硫化氢对多柔比星致心肌细胞凋亡的保护作用

摘    要:目的:研究硫化氢对多柔比星引起的心肌细胞凋亡的保护作用及其机制.方法:以硫氢化钠(NaHS)作为供体,先用不同浓度NaHS处理心肌细胞,再加入多柔比星(DOX)致其凋亡.应用胎盘蓝染色测量细胞的存活率;应用Annexin V和PI双染流式术检测其凋亡情况;应用比色法检测凋亡相关蛋白CASPASE-3和CASPASE-9的活性.结果:分别以DOX(5μmol·L-1)+ NaHS(50μmol·L-1)、DOX(5μmol·L-1)+ NaHS(100 μmol·L-1)、DOX(5μmol·L-1)+ NaHS( 200 μmol·L-1)浓度处理的各组细胞存活率均较单用DOX组(5μmol·L-1)增高;以DOX(5μmol ·L -1)+ NaHS(200 μmol·L-1)处理的细胞凋亡率较单用DOX组(5μmol·L-1)降低,以DOX(5μmol·L-1)+ NaHS(200μmol·L-1)处理的细胞的caspase-3和caspase-9活性较单用DOX组(5μmoloL-1)降低,各项指标差异均有统计学意义(P<0.05).结论:硫化氢可以保护DOX引起的心肌细胞损伤,抑制DOX引起的心肌细胞凋亡.

Abstract:Objective:To investigate the protective effects of hydrogen sulfide on doxorubicin - induced cardiomyocyte apoptosis. Method:With sodium hydrosulfide(NaHS) as the donor,cardiorayocytes were pretreated by NaHS with different concentrations before adding doxorubicin(DOX) to induce apoptosis.The viability of cardiomyocytes was measured with trypan blue staining.Apoptosis was assessed by Annexin V-FITC/propidium iodide double staining and the apoptosis-related protein CASPASE-3 and CASPASE-9 were determined by colorimetric method.Result:Compared with the group treated by DOX(5μmol·L~(-1) )alone,the survival rates of the cells in the groups treated by DOX(5μmol·L~(-1) )plus NaHS(50μmol·L~(-1)),DOX(5μmol·L~(-1) )plus NaHS(100μmol·L~(-1) ) and DOX (5μmol·L~(-1) )plus NaHS(200μmol·L~(-1) ) were all improved(P 0.05).The apoptosis ratio of the cells treated by DOX(5μmol·L~(-1)) plus NaHS(200μmol·L~(-1)) was significandy decreased(P 0.05),and the activity of CASPASE-3 and CASPASE-9 was also decreased(P 0.05).Conclusion:Hydragen sulfide can reduce doxorubicin cardiotoxicity by decreasing the cardiomyocyte apoptosis.
