HPLC法快速测定雷公藤制剂中雷公藤红素的含量

摘    要:目的:建立雷公藤制剂中雷公藤红素快速提取方法,用HPLC法测定其含量,并比较市售5个不同生产厂商6个批次的雷公藤制剂中雷公藤红素含量。方法:样品以甲醇溶解后超声提取直接上样检测,采用Aglient Zorbax EclipseXDB-C8柱(150 mm×4.6 mm,5μm);流动相:甲醇-1%醋酸(83:17);流速:1 ml·min-1;检测波长:425 nm。结果:雷公藤红素保留时间在6.7 min左右,线性范围为6.33~202.60μg·ml-1(r=0.999 6),平均加样回收率99.33%,RSD为2.04%(n=6);雷公藤制剂中雷公藤红素含量每片在17.67~400.62μg。结论:本法简便快捷,准确可靠,适合于雷公藤制剂中雷公藤红素快速定量分析。不同生产厂商、不同批次的雷公藤制剂中雷公藤红素含量存在较大差异。

Abstract:Objective: To establish a rapid extraction method and determine the content of tripterine in six batches tfipterygium preparations from five different manufacturers by HPLC. Method: The samples were dissolved with methanol by ultrasonic extraction and an external standard method was developed by HPLC with Zorbax Eclipse XDB-C8 column (150 mm ×4. 6 mm,5μm). The mobile phase consisted of methanol and 1% glacial acetic acid solution (83: 17, v/v). The detection wavelength was 425 nm and the flow rate was 1.0 ml·min^-1. Result: Retention time of tripteriue was 6. 7 min. The linear range for tripterine was 6. 33-202. 60μg·ml-1 ( r = 0. 999 6) with average overall recovery of 99. 33%. The content of tripterine in tripterygium preparations was between 17.67-400. 62μg per tablet. Conclusion : The method was fast and accurate, which could be used for the quantitative analysis of tfipterine in tripteryglum preparations. The content of tripterine in different batch tripterygium preparations from different manufacturers has significant difference.
