摘    要:目的:构建一种新型的具有更低基础表达的Tet-on调控系统,以用于毒性蛋白及 RNA病毒的真核细胞调控合成.方法:对目前常用的Tet-on系统中的四环素调控蛋白(rtTA、tTS)的表达途径进行改进,使得在非诱导条件下调控蛋白的基础诱导活性进一步降低.结果:相比原有的Tet-on系统,新构建的Tet-on系统非诱导状态下的基础表达值下降了一半以上.结论:成功构建基础表达值更低的Tet-on调控系统,为真核调控表达毒性蛋白或合成 RNA病毒打下了基础.

Abstract:Objective:To construct a new-type Tet-on gene expression system with low basal expression for the regulable expression of toxic protein and synthesis of +RNA virus.Methods:The way of expression of the regulatory proteins(rtTA & tTS) in the commonly used Tet-on system was improved for further reduction of basal induced activity under non-induced condition.Results:The non-induced expression level of the newly constructed Tet-on system was reduced by more than one half compared with the original system.Conclus...
