p38抑制剂对辐射损伤小鼠中免疫细胞的作用

摘    要:目的:探讨利用p38抑制剂SB203580(SB)抑制p38通路对6Gy受照小鼠免疫细胞辐射损伤的作用。方法:取雄性C57BL/6小鼠30只,随机分为对照组、照射组和SB组,每组10只。SB组和照射组小鼠接受以6Gy137Csγ射线的全身照射。SB组小鼠照射24h后腹腔注射SB(15mg/kg),每2d1次,共给药5次,其余2组小鼠腹腔注射对照溶液。小鼠受照10d后处死,取外周血计数,流式细胞仪检测CD4、CD8、B220表达,取骨髓细胞测定ROS水平。结果:照射组外周血白细胞(WBC)、CD8、B220细胞比例较对照组明显下降,骨髓ROS水平升高(P<0.01)。SB组外周血CD8比例较照射组上升,骨髓ROS水平下降(P<0.01)。结论:SB对小鼠6Gy照射后造血免疫损伤有一定的缓解作用,其机制可能与其降低照射后骨髓细胞ROS水平有关。

Abstract:Objective:To observe the effects of p38 inhibitor SB203580 (SB) on the immune cells in mice exposed to 6 Gy irradiation. Methods:Thirty C57BL/6 mice were randomly divided into control group, irradiated group and SB group (n=10 for each). Mice of control group received sham irradiation, and the rest accepted 6 Gy total body irradiation.
