酚红对无血清悬浮培养条件下肿瘤干细胞微球形成的影响

摘    要:目的 研究酚红对无血清悬浮培养条件下肿瘤干细胞微球形成的影响.方法 将人胶质瘤细胞U251、人乳腺癌细胞MCF-7、人肝癌细胞SMMC-7721、人宫颈癌细胞Hela等细胞分为酚红组和无酚红组,分别悬浮培养于添加了生长因子的DMEM-F12培养基中,观察5d后肿瘤干细胞微球的形成情况,并计算微球的形成率.结果 U215无酚红组细胞的微球形成率为3.38%,明显高于酚红组的(2.73%);MCF-7、SMMC-7721、Hela无酚红组细胞的微球形成率分别为6.81%、7.75%、7.18%,酚红组的细胞均无微球形成.结论 无血清悬浮培养肿瘤干细胞应采用无酚红培养基.

Abstract:OBJECTIVE To study the effects of phenol red on the formation of the microspheres of tumor stemcells under the condition of serum-free suspension culture.METHODS Human glioma cells U251,human breast cells MCF-7,human liver carcinoma cells SMMC-7721 and hunan cervical cancer cells Hela were respectively divided into two groups(one group was cultured with the phenol red suspension culture,and the other was without the phenol red suspension culture).Cell in both groups were cultured in DMEM-F12 medium with growth factors.After five days,the formation of microspheres was observed and the formation rate was calculated.RESULTS The formation rate of microspheres in U215 without phenol red group was 3.38%,it was significantly higher than the U215 with phenol red group in which the formation rate was 2.73%.The formation rates of microspheres of MCF-7,SMMC-7721 and Hela without phenol red was 6.81%,7.75% and 7.18%,respectively.But there were no microspheres formed in the group with phenol red.CONCLUSION Phenol-free red medium should be used in non-serum suspension stem cells culture.
