桂花提取物Osthin的免疫功能研究

摘    要:目的 通过对桂花提取物Osthin的免疫功能进行研究,促进其在食品、保健品等领域的开发。方法 将SPF级雌性昆明小鼠160只随机分为对照组和低、中、高剂量组(50、150、450 mg/kg bw)。灌胃Osthin 4周,分别进行脏器/体重比值测定、抗体生成细胞实验、脾淋巴细胞转化实验、迟发型变态反应实验、小鼠碳廓清实验、血清溶血素测定和脾细胞的Treg/Th17比值分析。结果 与植物油对照组相比,一定剂量的桂花提取物Osthin对小鼠的胸腺系数(F = 8.440,P<0.01)和脾脏系数(F = 4.710,P<0.01)有增强作用;对脾淋巴细胞增殖能力有促进作用(F = 5.991,P<0.01)以及可增强迟发型变态反应(F = 4.213,P<0.05);提高小鼠的吞噬指数a(F = 3.901,P<0.05);使抗体生成细胞数增加(F = 5.192,P<0.01)。同时,随着桂花提取物Osthin浓度的升高,小鼠脾细胞的Th17细胞比例下降(F = 5.203,P<0.05)、Treg细胞比例上升(F = 7.369,P<0.05),Treg/Th17比值也随之增大(F = 8.377,P<0.01)。结论 桂花提取物Osthin具有较好的免疫增强作用,可能通过影响Treg/Th17的平衡调节细胞免疫功能。

Abstract:Objective To study the immune function of Osmanthus fragrans extract Osthin so as to promote its development in the fields of food and health products. Methods A total of 160 SPF female Kunming mice were randomly divided into control group and low, medium and high dose groups(50, 150, 450 mg/kg·bw). Osthin was gavaged for 4 weeks, and the organ/body weight ratio assay, antibody-producing cell assay, splenic lymphocyte transformation assay, delayed metaplasia assay, mouse carbon contour assay, serum hemolysin assay and Treg/Th17 ratio analysis of splenocytes were performed. Results Compared with the vegetable oil control group, a certain dose of Osmanthus fragrans extract Osthin had an enhanced effect on the thymus coefficient(F=8.440, P<0.01) and spleen coefficient(F=4.710, P<0.01) of mice. It also boosted splenic lymphocyte proliferation(F=5.991, P<0.01), enhance delayed-type allergy(F=4.213, P<0.05), increase mouse phagocytic index a(F=3.901, P<0.05), and increase the number of antibody-producing cells(F=5.192, P<0.01). At the same time, with the increase of the concentration of Osthin of Osmanthus fragrans extract, the proportion of Th17 cells in mouse splenocytes decreased( F=5.203, P<0.05), the proportion of Treg cells increased(F=7.369, P<0.05), and the ratio of Treg/Th17 also increased(F=8.377,P <0.01). Conclusion Osthin, an extract of Osmanthus fragrans, has a good immune-enhancing effect and may regulate cellular immune function by affecting the balance of Treg/Th17.
