的溶血活性和细胞毒性均降低,诱导人类脐静脉内皮细胞(human umbilical veinendothelial cells,HUVEC)凋亡、胞内钙离子内流和钾离子外流等作用均受到抑制。实验结果表明,在创伤弧菌溶细胞素的活性发挥中,rVvhA的V201和V289两个氨基酸与该蛋白质损伤靶细胞时引起胞内外离子平衡失调有关,并能影响该蛋白质的溶血活性和细胞毒作用。  相似文献

目的:探讨JNK抑制剂SP600125诱导巨核细胞白血病系多倍体化的调控机制。方法:用SP600125和3种抑制剂(PD184352、U0126和LY294002)处理Dami和CMK细胞,用点突变技术对核糖体蛋白S6激酶1(S6K1)C端自抑制假底物结构域和疏水基序突变构建的S6K1质粒转染细胞,流式细胞术分析细胞的DNA倍性,Western印迹检测S6K1、MAPK和Akt蛋白的表达及磷酸化修饰位点的变化。结果:当单独使用3种抑制剂时,对Dami和CMK细胞的倍性无影响。当SP600125与3种抑制剂联合时,尽管PD184352下调了p44/42 MAPK在Thr202/Tyr204位点的磷酸化,但其并不抑制SP600125诱导的Dami和CMK细胞的多倍体化;相反,U0126抑制SP600125诱导的Dami和CMK细胞的多倍体化,但并不下调p44/42 MAPK的磷酸化;而LY294002增加Akt的磷酸化,并阻断SP600125诱导的Dami和CMK细胞的多倍体化。这3种抑制剂均在SP600125诱导的多倍体Dami和CMK细胞中部分抑制S6K1的Thr421/Ser424磷酸化,但并不增加S6K1的Thr389磷酸化。转染S6K1突变质粒的Dami细胞,并没有对SP600125诱导的多倍体化产生影响,同时,无论是模拟磷酸化或去磷酸化的Thr389突变均对SP600125诱导的多倍体化无影响,且未显现出与LY294002的协同作用。C端自抑制假底物结构域突变质粒(S6K1-D3E)可以进一步阻断SP600125诱导的已经被LY294002部分阻断的Dami细胞多倍体化。结论:S6K1的C端自抑制假底物结构域磷酸化与Akt去磷酸化协同调节SP600125诱导巨核细胞白血病系的多倍体化。  相似文献

不久前,由国家自然科学基金资助的两个课题:茶叶防癌作用的研究、绿茶抗氧化剂(儿茶素)抑制癌变机制的研究分别通过了成果鉴定。在此基础上,由中国预防医学科学院营养与食品卫生研究所发起和主持,又召开了茶叶防癌和其他保健作用的专题研讨会。参加会议的有来自全国23个医学、农业和商业方面教学、科研等单位的近50名科研人员参加。会上广泛交流了茶叶防癌与保健作用的研究结果,并讨论了今后进一步深入研究的设  相似文献

Antimutagenic activity of green tea polyphenols   总被引:12,自引:0,他引:12  
For centuries green tea has been a widely consumed beverage throughout the world. It is known to contain a number of pharmacologically active compounds. In this study water extracts of green tea (WEGT) and their major constituents, green tea polyphenols (GTP), were examined for antimutagenic activity. WEGT and GTP were found to significantly inhibit the reverse mutation induced by benzo[alpha]pyrene (BP), aflatoxin B1 (AFB1), 2-aminofluorene, and methanol extracts of coal tar pitch in Salmonella typhimurium TA100 and/or TA98 in the presence of a rat-liver microsomal activation system. GTP also inhibited gene forward mutation in V79 cells treated with AFB1 and BP, and also decreased the frequency of sister-chromatid exchanges and chromosomal aberrations in V79 cells treated with AFB1. The addition of GTP during and after nitrosation of methylurea resulted in a dose-dependent inhibition of mutagenicity. Studies to define the mechanism of the antimutagenic activity of GTP suggest that it may affect carcinogen metabolism, DNA adduct formation, the interaction of ultimate carcinogen or the scavenging of free radicals.  相似文献

Diseases activate the innate immune response which causes ancillary damage to the human body. Peroxynitrite (OONO-) or its carbon dioxide derivatives cause oxidation/nitration and hence mutation to various body polymers e.g. DNA, RNA, protein, lipids and sugars. The control of the ancillary damage can come from antioxidants which inhibit control the amount of peroxynitrite available for damage. In this paper we have developed three different levels of antioxidant screening: (i) Peroxynitrite or SIN-1 reaction with luminol to produce light, and the inhibition of light by substances therefore represents antioxidation. (ii) Nicking of plasmid DNA occurs via oxidants: and is prevented by antioxidants. (iii) Detection of plasmid luciferase activity post-oxidation and infection indicates either prevention or repair of damage: via antioxidants. We found green tea and a number of its polyphenolic constituents effective only at the first level of antioxidation, while extracts of various fruit help at all levels antioxidation. In the final analysis, a combination of green tea extracts and fruits is suggested to produce more complete antioxidant protection.  相似文献

Transitional metals, as vanadium, are known to exert noxious effects by generating oxidative stress. Addition of antioxidants in the diet could decrease the cytotoxic effect related to the oxidative stress. The present study, carried out in Wistar rats, is a contribution to the evaluation of protective effects of green tea Camellia sinensis, which is known to be rich in antioxidant compounds (polyphenols...). Rats were divided into four groups: (C) was control, (V) was given ammonium metavanadate (AMV), (TH) was given herbal tea as drink (66 g/l) and TH + V was given tea and metavanadate. Group (TH) was given herbal tea one month before vanadium treatment. Metavanadate was daily i.p. injected (5 mg NH4VO3/kg body weight) for 10 days. (C) and (TH) groups received i.p. injections of 0.9% NaCl during the same period. Changes in lipid peroxidation levels (TBARS) in kidney, liver and testes, serum concentrations of vitamins E and A and superoxidismutase (SOD) and catalase (CAT) activities in blood cells were determined. One month pre-treatment with green tea, followed by 10 days of treatment (TH) did not change TBARS in liver and testes as compared to controls, but induced a clear decrease of TBARS in kidneys. Intraperitoneal administration of AMV to rats (V) induced a time-dependant increase of TBARS in kidney, liver and testes that was lowered in rats (V + TH) drinking tea. Vitamin E concentrations were found to be drastically decreased from day 1 to 10 in rats (V). Vitamin A concentration was decreased at day 10 only. Drinking tea lowered AMV inhibitory effects in rats (V + TH), and conversely an increase of vitamins A and E concentrations were found at day 10. SOD and catalase activities were found increased in the blood cells from day 1 to day 5 and conversely decreased at day 10. In contrast, associated to green tea, AMV did not affect SOD and catalase activities compared to controls.  相似文献

Positive health effects of tea (Camellia sinensis) on a wide range of physiological problems and diseases are well known and are in part due to its copious antioxidant content. The effect of black tea extract (BTE), which is rich in polyphenolic antioxidants, against the consequences of radiation exposure has not been properly identified. The functional properties of BTE were analyzed and its radioprotective effect on V79 cells was explored in the present study. BTE scavenged free radicals and inhibited Fenton reaction-mediated 2-deoxyribose degradation and lipid peroxidation in a dose-dependent fashion, establishing its antioxidant properties. The radioprotective effects of BTE on strand break induction in pBR322 plasmid DNA were 100 % at 80 μg/ml and higher. In V79 cells, BTE was effective in decreasing the frequency of radiation-induced micronucleated cells and the yields of reactive oxygen species (ROS) and also in restoring the integrity of cellular mitochondrial membrane potential significantly. BTE exerted maximum protection against radiation-induced damage in V79 at a dose of 5 μg/ml. Due to the functional properties of BTE-flavonoids, which have been identified by HPLC, it is envisaged that the key player in radioprotection is elimination of ROS.  相似文献

Previously, we have shown that green tea extract (GTE) lowers the intestinal absorption of lipids and lipophilic compounds in rats. This study was conducted to investigate whether GTE inhibits the intestinal absorption and biliary secretion of benzo[a]pyrene (BaP), an extremely lipophilic potent carcinogen, present in foods as a contaminant. Male rats with lymph or bile duct cannula were infused at 3.0 ml/h for 8 h via a duodenal catheter with lipid emulsion containing (14)C-BaP with or without GTE in PBS buffer. Lymph and bile were collected hourly for 8 h. The (14)C-radioactivities in lymph, bile and intestine were determined and expressed as % dose infused. Results showed that GTE drastically lowered the lymphatic absorption of (14)C-BaP (7.6±3.2% in GTE-infused vs. 14.4±2.7% dose/8 h in control rats), with a significantly higher amount of (14)C-radioactivity present in the small intestinal lumen and cecum in rats infused with GTE. GTE also markedly increased the hourly rate (3.9±0.1% dose/h in GTE-infused vs. 3.0±0.1% dose/h in control rats) and the total biliary secretion of (14)C-BaP (31.5±0.8% dose/8 h in GTE-infused vs. 24.3±0.4% dose/8 h in control rats). The findings provide first direct evidence that GTE has a profound inhibitory effect on the intestinal absorption of BaP and promotes the excretion of absorbed BaP via the biliary route. Further studies are warranted to investigate whether green tea could be recommended as a dietary means of ameliorating the toxicity and carcinogenic effect of BaP.  相似文献

Several reports have attributed to green tea chemopreventive and therapeutic properties. Epidemiological studies have linked the regular use of green tea to a reduced incidence of breast and colon carcinomas. Tea contains several antioxidants, including polyphenols of the catechin (green tea) and theaflavin (black tea) groups. Green tea derivatives have been shown to act in vitro and in vivo as anti-inflammatory, anti-viral and anti-tumor drugs. Despite the extensive body of data only few studies have investigated the molecular mechanisms underlying these effects. In this brief review we focus on the inhibitory activity of catechins derived from green tea toward proteases involved in tumor invasion.  相似文献

The genotoxicity of 15 polycyclic aromatic hydrocarbons was determined with the alkaline version of the comet assay employing V79 lung fibroblasts of the Chinese hamster as target cells. These cells lack the enzymes necessary to convert PAHs to DNA-binding metabolites. Surprisingly, 11 PAHs, i.e., benzo[a]pyrene (BaP), benz[a]anthracene, 7,12-dimethylbenz[a]anthracene, 3-methylcholanthrene, fluoranthene, anthanthrene, 11H-benzo[b]fluorene, dibenz[a,h]anthracene, pyrene, benzo[ghi]perylene and benzo[e]pyrene caused DNA strand breaks even without external metabolic activation, while naphthalene, anthracene, phenanthrene and naphthacene were inactive. When the comet assay was performed in the dark or when yellow fluorescent lamps were used for illumination the DNA-damaging effect of the 11 PAHs disappeared. White fluorescent lamps exhibit emission maxima at 334.1, 365.0, 404.7, and 435.8 nm representing spectral lines of mercury. In the case of yellow fluorescent lamps these emissions were absent. Obviously, under standard laboratory illumination many PAHs are photo-activated, resulting in DNA-damaging species. This feature of PAHs should be taken into account when these compounds are employed for the initiation of skin cancer. The genotoxicity of BaP that is metabolically activated in V79 cells stably expressing human cytochrome P450-dependent monooxygenase (CYP1A1) as well as human epoxide hydrolase (V79-hCYP1A1-mEH) could not be detected with the comet assay performed under yellow light. Likewise the DNA-damaging effect of r-7,t-8-dihydroxy-t-9,10-epoxy-7,8,9,10-tetrahydrobenzo[a]pyrene (anti-BaPDE) observed with the comet assay was only weak. However, upon inhibition of nucleotide excision repair (NER), which is responsible for the removal of stable DNA adducts caused by anti-BaPDE, the tail moment rose 3.4-fold in the case of BaP and 12.9-fold in the case of anti-BaPDE. These results indicate that the genotoxicity of BaP and probably of other compounds producing stable DNA adducts are reliably detected with the comet assay only when NER is inhibited.  相似文献

With the use of the spin trapping methods, the scavenging effects of the extracts of green tea and other natural foods are studied. In stimulated polymorphonuclear leukocytes (PMN) system, water extract fraction 6 (F6) from green tea and green tea polyphenols (GTP) have the strongest scavenging effect on the active oxygen radicals, much stronger than vitamin C (Vc) and vitamin E (VE). Rosemary antioxidants (RA) and Curcumin (Cur) have weaker scavenging effects than Vc, but stronger than VE. In Fenton Reaction, Cur has the strongest scavenging effect (69%) on hydroxyl radicals. In irradiation, riboflavin system F6(74%) and GTP(72%) have very strong scavenging effects that are weaker than Vc, but much stronger than VE (23%). With the use of spin probe oxymetry, the oxygen consumption in respiratory burst of stimulated PMN were measured when the antioxidants existed in these systems. The results demonstrated that these antioxidants did not affect the respiratory burst of human polymorphonuclear leukocytes stimulated with PMA.  相似文献

Oxidants have been shown to be involved in alcohol-induced liver injury. This study was designed to test the hypothesis that the antioxidant polyphenolic extract of green tea, comprised predominantly of epigallocatechin gallate, protects against early alcohol-induced liver injury in rats. Male Wistar rats were fed high-fat liquid diets with or without ethanol (10-14 g kg(-1) day(-1)) and green tea (300 mg kg(-1) day(-1)) continuously for 4 weeks using an intragastric enteral feeding protocol. Mean body weight gains (approximately 4 g/day) were not significantly different between treatment groups, and green tea extract did not the affect average concentration or the cycling of urine ethanol concentrations (0-550 mg dl(-1) day(-1)). After 4 weeks, serum ALT levels were increased significantly about 4-fold over control values (35+/-3 IU/l) by enteral ethanol (114+/-18); inclusion of green tea extract in the diet significantly blunted this increase (65+/-10). Enteral ethanol also caused severe fatty accumulation, mild inflammation, and necrosis in the liver. While not affecting fat accumulation or inflammation, green tea extract significantly blunted increases in necrosis caused by ethanol. Furthermore, ethanol significantly increased the accumulation of protein adducts of 4-hydroxynonenal, a product of lipid peroxidation and an index of oxidative stress; green tea extract blocked this effect almost completely. TNFalpha protein levels were increased in liver by alcohol; this phenomenon was also blunted by green tea extract. These results indicate that simple dietary antioxidants, such as those found in green tea, prevent early alcohol-induced liver injury, most likely by preventing oxidative stress.  相似文献
