PGs are important mediators of the ovulatory process and prostaglandin G/H synthase-2 (PGHS-2) is a key rate-limiting enzyme in the PG biosynthetic pathway. To determine whether PGHS-2 is regulated in equine follicles before ovulation and, if so, to characterize its time course of induction, preovulatory follicles were isolated during estrus, 0, 12, 24, 30, 33, 36, and 39 h after an ovulatory dose of hCG (n = 5 follicles/time point). Cellular extracts were obtained from preparations of follicle wall (theca interna with attached granulosa cells), isolated granulosa cells, and theca interna and were analyzed by Western blot using specific anti-PGHS antibodies. Immunohistochemistry was used to characterize the in situ localization of PGHS-2 protein in preovulatory follicles, and follicular fluid concentrations of PGE2 and PGF were determined. The results showed the induction of PGHS-2, but not PGHS-1, in equine follicles before ovulation. The PGHS-2 protein (72,000 mol wt) was undetectable 0, 12, and 24 h post-hCG, first became apparent at 30 h, and reached maximal levels 39 h after hCG treatment. The induction of follicular PGHS-2 was localized exclusively in granulosa cells, and a pronounced staining was observed in the perinuclear region. Follicular fluid concentrations of PGE2 and PGF were low and not different between 0-33 h, but levels were increased at 36 and 39 h post-hCG (P < 0.01). Thus, the time course of PGHS-2 induction in equine follicles (30 h post-hCG) is clearly distinct from those previously observed in rat (4 h post-hCG) and bovine (18 h post-hCG) preovulatory follicles. Interestingly, in all three species, the interval from PGHS-2 induction to follicular rupture is highly conserved (approximately 10 h). Therefore, the progressively delayed expression of PGHS-2 in species with longer ovulatory processes supports its role as a molecular determinant of the species-specific length of the ovulatory process.  相似文献

The profiles of insulin-like growth factor-binding proteins (IGFBPs) in follicular fluid have been characterized in a number of mammals (rats, pigs, sheep, cattle, humans) and are good indicators of follicular status. We studied the IGFBP profiles of equine serum and ovarian follicular fluid recovered at various stages of the follicular phase. The levels of IGFBPs were related to the morphology and the steroidogenic activity of the follicles. Follicular fluids were recovered by ultrasound-guided follicular aspiration. In the first experiment, the dominant follicles of 10 mares were partly punctured (aspiration of 0.5-2.2 ml of fluid) once at the early dominant stage (22-25 mm in diameter) and a second time at the preovulatory stage (PO), 34 h after induction of ovulation. Among these 10 PO follicles, 5 were classified as healthy whereas the other 5 were classified as hemorrhagic, as assessed by ultrasonic morphology and subsequent ovulation or not. In another group of mares (n = 5), the largest follicle was punctured once at the late dominant stage (33-35 mm in diameter) and then at the PO stage, 34 h after induction of ovulation. Serum was prepared at each puncture session. In the second experiment, follicular fluid was recovered from the dominant and contemporary cohort subordinate follicles (n = 5 mares). Samples were individually assayed for estradiol-17beta and progesterone content by RIA, and IGFBPs were studied by using Western ligand blotting and densitometry. Equine serum and follicular fluid displayed IGFBP at 42-44 kDa (likely corresponding to IGFBP-3), 28-32 kDa (likely corresponding to IGFBP-5), 24 kDa (likely corresponding to IGFBP-4), and 35 kDa, identified as IGFBP-2 by immunoblotting, plus one band at 120 kDa. IGFBP were clearly more abundant in serum than in fluid from healthy follicles. In the follicular fluid, 42-44-kDa IGFBP was the major binding protein, and its level was almost constant at the various physiological statuses studied. Follicular development of the dominant follicle in each mare was characterized by a decrease in intrafollicular IGFBP-2 and 28-32-kDa IGFBP levels before LH stimulation and by an increase in IGFBP-2 after LH stimulation. Follicular regression of large follicles, as well as subordinate ones, was characterized by a low level of intrafollicular estradiol-17beta and was associated with an increase in IGFBP-2, 24-kDa IGFBP, and 28-32-kDa IGFBP intrafollicular levels. Taking these results together, we have demonstrated clear correlations between the intrafollicular levels of estradiol-17beta and IGFBP-2 and 28-32-kDa IGFBP. Therefore, follicular growth and regression in the mare are associated with specific changes in IGFBP levels. These changes could be of crucial importance for follicular development in ovulation or atresia.  相似文献

Our objective was to correlate hormonal changes with the timing and onset of estrus in heifers before and after luteolysis was induced with PGF2 alpha at two stages of the estrous cycle: d 6 to 9 (early; n = 10) or d 14 to 15 (late; n = 10). Blood was collected at intervals of 2 or 12 h to quantify serum concentrations of progesterone, estradiol-17 beta, and LH while heifers were observed visually for estrus and monitored for standing activity by pressure-sensitive, radiotelemetric devices. Although the concentrations of estradiol-17 beta that were associated with the putative appearance of the first dominant follicle declined before luteolysis was induced early in the cycle, some heifers that were given PGF2 alpha were in estrus as early as 35 h. Compared with heifers treated late in the estrous cycle, heifers that were treated early in the cycle produced less progesterone before PGF2 alpha treatment and had greater peak concentrations of estradiol-17 beta at estrus. In addition, heifers that were treated early in the cycle had shorter intervals from PGF2 alpha treatment to estrus, to peak estradiol-17 beta, and to peak LH and to initiation of estrus after the peak in estradiol-17 beta than did heifers treated later in the cycle. The increase in estradiol-17 beta associated with the putative first-wave follicle of the subsequent cycle and the duration of that cycle in early cycle heifers was less than after late cycle luteolysis. Results indicated that greater concentrations of estradiol-17 beta during estrus may be related to the durations of previous cycles and less progesterone exposure before luteolysis. The onset of estrus corresponded closely to, but preceded, the preovulatory LH surge by approximately 3 h.  相似文献

Nuclear and cytoplasmic exchange assays were developed and validated to quantify receptors for estradiol-17 beta (E217 beta) and progesterone (P4) in hypothalamic and pituitary tissues of gilts before, during, and after treatment with pregnant mare's serum (PMS) and human chorionic gonadotropin (hCG). Prepubertal gilts, 5 months old, were assigned randomly to four treatments. One group of gilts received 500 IU PMS (Day 0) and were sacrificed 2 days later (2 days post-PMS); another group received 500 IU PMS on Days 0 and 2, and were sacrificed 4 days from the initial injection (4 days post-PMS). A third group of gilts received PMS (500 IU) on Days 0 and 2, 1000 IU hCG on Day 4, and were sacrificed 5 days after hCG (5 days post-hCG). Controls were given saline on Days 0, 2 and 4 and sacrificed on Day 6. In pituitary tissues, there were no significant changes in numbers of cytoplasmic E2 17 beta receptors, cytoplasmic P4 receptors, nuclear P4 receptors or nuclear E2 17 beta receptors among the control, 2 days post-PMS, 4 days post-PMS or 5 days post-hCG treatment groups. In hypothalamic tissues, no differences in cytoplasmic E2 17 beta receptors, cytoplasmic P4 receptors or nuclear P4 receptors were found among any of the treatments. Nuclear receptors for E2 17 beta in hypothalamic tissues were greater, however, in gilts 2 days post-PMS (P less than 0.05) than in controls or 5 days post-hCG gilts, but they were not different from gilts 4 days post-PMS. Follicular development and serum concentrations of E2 17 beta followed the expected patterns after PMS; only ovaries from hCG-treated pigs contained corpora lutea. Because the PMS-hCG regimen simulated the onset of puberty, it seems that gilts attain puberty without a significant change in the number of receptors for E2 17 beta and P4 in the pituitary or hypothalamus.  相似文献

Prostaglandins (PGs) are important mediators regulating uterine functions during the reproductive process. The objective of this study was to examine, in myocytes from the circular and longitudinal layers of bovine myometrium, the relative levels of mRNA and proteins corresponding to the gene expression of key enzymes (phospholipase A2; prostaglandin G/H synthase-1 [PGHS-1]; prostaglandin G/H synthase-2 [PGHS-2]; prostaglandin I2 synthase) involved in PG biosynthesis. We examined the influence of estradiol-17beta and progesterone on the expression and activity of these enzymes. Treatment of myocytes with progesterone (P4: 10 nM, 24 h) in the absence or presence of estradiol-17beta (E2: 1 nM, 72 h) suppressed PG biosynthesis by approximately 60% in both myometrial layers. No significant effect was observed after E2 treatment. The combined effect of E2 and P4 on PG accumulation was correlated with the modulation of PGHS-2 protein and mRNA levels in the two myometrial layers without affecting other enzymes of the PG cascade. Selective or nonselective inhibition of PGHS activity with CGP 28238 (PGHS-2-specific; a product from Ciba-Geigy: 6-[2, 4-difluorophenoxy]-5-methyl-sulfonylamino-1-indanone) or indomethacin (PGHS-1 and -2) reduced prostacyclin accumulation (measured as 6-keto-PGF1alpha in the culture medium) in a dose-dependent manner in the two myometrial layers. A significant inhibitory effect was obtained at a low concentration of indomethacin (1 nM, p < 0.05) compared to CGP 28238 (10 nM, p < 0. 05). In both myometrial layers, the maximal effect of indomethacin and/or CGP 28238 on PG accumulation was observed at 100 nM and represented 85% and 65% inhibition, respectively. In the presence of phorbol 12-myristate (100 nM), CGP 28238 (10 nM) significantly suppressed PGHS-2 mRNA level by 44.80 +/- 7.67% (p < 0.01) and 27.83 +/- 7.62% (p < 0.05) in the longitudinal and circular layer, respectively. In contrast, indomethacin did not have any significant effect. These data constitute the first quantitative analysis of key enzymes involved in PG biosynthesis in separated myometrial layers. Furthermore, the results provide interesting information on the CGP 28238 drug modulating both enzymatic activity and mRNA expression of PGHS-2.  相似文献

The objective of the present study was to determine the duration of elevated concentrations of progesterone necessary to induce atresia of persistent ovarian follicles. Heifers were administered 25 mg of PGF2alpha on d 6 and 7 (d 0 = d of synchronized estrus) and a norgestomet implant from d 6 to 14. Ovaries were monitored by ultrasonography, and blood samples were collected on d 3, 5, 7, 9, 11, and 12 and daily from d 14 until ovulation. On d 12, heifers received either two progesterone-releasing intravaginal devices (PRID) for 6 h (6-h; n = 5), two PRID for 24 h (24-h; n = 5), or no treatment (CON; n = 5). Blood samples were collected at 15-min intervals from h -6 to 30 (PRID insertion = h 0) and analyzed for concentrations of LH. Characteristics of LH secretion were determined for consecutive 6-h periods (Period 0 to 5). Hourly blood samples, collected from h 0 to 29, were analyzed for concentrations of 17beta-estradiol (estradiol) and progesterone. The dominant ovarian follicles present on d 7 increased in size to 15.4+/-.3 mm on d 12 ("persistent follicle"). Following removal of the PRID and norgestomet implants, atresia of persistent follicles and ovulation of new follicles were induced in one of five and in four of five heifers in the 6-h and 24-h treatments, respectively. Persistent follicles ovulated after withdrawal of norgestomet in all other heifers. Concentrations of progesterone were increased from h 1 to 7 in the 6-h and h 1 to 26 in the 24-h treatment. Frequency of LH pulses was reduced (P < .05) during Periods 1 to 2 in the 6-h and Periods 1 to 5 in the 24-h treatment relative to the CON treatment. By h 10, concentrations of estradiol in the 6-h and 24-h treatments were lower (P < . 05) than in the CON treatment. This suppression continued through h 29 in the 24-h treatment (P < .05), whereas concentrations in the 6-h treatment were intermediate to those of the CON and 24-h treatments after h 14. Suppression of pulsatile LH release and estradiol secretion was evident with 6 and 24 h of treatment with progesterone, but only the 24-h treatment induced atresia of persistent follicles in a majority of the heifers.  相似文献

The porcine antral follicles, 3-6 mm in diameter, were dissected from the ovaries of mature pigs, and then granulosa and cumulus cells were isolated from each follicle. In atretic follicles, high activity of neutral Ca2+/Mg2+-dependent endonuclease and DNA ladder formation, estimated by electrophoresis, were noted in granulosa cells but not in cumulus cells. Extremely low activity of the endonuclease and no DNA ladder formation were observed in both types of cells obtained from healthy follicles. Moreover, apoptotic cells were observed histochemically among granulosa cells only. A good correlation (r = 0.987) between the endonuclease activity of granulosa cells and the progesterone/estradiol ratio of follicular fluid in each follicle was found. These results suggest that apoptosis occurs in granulosa cells but not cumulus cells in the atretic antral follicles in pigs.  相似文献

To determine the source of circulating inhibin and estradiol-17beta during the estrous cycle in mares, the cellular localization of the inhibin alpha, betaA, and betaB subunits and aromatase in the ovary was determined by immunohistochemistry. Concentrations of immunoreactive (ir-) inhibin, estradiol-17beta, progesterone, LH, and FSH in peripheral blood were also measured during the estrous cycle in mares. Immunohistochemically, inhibin alpha subunits were localized in the granulosa cells of small and large follicles and in the theca interna cells of large follicles, whereas inhibin betaA and betaB subunits were localized in the granulosa cells and in the theca interna cells of large follicles. On the other hand, aromatase was restricted to only the granulosa cells of large follicles. Plasma ir-inhibin concentrations began to increase 9 days before ovulation; they remained high until 2 days before ovulation, after which they decreased when the LH surge was initiated. Thereafter, a further sharp rise in circulating ir-inhibin concentrations occurred during the process of ovulation, followed by a second abrupt decline. After the decline, plasma concentrations of ir-inhibin remained low during the luteal phase. Plasma estradiol-17beta concentrations followed a profile similar to that of ir-inhibin, except during ovulation, and these two hormones were positively correlated throughout the estrous cycle. Plasma FSH concentrations were inversely related to ir-inhibin and estradiol-17beta. These findings suggest that the dimeric inhibin is mainly secreted by the granulosa cells and the theca cells of large follicles; granulosa cells of small follicles may secrete inhibin alpha subunit, and estradiol-17beta is secreted by the granulosa cells of only large follicles in mares.  相似文献

The objects of this study were to monitor the development of the cumulus complex and nuclear maturation in oocytes recovered from preovulatory follicles following treatment to induce ovulation and to investigate the in vitro maturation competence of oocytes recovered from smaller nonpreovulatory follicles of varying size. All follicles > or =5 mm in pony mares were individually punctured at 0, 6, 12, 24 and 35 h after an injection of LH to induce ovulation. The recovery rates of oocytes were 64% from 55 preovulatory follicles, 22% from 32 subordinate follicles and 52% from 227 small follicles. Cumulus expansion of the preovulatory oocytes occurred at 12 h post LH treatment while the metaphase I and II components of nuclear maturation were not completed until 24 and 35 h post LH respectively. For nonpreovulatory follicles, the frequency of atresia and oocyte competence for in vitro nuclear maturation both increased with increasing follicular size.  相似文献

Follicular growth, lifespan of the corpus luteum, and antioxidant status of lactating Holsteins that experienced heat stress were monitored. Eleven multiparous cows, 60 to 110 d in milk, were maintained from 0800 to 1800 h daily in environmental chambers from d 11 to 21 of the estrous cycle. Cows were randomly assigned to a heat stress (mean dry bulb temperature peaked at 38.3 degrees C) or control treatment (mean dry bulb temperatures varied from 20.8 to 25.6 degrees C). Rectal temperature and respiration rates of heat-stressed cows were higher at 1600 h than were those of control cows. The length of the estrous cycle and the interval from estrus until luteolysis were not different between treatments. Two of 6 control cows and 1 of 5 heat-stressed cows had extended cycles (> 24 d). Heat-stressed cows had more class 1 (2 to 5 mm) follicles from d 11 to 15 of the estrous cycle. Numbers of class 2 (6 to 9 mm) and class 3 (> or = 9 mm) follicles were similar between treatments. Plasma progesterone concentrations were higher for heat-stressed cows until d 19 of the estrous cycle. Treatment did not affect concentrations of alpha-tocopherol, beta-carotene, retinol, retinyl palmitate, or total protein in plasma or concentrations of malondialdehyde in muscle. In conclusion, heat stress did not extend luteal function or the length of the estrous cycle of lactating Holstein cows but did affect follicular growth and progesterone concentrations in plasma. Heat stress did not appear to increase lipid peroxidation or decrease lipid-soluble antioxidant concentrations in blood.  相似文献

Our objective was to examine the role of progestin type on serum concentrations of progesterone (p4) and estradiol-17 beta (E2), ovarian follicular dynamics, and fertility in cattle in the presence or absence of a corpus luteum (CL) in an estrus synchronization scheme using progestin and PGF2 alpha. In Exp. 1, 325 cows and heifers were given one injection of PGF2 alpha (d 0) and then assigned randomly within parity to five treatments: to receive a second PGF2 alpha injection 14 d later (control); to receive norgestomet (NORG) for 7 d beginning on d 8, with a second PGF2 alpha injection given either 1 d (NORG + no CL) or 6 d (NORG + CL) after insertion; or to receive a P4-releasing intravaginal device (PRID) in lieu of norgestomet at comparable times. Presence or absence of a CL was based on concentrations of serum P4 on d 14. Pregnancy rates after insemination were greater (P < .01) with luteal treatments than with nonluteal treatments. Embryonal survival between two stages of pregnancy was 87.6%. In Exp. 2, ovarian structures in 50 cows were examined daily using ultrasonography and the same five treatments. Diameter of the ovulatory follicle was greater (P < .05) with the nonluteal treatments (NORG and PRID + no CL) than with the control and luteal treatments (PRID and NORG + CL). Replacement of the dominant follicle during progestin treatment was altered by treatment (luteal status) and stage of the estrous cycle. Fertility was not enhanced by exogenous progestins when a CL was present. In the absence of a CL, progestin (P4 less than NORG at the doses used) reduced fertility by increasing E2 and the diameter of the ovulatory follicle and decreasing turnover of dominant follicles.  相似文献

Vascular endothelial growth factor (VEGF) is a cytokine that induces angiogenesis. Angiogenesis is a prominent histologic component of the luteinization process. Luteinization is also characterized by granulosa cell progesterone secretion in response to the luteinizing hormone (LH) surge. Local VEGF production in human pre-ovulatory follicles, induced by LH, was postulated to be a luteinization mediator in women. To investigate this hypothesis, serum and fluid from the dominant follicle of 31 healthy regularly cycling multiparous women undergoing laparoscopic sterilization were obtained. VEGF was measured by enzyme-linked immunosorbent assay, and LH and progesterone were measured by radioimmunoassay. Follicle aspiration was performed at a median of 13 days from the last menstrual period (range 11-17 days). The median pre-ovulatory follicle diameter was 16 mm (range 11-23 mm). Follicle fluid VEGF concentrations (mean 6900 pg/ml, range 1200-17 100 pg/ml) were correlated positively with follicle fluid progesterone concentrations (mean 10 176 nmol/l, range 636-66780 nmol/l, r=0.62, P=0.002). This correlation was even tighter (r=0.87, P < 0.0001) when only samples from the 22 women in the earliest stages of follicle luteinization were considered. In these women serum LH concentrations were also correlated with follicle fluid VEGF concentrations (r=0.51, P=0.02). Our findings demonstrate the close dynamic relationship between VEGF production and early luteinization in human follicles during normal non-stimulated cycles.  相似文献

Granulosa cells from small (less than 2mm) antral porcine follicles were grown in culture to study the effects of various hormones on growth, morphology and progesterone secretion. Culture medium 199D + 4% serum was found to be most suitable since it maintained a fairly constant cell population. Estradiol (1mug/ml) and human FSH stimulated cell growth. LH and FSH stimulated progesterone secretion and induced morphological changes associated with luteinization. Estradiol (0.1 mug/ml) inhibited progesterone secretion by granulosa cells.  相似文献

It is well documented that prostaglandin F2 alpha (PGF2 alpha) inhibits progesterone production in luteal cells, but its mode of action is uncertain. It has recently been suggested that PGF2 alpha acts by activating the calcium and phospholipid-dependent protein kinase, protein kinase C (PKC). This hypothesis has been tested by comparing the site and mode of action of PGF2 alpha, a PGF2 alpha analogue (cloprostenol) and the PKC activator phorbol myristate acetate (4 beta PMA) in human granulosa-lutein cells. PGF2 alpha and cloprostenol exerted similar concentration-dependent inhibitory actions on gonadotrophin-stimulated cyclic AMP (cAMP) accumulation and progesterone production by human granulosa-lutein cells. The similarity in the actions of PGF2 alpha and cloprostenol in human granulosa-lutein cells suggests that they can be used interchangeably to study the role of PGF2 alpha in the regulation of steroidogenesis in the human ovary. Gonadotrophin-stimulated cAMP accumulation and progesterone production was also concentration-dependently inhibited by 4 beta PMA. In addition, cloprostenol and 4 beta PMA also inhibited dibutyryl cAMP-stimulated progesterone production, suggesting that these compounds inhibit LH action at sites before and after the generation of cAMP. The pre-cAMP site of action can be localised to the stimulatory G-protein (Gs) as both compounds inhibited cholera toxin-stimulated cAMP accumulation without affecting forskolin-stimulated cAMP accumulation. The post cAMP site of action can be localised to actions on cholesterol side chain cleavage enzyme, as both cloprostenol and 4 beta PMA inhibited 22R hydroxycholesterol-supported progesterone production without affecting pregnenolone-supported progesterone production. The finding that cloprostenol and 4 beta PMA interact with the steroidogenic cascade in a similar manner is indicative of a shared common mediator of their actions in human granulosa-lutein cells, i.e. PKC. The inhibitory actions of PGF2 alpha and 4 beta PMA on hLH-stimulated progesterone production were abolished in the presence of the PKC inhibitor, staurosporine. In addition, in PKC-depleted cells (achieved by exposure to 4 beta PMA for 20 h) the inhibitory actions of PGF2 alpha and 4 beta PMA were abolished. These results support the hypothesis that the inhibitory actions of PGF2 alpha are mediated by PKC in human granulosa-lutein cells.  相似文献

Follicular fluids were obtained from 180 follicles of 15 women undergoing follicular aspiration for in-vitro fertilization. Follicular development was induced by a combination of buserelin acetate and human menopausal gonadotrophin. Endothelin-1 (ET-1) concentrations in human follicular fluids were determined by specific radioimmunoassay. ET-1-like immunoreactivity (ET-1-LI) ranged from 338 to 928 pg/ml. ET-1-LI concentrations in follicular fluids obtained from immature (< 15 mm) follicles were significantly higher than those from mature (15-25 mm) and post-mature (> or = 25 mm) follicles. No correlation was found between the concentration of ET-1-LI, on the one hand, and that of oestradiol, progesterone, testosterone, prolactin, luteinizing hormone, insulin-like growth factor-I, prostaglandin E2 or platelet activating factor on the other, in follicular fluids. However, a significant positive correlation was observed between ET-1-LI concentration and follicle stimulating hormone and IGF-II concentrations, respectively. These data suggest that the high concentration of ET-1 found in follicular fluids may play some physiological role in follicular development.  相似文献

Cattle with low (subluteal) levels of plasma progesterone develop a persistent dominant follicle; plasma estradiol and LH pulse frequency are elevated, and fertility subsequent to the ovulation of a prolonged dominant follicle is compromised. The hypotheses were 1) that prolonged dominant follicles produce more estradiol because they have theca and granulosa cells with an enhanced capacity to produce androgen and estradiol, respectively, and 2) that these changes in steroidogenic capacity are paralleled by concomitant changes in mRNA for the appropriate steroidogenic enzymes. Prolonged dominant follicles were induced by treating Holstein heifers with exogenous progesterone via an intravaginal controlled internal drug-release device (CIDR) from Day 14 to 28 of the cycle. Prolonged dominant follicles were collected just before (CIDRb, Day 28; n=4) or 24 h after (CIDRa, Day 29; n=4) CIDR removal, and their steroidogenic capacity was compared to that of growing, control dominant follicles obtained just before (CONTb, n=4) or 24 h after (CONTa, n=4) a luteolytic injection of prostaglandin F2alpha during the late luteal phase. After natural luteolysis, CIDR heifers maintained subluteal concentrations of progesterone (1-2 ng/ml) and had higher estradiol and LH pulse frequency than control heifers, as expected. In CIDR heifers, prolonged dominant follicles were present on the ovary for a longer time, reached a larger diameter, and had more granulosa cells and a larger mass of theca than dominant follicles from control heifers (p < 0.05). Concentrations of steroids in follicular fluid, estradiol secretion by granulosa cells in vitro, and levels of mRNA for steroidogenic enzymes in theca and granulosa cells provided no evidence for greater capacity of theca and granulosa cells of CIDR follicles to produce androgen and estradiol. In fact, follicular fluid estradiol and mRNA for P450 aromatase were higher after luteolysis than before in control animals (p < 0.05) but not after CIDR removal in treated animals. Therefore, the data do not support the hypotheses. Rather it is suggested that prolonged dominant follicles produce more estradiol because they have more granulosa cells and a larger mass of theca than control dominant follicles. In contrast, progesterone concentrations in the follicular fluid increased in CIDRa relative to CIDRb follicles (p < 0.05), a change that did not occur in control follicles; and granulosa cells from CIDRa follicles secreted more progesterone than granulosa cells from any other group. The increased capacity of CIDRa follicles to secrete progesterone suggests premature luteinization, which could contribute to decreased fertility in cattle that ovulate a prolonged dominant follicle.  相似文献

The cell cycle characteristics of mouse cumulus granulosa cells were determined before, during and following their expansion and mucification in vivo and in vitro. Cumulus-oocyte complexes (COC) were recovered from ovarian follicles or oviducts of prepubertal mice previously injected with pregnant mare serum gonadotrophin (PMSG) or a mixture of PMSG and human chorionic gonadotrophin (PMSG+hCG) to synchronize follicle differentiation and ovulation. Cell cycle parameters were determined by monitoring DNA content of cumulus cell nuclei, collected under rigorously controlled conditions, by flow cytometry. The proportion of cumulus cells in three cell cycle-related populations (G0/G1; S; G2/M) was calculated before and after exposure to various experimental conditions in vivo or in vitro. About 30% of cumulus cells recovered from undifferentiated (compact) COC isolated 43-45 h after PMSG injections were in S phase and 63% were in G0/G1 (2C DNA content). Less than 10% of the cells were in the G2/M population. Cell cycle profiles of cumulus cells recovered from mucified COC (oviducal) after PMSG+hCG-induced ovulation varied markedly from those collected before hCG injection and were characterized by the relative absence of S-phase cells and an increased proportion of cells in G0/G1. Cell cycle profiles of cumulus cells collected from mucified COC recovered from mouse ovarian follicles before ovulation (9-10 h after hCG) were also characterized by loss of S-phase cells and an increased G0/G1 population. Results suggest that changes in cell cycle parameters in vivo are primarily mediated in response to physiological changes that occur in the intrafollicular environment initiated by the ovulatory stimulus. A similar lack of S-phase cells was observed in mucified cumulus cells collected 24 h after exposure in vitro of compact COC to dibutyryl cyclic adenosine monophosphate (DBcAMP), follicle-stimulating hormone or epidermal growth factor (EGF). Additionally, the proportion of cumulus cells in G2/M was enhanced in COC exposed to DBcAMP, suggesting that cell division was inhibited under these conditions. Thus, both the G1-->S-phase and G2-->M-phase transitions in the cell cycle appear to be amenable to physiological regulation. Time course studies revealed dose-dependent changes in morphology occurred within 6 h of exposure in vitro of COC to EGF or DBcAMP. Results suggest that the disappearance of the S-phase population is a consequence of a decline in the number of cells beginning DNA synthesis and exit of cells from the S phase following completion of DNA synthesis. Furthermore, loss of proliferative activity in cumulus cells appears to be closely associated with COC expansion and mucification, whether induced under physiological conditions in vivo or in response to a range of hormonal stimuli in vitro. The observations indicate that several signal-transducing pathways mediate changes in cell cycle parameters during cumulus cell differentiation.  相似文献

We report the establishment and preliminary characterization of a stable steroidogenic granulosa cell line, JC-410. This cell line was obtained by spontaneous immortalization of a primary culture of porcine granulosa cells. Cultured JC-410 cells produced less progesterone than granulosa cells in primary culture. Progesterone synthesis by JC-410 cells was approximately 10% and 1% of the amount produced by granulosa cells from small and medium sized follicles, respectively. Although FSH and LH did not change progesterone levels in cultured JC-410 cells, forskolin and cholera toxin induced a 2.6- and 2.75-fold increase, respectively, versus control. The JC-410 cells responded to 0.1, 1 and 5 mM cAMP with an increase in progesterone synthesis of 2.5-, 28- and 49-fold versus control, respectively, after a 24 h incubation. No detectable levels of estradiol-17beta were found in JC-410 cells after 48 h in culture. However, addition of 0.01, 0.1 and 1 microM androstenedione elevated the levels of estradiol-17beta to 0.028, 0.3 and 1.21 pg/microg protein, respectively. The level of expression of 3betaHSD, aromatase and P450scc genes in JC-410 cells is of similar magnitude to the level of expression in granulosa cells in primary culture. The JC410 cells have been maintained in culture for more than one year during which their population doubled over 100 times. We conclude that JC-410 is a stable cell line that lost responsiveness to the gonadotropins during the process of immortalization, but retained its steroid biosynthetic capability and the expression of key steroidogenic genes. These characteristics may reflect features of cells arrested in an early stage of granulosa cell differentiation.  相似文献
