楸树内生真菌抗癌性能研究

摘    要:在新抗癌药物筛选过程中获得一种具有明显抗癌能力的楸树内生真菌,命名为FSN002.采用形态学和分子生物学方法判断该内生真菌为短枝木霉.FSN002发酵产物能够有效抑制肝癌、肺癌等多种癌症细胞,同时对正常肝细胞几乎没有明显的细胞毒性.FSN002发酵产物的有效抗癌成分为一种次生代谢物小分子楸灵素,有望开发成一种拥有自主知识产权的高效低毒抗癌药物.

Abstract:An entophytic fungus named FSN002 was isolated from the inner bark of Juglans mandshurica Maxim.Based on the morphology identification and internal transcribed spacer(ITS) sequence,it is clear that this fungus belongs to the Deuteromycotina,Hyphomycetes,Moniliales,Trichoderma longibrachiatum.The fermentation broth of FSN002 shows high anti-tumor activity against liver cancer,lung cancer,while the inhibiting effect against hepatic cell(strain HL-7702) is only one quarter of that against hepatocellular carcinoma cell(HepG2) at its IC50 concentration.Therefore,the fermentation of FSN002 provides a possible way to produce anticancer drug with higher efficiency and lower toxicity.
