橙皮苷对缺血再灌注损伤模型大鼠心肌组织保护作用的研究

摘    要:目的:研究橙皮苷对缺血再灌注损伤模型大鼠心肌组织的保护作用。方法:结扎大鼠左冠状动脉前降支复制心肌缺血再灌注损伤模型,60只Wistar大鼠均分为对照、模型和橙皮苷高、中、低剂量组。光镜下观察心肌组织病理变化,通过免疫组化法检测Fas、半胱天冬氨酸蛋白酶-3(Caspase-3)蛋白表达情况。结果:光镜下对照组心肌组织无病理损伤;模型组心肌组织呈严重炎性细胞浸润,并见多处坏死;橙皮苷高、中、低剂量组心肌细胞变性坏死程度显著减轻。与对照组比较,模型组心肌组织Fas和Caspase-3蛋白表达显著增强(P<0.05);与模型组比较,橙皮苷高、中、低剂量组心肌组织Fas、Caspase-3蛋白表达均显著下降(P<0.05)。结论:橙皮苷预处理对大鼠冠状动脉结扎后再灌注心肌损伤具有一定的保护作用。

Abstract:OBJECTIVE: To study the protective effects of hesperidin against myocardium injury induced by ischemia-reperfu- sion in rats. METHODS: Myocardial ischemia-reperfusion model was produced in rats by ligation of the left anterior descending coronary artery. 60 Wistar rats were divided into control group, model group, and hesperidin high-dose, medium-dose and low-dose groups. The pathologic changes and the expressions of Fas and Caspase-3 of myocardial tissues were observed by light mi- croscopy and immunohistochemistry, respectively. RESULTS: Light microscopic examination showed that the myocardial tissue of control group had no pathological changes; serious inflammatory cells infiltration and the multiple necrosis of myocardial tissue were found in model group. Degeneration and necrosis of myocardial cell were relieved in hesperidin high-dose, medium-dose and low-dose groups. Compared with control group, the expressions of Fas and Caspase-3 increased significantly in model group (P〈 0.05). Compared with model group, the expressions of Fas and Caspase-3 decreased significantly in hesperidin high-dose, medi- um-dose and low-dose groups (P〈0.05). CONCLUSION: Hesperidin pretreatment shows a significantly protective effect on myo- cardial ischemia-reperfusion injury in rats.
