雌激素对雄性大鼠大脑中动脉闭塞神经保护作用的研究

摘    要:目的探讨雌激素对永久性大脑中动脉闭塞的神经保护作用及其可能机制。方法将大鼠随机分为TTC染色组和凋亡组,其中每组又分为生理盐水给药组、雌二醇给药组和假手术组。采用线拴法制作大鼠大脑中动脉闭塞(MCAO)模型。各组动物均在线栓插入颈内动脉24h后处死,断头取脑。TTC染色组分别于4、8、24h行神经功能评分,并在末次评分后断头处死,行TTC染色,观察脑梗塞体积。凋亡组进行免疫组化染色,分别计数大脑皮质和纹状体区每平方毫米的bcl-2和bax的阳性细胞数。结果雌二醇在1mg/kg腹腔注射给药时能显著缩小大鼠脑梗塞体积。在鼠脑缺血24h雌二醇有效促进了抗凋亡基因bcl-2的表达,抑制了促凋亡基因bax的表达。结论雌激素对雄性大鼠缺血性脑损伤具有明显的神经保护作用。对基因和非基因因素的影响是神经保护作用的可能机制。

Abstract:Objective To observe the neuroprotective role and its mechanism of estrogens in male Wistar rats with middle cerebral artery occlusion. Methods Male Wistar rats were randomly divided into TIC stain group and apoptotic group. Each group was subdivided into saline group,estradiol group and control group. The models of cerebral isehemia were made by occlusion middle cerebral artery using an intraluminal filament method. One week later,all rats were subjected to permanent middle cerebral artery occlusion and were killed at 24h after cerebral ischemia. The size of infarction was showed by TIC stain. Immunohisto-chemistry was used to show the expression of bcl-2 and bax. The bel-2 and bax positive cells were counted in the cerebral cortex and corpus striatum. Results Intraperitoneal injection of 1mg/Kg estradiol could decrease the infarction volume. The expression of bcl-2 was promoted and the expression of bax was inhibited by estradiol at 24 h of cerebral isehemia. Conclusion E2 may play a protective role on cerebral ischemia in rats. Regulation of gene expression and other factors is involved in its mechanism.
