| 摘 要： | 目的：建立枸杞子中甜菜碱含量的HPLC-ELSD测定方法。方法：采用Hypersil NH2色谱柱（250 mm×4.6 mm,5μm）,流动相为甲醇-四氢呋喃-0.2%三氟乙酸（30：60：10）,流速为1.0 mL.min-1,柱温为25℃。结果：甜菜碱进样量为1.28～12.8μg时,其与色谱峰面积具有良好的线性关系（r=0.999 2）;平均加样回收率为98.60%,RSD为2.8%（n=6）;测得样品中甜菜碱含量为0.528 1～1.205 4 mg.g-1。结论：该HPLC-ELSD法简便,准确度高,可用于枸杞子的质量控制。

| Abstract: | Objective:To establish a HPLC-ELSD method for the determination of the content of betaine in Lycii Fructus.Methods:The analysis was performed on Hypersil NH2 column(250 mm×4.6 mm,5 μm).The mobile phase was composed of methanol,tetrahydrofuran and 0.2% trifluoroacetic acid with the volume ratio of 30 ∶60 ∶10.The flow rate of mobile phase was 1.0 mL ·min-1 and the column tempe-rature was and 25 ℃.Results:The calibration curve showed a good linearity with the range of 1.28~12.8 μg(r=0.999 2) for betaine.The average recovery was 98.60%,with the RSD of 2.8%(n=6).The content of betaine is determined as 0.528 1~1.205 4 mg ·g-1.Conclusion:The method is simple and accurate.It can be used to control the quality of Lycii Fructus. |
