Two new lignan glycosides, 2'-hydroxyl asarinin 2'-O-β-D-glucopyranoside (cuscutoside C, 1) and 2'-hydroxyl asarinin 2'-O-β-D-apiofuranosyl-(1?→?2)-[β-D-glucopyranosyl-(1?→?6)]-β-D-glucopyranoside (cuscutoside D, 2), were isolated from the seeds of Cuscuta chinensis Lam., along with six known compounds, 2'-hydroxyl asarinin 2'-O-β-D-glucopyranosyl-(1?→?6)-β-D-glucopyranoside (3), 2'-hydroxyl asarinin 2'-O-β-D-apiofuranosyl-(1?→?2)-β-D-glucopyranoside (cuscutoside A, 4), kaempferol 3,7-di-O-β-D-glucopyranoside (5), 5-caffeoyl quinic acid (6), 4-caffeoyl quinic acid (7), and cinnamic acid (8). Their structures were elucidated on the basis of spectroscopic analyses including HR-ESI-MS, ESI-MS/MS, (1)H and (13)C NMR, HSQC, HMBC, and TOCSY.  相似文献

目的研究菟丝子(Cuscuta chinensisLam.)的化学成分。方法采用多种色谱法进行分离纯化,根据理化性质及波谱数据进行化合物的结构鉴定。结果从菟丝子的体积分数为70%的乙醇溶液提取物中分离得到9个化合物,分别鉴定为对羟基反式桂皮酸十八烷基酯(1)、3-O-β-D-吡喃葡萄糖-5-羟基桂皮酸甲酯(2)、山柰酚(3)、紫云英苷(4)、金丝桃苷(5)、紫云英苷-6″-O-没食子酸酯(6)、槲皮素-3-O-(6″-没食子酰基)-β-D-葡萄糖苷(7)、β-谷甾醇(8)、胡萝卜苷(9)。结论化合物1、2、6、7首次从该属植物中分离得到。  相似文献

目的用HPLC测定菟丝子中金丝桃苷与槲皮苷的含量。方法采用ShimadzuC18柱(150mm×4.6mm,5μm),流动相为甲醇-0.2%H3PO4水溶液(45∶55),流速1.0ml·min-1,检测波长350nm,柱温为室温,用外标法定量,测定菟丝子中金丝桃苷与槲皮苷的含量。结果金丝桃苷的线性范围0.50~2.50μg(r=0.9998),回收率95.90%,RSD=0.93%;槲皮苷的线性范围0.40~2.00μg(r=0.9996),回收率99.19%,RSD=2.39%。结论所建方法简便,快速,线性关系良好。  相似文献

酸性菟丝子多糖的化学特征和免疫活性(英文)   总被引:3,自引:0,他引:3  
目的:研究从菟丝子(Cuscuta chinensis Lam)中提取分离的酸性纯多糖CHC-1的结构特征及免疫活性.方法:利用化学方法和NMR光谱分析方法对CHC-1的结构特征进行了解析,并研究了CHC-1对小鼠抗体生成以及对淋巴细胞增殖率的影响.结果:CHC-1的分子量大于1.0×10~6.糖组份分析结果显示CHC-1是由Rha,Ara,Gal和GalA组成,比例为:0.8:1.0:1.5:0.3.甲基化结果和~1H,~(13)C NMR分析进一步确定了CHC-1中各残基的连接方式.CHC-10.1 g/L能在体外明显升高T,B细胞的增殖率,CHC-125,50 mg/kg显著增加小鼠的脾重,淋巴细胞的增殖率以及促进抗体生成.结论:CHC-1为一种多分支的杂多糖,并具有免疫增强活性.  相似文献

目的 研究菟丝子提取物的体外抗氧化活性.方法 采用热水提取、酶-Sevage法除蛋白、不同比例乙醇分级沉淀等方法获得菟丝子多糖提取物;经95%乙醇回流提取,2乙酸乙酯、正丁醇萃取,得到乙酸乙酯和正丁醇提取物.利用1,1-二苯基-2-苦苯肼自由基(DPPH·)和超氧阴离子自由基(O2-·)分别测定了各组分的抗氧化活性.结果 菟丝子提取物均具有一定的抗氧化活性,且呈显著的量效关系.菟丝子乙酸乙酯、正丁醇提取物清除自由基的能力大于氧化型谷胱甘肽,小于还原型谷胱甘肽;菟丝子多糖清除自由基的能力均小于氧化型谷胱甘肽,其中90%醇沉多糖清除自由基的能力较强.结论 菟丝子乙酸乙酯、正丁醇提取物以及90%醇沉多糖是天然抗氧化剂的良好来源,可以进一步分离纯化.  相似文献

Their structures were determined by various spectroscopic methods. 菟丝子多糖对衰老小鼠脂质过氧化的影响   总被引:4,自引:1,他引:4  
目的:观察菟丝子多糖对衰老小鼠模型丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSHPX)和脑组织中脂褐质(LF)的影响。方法:用d半乳糖制备衰老小鼠模型,随机分为模型组(不给药)、阳性对照组(灌注维生素E200mg·kg-1,qd)、3个剂量菟丝子多糖组(分别灌注100,200,400mg·kg-1菟丝子多糖,qd),共7wk。另设空白对照组。结果:模型小鼠血清、肝、肾中MDA含量升高,SOD及GSHPX活性降低,脑中LF升高。而100,200,400mg·kg-1·d-1菟丝子多糖组能不同程度对抗模型小鼠脑组织中LF升高及血清、肝、肾组织中MDA升高,其量效关系均呈负相关,r分别为-0.916,-0.939,-0.950及-0.911,P<0.01;能不同程度抑制血清、肝、肾组织中SOD活力下降,其量效关系均呈正相关,r分别为0.898,0.939及0.946,P<0.01;能不同程度抑制血清、肝、肾组织中GSHPX活力下降,其量效关系均呈正相关,γ分别为0.947,0.969及0.947,P<0.01。结论:菟丝子多糖有显著清除氧自由基及抗脂质过氧化作用。  相似文献

Nine compounds were isolated from the leaves of Anthocephalus chinensis by column chromatography on silica gel and Sephadex LH-20, and their structures were elucidated by spectroscopic techniques as clethric acid-28-O-β-d-glucopyranosyl ester (1), mussaendoside T (2), β-stigmasterol (3), hederagenin (4), ursolic acid (5), clethric acid (6), 3β,6β,19α,24-tetrahydroxyurs-12-en-28-oic acid (7), mussaendoside I (8), and cadambine (9). Compounds 1 and 2, and 7 and 8 were isolated from the plants of this genus for the first time, and compounds 1 and 2 were new triterpenoid glycosides.  相似文献

首次从中药菟丝子 (Cuscuta chinedsis Lam.) 中得到一均一酸性多糖H3,并采用化学及光谱方法对其结构特征进行了研究。结果表明该多糖为一多分枝的复杂多糖,由1,6-linked- β -D Galp, 1,4-linked-β-D Galp, 1,4-linked-β-D GalA, 1,3, 6-linked-β-D Galp 及1,2,4-linked Rhap 组成,分枝点位于1,2,4-linked Rhap 的O-2 或 O-4以及1,3,6-linked-β-D Galp 的O-3位,并以1-linked Galp 为末端。侧链由1-linked Araf, 1,5-linked Araf 和 1,3,5-linked Araf 构成,并连接于主链的1,6-linked Galp 的O-3位。  相似文献

Two new lignan glycosides forsythiayanoside A (1) and forsythiayanoside B (2), one new stereoisomer of lignan glycoside (+)-8-hydroxyepipinoresinol-4-O-β-d-glucopyranoside (3), together with seven known compounds (410), were isolated from the fruits of Forsythia suspense (Thunb.) Vahl. Compounds 4 and 10 were isolated from this species for the first time. The structures of 13 were elucidated on the basis of chemical and spectral analyses, including 1D and 2D NMR data and HR-ESI-MS. The absolute configurations were determined by the circular dichroism method. All isolates were tested for their cytotoxicities against five human cancer cell lines (A549, Colo-205, Hep-3B, HL60, and KB). In particular, compound 3 showed significant cytotoxicity with IC50 values of 9.48, 7.75, 0.59, 4.06, and 38.38 μM, respectively.  相似文献

H3, a homogeneous acidic polysaccharide was obtained from the seeds of Cuscuta chinensisLam. Its structure was characterized for the first time by chemical and spectroscopic methods to be a highly branchedheteropolysaccharide with mean molecular weight of more than lxl0^6. It was composed of 1,6-1inked-β-D Galp, 1,4-linked-β-D Galp, 1,4-1inked-β-D GalA, 1,3,6-1inked-β-D Galp and 1,2,4-1inked Rhap, with branching points at O-2or 0-4 of 1,2,4-1inked Rhap and 0-3 of 1,3,6-1inked-β-D Galp. Its side chains included 1-1inked Araf, 1,5-1inked Arafand 1,3,5-1inked Araf at O-3 of 1,6-1inked Galp in the main chain.  相似文献

目的用高效液相色谱法测定菟丝子中对-香豆酸的含量。方法采用HypersilC18色谱柱(250mm×4.6mm,5μm),流动相:乙腈-0.1%冰醋酸水溶液(25∶75),流速:1.0mL.m in-1,检测波长:308nm。结果对-香豆酸在2.0~40.0μg.mL-1浓度范围内线性关系良好,r=0.9993,平均回收率为99.9%,RSD为1.9%。结论本法简便、准确,可用于菟丝子中对-香豆酸的含量测定。  相似文献

目的观察菟丝子提取物(CCLE)对大鼠心肌缺血/再灌注损伤(MI/RI)的保护作用。方法 30只SD大鼠随机分为3组(n=10):假手术组、生理盐水对照组、菟丝子提取物组。结扎左冠状动脉前降支,建立大鼠心肌缺血/再灌注损伤模型,记录各组心电图,HE染色观察心肌组织细胞形态,测定血清肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)、谷草转氨酶(AST)的含量变化。结果与对照组相比,CCLE可使MI/RI大鼠:①心率(HR)减慢(再灌注期各时间点P<0.05或P<0.01);②再灌注60、90、120 min时ST段明显降低(P<0.05或P<0.01);③心肌梗死程度明显减轻;④心肌酶CK、CK-MB、LDH、AST的含量(U.L-1)比对照组分别降低3 360(P<0.01)、5 478(P<0.01)、922(P<0.05)、235(P<0.01)。结论 CCLE对再灌注损伤心肌有明显的保护作用。  相似文献

目的:基于PI3K/PTEN/VEGF通路探讨菟丝子总黄酮(TF)体外抗肿瘤作用机制,考察合适的给药浓度,为菟丝子药材的临床合理应用提供参考。方法:以人肝癌细胞HepG2为实验对象,CCK-8法检测TF给药后的细胞活性,计算其IC 50值;Annexin V-FITC/PI双染法分析细胞凋亡坏死比例;实时荧光定量PCR (qPCR)法检测胞内磷脂酰肌醇激酶(PI3K)、人类第十号染色体缺失的磷酸酶及张力蛋白同源基因(PTEN)、血管内皮细胞生长因子(VEGF) mRNA的相对表达;酶联免疫吸附法(ELISA)检测缺氧诱导因子-1α(HIF-1α)、血管内皮细胞生长因子受体2、3(VEGFR-2、VEGFR-3)表达变化趋势。结果:相对阴性对照组,给药组细胞抑制率和凋亡率均具有良好的量-效正相关;qPCR结果显示TF给药可以显著的上调PTEN mRNA,下调PI3K、VEGF mRNA;ELISA结果显示HIF-1α、VEGFR-2、VEGFR-3蛋白的表达在给药后均有下降(P<0.01)。结论:菟丝子总黄酮有较好的体外抗肝癌药效,其作用可能与上调PTEN的表达对PI3K-Akt通路进行负调控诱导细胞发生凋亡、下调HIF-1α以及VEGF相关基因和受体蛋白的表达等作用机制共同发挥抗肿瘤作用。  相似文献

目的:研究菟丝子提取物对自然衰老小鼠的抗衰老作用。方法:给自然衰老小鼠灌胃菟丝子提取物,先用Y-型电迷宫检测小鼠的学习记忆能力,后应用生化方法检测老年小鼠脑组织内超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)的活力和丙二醛(MDA)含量。结果:小鼠行为实验结果表明,菟丝子提取物能使小鼠Y-型电迷宫测试正确次数显著降低,但对记忆的保持力影响不大。小鼠脑组织抗氧化酶的活力实验结果表明,菟丝子提取物能提高小鼠脑组织SOD、GSH-PX活力,降低MDA的含量。结论:菟丝子提取物抗衰老作用可能通过提高脑组织抗氧化酶SOD、GSH-PX活力,降低脂质过氧化物MDA含量而实现。  相似文献

目的:建立同时测定蒙药菟丝草中槲皮素、金丝桃苷和山柰酚3种成分含量的方法。方法:采用高效液相色谱法。色谱柱为Hypersil ODS-2(300 mm×4.6 mm,5μm),流动相为甲醇-水(58∶42,V/V),检测波长为281 nm,柱温为30℃,流速为1.0 ml/min。结果:槲皮素、金丝桃苷和山柰酚的质量浓度分别在1050、2.550、2.530、2.530、2.530μg/ml范围内与各自峰面积积分值呈良好的线性关系(r分别为0.999 7、0.999 5、0.999 6);精密度、稳定性、重复性试验的RSD<2%;平均加样回收率分别为98.90%、97.60%、97.39%,RSD分别为1.23%、1.37%、1.40%(n均为6)。结论:该方法简便、快速、准确,可为蒙药菟丝草的质量评价提供依据。  相似文献
