补充NO前体L-Arg对耐力训练大鼠疲劳状况的影响

摘    要:目的 :(1)观察耐力运动和补充外源性NO前体L -Arg(左旋精氨酸 )对机体各组织NO生成能力的影响 ;(2 )观察补充外源性NO前体L -Arg对机体运动疲劳状况的影响。方法 :将 30只SD大鼠随机分成安静组、运动对照组和运动用药组 ,运动组大鼠进行 4周跑台耐力训练和一次力竭运动 ,其中运动用药组每天给予 4 0mg每kg体重L -Arg灌胃 ,测定了力竭运动后即刻大鼠组织NO生成能力以及血清生化指标的变化。结果 :(1)所有运动组大鼠血清、肌肉、心肌、肝脏NOS活性均有升高趋势 ,尤其以血清、心肌和肝脏的升高具有显著性意义 ;(2 )运动用药组大鼠血清血尿素 (BUrea)、谷丙转氨酶 (ALT)、乳酸脱氢酶 (LDH)均较运动对照组有升高趋势。结论 :(1)补充外源性L -Arg对机体整体生成NO的能力 ,尤其是心脏等器官生成NO的能力可能有促进作用 ;(2 )补充外源性NO前体L -Arg会增加耐力运动中体内自由基损伤作用需进一步研究

Abstract:Objective:The present study aimed at investigating the effects of L-arginine,a nitric oxide (NO) precursor,on NO produced from tissues and fatigue in endurance treadmill trained rats.Methods:30 SD rats were randomly divided into 3 groups:sedentary group,exercise control group and exercise L-Arg group. The rats in both exercise groups were given 4 weeks of endurance training on treadmill at speed of 34m/min,60m/day,6 days/week,then followed an exhausted running.At the meantime,the rats in exercise L-Arg group were administed L-Arg 40mg/kg body weight by lavage every day.The NO and nitric oxide synthase(NOS)in serum,vastus lateralis,myocardium and liver,and some fatigue indexes,such as creatine kinase(CK),blood urea(BUrea),alanine aminotransferase(ALT),lactate dehydrogenase(LDH)in serum were determined immediately after the exhausted running.Results:(1)The NOS in serum,myocardium and liver in both exercise groups increased significantly,compared with sedentary group,but no difference was observed between two exercise groups,except for in myocardium.(2)The serum CK,BUrea,ALT and LDH in Exercise L-Arg Group were higher than those in exercise control proup,but no statistical significance.Conclusion:It needed to study further that supplementation of L-Arg could promote the production of NO in many tissues and increased the tissue damage induced by free radical during endurance training.
