辅助性T细胞9在多发性大动脉炎中的潜在作用

摘    要:目的检测辅助性T细胞9(Th9)及相关细胞因子白细胞介素9(IL 9)在多发性大动脉炎中的改变并探讨二者在多发性大动脉炎中的潜在作用。方法采集11例活动期多发性大动脉炎患者和年龄、性别匹配的10例健康体检者的外周血单个核细胞(PBMC)和血清。采用酶联免疫吸附试验检测血清IL 9水平,流式细胞术检测PBMC中Th9细胞百分数。结果多发性大动脉炎患者组血清IL 9水平为(10.3±4.5)ng/L,健康对照组(健康体检者)血清IL 9水平为(3.0±2.0)ng/L(P〈0.05)。活动期多发性大动脉炎患者PBMC中CD+4IL 9+T细胞数为(7.81±3.17)%,CD+4PU.1+T淋巴细胞数为(4.90±1.23)%;CD+4 T细胞中IL 9+PU.1+T细胞(Th9细胞)数为(8.25±3.73)%,而健康对照组外周血CD+4IL 9+T细胞、CD+4PU.1+T细胞及Th9细胞均未检出(P〈0.05)。IL 9、Th9细胞与多发性大动脉炎患者红细胞沉降率、C反应蛋白及美国国立卫生研究院大动脉炎活动性积分无相关性。结论Th9细胞及IL 9可能参与了多发性大动脉炎的免疫紊乱。

Abstract:ObjectiveTakayasu′s arteritis (TA) is a kind of global arteritis predominantly involves aorta and the branches. CD+4 T cells play a vital role in the pathogenesis of TA. However, it is still not clear how the Th9 cells which is named as the secretion of interleukin (IL) 9, a new subset of CD+4 T cells, participate in immune disorders of TA. This study was to identify the alternations of Th9 cells and the cytokine IL 9 in patients with TA. MethodsPeripheral blood samples were taken from 20 patients with TA and 10 healthy donors were as controls. IL 9 levels were tested by enzyme linked immunoabsorbent assay. CD+4 T cell subsets were analyzed by flow cytometry. Results IL 9 levels in sera of TA patients were elevated obviously than those in healthy donors (10.3±4.5)μg/L vs (3.0±2.0)μg/L, P〈0.05). Frequencies of CD+4IL 9+T cells were (7.81±3.17)%; CD+4PU.1+T cells were (4.90±1.23)%; CD+4IL 9+PU.1+T cells (Th9 cells) were (8.25±3.73)% in TA patients, but all of these subsets of CD+4 T cells were absent in healthy donors (P〈0.05).ConclusionPeripheral IL 9 producing CD+4T cells and IL 9 may be involved in the immune disturbance of Takayasu′s arteritis.
