Koumiss is a type of famous fermented mare milk and considered an important nutritious beverage in central Asian countries. However, the production of koumiss cannot meet public demand in the market due to availability of mare milk. In the present study, 52 lactic acid bacteria and 20 yeast strains from traditional homemade Kazakhstan koumiss were isolated and identified. The isolates were used in a trial that included fermented cow milk, and the flavor profiles, color, and taste to determine their contribution in the co-fermentation of cow milk. Based on the sensory evaluation, KZLAB13 and KZY10 strains were selected as the best cofermentation combinations. The optimal fermentation conditions were confirmed as the ratio of the starter culture 2.4:1.6 % (vol/vol) KZLAB13 strain to KZY10 strain and a temperature of 36°C for 16 h using response surface methodology. After evaluating the quality of the optimized cow-milk koumiss compared with the Kazakhstan koumiss, results suggested that cow milk fermented by these 2 strains possessed a promising taste, flavor, and physicochemical and rheological properties. Altogether, our results showed that cow milk fermented with a combination of KZLAB13 and KZY10 strains can simulate the taste, flavor, and quality of traditional koumiss. Our study provided a novel alternative to mare-milk koumiss and could be used in dairy programs to fulfill the needs of people.  相似文献

Set yoghurt was produced from caprine milk (A), 70% caprine-30% bovine (B) and 50% caprine–50% bovine milk (C) mixtures, and stored for 14 days at ±4°C. Two methods of fortification, namely ultrafiltration (UF) and the addition of bovine skim milk powder (SMP), were applied to the milk mixtures. Some chemical, physical, microbiological and sensory properties of the six samples were analysed on the 1st, 7th and 14th days of storage. The effects of milk type, concentration method and storage period on the physicochemical and microbiological properties of the samples were investigated statistically.  相似文献

Individual milk samples from cows given different amounts of supplemental zinc were analysed for somatic cell count (SCC) and milk composition. The concentrations of immunoglobulins (IgA, IgG1, IgG2 and IgM), lactoferrin (LF), bovine serum albumin as well as total fat, total protein and lactose were analysed. The growth of starter cultures in milk samples was studied by the conductance technique used as a model for processability of milk. The concentrations of IgA, IgG2, IgM and LF as well as conductance measurements were found to be markers of processability in addition to SCC. Relatively low SCC in milk affected the growth of starter culture with a delayed start of the fermentation process up to 2–4 h. Supplemental zinc influenced the concentration of IgA, IgG2, IgM and LF while no effect on SCC was observed. The lag phase for growth of starter cultures was prolonged in milk from cows given additional zinc.  相似文献

The effect of goats’ milk lactoperoxidase (LP) system on the activity of commercially available mesophilic cheese starter cultures was investigated. The growth and acid production of the starter cultures were measured at 2 h intervals for 8 h in goats’ milk kept at 30°C. Most of the starter cultures examined were found to be sensitive to the LP system, but varied in their susceptibility to inhibition. The activity of the mixed starter cultures CHN11, CHN22, CHN19, DCC240 and Flora Danica Normal was strongly inhibited by the LP system. However, the mixed starter culture LL 50C showed resistance to the LP system. The single strain culture Lactococcus lactis subsp. lactis NCDO 605 was inhibited by the LP system. However, the cultures Lactococcus lactis subsp. diacetylactis NCDO 176 and Leuconostoc mesenteroides subsp. cremoris ATCC 33313 were insensitive to the LP system. The results of this study indicate the need for routine screening of starter cultures for resistance to the LP system before using them for cheesemaking from goats’ milk preserved by the LP system.  相似文献

One goal of animal breeding is to increase the economic output through increased production, improved milk quality, and cow health. The objective of this study was to evaluate genetic progress in relation to milk composition, processability, and yield as a correlated response to selection for the Swedish breeding objective. Dairy cows with high genetic merit, classified as elite dairy cows, of the Swedish Red and Swedish Holstein breeds were used. Milk samples were collected on the farm level in winter and summer from a research herd at Nötcenter Viken, a bovine research farm in Sweden. Comparisons were made with milk from a Swedish Red herd, a Swedish Holstein herd, and a Swedish dairy processor in the same geographical area. Protein, lipid, and carbohydrate profiles as well as minerals were analyzed, and technological properties, including rennet-induced gelation characteristics, lipid oxidation, total antioxidant capacity, and fat globule size, were determined. Higher yields were found for elite cows for components of the protein, lipid, and carbohydrate profiles as well as for minerals, implying genetic progress in relation to milk yield; however, the content of some milk components (e.g., lipid and whey protein contents) had decreased on average. Milk from the elite cows had good gelation characteristics, but was more susceptible to lipid autooxidation and had a lower total antioxidant capacity. These results demonstrate that milk composition and processing characteristics could be used to adjust breeding practices to optimize the quality and stability of milk and dairy products.  相似文献

Bovine and caprine milk gels were made by GDL-acidification and yogurt fermentation (using 'ropy' and 'non-ropy' starter cultures). The respective gelation processes were monitored rheologically using dynamic oscillatory testing. The bovine fermented systems produced gel structures with about half the strength of the equivalent chemically acidified gels. The fermented caprine milk systems produced gel structures some eight to 10 times weaker than the equivalent acidified systems. In all cases the caprine systems were weaker than the bovine gels despite having higher protein contents. In all cases the 'ropy' milk systems followed somewhat different gelation patterns and formed weaker gels than the equivalent 'non-ropy' and GDL-acidified systems. These data suggested that the starter culture material (biomass and extracellular polysaccharides) may have interfered with the protein–protein interactions during yogurt fermentation. This produced weaker gel structures, possibly by a modified gelation mechanism .  相似文献

Koumiss is a slightly alcoholic fermented milk beverage, originally obtained by using natural mix starters (lactic acid bacteria and yeast). Seven Lactobacillus strains from lyophilized koumiss were isolated and identified as L. salivarius, L. buchneri and L. plantarum. The process of lactic acid fermentation caused by koumiss strains was faster (9–13 h) than that with other lactobacilli. The conversion ratio of glucose to lactic acid ranged from 47% to 79% and was strain dependent. All strains were resistant to low pH. Three of the strains isolated were viable during prolonged cold storage in fermented milk (3 weeks at 4°C).  相似文献

The effects of the starter culture biomass material and its exopolysaccharides (EPS) on the structure of bovine and caprine acid gels were investigated. Inactivated cell material and/or extracellular polysaccharide, which had been separated from yogurt starter cultures, were added to both types of milk. The milk systems were acidified using glucono-δ-lactone (GDL) and the gelation processes were monitored using dynamic (oscillatory) rheological testing. The addition of cell material had no effect on the bovine acid gel, whereas the addition of polysaccharide produced a weaker gel structure. The addition of cell material (> 5 × 108 cells/mL) and polysaccharide (0.35% w/v) weakened the caprine acid gel system. The treated gels were some three to eight times weaker than the untreated samples, suggesting that the starter culture metabolites and especially the EPS may act by interfering with the gel structures, particularly in the cases of milk systems such as caprine yogurt or low protein content bovine yogurt.  相似文献

Kefir starter culture was used for the production of beverages, and some chemical and microbiological parameter changes were determined during 24 h of fermentation and then after 2, 7, 14, 21 and 28 days of storage at 4 °C. Three different substrates (milk, whey and soya milk) were used as fermentation media for the kefir starter culture. After the fermentation, the carbohydrate content and pH decreased, but the dry matter and fat content of the beverages were not significantly different from their substrates. During storage, lactic acid bacteria in the beverages decreased, while yeasts increased and carbohydrate, ethanol and pH changed significantly in the three beverages.  相似文献

The objective of this study was to evaluate the effect of lactoferrin addition to milk replacer varying in crude protein (CP) on dry matter intake, growth, and days medicated. Thirty-four Holstein heifer calves were assigned to 4 treatments in a 2 × 2 factorial arrangement of treatments in a randomized complete block design. Treatments were as follows: 562 g daily of a nonmedicated conventional milk replacer (20% CP:20% fat) feeding regimen with or without 1 g of supplemental bovine lactoferrin (n = 9 for both treatments) or a nonmedicated intensified milk replacer feeding regimen (28% CP:20% fat) fed on a metabolizable energy basis (0.2 Mcal/kg BW0.75) from d 2 to 9, and at 0.27 Mcal/kg BW0.75 from d 10 to 42 with or without 1g supplemental bovine lactoferrin (n = 8 for both treatments). Calves were fed pelleted starter (25% CP) in 227.5-g increments beginning on d 2 and had free access to water. Calves remained on the study for 14 d postweaning. Dry matter intake was determined daily. Growth measurements were taken weekly. Blood samples were taken twice weekly for determination of blood urea N. On d 10 of life, calves were subjected to a xylose challenge. Calves on conventional treatments ate more starter preweaning, during weaning, and postweaning. Preweaning, intensively fed calves had higher dry matter intakes. Weights of intensified-fed calves were greater at weaning. Intensified milk replacer-fed calves had greater average daily gain preweaning and overall and higher gain:feed ratios preweaning, but conventionally fed calves had higher gain:feed ratios during weaning. Intensified milk replacer-fed calves had greater hip heights during weaning and postweaning and greater heart girths preweaning, weaning, and postweaning. Days medicated were greater preweaning and overall for intensified-fed calves. There were no differences among treatments for xylose absorption. Calves on conventional treatments had increased blood urea nitrogen concentrations preweaning. There were no effects of lactoferrin on any experimental variable. Intensified milk replacer-fed calves consumed less starter but had higher average daily gains overall and larger frames and greater BW than conventionally fed calves. An intensified milk replacer feeding regimen promotes faster growth during the preweaning period when compared with calves fed conventional treatments, but supplemental bovine lactoferrin was not beneficial under these experimental conditions.  相似文献

Koumiss is a popular dairy product in many lands, traditionally prepared from mare milk with spontaneous fermentation. Mare milk and its fermented derivates are more expensive than cow milk and its fermented derivates, and the possibility exists for producers and dealers to adulterate equine products with bovine items. In this work, we described the development of a triplex real-time PCR based on species-specific TaqMan probes for identification of bovine and equine DNA in milks and dairy products. In addition, a novel designed endogenous control was simultaneously amplified to eliminate possible false negatives. With this methodology, bovine and equine DNA were specifically identified by employing developed primers and probes. The limits of detection of this method were 0.001 ng for cow milk, yogurt, and mare milk, and 0.005 ng for sour soup and koumiss, respectively. In addition, the triplex real-time PCR assay for authentication of animal-derived products was effectively validated using binary DNA and milk mixtures, exhibiting well in terms of specificity, sensitivity, and reproducibility. In short, the triplex PCR assay was verified to be a time-saving and money-saving technique for the identification of bovine and equine DNA in milks and dairy products.  相似文献

以鲜牛乳和浓缩牛奶蛋白为主要原料,以酸度和感官评价为评价指标,通过单因素试验和正交试验确定高蛋白饮用型纯益生菌发酵乳的最佳发酵工艺条件。结果表明,高蛋白饮用型纯益生菌发酵乳的最佳发酵工艺条件为浓缩牛奶蛋白添加量6.0%,发酵剂接种量2.0%、发酵温度43 ℃、二次无菌均质压力4 MPa。在此优化条件下,制得的高蛋白饮用型纯益生菌发酵乳感官评分达到89分,酸度129.1 °T,口感清爽顺滑,组织状态良好,理化及微生物指标均符合国家相关标准。  相似文献

研究了西藏地区牦牛乳的理化指标和微生物指标。从西藏3个不同地区共取48份牦牛乳样品.通过理化和微生物指标的检测及其统计分析,研究发现牦牛乳的营养成分为:脂肪7.14%(体积分数)、粗蛋白5.06%(质量分数)、干物质18.45%(质量分数)、乳糖5.00%(质量分数)和灰分0.81%(质量分数),并且不同地区各成分的质量分数也不同。牦牛乳的微生物检测结果为(对数值):细菌5.63mL^-1,芽孢菌1.76mL^-1,乳酸菌5.19mL,真菌3.03mL^-1,肠道菌3.27mL^-1,大肠杆菌2.09mL^-1和金黄色葡萄球菌3.52mL^-1,并且存在明显的地区差异性。  相似文献

The effects of spray-dried animal plasma in milk replacer without or with the addition of additives containing fructooligosaccharides and spray-dried serum on health, growth, and intake of Holstein calves was measured in two 56-d experiments. In experiment 1, 120 calves were fed milk replacer containing 0 or 20% of crude protein as spray-dried bovine plasma for 42 d and 30 to 60 g/d of additives containing whey protein concentrate or bovine serum for the first 15 d. Commercial calf starter was available from d 29, and water was available at all times. In experiment 2, 120 calves were fed milk replacer containing 0 or 16% of crude protein as spray-dried bovine plasma with 0 or 30 to 60 g/d of additive containing bovine serum for the first 15 d. Additive containing bovine serum also contained fructooligosaccharides, whey, and vitamin/mineral premix. In experiment 1, calves fed additive containing bovine serum tended to have fewer days with diarrhea, lower use of electrolytes, and improved BW gain from d 29 to 56. The addition of spray-dried bovine plasma to milk replacer did not influence any parameter measured. In experiment 2, calves fed additive containing bovine serum or milk replacer containing spray-dried bovine plasma had lower mortality (4.4 vs. 20%) and tended to have improved fecal scores and fewer days with scours. Antibiotic use was lower when calves were fed the additive. Indices of enteric health (incidence of scours and treatment with antibiotics and electrolytes) were improved when plasma was added to milk replacer throughout the milk feeding period or as an additive during the first 15 d of the milk feeding period, when calves were most susceptible to enteric pathogens. The addition of spray-dried animal plasma to milk replacer or the addition of additive containing spray-dried bovine serum and oligosaccharides may be a useful adjunct to animal management during periods of stress.  相似文献

Two batches of Tenerife cheese were produced from pasteurised goats’ milk, one with a commercial starter and another with an autochthonous starter consisting of three selected strains of lactic acid bacteria isolated from artisanal Tenerife cheese. Influence of starter on the characteristics of the cheeses was evaluated. Owing to the composition of the starters, lactobacilli and leuconostocs varied significantly between both cheeses. However, the type of starter did not significantly affect the physicochemical and proteolytic properties of the cheeses. In general, cheeses made with autochthonous starter received higher scores for sensory attributes, especially for aroma, than those made with commercial starter.  相似文献

Feed management is one of the principal levers by which the production and composition of milk by dairy cows can be modulated in the short term. The response of milk yield and milk composition to variations in either energy or protein supplies is well known. However, in practice, dietary supplies of energy and protein vary simultaneously, and their interaction is still not well understood. The objective of this trial was to determine whether energy and protein interacted in their effects on milk production and milk composition and whether the response to changes in the diets depended on the parity and potential production of cows. From the results, a model was built to predict the response of milk yield and milk composition to simultaneous variations in energy and protein supplies relative to requirements of cows. Nine treatments, defined by their energy and protein supplies, were applied to 48 cows divided into 4 homogeneous groups (primiparous or multiparous × high or low milk potential) over three 4-wk periods. The control treatment was calculated to cover the predicted requirements of the group of cows in the middle of the trial and was applied to each cow. The other 8 treatments corresponded to fixed supplies of energy and protein, higher or lower than those of the control treatment. The results highlighted a significant energy × protein interaction not only on milk yield but also on protein content and yield. The response of milk yield to energy supply was zero with a negative protein balance and increased with protein supply equal to or higher than requirements. The response of milk yield to changes in the diet was greater for cows with high production potential than for those with low production potential, and the response of milk protein content was higher for primiparous cows than for multiparous cows. The model for the response of milk yield, protein yield, and protein content obtained in this trial made it possible to predict more accurately the variations in production and composition of milk relative to the potential of the cow because of changes in diet composition. In addition, the interaction obtained was in line with a response corresponding to the more limiting of 2 factors: energy or protein.  相似文献

This experiment was conducted to investigate the effect of starter cultures on the physicochemical properties, texture, and consumer preferences of soft white cheese (SWC) made from camel (Camelus dromedarius) milk. The experiment was laid out in a completely randomized design with 5 treatments [starter cultures; i.e., 1 thermophilic (STI-12), 2 blended (RST-743 and XPL-2), and 2 mesophilic (R-707 and CHN-22) cultures]. Starter cultures STI-12 and RST-743 were inoculated at 37°C, whereas XPL-2, R-707, and CHN-22 were inoculated at 30°C. Camel milk inoculated using STI-12 and RST-743 cultures resulted in faster acidification than XPL-2, R-707, and CHN-22 cultures. Camel milk SWC made using STI-12 and CHN-22 cultures gave lower pH (4.54) and titratable acidity (0.59), respectively, whereas R-707 culture resulted in high cheese yield (13.44 g/100 g). In addition, high fat (20.91 g/100 g), protein (17.49 g/100 g), total solids (43.44 g/100 g), and ash (2.40 g/100 g) contents were recorded for SWC made from camel milk made using RST-743 culture. Instrumental analysis of cheese texture revealed differences in resistance to deformation in which camel milk SWC made using RST-743 culture gave higher firmness (3.20 N) and brittleness (3.12 N). However, no significant difference was observed among camel milk SWC adhesiveness made using different starter cultures. Consumer preference for appearance, aroma, taste, and overall acceptances of SWC were affected by inoculation of starter cultures. Considering curd firmness, cheese yield, compositional quality, and textures using STI-12, RST-743, and R-707, these cultures were found to be better for the manufacture of camel milk SWC.  相似文献

Data from 60 multiparous Holstein cows were used in a 12-wk continuous design feeding trial. Cows were allocated to 1 of 4 experimental treatments (T1 to T4). In T1 and T2, the total mixed ration (TMR) contained either corn silage from the genetically modified (GM) variety Chardon Liberty Link, which is tolerant to the herbicide glufosinate ammonium, or its near isogenic nonGM counterpart, whereas the TMR used in T3 and T4 contained corn silage from the commercially available nonGM varieties Fabius and Antares, respectively. The objectives of the study were to determine if the inserted gene produced a marked effect on chemical composition, nutritive value, feed intake, and milk production, and to determine if transgenic DNA and the protein expressed by the inserted gene could be detected in bovine milk. The nutritive value, fermentation characteristics, mineral content, and amino acid composition of all 4 silages were similar. There were no significant treatment effects on milk yield, milk composition, and yield of milk constituents, and the dry matter (DM) intake of the GM variety was not significantly different from the 2 commercial varieties. However, although the DM intake noted for the nonGM near-isogenic variety was similar to the commercial varieties, it was significantly lower when compared with the GM variety. Polymerase chain reaction analyses of milk samples collected at wk 1, 6, and 12 of the study showed that none of the 90 milk samples tested positive, above a detection limit of 2.5 ng of total genomic DNA/mL of milk, for either tDNA (event T25) or the single-copy endogenous Zea mays gene, alcohol dehydrogenase. Using ELISA assays, the protein expressed by the T25 gene was not detected in milk.  相似文献

Donkey milk is characterized by low contents of total solids, fat, and caseins, especially κ-casein, which results in formation of a very weak gel upon renneting. The objective of this study was to evaluate the effect of fortification of donkey milk with microbial transglutaminase (MTGase) for cheesemaking in relation to different enzyme addition protocols (patterns, PAT). Four independent trials were performed using MTGase (5.0 U/g of milk protein) according to the following experimental patterns: control (no MTGase addition); MTGase addition (40°C) 15 min before starter inoculation (PAT1); addition of MTGase to milk simultaneously with starter culture (40°C) (PAT2); and MTGase addition simultaneously with rennet (42°C) in acidified milk (pH 6.3) (PAT3). Evolution of pH during acidification, cheesemaking parameters, and proximal composition and color of cheese at 24 h were recorded. The protein fractions of cheese and whey were investigated by urea-PAGE and sodium dodecyl sulfate-PAGE. Addition of MTGase had no significant effect on moisture, protein, fat, or cheese yield. The addition of MTGase with rennet (PAT3) improved curd firmness compared with the control. Among the different patterns of MTGase addition, PAT3 reduced gel formation time, time between rennet addition and cheese molding, and weight loss of cheese at 24 h. The PAT3 treatment also resulted in the lowest lightness and highest yellowness color values of the cheese. Sodium dodecyl sulfate-PAGE of cheeses revealed that MTGase modified the protein pattern in the high-molecular-weight zone (range 37–75 kDa) compared with the control. Of the MTGase protocols, PAT3 showed better casein retention in cheese, as confirmed by the lanes of α- and β-caseins in the electropherogram of the whey, which was subtler for this protocol. In conclusion, MTGase may be used in cheese production from donkey milk to improve curd firmness; MTGase should be added simultaneously with the rennet.  相似文献
