泽泻汤治疗高血压药理机制的网络分析

摘    要:目的: 利用网络药理学研究泽泻汤治疗高血压的作用机制。方法: 根据ADME计算方法,利用中药系统药理学技术平台(TCMSP)等数据库、反向分子对接服务器(DRAR-CPI)、人类基因组注释数据库(Gene Cards)预测和筛选泽泻汤的活性成分及其降血压作用靶点。借助Cytoscape 3.5.1软件构建泽泻汤活性成分-作用靶点网络,借助STRING平台构建靶蛋白互作网络,通过生物学信息注释数据库(DAVID)对靶点GO生物过程及KEGG信号通路进行分析。结果: 从泽泻汤中筛选出10个活性成分,作用于136个高血压相关靶点,涉及7个主要信号通路,参与10个主要生物过程。结论: 泽泻汤通过作用于醛固酮调节钠吸收、肾素血管紧张素(RAS)系统,NO信号转导等信号通路及生物过程发挥降压作用。

Abstract:OBJECTIVE To use network pharmacology to study the mechanism of Zexie Decoction in treating hypertension.METHODS Based on the ADME computing method, Chinese medicine system pharmacology platform (TCMSP), reverse molecular docking server (DRAR-CPI), and human genome annotation database (Gene Cards), the active ingredients of Zexie Decoction and its targets of lowering blood pressure were predicted and screened. With the help of Cytoscape 3.5.1 software, the active ingredients-targets network of Zexie decoction was constructed, and the target protein interaction network was constructed based on STRING platform. The GO biological process and KEGG signal pathway of targets were analyzed by biological information annotation database (DAVID).RESULTS Ten active ingredients were screened from Zexie decoction. These active ingredients acted on 136 related targets of hypertension, involving 7 main signaling pathways and participating in the 10 main biological processes.CONCLUSION Zexie decoction plays a role in lowering blood pressure by acting on aldosterone regulating sodium absorption, the renin angiotensin system (RAS), NO signal transduction and other biological processes.
