蒺藜总皂苷对大鼠缺血再灌注损伤心肌ICAM-1表达及中性粒细胞浸润的影响

摘    要:目的观察蒺藜总皂苷对缺血再灌注损伤心肌炎症因子ICAM-1表达及中性粒细胞浸润的影响。方法可逆性冠脉左前降支结扎缺血45min再灌注2h复制MI/R模型,SD雄性大鼠32只,随机分成假手术组、模型组、GSTT大剂量组、GSTT小剂量组,每组8只。HE染色观察心肌中性粒细胞浸润,测定心肌MPO活性,免疫组化测定心肌组织ICAM-1的表达。结果HE染色镜下观察模型组心肌中性粒细胞浸润严重,而GSTT大、小剂量组则较轻;GSTT大、小剂量组MPO活性显著降低(P〈0.01或0.05);缺血45min再灌注2h心肌组织ICAM-1无明显表达。结论GSTT对MI/R炎症损害有保护作用,可降低IVIPO活性,减轻中性粒细胞浸润。

Effects of GSTT on ICAM-1 expression and the infiltration of neutrophils in rats with myocardial ischemia/reperfusion injury

Abstract:Objective To investigate the effects of GSTT on expression of inflammatory molecule ICAM-1 and the infiltration of neutrophils in rats after myocardial ischemia/reperfusion (MI/R) injury. Methods A model of myocardial ischemia 45min followed by 2h reperfusion was made by reversibly ligating coronary left descending artery. 32 Sprague Dawley (SD) rats were divided randomly to 4 groups, ie: sham-operated group, model group, large and dosage of GSTT. HE staining was used to observe infiltrating neutophils. The activity of MPO was measured, too. Immunohistochemistry was used to determine ICAM-1 expression in myocardium. Results Light microscopy showed that the infiltration of neutrophils in both large and low dosage of GSTT group was less serious than that in model group. Both large and low dosage of GSTT significantly decreased MPO activity (P<0.01, P<0.05). ICAM-1 expression was not detected in myocardium after ischemia 45min followed by 2h reperfusion. Conclusion GSTT could protect myocardium from inflammatory injury during MI/R. It had the actions of decreasing MPO activity and ameliorating neutrophil infiltration.
