芦荟多糖对糖尿病足溃疡大鼠创面愈合的影响

摘    要:目的研究芦荟多糖对糖尿病足溃疡大鼠创面愈合的影响及相关机制。方法采用腹腔注射1%链脲佐菌素(STZ)60 mg·kg-1的方法构建糖尿病大鼠模型,另选取15只大鼠为空白组,腹腔注射等量柠檬酸-柠檬酸钠缓冲液;糖尿病大鼠模型构建成功后制备糖尿病足溃疡创面模型。将糖尿病足溃疡大鼠随机分为模型组、实验组和对照组,各15只。实验组于创面外敷浸湿10%芦荟多糖溶液的纺纱,对照组外涂重组牛碱性成纤维细胞生长因子,空白组和模型组外敷浸湿生理盐水的纺纱,每日换药2次,直至创面完全愈合。观察各组糖尿病大鼠创面愈合情况,用酶联免疫吸附法测定糖尿病足溃疡大鼠血清碱性成纤维因子(bFGF)、血小板衍生生长因子-BB(PDGF-BB)及白细胞介素-1(IL-1)、肿瘤坏死因子-α(TNF-α)水平;用蛋白质印迹(WB)法检测创面组织Wnt/β-catenin蛋白表达。结果治疗后14 d,空白组、模型组、实验组和对照组糖尿病足溃疡大鼠的创面愈合率分别为(95.46±2.14)%,(65.15±2.48)%,(89.45±3.15)%,(85.46±2.75)%,与模型组比较,其他各组差异均有统计学意义(P<0.05)。与空白组相比,模型组大鼠血清bFGF、PDGF-BB水平降低,而血清IL-1、TNF-α水平升高(均P<0.05);与模型组比较,实验组和对照组血清bFGF、PDGF-BB水平升高,而血清IL-1、TNF-α水平降低(均P<0.05)。治疗后14 d,空白组、模型组、实验组和对照组大鼠创面组织R-脊椎蛋白3(Rspo3)蛋白表达量分别为1.15±0.21,0.45±0.14,0.89±0.12,0.96±0.10;糖原合成激酶-3β(GSK-3β)蛋白表达量分别为0.24±0.03,1.16±0.34,0.49±0.05,0.52±0.12;β-连环素(β-catenin)蛋白表达量分别为0.99±0.10,0.36±0.05,0.67±0.04,0.82±0.06,与模型组比较,差异均有统计学意义(均P<0.05)。结论芦荟多糖可降低糖尿病足溃疡大鼠血清炎性因子水平,升高生长因子水平,同时可调控Wnt/β-catenin通路蛋白表达进而促进大鼠创面愈合。

Abstract:Objective To investigate the effect of aloe polysaccharide on wound healing of rats with diabetic foot ulcer and its mechanism.Methods The diabetic rat model was established by intraperitoneal injection of 1%streptozocin(STZ)60 mg·kg-1 in SD rats.Another 15 rats were selected as blank group,and the same amount of citric acid-sodium citrate buffer was injected intraperitoneally.The diabetic foot ulcer wound model was prepared after the successful establishment of diabetic model.Rats were randomly divided into model group,test group and control group,with 15 rats in each group.Test group was treated with 10%aloe polysaccharide solution for external application,while control group was coated with recombinant bovine basic fibroblast growth factor.Blank group and model group were treated with wet saline spinners for 2 times a day until the wound healed completely.The wound healing of diabetic rats in each group was observed;serum basic fibroblast growth factor(bFGF),platelet-derived growth factor-BB(PDGF-BB),interleukin-1(IL-1),tumor necrosis factor-α(TNF-α)were measured by enzyme-linked immunosorbent assay(ELISA)kit;Wnt/β-catenin protein expression in wound tissue of diabetic rats were measured by Western blot method.Results 14 d after treatment,the wound healing rates in blank group,model group,test group and control group were(95.46±2.14)%,(65.15±2.48)%,(89.45±3.15)%,(85.46±2.75)%;compared with model group,there were significant differences in other groups(P<0.05).Compared with blank group,the levels of bFGF and PDGF-BB in model group were decreased,and the levels of IL-1 and TNF-αwere increased(all P<0.05).Compared with model group,the levels of b FGF and PDGF-BB in test group and control group were increased,and the levels of IL-1 and TNF-αwere decreased(all P<0.05).The expression of R-spine protein 3(Rspo3)in wound tissues of rats in blank group,model group,test group and control group were 1.15±0.21,0.45±0.14,0.89±0.12,0.96±0.10;and the expression of glycogen synthetic kinase-3β(GSK-3β)were 0.24±0.03,1.16±0.34,0.49±0.05,0.52±0.12;andβ-catenin(β-catenin)were 0.99±0.10,0.36±0.05,0.67±0.04 and 0.82±0.06,the difference was statistically significant compared with model group(all P<0.05).Conclusion Aloe polysaccharide could decrease the levels of serum inflammatory factors and increase the levels of growth factors in diabetic foot ulcer rats,and regulate the expression of Wnt/beta-catenin pathway protein to promote wound healing in rats.
