Summary The conductiveness of tropical savanna soils in Nigeria to the damping-off of tomato seedlings induced bySclerotium rolfsii was studied using soil from different localities in the savanna zones of the country. The results showed that soils low in percentage clay content were more conducive to the disease than those with high clay contents. Statistical analysis also revealed a high negative correlation (r=−0.91) between disease conduciveness indices and percentage clay contents of the various soils, indicating that as clay content of soil increased, disease decreased.  相似文献

Summary The hyphal tip ofSclerotium rolfsii was examined after fixation by freeze substitution. The Spitzenkörper consisted of a dense mass of apical vesicles and microvesicles surrounding a vesicle-free zone. Linear arrangements of microvesicles were occasionally observed within the Spitzenkörper. Abundant microfilaments were seen within the Spitzenkörper region, often in close association with apical vesicles and microvesicles. Microtubules passed through the Spitzenkörper and terminated at the plasmalemma at the extreme hyphal apex. Filasomes were mostly observed within the apical region and were in close proximity to the plasmalemma. Rough ER, mitochondria, microtubules, and vacuoles were abundant in the subapical region and were usually oriented parallel to the long axis of the hypha. Ribosomes were aligned on the outer surfaces of mitochondria. Golgi body equivalents were observed throughout the subapical region and appeared as inflated cisternae of varying shapes and electron opacities. Populations of microorganisms from soil treated with guanidine thiocyanate, guanylurea sulfate, thiourea, or furfural were compared with those of untreated soil. The materials effected quantitative and/or qualitative changes in composition of the soil microflora depending on the compound used. Guanidine thiocyanate (Gt) significantly (p0.05) increased total fungal populations relative to populations of other treatments. Populations of Penicillium purpurogenum were markedly higher in Gt-treated soil. Gt also increased total bacterial populations, and was the only compound that increased actinomycete populations. The relative percentage of Trichoderma harzianum was significantly higher in soil treated with thiourea than in the other treatments. Furfural increased the percentage of P. purpurogenum with respect to total fungi, and was as effective as guanylurea sulfate in increasing chitinolytic bacteria and those in the Pseudomonas cepacia-group. Thiourea most effectively promoted proliferation of coryneform bacteria. Chitinolytic fungi increased synergistically when Gt and guanylurea sulfate were applied in combination.  相似文献

Screenhouse studies were conducted to investigate the effects of Fusarium oxysporum f. sp. glycines and Sclerotium rolfsii on the pathogenicity of Meloidogyne incognita race 2 on soybean and the influence of the nematode on wilt incidence and growth of soybean. The interaction of each fungus with the nematode resulted in reduced shoot and root growth. Final nematode population was also reduced with concomitant inoculation of nematode and fungus or inoculation of fungus before nematode. While M. incognita suppressed wilt incidence in two nematode-susceptible cultivars of soybean (TGX 1485-2D and TGX 1440-IE), it had limited effect on wilt incidence in the nematode resistant cultivar of soybean (TGX 1448-2E). When F. oxysporumwas inoculated with the nematode, the mean number of nematodes that penetrated soybean roots decreased by 75% in TGX 1448-2E, 68% in TGX 1485-1D and 65% in TGX 1440-1E. Similarly when the soil was treated with S. rolfsii, the number decreased by 78% in TGX 1448-2E, 77% in TGX 1485-1D and 68% in TGX 1440-1E. The nematode did not develop beyond second-stage juvenile in TGX-1448-2E.  相似文献

Tree root systems may improve soil fertility through carbon inputs, uptake of leachable nutrients and maintenance of soil biomass, but can at the same time reduce crop yields by competition for water and nutrients. Quantitative information about the positive and negative effects of tree roots and their changes in space and time are necessary for the optimization of agroforestry associations. An alley cropping experiment was layed out as a randomized complete block design on a Plinthic Lixisol/Ferralic Cambisol with Gliricidia sepium hedgerows at 5 m distance, including a sole cropping control. The development of root systems was monitored by sequential soil coring (eight samplings) during one year, with maize and groundnut as crops. Additional information is presented from a single sampling for rice during the foregoing year. Pronounced fluctuations of live root length density indicated an important variability in the nutrient and water uptake capacity of the vegetation. At low total root length density, the hedgerows affected the root development in the agroforestry plots directly by the presence of their root systems. At high root length density, they affected root development mainly by improving crop root growth and influencing the composition of the spontaneous vegetation. The root length density of the hedgerows was too low to compete with the crops for soil resources. The hedgerows tended to increase root length densities in the subsoil when few roots were present, thus possibly reducing the risk of nutrient leaching. However, the length density of the perennial root systems decreased during the cropping season, presumably as an effect of repeated pruning, and attained minimum values almost at the same time as the crops. Trees with denser root systems which are less frequently pruned may be more efficient in achieving closer nutrient cycles, though at the cost of higher root competition with crops.  相似文献

【目的】解析不同连作年限花魔芋软腐病株、健株根域的丛枝菌根真菌(arbuscular mycorrhizal fungi,AMF)群落多样性。【方法】使用AMF 18S SSU rRNA基因特异引物AMV4.5NF/AMDGR对正茬及连作2年和3年的软腐病株、健株魔芋根系和根际土壤DNA扩增建库,通过高通量测序和生物信息学分析探究魔芋软腐病与其根域AMF群落多样性的关系。【结果】魔芋根系具有明显的AMF菌丝、泡囊和丛枝等结构。在相同连作年限条件下,健株根系AMF总侵染率、侵染强度和孢子密度均显著高于病株(P<0.05);在不同连作年限条件下,病株根系AMF总侵染率和侵染强度随连作年限延长而降低。从所有样品中共鉴定到9属53种AMF,其中有49个已知种和4个新种。球囊霉属(Glomus)和类球囊霉属(Claroideoglomus)是AMF群落的优势属,其AMF种分别占总AMF种数的41.5%和26.4%;丰度最高的Paraglomus sp.VTX00308是所有样品的共有种。连作、软腐病及二者的交互作用显著影响根系AMF群落的Shannon指数和Simpson指数及根际土壤AMF的Chao1指数(P<0.05)。通过丰度差异分析发现6个在连作软腐病发生后丰度差异显著的AMF种(P<0.05);NMDS分析表明,不同连作年限的魔芋软腐病株与健株之间的根域AMF菌种组成、相对丰度和群落结构存在差异。相关性分析表明,软腐病发病率和病情指数与魔芋根系和根际土壤AMF的Shannon指数、根系AMF的Chao1和Simpson指数以及AMF总侵染率、侵染强度和孢子密度极显著负相关(P<0.01)。【结论】比对健株,连作魔芋软腐病株根际土壤AMF孢子密度以及根系AMF侵染率、种数和多样性均降低,其群落结构显著改变。  相似文献

毛鸡骨草是治疗肝病的重要中草药之一,而连作障碍致其产量和品质下降。从主产区广西玉林分别采集正茬、重茬和连作三年的毛鸡骨草根际土壤样品,以种植前水稻土壤为对照,采用Biolog GN2 平板碳源分析技术,研究连作条件下根际土壤微生物群落功能多样性变化特征,以揭示其连作障碍的原因。结果表明,连作导致根际土壤微生物功能多样性显著降低,微生物利用各类碳源的能力下降。主成分分析(PCA)表明,不同种植年限的毛鸡骨草根际土壤微生物群落结构之间存在显著差异,且均与种植前对照明显不同,而同一种植年限不同生长期土壤微生物碳代谢群落结构虽有差异,但并不显著。相关分析表明,其产量与微生物功能多样性指数(如Shannon,Simpson,Mclntosh)以及平均吸光值(AWCD)值正相关,而与第一主成分(PC1)呈显著负相关,说明连作条件下其产量的降低在一定程度上与土壤微生物功能多样性的降低和群落结构的改变有关。  相似文献

From comparison of the alignments of the internally transcribed spacers (ITS) of ribosomal DNA from Ganoderma associated with oil palm basal stem rot (BSR) and other Ganoderma species, two specific primer pairs were selected to provide a specific DNA amplification of pathogenic Ganoderma in oil palm. Each primer pair produced a single PCR product of about 450 bp (for primer pair IT1–IT2) and 334 bp (for primer pair IT1–IT3) when oil palm Ganoderma DNA was used. No PCR amplification product was observed when other Ganoderma species DNA was used in PCR amplification with these primer pairs. Three specific restriction enzyme sites were identified in the ITS and intergenic spacer (IGS1) regions. The restriction enzymes MluI, SacI and HinfI were used to digest the ITS-PCR product and restriction enzymes TfiI, ScaI and HincII were used to digest the IGS1-PCR product. Of the three restriction enzymes used in each rDNA region, MluI specifically digested the ITS regions, and TfiI specifically digested the IGS1 region of oil palm Ganoderma. Analysis of the published ITS nucleotide sequences of 31 Ganoderma species showed that the MluI restriction site was not present in other Ganoderma species. The use of both specific primers and restriction enzyme analysis can be applied as a standard protocol to identify pathogenic Ganoderma in oil palm. In this study, the use of specific primers and PCR-RFLP analyses of the rDNA gave consistent results for the characterisation of pathogenic Ganoderma, and indicated that Ganoderma strains associated with BSR disease in oil palms belong to a single species.  相似文献

物种多样性(或同一物种遗传多样性)减少和氮富集都是影响陆地生态系统进程的主要因素,它们之间的交互作用是否对土壤微生物群落产生显著影响已成为研究者关心的主要科学问题。研究羊草基因型数目(1、2、4三种基因型数目组合)和氮添加(无氮添加、低氮添加和高氮添加3种水平)对土壤微生物群落的总磷脂脂肪酸(PLFA,Phospholipid Fatty Acid)含量、细菌PLFA生物标记含量、真菌PLFA生物标记含量、真菌/细菌比、以及基于每个PLFA生物标记相对含量百分比所得微生物群落的Shannon-Wiener多样性指数和Simpson优势度指数的影响。结果表明:氮添加对细菌PLFA生物标记含量,以及土壤微生物PLFA生物标记的Shannon-Wiener多样性指数和Simpson优势度指数具有显著影响(P0.05);基因型数目对所测变量无显著影响(P0.05),但基因型数目和氮添加的交互作用对细菌PLFA生物标记含量和真菌/细菌比具有显著影响(P0.05)。研究结果为全球变化背景下氮沉降及重要物种种群数量减少对土壤微生物群落的影响提供了科学数据,为合理解释群落动态变化提供了数据支持。  相似文献

The toxicity of the pyrethroid insecticide deltamethrin to a cereal aphid and a coccinellid beetle predator was assessed. Deltamethrin gave effective aphid control in winter wheat at dose-rates of 6.25, 3.13 and 1.56g a.i./ha. The direct exposure of adultCoccinella septempunctata L. (Coleoptera: Coccinellidae) to spray drops was estimated at a range of positions in a cereal crop canopy from volumetric analysis of fluorescent tracer deposits. These measurements were used to calculate exposure to deltamethrin at the three experimental dose-rates. Observations of coccinellid beetle distribution through a cereal crop canopy permitted a realistic range of direct contact doses tobe calculated and the toxic effects of these levels of exposure to be predicted from laboratory dose-response data. Estimated beetle mortalities from direct exposure were 19, 8 and 3% at the three experimental dose-rates.In situbioassays with adultC. septempunctata which exposed beetles continuously to deltamethrin residues on flag leaves, resulted in 100, 94 and 39% mortality respectively at these dose-rates during the 10 days after spray application. Additionalin situ bioassays exposed beetles to deltamethrin residues on flag leaves for 24 h and then transferred surviving beetles to the soil under the cereal crop canopy for a further 9 days. This resulted in 89, 69 and 29% beetle mortality respectively at the three dose-rates. Mortality predictions combining both direct contact and residual exposure were made for the three dose-rates.to determine the maximum impact of summer sprays of deltamethrin on adult coccinellid populations in cereals. These worst case predictions suggested that a reduction in dose-rate by as much as three quarters of the recommended application rate in UK cereals may be necessary to preserve approximately 60% of adultC. septempunctata in the crop over the 10 days after a deltamethrin spray application. The methodology described may be appropriate for estimating selective dose-rates for key enemies in a range of crops.  相似文献

Summary Rhodobacter capsulatus genes homologous to Klebsiella pneumoniae nifE, nifN and nifX were identified by DNA sequence analysis of a 4282 bp fragment of nif region A. Four open reading frames coding for a 51188 (NifE), a 49459 (NifN), a 17459 (NifX) and a 17472 (ORF4) dalton protein were detected. A typical NifA activated consensus promoter and two imperfect putative NifA binding sites were located in the 377 bp sequence in front of the nifE coding region. Comparison of the deduced amino acid sequences of R. capsulatus NifE and NifN revealed homologies not only to analogous gene products of other organisms but also to the and subunits of the nitrogenase iron-molybdenum protein. In addition, the R. capsulatus nifE and nifN proteins shared considerable homology with each other. The map position of nifX downstream of nifEN corresponded in R. capsulatus and K. pneumoniae and the deduced molecular weights of both proteins were nearly identical. Nevertheless, R. capsulatus NifX was more related to the C-terminal end of NifY from K. pneumoniae than to NifX. A small domain of approximately 33 amino acid residues showing the highest degree of homology between NifY and NifX was also present in all nifB proteins analyzed so far. This homology indicated an evolutionary relationship of nifX, nifY and nifB and also suggested that NifX and NifY might play a role in maturation and/or stability of the iron-molybdenum cofactor. The open reading rame (ORF4) downstream of nifX in R. capsulatus is also present in Azotobacter vinelandii but not in K. pneumoniae. Interposon-induced insertion and deletion mutants proved that nifE and nifN were necessary for nitrogen fixation in R. capsulatus. In contrast, no essential role could be demonstrated for nifX and ORF4 whereas at least one gene downstream of ORF4 appeared to be important for nitrogen fixation.  相似文献

Targeted gene replacement via homologous recombination (HR) is a conventional approach for the analysis of gene function. However, this event is rare in Beauveria bassiana, which hampers efficient functional analysis in this widely used entomopathogenic fungus. To improve homologous recombination frequency in B. bassiana, we investigated the effect of the ratio of homologous sequence to non-homologous sequence (HS/NHS) in gene disruption cassette upon the HR frequency by two gene loci BbNtl and BbThi, using the herpes simplex virus thymidine kinase as a negative selectable marker against ectopic transformants. Our data revealed that an increase of the ratio of HS/NHS achieved by either extending homologous sequence or decreasing non-homologous sequence could improve HR frequency in B. bassiana. We determined empirically that (1) at least 700 bp of homology to both sides of a target gene was needed to get a reasonable number of disruptants, e.g., 6.7‰ to 13.3‰ in B. bassiana. (2) When the ratio of HS/NHS was above 0.8, an acceptable HR frequency could be achieved for gene replacement in B. bassiana, while when the ratio was below 0.3, few gene disrupted mutants were obtained.  相似文献

The effects of female sex pheromone concentration and time of day on behavioral responses of male brown-banded cockroaches were monitored with a two-choice olfactometer. The duration of each behavior or behavioral event in a deterministic response sequence and the probabilities of behavioral transitions were analyzed. Pheromone concentration had a significant effect on all behaviors in the response sequence. The time of testing, relative to the entrainment photoperiodic regime, significantly affected behavioral events early in the sequence but not later-occuring behaviors. Although the probability of males choosing the pheromone rather than the solvent control was a function of the pheromone concentration, it was independent of the time of testing. By affecting the probability of behavioral transitions, the pheromone concentration and time of testing determined the number of insects exhibiting specific behaviors in the response sequence. The behavioral response sequence can be characterized by probabilities of behavioral transitions along with latencies of early behavioral acts.  相似文献

We report the nucleotide sequence of the core histone gene cluster from the Cnidarian Acropora formosa. This is the first histone gene cluster to be sequenced from a diploblastic organism and the predicted amino acid sequences most resemble those of sea urchin equivalents. Each of the Cnidarian histone genes has two conserved regions 3 of the coding sequences and these closely resemble those of the metazoan a-class histone genes. In A. formosa the core histone genes are arranged as opposed (H3/H4 and H2A/H2B) pairs, a pattern common to the nondeuterostome metazoa, and tandem repetition is the predominant pattern of organization in the Cnidarian. With the recent identification of several classes of homeobox genes in Cnidarians these features clearly align the Cnidaria with triploblastic metazoans, supporting a monophyletic origin of the metazoa.  相似文献

Acetobacter methanolicus MB58 can grow on methanol. Since this substrate exhibits to be energy deficient there must be a chance to oxidize methanol to CO2 merely for purpose of energy generation. For the assimilation of methanol the FBP variant of the RuMP pathway is used. Hence methanol can be oxidized cyclically via 6-phosphogluconate. Since Acetobacter methanolicus MB58 possesses all enzymes for a linear oxidation via formate the question arises which of both sequences is responsible for generation of the energy required. In order to clarify this the linear sequence was blocked by inhibiting the formate dehydrogenase with hypophosphite and by mutagenesis inducing mutants defective in formaldehyde or formate dehydrogenase. It has been shown that the linear dissimilatory sequence is indispensable for methylotrophic growth. Although the cyclic oxidation of formaldehyde to CO2 has not been influenced by hypophosphite and with mutants both the wild type and the formaldehyde dehydrogenase defect mutants cannot grown on methanol. The cyclic oxidation of formaldehyde does not seem to be coupled to a sufficient energy generation, probably it operates only detoxifying and provides reducing equivalents for syntheses. Uptake kinetics of arsenate were determined in arsenate tolerant and non-tolerant clones of the grassDeschampsia cespitosa under differing root phosphorus status to investigate the mechanism controlling the suppression of arsenate influx observed in tolerant clones. Influx was always lower in tolerants compared to non-tolerants. Short term influx of arsenate by the high affinity uptake system in both tolerant clones was relatively insensitive to root phosphorus status. This was in contrast to the literature where the regulation of the phosphate (arsenate) uptake system is normally much more responsive to plant phosphorus status. The low affinity uptake system in both tolerant and non-tolerant clones, unlike the high affinity uptake system, was more closely regulated by root phosphate status and was repressed to a much greater degree under increasing root phosphorus levels than the high affinity system.  相似文献

植物蒸腾速率与叶性状关系的环境响应,对探索植物碳水代谢关系和叶性状构建模式之间耦合的生理生态学机制具有重要的意义。选择甘肃省秦王川国家湿地公园的盐沼湿地为实验地,按研究区域滨藜(Atriplex patens)植株密度设置Ⅰ(16—21株/m~2)、Ⅱ(9—15株/m~2)、Ⅲ(9株/m~2)3个梯度,采用标准化主轴估计(standardized major axis estimation,SMA)方法,以叶脉密度和中脉直径分别表示叶脉性状,研究了种群密度影响下滨藜蒸腾速率与叶脉性状的关系。结果表明:随种群密度减小,滨藜的中脉直径、株高、盖度、光合有效辐射(PAR)、叶面积(LA)均逐渐降低,而叶脉密度、叶干重、蒸腾速率(T_r)和净光合速率(P_n)逐渐增加。在高密度(Ⅰ)和低密度(Ⅲ)中,滨藜的蒸腾速率和叶脉密度之间存在极显著正相关关系(P0.01),与中脉直径之间存在显著的负相关关系(P0.05);在中密度(Ⅱ)中,滨藜的蒸腾速率与叶脉密度呈显著正相关关系(P0.05),与中脉直径不存在相关关系(P0.05)。在小密度样地,蒸腾速率(T_r)较小,滨藜采取减小叶脉密度、增大中脉直径的策略,即在阴生环境中滨藜叶片需要大的中脉直径来支撑,同时较小的叶脉密度亦可满足其蒸腾需求,体现了密度制约下湿地植物的生物量分配格局和资源利用对策。  相似文献
