缺氧诱导因子-1α和血管内皮生长因子在腺样囊性癌中的表达和相关性研究

摘    要:目的研究缺氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)、微血管密度(MVD)在腺样囊性癌(ACC)中的表达和相关性,并探讨它们在腺样囊性癌侵袭和转移生物学行为的机理。方法应用免疫组化SP法,检测35例腺样囊性癌、13例多形性腺瘤和10例正常唾液腺组织中HIF-1α、VEGF的表达和MVD计数。结果HIF-1α、VEGF、MVD计数在正常唾液腺组织、多形性腺瘤、ACC中依次增加,组间差异具有统计学意义(P〈0.05) ACC中MVD计数随着HIF-1α、VEGF表达的增强而升高(P〈0.05) ACC中HIF-1α、VEGF的表达呈正相关,r=0.69,(P〈0.001)。结论HIF-1α、VEGF、MVD在ACC中的表达有相关性,HIF-1α通过影响肿瘤血管的生成,使ACC具有相应的侵袭和转移的生物学行为,HIF-1α、VEGF、MVD可以作为判断ACC侵袭、转移及预后的重要指标。

Abstract:Objective To investigate the expression of hypoxia inducible fector-1α(HIF-1α)、vasculare endothelial growth factor (VEGF)、microvessel density(MVD) and their relations in adenoid cystic carcinoma (ACC),and explore the invasion and metastasis biological behaviour by HIF-1α,VEGF,MVD in ACC.Methods SP immunochemistry was applied to detect the expression of HIF-1α、VEGF、MVD counted number in 35 cases of ACC,13 cases of pleomorphic adenoma,10 cases of normal salivary gland tissue.Results Positive expression rate of HIF-1α,VEGF,MVD enhanced in normal salivary gland tissue,pleomorphic adenoma and ACC stepply(P0.05);the expression levels of MVD tended to depend on that of HIF-1α、VEGF in ACC(P0.05);expression of HIF-1α positively correlated expression of VEGF,the coefficient of correlation was 0.69,P0.05.Conclusions The expression of HIF-1α、VEGF 、MVD was coherent in ACC,HIF-1α makes corresponding invasion and metastasis biological behaviour in ACC by tumor vessel formn,HIF-1α、VEGF 、 MVD can be considered as the significant index of invasion and metastasis and prognosis.
