The use of combusted natural gas as atmosphere for cultivation of the anaerobic bacterial flora from the cecum of mice is described. The combusted gas (inert gas) continuously flushes a glove box in which it is possible to cultivate a number of strict anaerobes from the cecum of mice.  相似文献

A thermophilic fungus, K1014, newly derived from a compost was selected on the basis of protease productivity as the only one of 81 isolates to produce high levels of acid protease. The fungus was named Penicillium duponti K1014 based on taxonomical studies. It grew in the temperature range of 28 to 58 C, and the optimum was 45 to 50 C. These temperature characteristics showed that the fungus was the most strongly thermophilic of all the fungi next to Humicola lanuginosa. When P. duponti K1014 was grown on moistened wheat bran, maximal accumulation of acid protease occurred after 2 days at 45 to 50 C. The addition of ammonium salts, but not nitrate, was effective for the production of the acid protease. The acid protease of P. duponti K1014 was stable at 60 C for 1 hr and retained more than 65% of original activity after the treatment for 1 hr at 70 C at pH 4.7. This thermal property was different from those of the ordinary acid proteases, indicating that the enzyme is a thermostable protein.  相似文献

目的:研究嗜热脂肪土芽孢杆菌CHBl产酶特性。方法:以蛋白酶活力为主要指标,考察温度、pH、接种量、装量等条件对CHB1产蛋白酶的影响。结果:CHB1适宜的产蛋白酶条件为:装量40mLt250mL,pH8.0,接种量5%,温度58℃,转速180r/min,时间36~44h,Tween-80对CHB1产蛋白酶具有抑制作用。结论:优化后蛋白酶产量有较大提高,最高酶活力达48U/mL,是所报道多数嗜热细菌产蛋白酶量的2—12倍。CHB1蛋白酶特性的研究及产蛋白酶量的提高,有利于揭示CHB1在堆肥化中的作用机理,提高堆肥效果。  相似文献

A crystalline acid protease produced by a strain of Acrocylindrium in a submerged culture was prepared by treatment with acetone (60%), salting out with ammonium sulfate (saturated) and, after chromatography on Duolite GS-101 column, dialysis against distilled water. This preparation was homogeneous on sedimentation analysis, starch-gel electrophoresis and gel filtration with Sephadex G-75. The optimum pH was 2.0 for milk casein digestion and the pH stability was for 2.0~5.0 at 30°C for one day. The crystalline enzyme was completely stable below 50°C, but lost the activity at 70°G in ten minutes. The acid protease was almost equal to pepsin on specific activity when milk casein solution (pH 2.0) was used as substrate.  相似文献

A wood-destroying fungus, Trametes sanguinea, produced a potent acid protease in a submerged culture. Maximum proteolytic activity of the culture was attained after 140-hours cultivation in a medium containing dextrin and corn steep liquor. The acid protease was obtained in crystalline form from the mycelium-free culture filtrate by the following successive treatments: acetone precipitation, ionexchange column chromatography, ammonium sulfate fractionation, dialysis, and crystallization by acetone. Throughout the over-all process, the acid protease was purified approximately 30-fold with about 8% recovery of the original activity.  相似文献

探讨了液体发酵嗜热毛壳菌(Chaetomium thermophile)产生的内切β-葡聚糖酶的分离纯化及特性。粗酶液经硫酸铵分级沉淀,DEAE-Seplharose Fast Flow阴离子层析,Pheny1-Sepha-rose疏水层析,Sephacry1 S-100分子筛层析等步骤便可获得凝胶电泳均一的内切β-葡聚糖酶,经12.5%SDS-PAGE和凝胶过滤层析法分离纯化酶蛋白的分子量约为67.8kD的69.8kD。该酶反应的最适温度和pH分别为60℃和4.0-4.5在pH5.0条件下,该酶在60℃下稳定:70℃保温1h后,仍保留30%的活性;在80摄氏度的半衰期为25min,金属离子内切β-葡聚糖酶的活性影响较大,其中Na^ 对酶有激活作用;Fe^2 ,Ag^ ,Cu^2 ,Ba^2 ,Zn^2 等对酶有抑制作用。该酶对结晶纤维素有没水解能力。  相似文献

The culture filtrate of the crayfish plague fungus, Aphanomyces astaci (Saprolegniaceae), incubated in a peptone glucose medium was tested for chitinase activity under different conditions. The activities were assayed turbidimetrically using low-polymerized chitin as a substrate. Adsorption of chitinase was found to occur on chitin and probably on cellulose and sulphomethyl cellulose but not at all or only a little on some other cellulose derivatives. The pH optimum of the enzyme activity was found to lie at about pll 5.0–5.5. The stability was greatest near pH 6.5 and the highest degree of adsorption occurred at still higher pH values. Enzyme adsorption on the substrate seemed to protect the enzyme against inactivation by heating, shaking, and extreme pH-conditions. The chitinase activity was positively affected by the rest of the culture filtrate. Mercury, cobalt, and copper chlorides, and to a lesser degree some other metal salts, lowered the enzyme activity when present in the test medium. Cellobiose, but neither glucose nor N-acetyl glucosamine had a pronounced inhibiting effect on the activity. Neither cellobiose nor N-acetyl glucosamine seemed to affect chitinase adsorption on chitin. Some chelating and reducing compounds inactivated the culture filtrate. This activity-reducing effect of chelators was strongly prevented by EDTA in some cases.  相似文献

本研究为从云南腾冲热泉中分离纯化得到一株产高温蛋白酶的菌株并对其进行驯化培养,用以探究该菌株的生长条件及酶学特性,通过选择培养基筛选能够分解脱脂奶粉产蛋白酶的菌株,应用常规方法液体培养菌体,探究温度、pH、碳源、氮源对菌株生长情况的影响,并采用福林酚法测蛋白酶活性。并提取蛋白酶液对酶的最适pH、温度以及热稳定性、pH稳定性进行研究。结果发现通过含脱脂奶粉的固体培养基筛选得到一株产蛋白酶菌株A-2,经过生理生化试验和16S rDNA鉴定知该菌种属于Aneurinibacillus属。酵母粉、葡萄糖、55℃、pH值7.5分别为菌株生长的最适氮源、碳源、温度和pH。此外该菌株所产的蛋白酶最适温度为60℃,在pH值7~9具有较好的酶活性。因此,该菌株为嗜热芽孢杆菌,所产的碱性蛋白酶具有较高的耐受温度和pH稳定性,为进一步开发利用提供参考的价值。  相似文献

An acid protease produced by the thermophilic fungus Penicillium duponti K 1014 has been purified by consecutive ion-exchange and gel permeation chromatography, and crystallized from aqueous acetone solution. The purified endopeptidase gave a symmetrical schlieren peak by sedimentation velocity, and was found to be homogeneous upon disc gel electrophoresis at pH 9.5. The enzyme was most active at pH 2.5 against milk casein and showed high thermostability. An isoelectric point of 3.81 was found by isoelectric focusing. A minimum molecular weight of 41 590 was calculated from the amino acid composition, adopting an arginine content of one residue per mole of enzyme. This minimum molecular weight is in good agreement with the value of 41 000 previously found by gel permeation (Hashimoto, H., Iwaasa, T., and Yokotsuka, T. (1973), Appl. Microbiol. 25, 578). Besides the thermostability, the purified P. duponti protease differs from other well-characterized acid proteases in that it contains carbohydrate, 4.33% expressed as glucose. The enzyme was not affected by p-bromophenacyl bromide, but was completely inactivated by alpha-diazo-p-bromoacetophenone, diazoacetyl-DL-norleucine methyl ester, and diazoacetylglycine ethyl ester, in the presence of Cu2+. The complete inactivation of the protease by diazoacetyl-DL-norleucine methyl ester resulted in the specific incorporation of 1 mol of norleucine/mol of enzyme. On the basis of similar behavior of other acid proteases toward this inactivator, the results suggest the presence at the active site of an unusually reactive carboxyl group, involved in the catalytic function. The naturally occurring pepsin inhibitor of Streptomyces naniwaensis [Murao, S., and Satoi, S. (1970), Agric. Biol. Chem. 34, 1265] inhibited also the protease, at a threefold molar excess with respect to the enzyme.  相似文献

Streptomyces limosus was selected because it secreted a novel protease that catalyzed the synthetic reaction forming Pro-Pro-Pro from Pro-Pro. The protease was purified to an electrophoretically homogeneous state and an activity of more than about 20,000-fold that of the culture broth. The molecular mass of the enzyme was estimated to be 50 kDa by SDS-polyacrylamide gel electrophoresis. The enzyme was most active in alkaline pH for the synthetic reaction producing Pro-Pro-Pro from Pro-Pro, although for the hydrolytic reaction forming proline it was most active in neutral pH. The enzyme was inhibited by 1,2-epoxy-3-(p-nitrophenoxy)propane (EPNP) and diazoacetyl-DL-norleucine methyl ester (DAN). It can be considered that this enzyme belongs to the class of aspartic proteases. The substrate specificity indicates that this enzyme has a strong affinity for proline as a N-terminal amino acid of peptides.  相似文献

The thermophilic fungus Thermomyces lanuginosus, which is able to use dextran as primary carbon source for growth, excreted during the early phases of growth an enzyme activity capable of degrading dextran. The activity peaked at 22 h and decreased rapidly after the culture entered the stationary phase, probably caused by protease activity. Results from growth on a number of different carbon sources showed that polymer carbohydrates yielded the highest dextranase activities. On the basis of the substrate specificity and the release of glucose in the α-anomeric form from the hydrolysis of maltose, it is proposed that the enzyme responsible for the necessary degradation of dextran to smaller saccharides is an α-glucosidase. A protein disulfide isomerase (PDI) was purified to homogeneity from the thermophilic fungus Humicola insolens by a rapid three-step procedure, anion-exchange chromatography, concanavalin A-affinity chromatography, and reverse phase high performance liquid chromatography. Forty-oneμg of PDI was obtained from 100 g of wet mycelium. Concanavalin A-Sepharose chromatography is available for purification of the fungal PDI, indicating that the enzyme is also glycosylated like the yeast PDI. The fungal PDI exists as a dimer (2x60kDa), has a pI of 3.5, and is fairly heat-stable. The amino acid composition of the PDI is similar to those of yeast and bovine liver PDI, and the high content of acidic amino acid residues agrees with the lower acidic pI.  相似文献

肠道微生物分泌的蛋白酶可促进家蚕对桑叶养分的消化吸收,枯草芽孢杆菌是家蚕肠道内一种重要的产蛋白酶菌株。为提高枯草芽孢杆菌蛋白酶的高效利用,对该菌株适宜发酵条件及酶学性质进行了研究。结果表明:各因素对枯草芽孢杆菌产酶活性影响的大小顺序依次为:pH值〉培养温度〉培养时间〉装液量;最适的产酶条件为:pH=7,培养温度:30 ℃,培养时间:36 h;对枯草芽孢杆菌产蛋白酶进行初步提纯后并研究得出该酶反应的最适pH 10.0,最适反应温度为:60 ℃;该酶为碱性蛋白酶、不耐高温、不耐酸,但在35 ℃条件下热稳定性较好。  相似文献

Some physicochemical properties, amino acid composition, and substrate specificity of the acid protease of Rhodotorula glutinis K-24 were determined. The molecular weight was estimated to be about 30,000 by the sedimentation equilibrium method. This value also coincided with the data obtained from Andrews’s method.

The isoelectric point was pH 4.5, and the amino terminal amino acid was identified to be alanine. The enzyme contained 14.5% of nitrogen and was composed of 285 residues of amino acid. Substrate specificity toward synthetic peptides was similar to that of pepsin, but its activity was considerably weak.

The enzyme was inactivated by diazoacetyl glycine ethylester, p-bromophenacyl bromide, et al., which attacked the active center of pepsin.  相似文献

Among some 38 strains of the genus Penicillium we investigated seven wild-type strains (P. daleae IFO-6087, P. frequentans AHU-8328, P. funiculosum IAM-7013, P. janthinellum IFO-8070, IAM-7026, P. lividum IAM-7200, and P. oxalicum AHU-8336) that were found to be excellent strains for a new type of acid carboxypeptidase production in a surface koji culture at 25 C. The production of acid carboxypeptidase was determined in various culture conditions in a koji culture. The maximum yields of acid carboxypeptidase were obtained by P. janthinellum IFO-8070. Partial purification and isolation of the acid carboxypeptidase from strains of Penicillium were performed with gel filtration on Bio-Gel P-100. Characterization studies indicate that the acid carboxypeptidases from P. daleae IFO-6087, P. funiculosum IAM-7013, P. janthinellum IFO-8070, and P. oxalicum AHU-8336 have some properties similar to those of the enzyme of Aspergillus saitoi with regard to the hydrolysis of several peptides at acidic pH range but have other slightly different properties with regard to stability, pH optima, inhibitors, and molecular weights.  相似文献

Some physical and chemical properties of the extracellular lipase from the thermophilic fungus, Humico la lanuginosa S–38, were investigated. The results were as follows: Sedimentation coefficient was 2.4 × 10?13 (cm-g/sec-dyne); diffusion coefficient was 8.8 × 10?7 (cm2/sec); and frictional coefficient was 1.22. Molecular weight was 27,500±500 and α-helix content was 18.9%. The number of amino acid residues contained in 1 mole of protein of Humicola lipase was 224. Sugar and lipid were not detected. The effect of calcium ion and denaturing reagents, such as urea, sodium dodecyl sulfate and dithiothreitol, on the thermostability of Humicola lipase was examined. It was concluded that the thermostability of Humicola lipase was not influenced by protective cofactors but was attributable to the enzyme itself. Some properties of enzyme structure which were concerned with the thermostability of Humicola lipase are also discussed.  相似文献

Tannase isolated from Penicillium chrysogenum was purified 24-fold with 18.5% recovery after ammonium sulfate precipitation, DEAE-cellulose column chromatography, and Sephadex G-200 gel filtration. Optimum enzyme activity was recorded at pH 5.0 to 6.0 and at 30 to 40°C. The enzyme was stable up to 30°C and within the pH range of 4.0 to 6.5. The Km value was found to be 0.48 × 10−4 M when tannic acid was used as the substrate. Metal salts at 20 mM inhibited the enzyme to different levels.  相似文献
